RESUMO

Neuronal computation is achieved through connections of individual neurons into a larger network. To expand the repertoire of endogenous cellular communication, we developed a synthetic, photon-assisted synaptic transmission (PhAST) system. PhAST is based on luciferases and channelrhodopsins that enable the transmission of a neuronal state across space, using photons as neurotransmitters. PhAST overcomes synaptic barriers and rescues the behavioral deficit of a glutamate mutant with conditional, calcium-triggered photon emission between two neurons of the Caenorhabditis elegans nociceptive avoidance circuit. To demonstrate versatility and flexibility, we generated de novo synaptic transmission between two unconnected cells in a sexually dimorphic neuronal circuit, suppressed endogenous nocifensive response through activation of an anion channelrhodopsin and switched attractive to aversive behavior in an olfactory circuit. Finally, we applied PhAST to dissect the calcium dynamics of the temporal pattern generator in a motor circuit for ovipositioning. In summary, we established photon-based synaptic transmission that facilitates the modification of animal behavior.

Rab1 is a highly conserved small GTPase that exists in humans as two isoforms: Rab1A and Rab1B, sharing 92% sequence identity. These proteins regulate vesicle trafficking between the endoplasmic reticulum (ER) and Golgi and within the Golgi stacks. Rab1A and Rab1B may be oncogenes, as they are frequently dysregulated in various human cancers. Moreover, they contribute to the progression of Parkinson's disease. The availability of high-quality antibodies specific for Rab1A or Rab1B is essential to understand the distinct functions of these Rab1 proteins in both health and diseaseand to enhance the reproducibility of research involving these proteins. In this study, we characterized seven antibodies targeting Rab1A and five antibodies targeting Rab1B for Western Blot, immunoprecipitation, and immunofluorescence using a standardized experimental protocol based on comparing read-outs in knockout cell lines and isogenic parental controls. These studies are part of a much larger, collaborative initiative seeking to address the antibody reproducibility issue by characterizing commercially available antibodies for human proteins and publishing the results openly as a valuable resource for the scientific community. While uses of antibodies and protocols vary between laboratories, we encourage readers to use this report as a guide to select the most appropriate antibodies for their specific needs.

At least one in five of all planetary nebulae are the product of a common envelope (CE) interaction, where the companion in-spirals into the envelope of an asymptotic giant branch (AGB) star ejecting the nebula and leaving behind a compact binary. In this work we carry out 3D smoothed particle hydrodynamics simulations of the CE interaction between a 1.7 M⊙ AGB star and a 0.6 M⊙ companion. We model the AGB structure using a 1D stellar model taken at the seventh thermal pulse. The interaction takes place when the giant is on the expanding phase of the seventh thermal pulse and has a radius of 250 R⊙. The post-CE orbital separations varies between 20 and 31 R⊙, with the inclusion of recombination energy resulting in wider separations. Based on the observed short in-spiral time-scales, we suggest that thermal pulses can trigger CEs, extending the ability of AGB stars to capture companions into CEs, that would lead to the prediction of a larger population of post-AGB, post-CE binaries. Simulations that include a tabulated equation of state unbind a great deal more gas, likely unbinding the entire envelope on short time-scales. The shape of the CE after the in-spiral is more spherical for AGB than red giant branch stars, and even more so if recombination energy is included. We expect that the planetary nebula formed from this CE will have different features from those predicted by Zou et al. 2020.

Necrotizing enterocolitis (NEC) is a serious condition related to prematurity and the initiation of enteral feeding. In this article, we review the evidence published in recent years on necrotizing enterocolitis risk factors (prematurity, feeding with low-weight formula, existence of intestinal dysbiosis) and protective factors (human milk or donated milk supply, supplementation of human milk with oligosaccharides, probiotics administration, and the determination of disease predictive biomarkers). A systematic review was conducted of preventive, risk and predictive factors for necrotizing enterocolitis in neonates prior to 37 weeks' gestational age, based on a literature search for clinical trials, meta-analyses, randomized controlled trials and systematic reviews published between January 2018 and October 2021. For this purpose, the PubMed, MEDLINE, and Cochrane Library databases were consulted. The literature search obtained 113 articles, of which 19 were selected for further analysis after applying the inclusion and exclusion criteria. The conclusions drawn from this analysis were that adequate knowledge of risk factors that can be prevented or modified (such as alteration of the intestinal microbiota, oxidative stress, metabolic dysfunction at birth, or alteration of the immunity modulation) can reduce the incidence of NEC in premature infants. These factors include the supplementation of enteral nutrition with human milk oligosaccharides (with prebiotic and immunomodulatory effects), the combined administration of probiotics (especially the Lactobacillus spp and Bifidobacterium spp combination, which inhibits bacterial adhesion effects, improves the intestinal mucosa barrier function, strengthens the innate and adaptive immune system and increases the secretion of bioactive metabolites), the supplementation of human milk with lactoferrin and the use of donated milk fortified in accordance with the characteristics of the premature newborn. The determination of factors that can predict the existence of NEC, such as fecal calprotectin, increased TLR4 activity, and IL6 receptor, can lead to an early diagnosis of NEC. Although further studies should be conducted to determine the values of predictive biomarkers of NEC, and/or the recommended doses and strains of probiotics, lactoferrin or oligosaccharides, the knowledge acquired in recent years is encouraging.

La cavidad oral es el sitio más común de carcinoma de cabeza y cuello de células escamosas; enfermedad con gran riesgo de morbilidad y mortalidad a nivel mundial. Aunque la primera opción de tratamiento para paciente con cáncer de la cavidad oral sigue siendo la resección quirúrgica, muchos pacientes que presentan enfermedad avanzada son tratados con un enfoque multidisciplinario. Los pacientes con metástasis linfática, aunque se mantengan los márgenes quirúrgicos negativos, se ha encontrado que pueden tener alto riesgo de recidiva local-regional y muerte por enfermedad, y se recomienda con más frecuencia recibir postoperatorio de radiación, así como la quimioterapia sistémica. La base de este enfoque, así como los avances científicos para futuros tratamientos a los pacientes con alto riesgo de cáncer de cavidad oral; lo cual seria para ellos de gran importancia poder ofrecerles mayor calidad de vida y disminuir el riesgo de mortalidad.


The oral cavity is the most common site of head and neck squamous cell carcinoma, a disease with a high risk of morbidity and mortality worldwide. Although the first treatment option for patients with oral cavity cancer is still surgical resection, many patients with advanced disease are treated with a multidisciplinary approach. Patients with lymphatic metastasis, in spite of keeping surgical margins negative, have been found to have high risk of locoregional recurrence and death from disease, and it is recommended that they receive postoperative radiation and systemic chemotherapy more often. The basis of this approach, as well as scientific and focus on future treatments for patients at high risk of cancer cavity that offer better quality of life and reduce the risk of mortality.
