RESUMO

A liquid chromatographic (LC) method is proposed for measuring 0.485-0.970% morantel tartrate in cattle feeds. The drug is leached from feed, diluted, separated from interfering substances on a silica column, and measured in the effluent stream by 313 nm spectrophotometric detection. Two potential degradation products, i.e., cis-isomer of morantel tartrate and N-(3-methylaminopropyl)-trans-3-(3-methyl-2-thienyl)acrylamide, and a related anthelmintic, i.e., pyrantel tartrate, do not interfere. Average recovery of drug from liquid spiked samples and laboratory blends was 98-100% with a maximum coefficient of variation (CV) of 2.3%. Results for pelleted and crumbled commercial scale feeds ranged from 94 to 102% of label claim, with a maximum CV of 1.5%.

RESUMO

A method for the detection and quantitation of sodium salinomycin in feeds by a liquid chromatographic, post-column reaction system is presented. Sodium salinomycin is leached from feed with hexane, isolated as a dried residue, and dissolved in mobile phase for analysis. The drug is separated from interfering substances on a silica LC column, combined with vanillin reagent in a mixing tee, reacted with vanillin at elevated temperature in a coil, and detected in the effluent stream at 527 nm. Response, as peak area, is linear with concentration. Feed components, other common veterinary drugs, and closely related compounds do not interfere. Average recovery and coefficient of variation (CV) values for liquid spiked feeds and supplements were 101 +/- 3.5% and 100 +/- 0.3%, respectively. Results for laboratory blends and commercial scale feeds were within 6% of intent with CV values ranging from 0.4 to 5.1%.

A new method for the determination of pyrantel tartrate in swine feed an supplements has been developed because the current official AOAC method is not applicable to feeds co-medicated with tylosin. The new method involves: (a) leaching of drug from feed with methanolic NaCl solution, (b) removal of interfering substances by ion pair liquid-liquid extraction and high performance liquid chromatography, and (c) quantitation of pyrantel tartrate by monitoring the ultraviolet absorption of the effluent stream at 313nm. The method of standard addition is used to compensate for the effect of the feed matrix on drug recovery. No interference is encountered from tylosin, carbadox, lincomycin, non-drug components of feeds and supplements, or potential degradation products of pyrantel tartrate, i.e., cis isomer of pyrantel tartrate and (E)-N-(3-methylaminopropyl)-2-thiopheneacrylamide. Results for the assay of 3 lots each of feeds and supplements containing 0.0106 and 0.106% pyrantel tartrate, respectively, were within +/-4% of label claim. Coefficients of variation ranged from 1.6 to 1.8% for feeds and from 1.9 to 3.9% for supplements.

The official first action method for carbadox in swine feed, 42.C01-42.C04, was modified in 2 respects. First, the samples were leached overnight at room temperature instead of boiled for 1 hr. This change avoided problems with overheating and excessive evaporation. Second, the dilution scheme for samples spiked with carbadox standard solution was changed to give absorbance values that were within the optimum working range of all types of spectrophotometers. The modified procedure was collaboratively studied by 21 laboratories. The repeatability standard deviation (sigma0) and reproducibility standard deviation (sigmax) were sigma0 = 0.00029% and sigmax = 0.00056% (8.9% of grand mean) for feeds containing 0.00617% carbadox; and sigma = 0.0012% and sigmax = 0.0019% (9.3% of grand mean) for feeds containing 0.0198% carbadox. The between-laboratory variance ratio was significant for feeds containing 0.0198% carbadox. The mean per cent of intent values for feeds containing 0.00617% carbadox and 0.0198% carbadox were 102% and 104%, respectively. In general, the statistical results were comparable to those previously obtained for the official first action method. Consequently, the modified procedure is not recommended as a replacement for the official first action method.
