RESUMO

Hepatocellular carcinoma (HCC) is one of the major causes of cancer-related death worldwide. However, the molecular mechanism underlying HCC carcinogenesis remains to be further elucidated. Up-frameshift protein 1 (UPF1) is a RNA/DNA-dependent ATPase and ATP-dependent RNA helicase. Here, we explored the expression and function of UPF1 in HCC. In this study, we demonstrated that UPF1 expression was significantly reduced in hepatocellular carcinoma (HCC) tissues compared with the adjacent normal tissues. And further functional assays revealed that knockdown of UPF1 promoted HCC cells growth and invasion. Furthermore, we found that UPF1 could bind to long non-coding RNA urothelial cancer associated 1 (UCA1) and was negatively correlated with UCA1. UCA1 expression also affected HCC growth and invasion. Knockdown of UCA1 ameliorated the effect of UPF1 knock down on HCC growth and invasion. Knockdown of UPF1 enhances glycolysis in HCC. Taken together, our results provided new insights for finding novel therapeutic targets for hepatocellular carcinoma progression.

AIMS: The purpose of this study was to assess the effects of adjuvant interventions for radioiodine in patients with thyroid cancer. MATERIALS AND METHODS: A systematic search was undertaken on July 9, 2014. RevMan 5 software was used to synthesize data. RESULTS: 13 randomized controlled trials were included. The pooled risk ratio of 0.99 (95% confidence interval (CI): 0.96-1.02, p = 0.58) indicated no significant difference in successful thyroid remnant ablation between recombinant human thyroid-stimulating hormone (rhTSH) and thyroid hormone withdrawal in 7 trials. The percentage of patients who had successful ablation was significantly higher in the oral-lithium group than in the control group (p = 0.017). A computerized decision aid improves informed decision-making in patients with early-stage papillary thyroid cancer (PTC) who are considering adjuvant radioiodine treatment (p < 0.001). Amifostine pretreatment did not prevent parenchymal damage to the major salivary gland function after radioiodine treatment (p = 0.2461). CONCLUSIONS: The present systematic review suggests that rhTSH, lithium, and computerized decision aids maybe act as beneficial adjuvant interventions for radioiodine in patients with thyroid cancer; however, amifostine does not exhibit helpful effects in thyroid cancer patients treated with radioiodine.

OBJECTIVE: To explore the clinical characteristics and prognosis of patients with idiopathic sudden sensorineural hearing loss (ISSHL) with vertigo. METHOD: By analyzing the clinical data of 271 ISSHL patients, they were divided into without vertigo group (n = 169) and vertigo group (n = 102). In vertigo group, 34 cases were patients with benign paroxysmal positional vertigo (BPPV) secondary to the ISSHL. All patients received conventional treatment. According to the types of BPPV, patients with secondary BPPV received Epley maneuver or Barbecue roll maneuver. By analyzing the results of the pure tone audiometry test and treatment outcomes of the patients, we summarized the clinical characteristics of ISSHL patients with vertigo. RESULT: The audiometric curves of ISSHL with vertigo group were mainly at high frequency. The degrees of hearing loss of these patients were severe and profound. After treatment, the improvement of hearing threshold for ISSHL with vertigo group was lower than that for ISSHL without vertigo group. What's more, the rate of recovery, success and total effective of audition for ISSHL with vertigo group was also obviously lower than that for ISSHL without vertigo group. Of all the patients with BPPV, 27 cases of posterior semicircular canal and 7 cases of lateral semicircular canal were identified. All patients with BPPV were diagnosed as the same ears as the ISSHL. CONCLUSION: ISSHL with vertigo group lost hearing more severely than ISSHL without vertigo group. Also, the improvement of hearing and the effective after treatment were really poor. The symptoms of ISSHL with BPPV group improved and eased significantly than that of ISSHL without BPPV group. The major of BPPV secondary to the ISSHL occurs in the posterior semicircular canal. The canalith repositioning is an effective therapy to the secondary BPPV.

OBJECTIVE: To investigate the pathological changes of chronic intermittent hypoxia on upper and lower respiratory tissue in experimental rats. METHODS: A total of 40 female SD rats were divided into 2 groups. Twenty rats were fed in normal state (control group), and 20 rats were fed in hypoxia environment (hypoxia group). The pathological changes of upper and lower respiratory tissue were observed under optical microscope. RESULTS: Chronic intermittent hypoxia resulted in irreversible changes both at upper and lower respiratory tract in rats. The thickness of the lamina propria in soft palate was significantly increased in hypoxia group (125.85 ± 6.34) µm vs. (57.26 ± 4.67) µm (t=36.330, P<0.01). Lung pathological examination in hypoxia group showed pulmonary interval thickening (2.15 ± 0.49) µm vs. (0.45 ± 0.12) µm (t=14.132, P<0.01). CONCLUSIONS: This study confirmed that long-term hypoxia can lead to organization reconstruction in upper and lower respiratory tract in rats. In OSAHS patients, it is suggested that earlier intervention could alliviate the pathological changes in respiratory system.

OBJECTIVE: To explore the role of NF-κB p65 and p38 MAPK in lung injury in rats suffered chronic intermittent hypoxia.
 METHODS: Experiment were divided into a control group and a hypoxia group (n=20). After 5 weeks of hypoxia treatment, the expression of NF-κB p65 and p38 MAPK in lung tissues of rats were detected by optical microscope and immunohistochemical techniques and Western blot.
 RESULTS: Pathological examination showed pulmonary interval thickness was (0.45 ± 0.12) µm and (2.15 ± 0.49) µm in the control and hypoxia group, respectively (P<0. 05). Compared with the control group, the levels of both NF-κB p65 and p38 MAPK were increased in the hypoxia group (P<0. 01), which were confirmed by Western blot.
 CONCLUSION: NF-κB p65 and p38 MAPK may play important roles in lung injury reconstruction in of the rats treated by chronic intermittent hypoxia.

OBJECTIVE: Detection and significance of CXCL8, IL-10 in serum and tissue of the patients' with OSHAS. METHOD: All subjects take serum CXCL8 and IL-10 levels were determined by ELISA. All subjects take the same position of the soft palate tissues in the operation. CXCL8 and IL-10 in the tissue expression was measured by immune histochemistry. RESULT: (1) Serum CXCL8 levels in OSHAS is higher than that in the control group (P < 0.05). (2) Serum IL-10 levels in OSHAS is lower than that in the control group (P < 0.05). (3) Tissues CXCL8 expression was correlated with serum CXCL8 (r = 0.474, P < 0.05). (4) Tissues IL-10 expression wasn't any correlated with serum IL-10. (5) Tissues IL-10 expression was correlated with tissues CXCL8 (r = 0.482, P < 0.05). CONCLUSION: OSAHS may be a disease associated with inflammation, and closely relate with CXCL8 and IL-10.
