RESUMO

Serotonergic system is involved in the regulation of physiological functions and behavioral traits including cognition, memory, aggression, stress coping, appetite and immunomodulation. Serotonin exerts its functions via binding distinct serotonin receptors which are classified into 7 groups. Salmonid exhibits expanded functional gene copies due to salmonid-specific whole genome duplication. However, serotonin receptor (htr) repertoire is not fully identified in rainbow trout (Oncorhynchus mykiss). In this study, we identified 39 htr genes, including 14 htr1, 4 htr2, 4 htr2 like, 3 htr3, 4 htr4, 2 htr5, 2 htr6, and 6 htr7 subtypes. We investigated physiological functions of serotonin receptors in response to bacterial pathogens exposure and salinity changes. We showed htr1, htr2, htr4 and htr7 subtypes were associated with immunomodulation in response to Vibrio anguillarum or Aeromonas salmonicida infection. Saltwater (salinity of 15) transfer significantly altered htr1, htr2, htr4, and htr7 subtypes, suggesting trout Htr was associated with osmoregulation. We further showed residues interacted with inverse agonist (methiothepin) and serotonin analogue (5-Carboxamidotryptamine) were conserved between trout and human, suggesting exogenous ligands targeting human HTRs might have a role in aquaculture. This study showed duplicated trout Htrs might be physiologically neofunctionalized and potentially exhibit pleiotropic effects in regulating immunomodulation and osmoregulation.

Human activities can cause zinc (Zn) contamination of aquatic environments. Zn is an essential trace metal, but effects of environmentally relevant Zn exposure on the brain-intestine axis in fish are poorly understood. Here, six-month-old female zebrafish (Danio rerio) were exposed to environmentally relevant Zn concentrations for six weeks. Zn significantly accumulated in the brain and intestine, causing anxiety-like behaviors and altered social behaviors. Zn accumulation altered levels of neurotransmitters, including serotonin, glutamate, and γ-aminobutyric acid, in the brain and intestine, and these changes were directly associated with changes in behavior. Zn caused oxidative damage and mitochondrial dysfunction, and impaired NADH dehydrogenase, thereby dysregulating the energy supply in brain. Zn exposure resulted in nucleotide imbalance and dysregulation of DNA replication and the cell cycle, potentially impairing the self-renewal of intestinal cells. Zn also disturbed carbohydrate and peptide metabolism in the intestine. These results indicate that chronic exposure to Zn at environmentally relevant concentrations dysregulates the bidirectional interaction of the brain-intestine axis with respect to neurotransmitters, nutrients, and nucleotide metabolites, thereby causing neurological disorder-like behaviors. Our study highlights the necessity to evaluate the negative impacts of chronic environmentally relevant Zn exposure on the health of humans and aquatic animals.

To investigate the effects of mariculture on the sources, distribution and preservation of sedimentary organic carbon (SOC), sediments from an mariculture area in Ailian Bay, China, and a control area were analyzed for grain size composition, total organic carbon (TOC), total nitrogen (TN), carbon/nitrogen (C/N) ratio, and stable carbon and nitrogen isotopic composition (δ13C and δ15N). The sedimentary type of sediments in study area was clay silt. TOC, TN, C/N, δ13C, and δ15N ranged from 0.58 to 1.21%, 0.06-0.17%, 6.29-9.82, -23.20 to -18.50‰, and 6.17-7.38‰, respectively, and followed similar spatial patterns. TOC, TN and δ13C were higher in mariculture area than in control area. Biodeposit and kelp OC contributions greater of SOC in mariculture area than the control area (biodeposits: 20.10 ±â€¯4.84 to 6.2 ±â€¯1.3%; kelp: 15.3 ±â€¯6.63 to 5.2 ±â€¯0.84%). Overall, mariculture activities significantly influence the sources, distribution and preservation of SOC.

Acute heat stress is common in aquaculture and can affect diverse physiological processes in fish; however, different species of fish have various mechanisms for heat stress adaptation. In this study, we profiled the transcriptome responses of the Atlantic salmon (Salmo salar) to heat stress at 23 °C for 6 or 24 h, compared with that of fish at a normal temperature of 13 °C. The liver was selected as the target tissue for this analysis. A total of 243 and 88 genes were differentially expressed after 6 and 24 h of heat stress, respectively. Of these, only 22 were common to both time points, and most of these common genes were molecular chaperones such as heat shock cognate 71 kDa protein and heat shock protein 90-alpha. Genes such as activating transcription factor 6, calreticulin, protein disulfide isomerase A3, and protein kinase R-like endoplasmic reticulum kinase-eukaryotic initiation factor 2-alpha were only up-regulated after 6 h of heat stress; most of these genes are involved in the endoplasmic reticulum stress pathway. Indeed, endoplasmic reticulum stress was identified at 6 h but not at 24 h, suggesting that stress response plays an important role in the adaptation of Atlantic salmon to acute heat stress. Other up-regulated genes at 6 h were related to the insulin and nucleotide oligomerization domain-like receptor signaling pathways, which directly eliminate misfolded proteins and sustain sugar and lipid homeostasis. At 24 h, heat stress influenced the expression of steroid and terpenoid backbone biosynthesis, which may influence the sexual development and differentiation of Atlantic salmon. Overall, our results elucidate the transcriptome mechanisms that contribute to short-term heat tolerance in the liver of Atlantic salmon.

Rainbow trout (Oncorhynchus mykiss) is a cold-water fish species with optimal temperature of 12-18 ℃. Temperature stress is a common issue in commercial culture of the Salmonid fish in summer. Both maple trout and steelhead trout are rainbow trout breeding species with good quality. The purpose of this study was to explore the impact of acute temperature stress on antioxidant responses of two kinds of rainbow trout. The juvenile maple trout was diploid, and the juvenile steelhead trout was triploid. The fishes were exposed to four different temperature treatments: 13, 17, 21, and 25 ℃, and hepatic samples were collected at 0, 1, 6, and 12 h. After acute heat stress, the fish were transferred to the tank with optimal temperature (13 ℃) and hepatic samples were collected after 1, 12, 24, and 48 h. The biomarkers, superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), lipid peroxidation (LPO), malondialdehyde (MDA), and heat shock protein 70 kDa (HSP70) were measured. Results showed that the activities of SOD, CAT, and GPx in both maple trout and steelhead trout in 17 ℃ treatment had no significant increase after acute temperature stress. The SOD activities of maple trout and steelhead trout in 21 ℃ treatment significantly increased in response to acute temperature stress, but the SOD activity of maple trout returned to normal during the recovery process. The activities of SOD, CAT, and GPx for both species in 25 ℃ treatment were significantly increased after acute temperature stress, which returned to normal for 24 h recovery. The amount of HSP70 produced by maple trout at 17, 21, and 25 ℃ treatments was significantly higher than that at 13 ℃ treatment, while that of steelhead trout was significantly higher for the 21 and 25 ℃ treatments than that at 13 ℃ treatment. A variety of antioxidant parameters were analyzed by integrated biomarker response (IBR) values. Results showed that the antioxidant response of maple trout was significantly higher than that of steelhead trout in 17 ℃ and 21 ℃ treatments, but significantly lower than steelhead trout in 25 ℃ treatment. Such a result indicated that the antioxidant responses of different ploidy rainbow trouts were different under acute stress at different temperatures.

The concentrations of typical heavy metals including Cu, Hg, Pb, Zn, Mn, Cd, Cr and As in the surface sediment and suspended particulate matter (SPM) in Jinghai Bay, Rongcheng city were tested. The degree of heavy metal pollution in the surface sediment was assessed according to the national standard of marine sediment quality. The potential ecological risk (PER) of heavy metals to marine ecosystem in Jinghai Bay was assessed using PER coefficient and risk index. The results showed that the levels of heavy metals and PER in the surface sediments were low enough to meet the requirement of the first class of the sediment quality standards except the levels of Cu and Zn in the sampling site S2 and Cd in S3, S4, S5 and S6 which met the requirements of the second class of the sediment quality standards. The PER coefficients of heavy metals increased following the sequence of Zn < Cr < Pb < Cu < As

An in situ monitoring of the sediment characteristics and macrobenthic communities was undertaken at a marine fish culture site in subtropical waters of Hong Kong before and after the deployment of biofilters which were made of cement concrete artificial reef (AR) structures. According to the distance to the boundary of the fish cages, 6 points were selected as sampling stations: 2 at the fish cages, 2 near the boundary of the fish culture area, and 2 reference sites further away from the culture area. Bimonthly sediment samples were collected for analysis of silt-clay fraction (SCF), moisture content (MC), total organic carbon (TOC), total Kjeldahl nitrogen (TKN) and total phosphorus (TP). The macrobenthos (>0.5mm) present in the sediment were sorted, identified and enumerated. TOC, TKN and TP levels at the fish cage stations were consistently higher than those at the reference stations over the 1-year pre-AR and 2-year post-AR deployment monitoring period. The diversity of macrofauna was significantly reduced at the fish cage stations relative to the reference sites. The intermediary stations near the fish culture area showed a transitional state of disturbance. Over the 2-year post-AR deployment period, TOC, TKN and TP showed a decreasing trend at the fish cage and intermediary stations. More diverse macrofaunal communities were recorded at the fish cage stations, with species diversity H'increasing from 0-1 at the beginning of the AR deployment to H'>2 at the end of the study. The present results demonstrated that artificial reefs can improve the benthic abiotic environment and biotic conditions beneath fish rafts which are deteriorated due to farming activities.

By analysis of the fatty acid profiles in mussel tissues and suspended particulate matter (SPM) in water, the present study showed a significant relationship of the trophic linkage between mussels and the SPM. At seven locations from inner to outer areas along the eutrophic Tolo Harbour and Tolo Channel, Hong Kong, the composition (as percentage of total fatty acids) of both monoenoic and polyunsaturated fatty acids in the tissues of green-lipped mussels Perna viridis and SPM in water had significant correlation (p<0.01). In particular, the composition of docosahexaenoic acid (DHA, 22:6n3) (as percentage of total fatty acids) in mussel tissues was statistically correlated with that in SPM (p<0.01), implying that mussels incorporate DHA, the biomarker of dinoflagellates. Principal component analysis further demonstrated that the fatty acid profiles of SPM were different among locations in the harbour, mid and outer channel of the study area, so were the mussel tissue fatty acid profiles. Cluster analysis of phytoplankton data also revealed the dominance of diatoms in the inner harbour and channel areas, whereas dinoflagellates were abundant in the outer channel waters. The possible implications of using benthic suspension feeders such as green-lipped mussels P. viridis for controlling phytoplankton abundance in coastal waters are discussed.

To study the correlation between nutrient enrichment derived from fish farming activities and changes in macrobenthic assemblages, a one-year field study was conducted in Kau Sai Bay marine fish culture zone of Hong Kong. Bimonthly sediment samples were collected at six stations: two at the fish cages, two near the boundary of the fish culture area, and two reference sites further away from the culture area. Sediment physico-chemical characteristics in terms of silt/clay fraction, moisture content, total organic carbon (TOC), total Kjeldahl nitrogen (TKN) and total phosphorus (TP) were analyzed. The macrobenthos (>0.5 mm) present in the sediment were sorted, identified and enumerated. On average, TOC, TKN and TP levels at the fish cage stations were 82.8%, 128.5% and 1315.7% higher than those at the reference stations, respectively. As a result, the N:P molar ratio was greatly reduced from 8.75 at the reference stations to 1.83 at the fish cage stations. Univariate and multivariate analyses revealed that diversity of macrofauna was significantly reduced and community structure differed at the fish cage stations relative to the reference sites. The intermediary stations near the fish culture area showed a transitional state of disturbance. Faunal diversity was negatively correlated with nutrient level, reflecting the adverse impacts of nutrient enrichment derived from fish farming activities on the benthic assemblages. Whilst in subtropical Asia-Pacific trash fish is the major feed for fish culture resulting in a higher nutrient loading and nutrient ratio accumulated in the sediment beneath the fish rafts, the effects of nutrient enrichment on macrobenthic assemblages are comparable to that in temperate waters owing to relatively high sediment metabolism rate and smaller fish culture scale in Hong Kong.
