RESUMO

In RBL-2H3 rat basophilic leukemia cells, Fc epsilon R1 crosslinking by multivalent antigen stimulates phosphatidylinositol (PI) turnover and Ca2+ influx and causes functional responses that include secretion, membrane ruffling and actin polymerization. Here, we show that the tyrosine kinase inhibitor, genistein, inhibits antigen-induced PI turnover, determined from assays of 1,4,5-inositol trisphosphate production, and impairs receptor-mediated secretion, ruffling and actin polymerization. Genistein has little effect on several functional responses to stimuli that bypass PI hydrolysis (ionomycin-induced secretion, phorbol ester-induced ruffling) but it inhibits phorbol ester-induced actin polymerization. These data implicate a common tyrosine kinase-dependent event, most likely the activation of phospholipase C gamma, in the Fc epsilon R1-mediated stimulation of PI turnover, secretion and ruffling. There may be additional tyrosine kinase-mediated events in the actin assembly pathway.

RESUMO

A study was done to determine perceived importance of medication information and the kind of information provided to patients in an outpatient setting. A scale describing 14 points of medication information was used prior to the physician's visit in one study phase and afterwards in the second phase. Results showed that patients perceived all 14 items to be important but rated precautions to observe, drug interactions, and adverse effects most highly. Also, less than half the time was information considered highly important by patients provided by their physicians.

A clinical pharmacy service providing blood cholesterol testing and consultation was implemented in a community pharmacy. In this pilot study, 27 patients each paid $10 to have a serum cholesterol determination. All subjects completed a questionnaire to determine attitude toward such a service, intention to use it in the future, and willingness to pay for its use. A posttest patient satisfaction questionnaire was completed after blood-level results were reported and discussed with the patient. All subjects stated that they were strongly in favor of such a service, that they would use it, and that they would pay an average of $11.60 for the test. On the posttest questionnaire, patients expressed strong satisfaction with various aspects of the service, and the amount that they were willing to pay increased significantly to $14.35. It is recommended that community pharmacies implement such patient-oriented services.

RESUMO

A program which utilizes patient instructors (PI) to teach and evaluate interviewing skills of pharmacy students is described. The PIs were programmed with a history of either hypertension, chronic pulmonary disease, or congestive heart failure. Content areas within each history included past and present drug therapy, adverse drug reactions, drug interactions, as well as socioeconomic and other factors affecting therapy. The interviews were done with small groups of students and followed by PI evaluation of interview content and process. The majority of students felt the experience helped them assess and improve their interviewing skills and agreed the program should continue. The advantages of utilizing PIs to teach interviewing skills are discussed.

These studies were conducted to examine the basic characteristics of postextrasystolic potentiation (PESP) and the relationship of loading effects to PESP. Measurements of left ventricular (LV) and aortic pressures, the rate of pressure rise, and echocardiographically determined LV dimensions were made in anesthetized open-chest dogs. The hearts were paced, and timed extrasystoles were introduced that were followed by postextrasystoles (PES). PES's were elicited after an interval equal to either a full compensatory pause or a time when the diastolic properties of the LV could not be distinguished from control (isolength). Potentiation of contraction for the PES's introduced after an isolength pause was dependent on both the heart rate and the extrasystolic interval, whereas the PES's that occurred after a full pause showed no dependence on either of these intervals. PESP elicited during the isolength period was not dependent on either preload and afterload. It is concluded that PESP depends on the combination of heart rate and extrasystolic and postextrasystolic intervals. Further, PESP may be inaccurate in assessing contractile reserve unless the heart rate and extrasystolic interval are known and the PES is introduced after an isolength pause.

Twelve-day normal and dystrophic chick embryo breast muscle cells were cultured for up to 14 days, using normal embryo extract in the culture medium. Fluorodeoxyuridine was added on day 3 to suppress fibroblast overgrowth. Three of 6 experiments with normal cells were severely infected with avian leucosis/sarcoma (ALS) virus particles. The findings appear to lend support to the suggestion that the ALS virus is a mitochondriophage. Infected cultures demonstrated a depressed rate of total protein synthesis as reflected by incorporation of [3H]leucine. Extractable protein and total protein content were also depressed within several days after ALS particles were identified. These findings reinforce caution in acceptance of the common assumption that viral infections are an unavoidable and metabolically benign component of muscle cell cultures.

RESUMO

A pharmacy clinic was established to provide consultation and services to practitioners and patients in the ambulatory care department of a Veterans Administration hospital. Among the services provided by the pharmacist are drug therapy consultations and monitoring, drug use review, drug information, and patient education. The pharmacy clinic assures the staff of the pharmacist's availability for consultation, allows the pharmacist to better organize the time required for consultation, makes the services of the pharmacist available to all outpatients, and provides a good training environment for students.
