RESUMO

Due to the effects of climate change, conditions tend to be increasingly extreme, with water availability being one of the main limiting factors in potato production. The objective of this study was to analyze the differential response of physiological and yield components in six potato varieties under water deficit conditions. For this purpose, a greenhouse trial was carried out with the varieties Agata, Agria, Kennebec, Monalisa, Sante and Zorba. The drought stress was applied in stressed plants 36 days after planting (DAP) by withholding water for 25 days. All measurements were taken at four different times: before stress (T0), 17 days (T1) and 24 days (T2) after stress and five days after re-watering. The physiological parameters evaluated were chlorophyll content and fluorescence, relative leaf water content, stomatal conductance, electrolytic leakage and water potential. After the drought period, the aerial part of half of the plants was cut to evaluate the produced biomass. At the end of the cycle yield components were determined. Stomatal conductance and water potential were the parameters that showed the highest differences between the two hydric conditions, and Monalisa was the variety with the best response in tuber production under stress conditions. Indirect selection based on parameters associated with water stress can be a useful tool in potato breeding programs for the identification of more tolerant varieties.

RESUMO

KEY MESSAGE: SNPs in candidate genes Pain - 1, InvCD141 (invertases), SSIV (starch synthase), StCDF1 (transcription factor), LapN (leucine aminopeptidase), and cytoplasm type are associated with potato tuber yield, starch content and/or starch yield. Tuber yield (TY), starch content (TSC), and starch yield (TSY) are complex characters of high importance for the potato crop in general and for industrial starch production in particular. DNA markers associated with superior alleles of genes that control the natural variation of TY, TSC, and TSY could increase precision and speed of breeding new cultivars optimized for potato starch production. Diagnostic DNA markers are identified by association mapping in populations of tetraploid potato varieties and advanced breeding clones. A novel association mapping population of 282 genotypes including varieties, breeding clones and Andean landraces was assembled and field evaluated in Northern Spain for TY, TSC, TSY, tuber number (TN) and tuber weight (TW). The landraces had lower mean values of TY, TW, TN, and TSY. The population was genotyped for 183 microsatellite alleles, 221 single nucleotide polymorphisms (SNPs) in fourteen candidate genes and eight known diagnostic markers for TSC and TSY. Association test statistics including kinship and population structure reproduced five known marker-trait associations of candidate genes and discovered new ones, particularly for tuber yield and starch yield. The inclusion of landraces increased the number of detected marker-trait associations. Integration of the present association mapping results with previous QTL linkage mapping studies for TY, TSC, TSY, TW, TN, and tuberization revealed some hot spots of QTL for these traits in the potato genome. The genomic positions of markers linked or associated with QTL for complex tuber traits suggest high multiplicity and genome wide distribution of the underlying genes.

RESUMO

The cDNA-AFLP technique can be used to monitor differential gene expression, but also for linkage mapping. Extending previous works, we have now constructed an integrated linkage map of the potato transcriptome based on the said technique that has a length of around 800 cM and contains nearly 700 transcriptome derived fragments (TDFs). At the same time, most of these markers are anchored to the bins of a highly saturated reference map in potato, combining in this way the information provided by different marker types. Moreover, we detected and confirmed an elevated degree of allelic fragments with this marker type, which was present in nearly half of all primer combinations and involved around 20% of all fragments. These properties were particularly useful to establish anchor points for integrating the individual parental linkage maps. Comparative expression profiling in different plant materials revealed that only a few additional TDFs were obtained which were specific for mature leaves or tubers compared to the TDFs present in whole in vitro plants. Since TDF markers are derived from coding regions, they generally also represent sequences with a biological function. In four case studies, co-migrating TDFs in different Solanum wild species always represented potential alleles based on elevated homologies among them. Two resistance gene homologs were identified by analysing TDFs, which were co-located with known QTLs.

RESUMO

With the present study we try to achieve an objective confirmation of the efficacy given by the mechanical suture in the closing of the pharyngeal defect following a total laryngectomy against the conventional manual suture with absorbable material. Therefore we have studied three random groups of patients: in the first one we include those who had a manual suture (n = 50), in the second one those who had performed a mechanical suture with closed technique (n = 38) and in the third one those who had a mechanical suture with an open technique (n = 12). We have checked that mechanical suture with a closed technique improves significantly the surgical length (212 minutes), the beginning of the oral feeding (10.73 days) and the time of in-hospital stay (13.5 days). Mechanical sutures with a closed technique also reduce the rate of complications such as fistulae (5.26%), infection (0%) and haemorrhage (0%).

RESUMO

We report a case of prostate carcinoma metastatic to the mandible that had been initially suspected as being a primary carcinoma of the parotid with lymph node metastasis. The present case underscores the diverse forms of manifestation of prostate carcinoma.

RESUMO

The case of a senile dementia patient 86 year old, and a supposed clinical history of swallowing a foreign body is presented. After a clinical study (esophagogram, flexible and rigid esophagoscopy and bronchoscopy) and finally the autopsy of the patient, we found and idiopathic esophago-bronchial fistula.
