RESUMO

La degeneración macular asociada a la edad (DMAE) constituye una de las principales causas de la pérdida de agudeza visual (AV) en los mayores de 50 años en el mundo, siendo la DMAE neovascular (DMAEn) la causante del 80% de los casos de pérdida de visión severa debido a esta enfermedad. Hace ya más de una década que se emplean los fármacos antifactor de crecimiento del endotelio vascular (anti-VEGF) para el tratamiento de esta enfermedad, cambiando drásticamente el pronóstico visual de estos pacientes. Sin embargo, los primeros estudios de los que se disponían datos de los resultados eran a corto plazo. En la actualidad existen ya diferentes series publicadas de los resultados de la DMAE a largo plazo tras el tratamiento con anti-VEGF, siendo el objetivo de la presente revisión sintetizar dichos resultados. El seguimiento medio de los estudios incluidos fue de 8,2 años (rango: 5-12 años). La AV inicial media fue 55,3 letras del Early Treatment Diabetic Retinopathy Study (ETDRS) (rango: 45,6-65) siendo la AV final media 50,1 letras (rango: 33,0-64,3), existiendo una pérdida media de 5,2 letras. Al final del seguimiento un 29,4% de los pacientes mantuvieron una AV>70 letras. El 67,9% de los pacientes se mantuvo estable al final del seguimiento (<15 letras de pérdida), existiendo una pérdida severa (≥15 letras) del 30,1%. La fibrosis y la atrofia fueron las principales causas de pérdida de AV a largo plazo, presentándose al final del seguimiento en un 52,5% y un 60,5%, respectivamente.(AU)

Gas chromatography (GC) is a separation technique commonly developed for targeted in situ analyses in planetary space missions. It is coupled with low-resolution mass spectrometry to obtain additional structural information and allow compound identification. However, ground-based analyses of extraterrestrial samples have shown the presence of large molecular diversities. For future targeted in situ analyses, it is therefore essential to develop new technologies. High resolution mass spectrometry (HRMS) is currently being spatialized using FT-orbitrap-MS technology. In this contribution, the coupling of gas chromatography with FT-orbitrap-MS is studied for targeted amino acid analyses. The method for enantioselective separation of amino acids was optimized on a standard mixture comprising 47 amino acid enantiomers. Different ionization modes were optimized, chemical ionization with three different reactive gasses (NH3, CH4 and NH3/CH4) and electron impact ionization at different electron energies. Single ion and full scan monitoring modes were compared, and detection and quantification limits were estimated by internal calibration under the optimized conditions. The GC-FT-orbitrap-MS demonstrated its ability to separate 47 amino acid enantiomers with minimal co-elution. Furthermore, due to the high mass resolution and accuracy of FT-orbitrap-MS, with mass extraction, the S/N is close to zero, allowing average LOD values of 10⁻7 M, orders of magnitude lower than conventional GC-MS techniques. Finally, these conditions were tested for enantioselective analysis of amino acids on an analog of a pre-cometary organic material showing similarities to that of extraterrestrial materials.

PURPOSE: To evaluate the validity and reliability of the Child Perceptions Questionnaire 11-14 (CPQ11-14) for children attending public educational institutions in Floridablanca (Colombia). METHODS: An evaluation study of diagnostic technology was conducted. The questionnaire was completed by 500 students aged 11-14 years who were selected by simple random sampling. The test-retest reproducibility was evaluated in 36 students chosen at random from one of the schools; the questionnaire was completed a second time by these students, with an interval of two weeks between administrations. Internal consistency was estimated using the Cronbach's alpha coefficient, and test-retest reproducibility was determined with the intraclass correlation coefficient (ICC). The Rasch Rating Scale Model was used to perform the validation; the structure of the categories, the quality of the items, the invariance and the one-dimensionality were analyzed; invariance was confirmed using differential item functioning (DIF). Informed consent was obtained from the parents or guardians, and assent was obtained from the minors. RESULTS: The mean age was 12.7 ± 1.1 years, and 210 (42.0%) participants were female. The Cronbach's alpha and ICC values were 0.91 and 0.63, respectively. The 37 items fit the Rasch model, but a uniform DIF was observed for an item that was eliminated. The questionnaire was not one-dimensional, and the proportion of variance explained by the model was poor (26.3%). CONCLUSION: The CPQ11-14 showed unsatisfactory test-retest reproducibility and excellent internal consistency; no unidimensionality was observed, but the items fit the Rasch model.

This study was designed to evaluate the effects of substituting corn grain with nonforage fiber sources in diets containing soybean meal (SBM) or canola meal (CM) as the primary protein source. Sixteen Holstein cows were assigned to a replicated 4 × 4 Latin square design with 4 periods of 28 d each. Treatments were arranged as a 2 × 2 factorial with 2 protein sources (SBM and CM) and 2 dietary starch concentrations (21 and 27% dry matter, DM). Diets were formulated to contain 16.5% CP, and the 21% starch diets were obtained by replacing corn grain with soybean hulls and beet pulp. Protein source × starch interactions were observed for DM intake (DMI), milk fat and protein concentrations, milk protein yield, milk urea nitrogen, and feed efficiency. Cows fed CM diets had a higher DMI when dietary starch concentration was 27% compared with 21%, but those cows had DMI similar to that of cows on SBM diets regardless of the starch concentration. Milk fat percentage was decreased in cows fed CM with 27% starch compared with cows fed CM with 21% starch and cows fed SBM with 27% starch. Milk protein percentage and yield and milk lactose percentage were least in cows fed CM with 21% starch compared with the other 3 diets, but feed efficiency was greater for cows fed CM with 21% starch. Milk urea nitrogen was least in cows fed CM with 27% starch compared with the other 3 diets. Cows fed diets with 27% starch produced 2.5 kg/d more milk and 1.9 kg/d more energy-corrected milk compared with cows fed 21% starch. Digestibility of DM and organic matter was higher in cows fed SBM diets than in cows on CM diets, and cows fed 27% starch showed greater DM and organic matter digestibility than cows on 21% starch. Digestibility of neutral detergent fiber and acid detergent fiber was greater in diets with SBM than in those with CM. Molar proportion of acetate was the lowest for cows fed CM with 21% starch compared with cows fed SBM with 21% starch, with the remaining cows fed being intermediate and similar. However, propionate was highest for cows fed CM with 21% starch than for cows fed SBM with 21% starch, but the remaining treatments were intermediate and similar. Isobutyrate was greater for cows fed CM with 21% starch, which resulted in the lowest acetate:propionate ratio compared with cows fed the remaining treatments. Overall, we confirmed that the interaction of protein with starch in CM diets can sustain similar cow performance as with the SBM diets. Those making decisions about starch concentration and protein source should consider feed price when SBM or CM and different starch levels are being formulated in diets for lactating dairy cows.

A meta-analysis was conducted to compare the effects of feeding dairy cows conventional sorghum silage (CSS) or conventional corn silage (CCS) compared with brown midrib sorghum silage (BMRSS) diets on dry matter intake (DMI), milk production, and milk composition. Data from 9 published articles (1984 to 2015) were used to contrast diets with CSS (7 means comparisons; 104 cows) or CCS (13 means comparisons; 204 cows) versus BMRSS diets. Statistical analysis was performed using fixed or random effects models with the Metafor package of R (https://www.R-project.org). The degree of heterogeneity was measured with the I2 statistic, and publication bias was determined with funnel plots and Egger's regression test. Other sources of heterogeneity of response were analyzed through meta-regression. Estimated effect size was calculated for DMI, milk production, and milk composition. No evidence of publication bias was observed for any variable tested. The highest degree of heterogeneity (I2 = 41.5 and 72.6%) was observed for DMI among dependent variables tested in both comparisons, indicating that intake responses to silage type are rather inconsistent; in contrast, milk production had the lowest degree of heterogeneity (I2 = 0%), supporting the idea that the responses of this variable to silage type were very consistent across studies. Compared with BMRSS diets, cows fed CSS diets exhibited decreased milk production (1.64 kg/d), milk fat concentration (0.09%), milk fat yield (0.08 kg/d), milk protein yield (0.04 kg/d), and milk lactose yield (0.16 kg/d) and tended to decrease DMI (0.83 kg/d). Compared with CCS diets, cows fed BMRSS diets increased milk fat concentration (0.10%), but decreased milk protein concentration (0.06%) and tended to increase lactose yield (0.08 kg/d). Meta-regression indicated that days in milk affected DMI and milk production when CSS diets were compared with BMRSS diets, and DMI when CCS diets were compared with BMRSS diets. Additionally, the inclusion rate of silage in the diet and dietary neutral detergent fiber affected yields of milk fat and lactose, respectively, when CCS and BMRSS diets were compared. Overall, lactation performance improved when cows were fed diets formulated with BMRSS compared with CSS, but performance was not different for cows fed BMRSS and CCS diets. However, the small sample size may have influenced these results by increasing the margin of the error and, concurrently, the power of the meta-analysis. Results of this analysis suggest that additional research is needed to explore the effects of days in milk and the inclusion rates of silages in the diets when comparing BMRSS with CSS or CCS.

The aim of this study was to evaluate repeated measures over the years to estimate repeatability coefficient and the number of the optimum measure to select superior genotypes in Annona muricata L. The fruit production was evaluated over 16 years in 71 genotypes without an experimental design. The estimation of variance components and the prediction of the permanent phenotypic value were performed using REML/BLUP proceedings. The coefficient of determination, accuracy, and selective efficiency increased when measures increased. The coefficient of determination of 80% was reached beyond 8 crop seasons with high accuracy and selective efficiency. Thus, the evaluation of 8 crop seasons can be suitable to select superior genotypes in the A. muricata L. breeding program. Predicted selection gain had a high magnitude for fruit production indicating that it is possible to take a progressive genetic advance for this trait over cycle breeding.

Betacyanins and phenolic compounds from acetonitrile:acidified water extracts of Alternanthera brasiliana and Alternanthera tenella were characterized and quantified using a high-performance liquid chromatography system coupled with diode array and electrospray mass spectrometry detection. Four betacyanins (amaranthine, isoamaranthine, betanin and isobetanin) were tentatively identified and quantified. Twenty eight phenolic compounds of four different families (hydroxybenzoic and hydroxycinnamic acids, flavones and flavonols) were separated and characterized on the basis of their accurate MS and MS/MS information out of which ten compounds were confirmed by authentic standards. These plant species could be considered as an especially rich source of natural bioactive compounds and potential food colorants. A. brasiliana showed the highest betacyanin and polyphenols content (89µg/g and 35,243µg/g, respectively). Among polyphenols, flavonols were the more abundant (kaempferol-glucoside, kaempferol-rutinoside and kaempferol-rhamnosyl-rhamnosyl-glycoside). Meanwhile, A. tenella showed a different polyphenols profile with flavones as major compounds (glucopyranosil-vitexin and vitexin). As a novelty, pentosyl-vitexin and pentosyl-isovitexin were detected for the first time in Alternanthera plants. Both A. brasiliana and A. tenella leaves showed high total polyphenol content and in vitro antioxidant activity (FRAP). These results provide an analytical base concerning the phenolic and betalains composition and the antioxidant properties of two members of the promising Alternanthera gender, for subsequent applications, such as functional food ingredients.

The objective of this study was to investigate the effect of substituting soybean meal products with yeast-derived microbial protein (YMP) on lactation performance in diets containing 2 forage-to-concentrate ratios. Sixteen Holstein cows (4 primiparous and 12 multiparous) were randomly assigned to multiple 4 × 4 Latin squares with a 2 × 2 factorial arrangement of treatments. Diets contained low (LF; 45% of diet DM) or high forage (HF; 65% of diet DM) and YMP at 0 (NYMP) or 2.25% (WYMP) of the diet. The forage mix consisted of 67% corn silage and 33% alfalfa hay on a DM basis. No interactions of forage and YMP were noted for any of the production parameters measured. Feed efficiency (energy-corrected milk/dry matter intake) was greater for cows fed NYMP compared with WYMP. Regardless of the addition of YMP, cows fed LF had greater dry matter intake and produced more milk than cows fed HF. In addition, cows fed LF produced more energy-corrected milk than those fed HF. Milk fat percentage was lower in cows fed LF compared with HF, whereas fat yield was similar between forage concentrations. Fat yield tended to decrease with feeding YMP. Interactions of forage and YMP were observed for propionate concentration, acetate and propionate proportion, and acetate-to-propionate ratio. A tendency for an interaction of forage and YMP was also noted for ruminal pH. Cows fed HF diets had greater ruminal ammonia and butyrate concentrations, as well as proportion of butyrate. Arterial concentrations of Ile, Leu, Met, Thr, and Val were greater in cows fed LF. Cows fed NYMP had greater arterial concentrations of Ile, Lys, Trp, and Val than cows fed WYMP. Substitution of soybean proteins with YMP did not improve performance or feed efficiency of high-producing dairy cows regardless of the forage-to-concentrate ratio of the diet.

The objective of this study was to determine if increased dietary fat from dried distillers grains with solubles (DDGS) in diets of growing heifers affected dry matter intake, average daily gain (ADG), growth performance, and nutrient digestibility. Thirty-three Holstein heifers (133±18 d old) were used in a 24-wk randomized complete block design. Treatments were (1) control (CON) containing ground corn and soybean products, (2) low-fat (LFDG) containing low-fat, high-protein DDGS and ground corn, and (3) high-fat (HFDG) with traditional DDGS. All diets contained 39.8% grass hay, 24.8% corn silage, and 1.5% vitamins and minerals. The HFDG diet was formulated to contain 4.8% fat compared with 2.8% in the CON and LFDG diets, which were greater in nonfibrous carbohydrate. Diets had a net energy gain of 1.0Mcal/kg of dry matter and were limit-fed at 2.45% of body weight. Heifers were weighed every 2wk and rations were adjusted accordingly. Heart girth, hip and wither heights, body length, and body condition score were recorded every 2wk. Total-tract digestion of nutrients was evaluated during wk16 using fecal grab sampling and an external marker. No treatments by time interactions were found. Dry matter intakes, body weights, ADG, and gain-to-feed ratio were similar among treatments; however, ADG averaged 0.96kg/d among treatments, which is greater than recommended. All body frame measurements and body condition scores were similar among treatments. Total-tract digestibilities of dry matter and organic matter were not different among treatments. However, crude protein and neutral detergent fiber digestibility were increased in the HFDG diet compared with the CON and LFDG diets. These results demonstrate that using DDGS or low-fat DDGS with corn in growing heifer rations can maintain performance. Utilizing the fat in DDGS as a dietary energy source in replacement of starch from corn did not influence growth performance or negatively affect nutrient digestion.

The objective of this research was to determine the subsequent effects on lactation and reproductive performance from feeding fat from distillers dried grains with solubles (DDGS) to growing dairy heifers. During the prepubertal growth phase, 33 Holstein heifers (133±18 d old) were used in a 24-wk randomized complete block design. Treatments included (1) a control diet (CON) containing ground corn and soybean products, 2) a diet with low-fat DDGS (LFDG), and (3) a higher-fat diet with traditional DDGS (HFDG). All diets contained 39.8% grass hay, 24.8% corn silage, and 1.5% vitamins and minerals. Previous results demonstrated that growth performance was maintained across treatments, but plasma cholesterol and fatty acids were greater and puberty may occur earlier in heifers fed HFDG. It was hypothesized that differences among treatments in metabolic profile and puberty may influence reproductive and first-lactation performance. Posttrial data on reproductive performance and milk production for the first 4 mo of lactation were collected for each heifer from dairy herd records. At 3wk prepartum and at calving, body weights, body condition scores, and body measurements were taken. No differences were observed among treatments for age at conception or age at calving. At calving, heifers fed the HFDG were shorter in withers height compared with heifers fed the other diets. Milk yields and components were similar or improved in heifers fed the distillers grains diets compared with heifers fed CON. Heifers fed LFDG had greater milk production and a tendency for greater milk protein yields compared with the heifers fed CON. Energy-corrected milk yields were similar among treatments. Feeding increased dietary fat from DDGS during the prepubertal growth phase did not negatively affect milk production, despite earlier attainment of puberty compared with other treatments. The overall ADG for all 3 treatments was 0.96kg/d during the prepubertal period, which is greater than recommended and may have compromised milk yield across treatments. Based on these findings, producers can feed either DDGS or LFDG with corn in replacement of soybean products and corn to prepubertal heifers and maintain or enhance subsequent reproductive and lactation performance. Dietary fat from DDGS can replace starch from corn as an energy source for prepubertal heifers without detriment to later performance.

Treatment diets were (1) control (CON) containing ground corn (15.9% of DM) and soybean products (17.9%), (2) low-fat (LFDG) containing low-fat DDGS (21.9%) and ground corn (11.9%), or (3) high-fat (HFDG) with traditional DDGS (33.8%). Diets were isonitrogenous and isocaloric, but the HFDG diet was formulated to contain 4.8% fat compared with 2.8% in the CON and LFDG diets. All 3 diets were limit-fed to 2.45% of body weight on a dry matter basis, and resulted in a mean average daily gain of 0.96kg/d across treatments. Every 4wk, jugular blood was collected for analysis of metabolites and metabolic hormones. During wk20 of the feeding period, blood samples were collected for analysis of plasma fatty acid profiles. When heifers weighed between 200 and 300kg of body weight, coccygeal blood samples were taken twice weekly for analysis of progesterone to determine if puberty had been reached. Plasma concentrations of nonesterified fatty acids were similar among treatments and consistent over the duration of the study. Plasma concentrations of ß-hydroxybutyrate, insulin, insulin-like growth factor-1, and leptin were similar among heifers fed each treatment diet, but increased over the duration of the feeding period. Serum concentrations of glucose tended to be less in heifers fed HFDG compared with heifers fed the CON diet. Glucose concentrations fluctuated throughout the feeding period, but no treatment by time interactions were noted. Plasma urea N concentrations were less in heifers fed LFDG compared with heifers fed HFDG and CON diets. The concentrations of plasma urea N increased over the duration of the feeding period, with no treatment by week interaction. Total plasma cholesterol was greater in heifers fed HFDG compared with the CON and LFDG diets, and a significant week effect and a week by treatment interaction were observed. Fatty acid profiles also differed among treatments based on the supply of fatty acids from the diet. Progesterone analysis indicated that heifers fed HFDG tended to be pubertal at a younger age than heifers on CON. These results demonstrate that dietary fat from DDGS can be used in high-plane of nutrition rations for growing heifers and maintain metabolic energy status compared with starch from corn, but alters the concentrations of different blood lipids.

The objective of this study was to examine the effects substituting soybean meal with a yeast-derived microbial protein (YMP) on rumen and blood metabolites, dry matter intake, and milk production of high-producing dairy cows. Sixteen Holstein cows (12 multiparous and 4 primiparous), 93 ± 37 DIM (mean ± SD) at the beginning of the experiment, were used in a 4×4 Latin square design with four 28-d periods. Cows were blocked by parity and production, with 1 square consisting of 4 animals fitted with rumen cannulas. Basal diets, formulated for 16.1% crude protein and 1.56 Mcal/kg of net energy for lactation, contained 40% corn silage, 20% alfalfa hay, and 40% concentrate mix. During each period, cows were fed 1 of 4 treatment diets corresponding to YMP (DEMP; Alltech Inc., Nicholasville, KY) concentrations of 0, 1.14, 2.28, and 3.41% DM. Soybean meal (44% CP) was replaced by YMP to attain isonitrogenous and isoenergetic diets. Dietary treatments had no effect on pH and on most ruminal volatile fatty acid concentrations, with the exception of isovalerate, which decreased linearly with the addition of YMP. Rumen ammonia concentration decreased linearly, whereas free amino acids, total amino acid nitrogen, and soluble proteins weighing more than 10 kDa showed a cubic response on rumen N fractionation. A quadratic response was observed in oligopeptides that weighed between 3 and 10 kDa and peptides under 3kDa when expressed as percentages of total amino acids and total nitrogen. Although nonesterified fatty acid concentration in blood did not differ between treatments, ß-hydroxybutyrate and plasma glucose increased linearly as YMP increased. Dry matter intake showed a cubic effect, where cows fed 1.14, and 3.41% YMP had the highest intake. Milk production was not affected by YMP, whereas a trend was observed for a quadratic increase for 4% fat-corrected milk and energy-corrected milk. Medium- and long-chain fatty acid concentrations in milk increased quadratically, which elicited similar effects on milk fat concentration and yield. Total solids percentage and yield, and milk urea nitrogen also showed quadratic effects as YMP increased in the diet. No effects were observed on feed efficiency, milk protein, and lactose percentage or yield. A complementary in vitro study demonstrated a quadratic tendency for apparent and true dry matter digestibility as YMP was added to the diet. It was concluded that the substitution of soybean meal with YMP increased the percentage of total solids in milk and tended to improve energy-corrected and fat-corrected milk production in high-producing dairy cows consuming high-forage diets.

Distillers grains with solubles (DGS) is the major coproduct of ethanol production, usually made from corn, which is fed to dairy cattle. It is a good protein (crude protein, CP) source (>30% CP) high in ruminally undegradable protein (approximately 55% of CP) and is a good energy source (net energy for lactation of approximately 2.25 Mcal/kg of dry matter). The intermediate fat concentration (10% of dry matter) and readily digestible fiber (approximately 39% neutral detergent fiber) contribute to the high energy content in DGS. Performance was usually similar when animals were fed wet or dried products, although some research results tended to favor the wet products. Diets can contain DGS as partial replacement for both concentrates and forages, but DGS usually replaces concentrates. Adequate effective fiber was needed to avoid milk fat depression when DGS replaced forages in lactating cow diets. Nutritionally balanced diets can be formulated that contain 20% or more of the diet dry matter as DGS. Such diets supported similar or higher milk production compared with when cows were fed traditional feeds. Although DGS can constitute more than 30% of diet dry matter, gut fill may limit dry matter intake and production in diets with more than 20% wet DGS and that also contain other moist feeds. The fiber in DGS, which often replaces high-starch feeds, does not eliminate acidosis but minimizes its problems. Distillers solubles, which are often blended with distillers grains to provide DGS, can be fed separately as condensed corn distillers solubles. Other distillers coproducts besides DGS such as high-protein distillers grains, corn germ, corn bran, and low-fat distillers grains are becoming available.

Eight multiparous and 8 primiparous Holstein cows were used in a replicated 4 x 4 Latin square design with 4-wk periods to determine the effects on dairy cow performance of feeding corn germ (CG) compared with dried distillers grains with solubles (DDGS) or corn oil (CO). Four isolipidic dietary treatments were formulated: a control diet, a 14% corn germ diet (CGD), a 30% dry distillers grains with solubles diet (DGD), and a 2.5% corn oil diet (COD). All diets were formulated to contain 6.0% fat, with the fat in the control diet provided by a ruminally inert fat source. Dry matter intake was decreased by feeding the COD compared with the CGD; however, no difference in dry matter intake was observed among the control diet, the DGD, and the COD. Dietary treatments had no effect on milk yield, energy-corrected milk, or 4% fat-corrected milk. Feeding CG had no effect on milk fat percentage when compared with the control diet; however, milk fat percentage tended to decrease with DDGS and decreased with CO when compared with the CGD. Milk protein percentage decreased when cows were fed the COD compared with the control diet. Feeding CO tended to decrease milk fat yield compared with CG; however, dietary treatments had no effect on milk protein and lactose yield. Feed efficiency was not affected by dietary treatments and averaged 1.55 kg of energy-corrected milk/kg of dry matter intake. Feeding DDGS and CO increased the concentration of vaccenic and conjugated linoleic acid in milk fat. Concentrations of monounsaturated and polyunsaturated fatty acids in milk were increased in response to feeding the 3 corn coproducts. Fat from CG appears to be relatively protected in the rumen when compared with that from DDGS and CO and therefore will not affect the production of milk fat to the degree of the more available fat in DDGS and CO.

Two experiments were conducted to evaluate the fermentation characteristics of ensiled wet corn distillers grains with solubles (WDG) alone or mixed with soybean hulls (SH) and the ability of the mixture to maintain growth performance in dairy heifers. The first experiment was an ensiling study using laboratory silos. Ensiled blends were 100% WDG, 85% WDG with 15% SH, and 70% WDG with 30% SH on an as-fed basis. Silos were opened for analysis on d 0, 3, 7, and 21. The pH was less in the 100% WDG compared with other treatments (P < 0.01), but all treatments had a pH near 4. Lactic acid concentration was greater in 100% WDG compared with the blends of WDG and SH (P < 0.01). Acetic acid was not found in 100% WDG and increased over time in the 2 blends (P < 0.01). Other differences between blends, such as DM, CP, ammonia N, fiber, and fat, were reflective of the different concentrations of WDG and SH in the blends. In the second experiment, the 70% WDG and 30% SH (as-fed) blend was ensiled in a silo bag and then evaluated as a feed for growing dairy heifer diets. Twenty-four heifers were used in a randomized complete block design and assigned to be fed 1 of 3 diets: 1) control, 2) low inclusion of WDGSH, and 3) a high inclusion of WDGSH. All treatment diets consisted of 50% brome grass hay on a DM basis. The control diet had 50% of the diet (DM basis) as a grain mix, which was composed of corn, soybean meal, and minerals. The low WDGSH diet contained 24.4% of the blend and 25.6% grain mix. The high WDGSH diet contained 48.7% of the blend and 1.3% mineral mix. Average daily gain and most of the body growth measures were similar among treatments. However, DMI decreased linearly (P < 0.01) as the WDGSH blends were fed, resulting in improved (P = 0.02) G:F. Results from these experiments indicated that WDG can be effectively ensiled with SH and sustain adequate growth rate when fed to growing dairy heifers.

The Pure water (P,T)-phase diagram is known in the form of empirical equations or tables from nearly a century as a result of Bridgman's work. However, few data are available on other aqueous systems probably due to the difficulty of high-pressure measurements. As an alternative, six approaches are presented here to obtain the food phase diagrams in the range of pressure 0.1-210 MPa. Both empirical and theoretical methods are described including the use of an artificial neural network (ANN). Experimental freezing points obtained at the laboratory of the authors and from literature are statistically compared to the calculated ones. About 400 independent freezing data points of aqueous solutions, gels, and foods are analysed. A polynomial equation is the most accurate and simple method to describe the entire melting curve. The ANN is the most versatile model, as only one model allows the calculation of the initial freezing point of all the aqueous systems considered. Robinson and Stokes' equation is successfully extended to the high pressures domain with an average prediction error of 0.4 degrees C. The choice of one approach over the others depends mainly on the availability of experimental data, the accuracy required and the intended use for the calculated data.

Oca (Oxalis tuberosa Mol.) is an under-utilized tuber crop from the Andean region. Cryopreservation would allow the safe and long-term preservation of the genetic resources of this crop. A protocol for the cryopreservation of in vitro grown shoots has been developed using the vitrification solution PVS2. Two genotypes were studied (G1 and G27). Nodal segments were cultured on MS medium and incubated at 10 degree C with 16 h photoperiod and 10 mol per square meter per second irradiance, for two weeks. Apices were then excised and cultured on MS+0.15 M sucrose for 3 days at 5 degree C in darkness. Subsequently, apices were immersed in a loading solution (liquid MS medium+2 M glycerol+0.4 M sucrose), and then treated with the vitrification solution PVS2 for 0 to 40 minutes. Cryovials were then immersed in liquid nitrogen. Four weeks after rewarming and culture on recovery medium, genotype G1 showed approximately 60 percent recovery (normal growth) with 20 min PVS2 treatment. Genotype G27 showed lower recovery (30 percent). Differential scanning calorimetry yielded a Tg midpoint for PSV2 solution of ca. -120 degree C. Calorimetric studies on apices at different stages of the cryopreservation protocol showed a change in calorimetric parameters consistent with a decrease in the amount of frozen water as the protocol advanced.

We performed genomic subtraction coupled to microarray-based gene expression profiling and identified the PDZ (postsynaptic density-95/Discs large/zona occludens-1)-binding kinase/T-LAK (lymphokine-activated killer T cell) cell originating protein kinase (PBK/TOPK) as a gene highly enriched in neural stem cell cultures. Previous studies have identified PBK/TOPK as a mitogen-activated protein kinase (MAPK) kinase that phosphorylated P38 MAPK but with no known expression or function in the nervous system. First, using a novel, bioinformatics-based approach to assess cross-correlation in large microarray datasets, we generated the hypothesis of a cell-cycle-related role for PBK/TOPK in neural cells. We then demonstrated that both PBK/TOPK and P38 are activated in a cell-cycle-dependent manner in neuronal progenitor cells in vitro, and inhibition of this pathway disrupts progenitor proliferation and self-renewal, a core feature of progenitors. In vivo, PBK/TOPK is expressed in rapidly proliferating cells in the adult subependymal zone (SEZ) and early postnatal cerebellar external granular layer. Using an approach based on transgenically targeted ablation and lineage tracing in mice, we show that PBK/TOPK-positive cells in the SEZ are GFAP negative but arise from GFAP-positive neural stem cells during adult neurogenesis. Furthermore, ablation of the adult stem cell population leads to concomitant loss of PBK/TOPK-positive cells in the SEZ. Together, these studies demonstrate that PBK/TOPK is a marker for transiently amplifying neural progenitors in the SEZ. Additionally, they suggest that PBK/TOPK plays an important role in these progenitors, and further implicates the P38 MAPK pathway in general, as an important regulator of progenitor proliferation and self-renewal.

Estudiamos la sensibilidad a antimicrobianos de las bacterias aisladas en esputos de enfermos con fibrosis quística en nuestro hospital durante los años 2001 y 2002. Los microorganismos más frecuentemente aislados fueron, por orden, Staphylococcus aureus (59,89%), Pseudomonas aeruginosa (49,45%), Stenotrophomonas maltophilia (4,9%) y Haemophilus influenzae (3,8%). El porcentaje de S. aureus resistentes a la meticilina fue de un 18%, y que no se encontró ninguna cepa con sensibilidad disminuida a los glucopéptidos. Los carbapenémicos fueron los antimicrobianos con mayor actividad frente a P. aeruginosa, aunque sin alcanzar el 100% de eficacia (AU)


We studied the antimicrobial susceptibility of bacteria isolated from sputum from patients with cystic fibrosis in our hospital during 2001 and 2002. The most frequently isolated microorganisms were Staphylococcus aureus (59.89%), Pseudomonas aeruginosa (49.45%), Stenotrophomonas maltophilia (4.9%) and Haemophilus influenzae (3.8%). The rate of methicillin-resistant S. aureus was 18%, and no strains with low susceptibility to glycopeptides were found. Carbapenems showed the highest activity against P. aeruginosa, although this did not reach 100% (AU)
