RESUMO

The objective of this study was to evaluate genetic variability and select putative mutants of Terra Maranhão plantain cultivar (AAB genome) subjected to gamma radiation based on agronomic data and inter simple sequence repeat molecular marker profiles using the Ward-MLM strategy. A total of 233 irradiated plants and 41 controls were assessed. The agronomic and molecular data were subjected to the Ward-MLM statistical algorithm in the SAS program. Cluster analysis was performed by the average distance method (UPGMA), based on the distance matrix of the Gower algorithm, and the cophenetic correlation coefficient calculated using the R software. The distance between the putative mutants ranged from 0.321 to 0.524, with an average distance of 0.426, and a cophenetic correlation coefficient of 0.79. Three putative mutants, which were selected based on the best agronomic traits and low height, will undergo further evaluation in the next stages of the banana breeding program at Embrapa. These results describe the first attempt of using combined data of Terra Maranhão plantain cultivar for the purpose of selecting mutants and assessing genetic variability.

The aim of this study was to select putative Terra Maranhão plantain cultivar mutants obtained by gamma radiation, with good agronomic traits and short height. A total of 315 buds were irradiated in vitro with gamma rays in doses of 20 Gy and were subcultivated and evaluated in the field over 2 production cycles. The clones were evaluated to select the best 10% of the plants. Cultivation was undertaken at a spacing of 3 x 4 m, and fertilization was carried out according to the technical recommendations for the crop. A total of 111 irradiated plants and 41 controls were evaluated in the field. Among the irradiated plants selected, genotypes that exhibited reduced height were observed. The genotypes Irra 04, Irra 13, Irra 19, and Irra 21 exhibited a height of 3.6 m, which was below the mean value of the controls selected. Other irradiated genotypes selected such as Irra 14 and Irra 16, with a height of 3.65 m, are promising because, in addition to reduced height, they exhibited good bunch weight and shorter period to flowering in relation to the mean value of the controls, which is a significant factor for the next stages in breeding. These results confirm the possibility of inducing mutations in Terra type banana plants to obtain desirable agronomic traits and short height.

We investigated 10 similarity (and disimilarity) coefficients in a set of 40 wild genotypes of Croton linearifolius subjected to analyses using hierarchical grouping methods, grouping methods by optimization and data projection in two-dimensional space. Genotypes were characterized by analyzing DNA polymorphism with the use of 15 ISSR and 12 RAPD markers. The distance measurements were compared by the Spearman correlation test, projection in two-dimensional space and grouping efficiency evaluation. The Spearman correlation coefficients between the 10 coefficients evaluated were significant (P < 0.001) and indicated significant changes in genotype ranking due to type of coefficient used (0.76 ≤ rs ≤ 1). Wide variation was also observed in the efficiency of clustering methods, where the unweighted pair group method with arithmetic mean was the most suitable (0.3 ≤ D ≤ 1.5 ; 0.41 ≤ rc ≤ 0.77; 5.99 ≤ S ≤ 12.61). Projection efficiencies in two-dimensional space showed high-stress values (65 < S < 89%). Similar to the results observed for hierarchical clustering methods and for projection in two-dimensional space, the formation of groups with grouping methods by optimization showed variations when using different coefficients. We believe that the results confirm the influence of coefficients in studies of genetic diversity, showing the need to use criteria and standards for selecting appropriate methods for genetic studies of the genus Croton.

UNLABELLED: IV droperidol inhibits epidural morphine-induced pruritus, but this effect disappears when the dose is increased from 2.5 to 5.0 mg. This study was performed to determine whether epidural droperidol would have a similar effect. In this double-blinded study, we enrolled 140 patients undergoing Cesarean delivery under epidural anesthesia who were randomly allocated to four groups. Anesthesia consisted of 150 mg of 0.5% bupivacaine with 1:200,000 epinephrine, with 2 mg of morphine and 0.0, 1.25, 2.5, or 5.0 mg of droperidol (Groups 1 to 4). During the postoperative period, patients were assessed for pruritus (absent, mild, moderate, or severe) and other untoward symptoms. The chi(2) test was used to compare the incidence of the side effects. For the analysis of pruritus, we used the Mantel-Haenszel test for linear association. Droperidol induced a dose-related reduction in the incidence of pruritus (P < 0.001). This reduction was independent of the incidence of somnolence, which increased with droperidol dose (P < 0.05 when the incidence of somnolence in Groups 1 and 4 was compared). We conclude that droperidol, in doses up to 5 mg epidurally, induces a dose-related reduction in the incidence of pruritus without inducing significant side effects. IMPLICATIONS: Epidural morphine is effective for pain control but yields some side effects, including pruritus, that can be severe. Studying patients undergoing Cesarean delivery, we found a dose-related reduction in the incidence of pruritus using epidural droperidol.

BACKGROUND AND OBJECTIVES: Because the mechanism of inhibition of epidural morphine-induced pruritus by droperidol is not clear, this study was undertaken to determine the effects of larger doses of droperidol or morphine, or both. METHODS: A double-blind study was performed in 210 ASA I or II patients undergoing cesarean delivery, who were randomly assigned to six groups. All patients received epidural anesthesia with 0.5% bupivacaine containing 1:200,000 epinephrine, to which 2 mg (groups 1, 2, and 3) or 4 mg (groups 4, 5, and 6) morphine sulfate was added. Just after delivery, 2.5 mg droperidol was given intravenously to groups 2 and 5, and 5 mg was given to groups 3 and 6. During the postoperative period, the patients were assessed for the occurrence and severity of pruritus (classified as absent, mild, moderate, or severe) or other untoward symptoms. The groups were compared for the incidence of pruritus by the Mann-Whitney nonparametric test. RESULTS: The incidence of pruritus was significantly reduced only when the control group. (no droperidol) was compared with the group that received 2.5 mg droperidol, both when 2 mg and when 4 mg morphine was used. A 5-mg dose of droperidol had no inhibitory effect. There was no difference in the incidence of pruritus between use of 2 mg and 4 mg morphine. Other untoward effects of morphine either could not be observed or occurred with an incidence unaffected by either dose of droperidol. CONCLUSION: Pruritus caused by epidural use of 2 or 4 mg of morphine is inhibited by the intravenous use of 2.5 mg droperidol but not by a larger dose.
