The roles of epigenetic regulation in graft-versus-host disease.

Allogeneic hematopoietic stem cell transplantation (aHSCT) is utilized as a potential curative treatment for various hematologic malignancies. However, graft-versus-host disease (GVHD) post-aHSCT is a severe complication that significantly impacts patients' quality of life and overall survival, becoming a major cause of non-relapse mortality. In recent years, the association between epigenetics and GVHD has garnered increasing attention. Epigenetics focuses on studying mechanisms that affect gene expression without altering DNA sequences, primarily including DNA methylation, histone modifications, non-coding RNAs (ncRNAs) regulation, and RNA modifications. This review summarizes the role of epigenetic regulation in the pathogenesis of GVHD, with a focus on DNA methylation, histone modifications, ncRNA, RNA modifications and their involvement and applications in the occurrence and development of GVHD. It also highlights advancements in relevant diagnostic markers and drugs, aiming to provide new insights for the clinical diagnosis and treatment of GVHD.

Effect of GO content on microstructure and mechanical properties of Ti6Al4V coating reinforced artificial joint.

In this study, we have innovatively proposed a method of in-situ synthesized TiC hard phase to improve the surface mechanical properties of artificial joint materials (Ti6Al4V). In order to explore the optimum graphene oxide (GO) addition, GO/Ti6Al4V composite powders with different proportions (0, 0.5, 1.0, and 1.5 wt.%) were prepared. The homogeneously dispersed GO/Ti6Al4V composite powder was prepared on Ti6Al4V substrate by laser cladding technology. The microstructure, phase composition, and mechanical behavior of GO/Ti6Al4V composite coatings were studied by scanning electron microscope (SEM), optical microscope (OM), energy dispersive spectrometer (EDS), tribometer, hardness tester, and surface profiler. The results showed that the addition of GO could significantly improve the mechanical properties of TC4 substrate. During the preparation of the coating, the grain size of in-situ TiC phase was nanoscale and was distributed between acicular martensite, which played a critical role in enhancing the mechanical properties of the coating. The TiC phase distributed between acicular martensite refine the grain size of α' phase and improve the cutting resistance of the coating. Nevertheless, excessive GO decreased the fluidity of the molten pool, and micro holes tended to generate in the coating, which had a negative impact on the mechanical properties of the coating. At the GO content of 0.5 wt.%, the microhardness of the GO/Ti6Al4V coating was 1.325 times that of pure Ti6Al4V. Under the friction environment of simulated body fluid solution, the average friction coefficient was approximately 0.307 and the wear rate decreased to 3.5 × 10-7 mm3/N · m.

Transcatheter closure of atrial septal defect associated with arrhythmogenic right ventricular cardiomyopathy: a case report and literature review.

Arrhythmogenic right ventricular cardiomyopathy is characterised by progressive, fibrofatty replacement of myocardium, and ventricular arrhythmias, and its prognosis is usually poor. Arrhythmogenic right ventricular cardiomyopathy associated with atrial septal defect is very rare, and this combination may make the diagnosis, treatment, and prognosis difficult. We present a case of a patient with this association who underwent interventional treatment with a septal defect occluder. Transcatheter closure of atrial septal defect in a patient with arrhythmogenic right ventricular cardiomyopathy is hitherto unreported. During a 3-year follow-up he remained relatively stable. We also review the cases reported in the medical literature describing this uncommon association between arrhythmogenic right ventricular cardiomyopathy and atrial septal defect or patent foramen ovale.

[Clinical observation on therapeutic effect of the pressing plantar reflex area with wooden needle for treatment of patients with insomnia].

OBJECTIVE: To investigate the therapeutic effect of wooden needle on the patients with insomnia. METHODS: One hundred and twenty patients with insomnia were randomly divided into a wooden needle group and a western medicine group, 60 cases in each group. In the wooden needle group, the patients were treated with wooden needle to press the plantar reflex areas, such as cerebellar, throid and cerebral areas. In the western medicine group, Alprazolam was taken orally. Before and after treatment, Pittsburgh Sleep Quality Index (PSQI) was used to evaluate the therapeutic effect of both groups. RESULTS: The total therapeutic effect was 100.0% in the wooden needle group, while it was 90.7% in the western medicine group, there was no significant difference between the two groups (P > 0.05). Compared with PSQI before and after treatment, there was difference in the both groups (All P < 0.01), but there was no difference between the two groups (P > 0.05). CONCLUSION: The therapeutic effect of wooden needle is similar to that of Alprazolam on the insomnia, indicats that wooden needle is a better therapy for treating insomnia.
