RESUMO

Long noncoding RNAs (lncRNAs) stand as indispensable regulators of initiation and development in melanoma (melanoma). However, the action molecular mechanisms linked to melanoma remain unclear. In the current study, the findings revealed that AGAP2-AS1 was considerably greater in melanoma than in healthy tissues and that the level of AGAP2-AS1 in cancer tissue was significantly linked to the cancerous TNM stage of patients. Individuals with high AGAP2-AS1 had a considerably shorter survival duration than patients with low AGAP2-AS1, regardless of progression-free survival or overall survival. Functionally, downregulating the expression of AGAP2-AS1 can inhibit the growth of melanocytes. Compared with the control group, AGAP2-AS1 knockdown increased Erastin-mediated iron death in melanoma cells. However, iron death inhibitor FERSINT-1 restored this effect, while Erastin induced melanoma cell death. Besides, intracellular iron and Fe2+ levels increased after AGAP2-AS1 knockdown in melanoma cells treated with Erastin compared with the si-NC group. In addition, AGAP2-AS1 silencing resulted in a significant decrease in glutathione (GSH) content in Erastin-treated melanoma cells. The mechanistic results suggest AGAP2-AS1 increases SLC7A11 mRNA stability through the IGF2BP2 pathway. In this investigation, we discovered new activities for AGAP2-AS1 and firstly discovered its mechanistic basis in ferroptosis and melanoma formation that might help in the search for potential therapy options in melanoma.

Objectives: This study aimed to identify the independent factors associated with depression in community-dwelling older adults in Wuhan, China. Methods: Four hundred and seventy older adults (aged ≥65 years) from four communities dwelling on Junshan Street in Wuhan, China were included in this study. Participants completed a questionnaire that asked questions pertaining to age, gender, educational level, income, living situation, care situation, social support, and social engagement. The 30-item Geriatric Depression Scale (GDS-30), the Fried frailty phenotype scale, the activities of daily living (ADL) scale, the mini nutritional assessment scale-short form (MNA-SF), and the Mini-cog scale were used to assess depression, frailty, self-care ability, malnutritional risk, and cognitive dysfunction, respectively. Differences in age, gender, educational level, income, living situation, care situation, social support, social engagement, ADL score, risk of malnutrition, frailty, and cognitive dysfunction between the non-depression (GDS-30 score <10 points) and depression groups (GDS-30 score ≥10 points) were compared using a chi-square test. Moreover, correlations between factors and depression were analyzed using Pearson's correlation. Then, significant variables (p < 0.05) from the chi-square test were included in a multivariable logistic regression model to identify the independent factors associated with depression. Results: The incidence of depression among the participants was 14.04%. Age (p < 0.001), educational level (p < 0.001), living situation (p < 0.001), social support (p = 0.001), ADL score (p = 0.023), frailty (p < 0.001), and cognitive dysfunction (p < 0.001) were all significantly associated with depression, in which age, poor social support, frailty, and cognitive dysfunction were identified as independent factors. Conclusion: Improving social support and effective interventions for frailty and cognitive dysfunction may help relieve depression in community-dwelling older adults.

RESUMO

Here, we prepared small graphite nanoflakes (SGN) by a new strategy of pulverization as the cathode of aluminum ion batteries (AIBs). Electrochemical measurements show that SGN has a very high discharge capacity, excellent rate performance and good cycling stability mainly due to its enlarged edge plane, reduced thickness and high crystallinity. This work provides a new route for preparing high performance graphite-based materials for AIBs.

RESUMO

A new method using TurboFlow on-line cleanup liquid chromatography combined with tandem mass spectrometry (MS/MS) has been developed for simultaneous determination of six progesterones including 19-norethindrone, 17alpha-hydroxyprogesterone, medroxyprogesterone, megestrol acetate, progesterone and melengestrol acetate in milk. Samples were extracted by acetonitrile. The analytes in extract were on-line purified directly on Cyclone-P purification column where the sample matrices were washed away. Subsequently, the analytes which were eluted from the extraction column onto Phenyl-Hexyl column were separated with a gradient elution, and detected with electrospray ionization mass spectrometry in the positive scan mode using multiple reaction monitoring (MRM). The isotope internal standards were employed for quantification. As a result, the linearities were satisfactory with the correlation coefficients of > 0.999 at concentrations ranging from 0.1 microg/L to 50 microg/L. Based on the repeated analysis of a known blank sample, the limit of quantification (LOQ) is 0.5 microg/kg. Average recoveries of the analytes fortified at three levels (1, 5 and 25 microg/kg) ranged from 90.8% to 107.5%, and the relative standard deviations (RSDs) ranged from 6.3% to 11.8%. This proposed method is simple, rapid, sensitive and highly selective, and can be applied to simultaneous identification an quantification of the six progesterones in milk.

Room temperature ferromagnetism has been observed in SnO2 nanowires synthesized by a chemical vapor deposition using Au layers as catalyst. The nanowires are homogeneous and single-crystalline grown along the [101] direction, with diameters ranging from 25 to 100 nm and length greater than 20 microm. The special magnetization reaches 0.114 emu/g for the nanowires with diameter of approximately 25 nm and reduces with increasing diameters. Branched SnO2 nanowires were prepared via a two-step vapor-liquid-solid approach, and an enhanced magnetization was obtained. To the contrary, the nanowires annealed at 1300 degrees C in air were completely transformed into the particles and exhibit weakened magnetization. These results demonstrate that the ferromagnetic properties of the samples depend on the surface-to-volume ratio of nanowires. With a combined study of photoluminescence, our results reveal that the oxygen vacancies at the surface of nanowires contribute to the ferromagnetism of SnO2 nanowires. This argument is further confirmed by a sequential annealing in a rich-oxygen atmosphere, then in a low vacuum condition.

SnO2 and Ni(x)Sn(1-x)O(2-delta) (x = 0.007-0.043) 1D nanostructures are fabricated using a catalyzer assisted chemical vapor deposition (CVD). The morphology of the 1D nanostructure is sensitive to the fabrication conditions. As the Ar flux rate is decreased from 50 sccm to 40 sccm, the 1D nanostructure changes from nanowire to nanobelt. All of the Ni(x)Sn(1-x)O(2-delta) 1D nanostructures exhibit room temperature ferromagnetism (RTFM). With the increasing x, magnetic moment per Ni ion increases at first, reaches a maximum of 3.33 microB in x = 0.025, then decreases. The results of annealing in vacuum and oxidizing atmospheres reveal that oxygen vacancies play a crucial role in introducing ferromagnetism, which implies that the origin of RTFM can be understood by the bound magnetic polaron model (BMP).

In order to get magnetic films with high permeability and high resistivity for applications in high frequency devices, (Fe65Co35)x(SiO2)1-x nano-granular films with low metal volume fraction were fabricated by radio frequency magnetron sputtering, and excellent soft magnetic properties have been acquired by annealing the sample with x = 0.38 at 260 degrees C in a magnetic field. The real part micro' of complex permeability is larger than 100 when frequency f is lower than 0.7 GHz, and the ferromagnetic resonance frequency f(r) is as high as 1.8 GHz. More interestedly, the resistivity of this sample reaches 13.4 momega x cm, 4 orders higher than that of pure FeCo alloy. Ferromagnetic resonance experiment and deltam-H curve study were employed to elucidate the mechanism of soft magnetic properties. The results reveal that the intergranule exchange coupling plays an important role in realizing good soft magnetic properties for metal-insulator granular films.
