RESUMO

Cell cultures isolated by the enzymatic method from the terminal placenta amnion consist mainly from epithelial cells, expressing cytokeratin-7, CD90, and CD73, are characterized by high viability and low proliferative potential. Adhesive cultures of umbilical (Wharton's jelly) cells, despite the fibroblast-like shape of the cells and expression of surface markers, intrinsic to mesenchymal stromal cells, are also characterized by high heterogeneity during the initial stages of culturing, judging by an appreciable share of cytokeratin-expressing cells. The terminal placenta chorionic villi can be a source of cells with the most typical morphology and immunophenotypical profile of the resident multipotent mesenchymal stromal cells, which retain high viability in vitro and have a high proliferative potential.

RESUMO

The expression of mRNA of cytokines and immunoregulatory molecules characterizing the interaction of mesenchymal stromal cells from chorionic villi of postpartum placenta and allogenic mononuclear blood cells was studied during 3-day co-culturing of these cells. The expression of foxp3, il2ra, and il10 mRNA in floating mononuclear cells increased from day 1 to 3 in co-culture, which can refl ect the process of induction of regulatory T cells in the lymphocyte population under the action of mesenchymal stromal cells.

The authors conducted histological, immunohistochemical, and morphometric studies of 47 endomyocardial biopsies and 33 hearts with a high morphological criterion for the diagnosis of arrhythmogenic right ventricular dysplasia after F.I. Marcus et al. (2010), as well as intravital and postmortem cardiac magnetic resonance imaging (MRI). The average residual cardiomyocyte area was 39.7 +/- 12.8%, and the average cardiomyocyte diameter was 10.4 +/- 1.5 microm. Muscle fiber atrophy, lipomatosis, and fibrosis in the right ventricle were detected in all cases, myocarditis in 44%, mitral valve prolapse in 42%, and muscle fiber dyscomplexation in the right ventricular in 33%. The most informative signs for the diagnosis of arrhythmogenic right ventricular dysplasia using MRI are right ventricular contraction dyssynchrony and those using a morphological study are a residual cardiomyocyte area and muscle fiber atrophy in the right ventricle. A decrease in or focal absence of gamma-catenin expression in the muscle fibers was observed only in 60% of the patients.

RESUMO

The novel synthetic derivative of carnosine, (S)-6-hydroxy-2,5,7,8-tetramethylchroman-2-carbonyl-ß-alanyl-L-histidine (S-Trolox™-Carnosine, STC) increases the resistance of rats to experimental acute hypobaric hypoxia (AHH) thus protecting brain from the oxidative damage. This effect is accompanied by better preservation of the acquired skills in Morris water maze possibly by increasing efficiency of the brain antioxidant system. In addition, STC caused an increase in life span of both male and female fruit fly Drosophila melanogaster whereas carnosine increased life span only in male fruit flies. The results indicate that development of the drug based on STC could be beneficial in neurology and gerontology.

RESUMO

To assess dynamics of structural-functional parameters of the circulatory system including the state of the cardiopulmonary baroreflex (CPBR) in patients with ischemic heart disease (IHD) and cardiac failure (CF) on losartan treatment, we studied 14 IHD men with CF (NYHA functional class II-III), mean age 54.6 +/- 7.1 years. Before and after losartan treatment the patients were examined using echocardiography, radiocardiography with 131-I albumin, occlusion plethysmography, 131-I hippuran clearance. CPBP was estimated by the change in circulation flow rate in the forearm in low body rarefaction by means of low pressure camera. Losartan was given in maximal tolerance dose (25-100 mg/day). Examination in the end of the treatment demonstrated diminished venous tone, increased blood flow in the forearm, reduced volume of circulating plasm, elevated hematocrit, higher ratio of early to late filling peaks of the left ventricle, progression of baroreflex dysfunction. Thus, long-term losartan treatment promoted improvement in peripheral vascular tonicity, diastolic function of left-ventricular myocardium, reduction in circulating plasm volume, progression of baroreflex dysfunction.

Inoculation of virus-specific cytotoxic lymphocytes (CTL) to pregnant mice infected with influenza virus was shown to result in a decrease of infectious virus concentration in the lungs and blood of mice as well as in the placentas and fetuses. With an increase in the dose of inoculated CTL, however, premature deliveries with stillbirths were observed as well as deaths of the mice, the highest frequency of fetal deaths being observed after infection of pregnant mice in the first half of pregnancy. It is not excluded that maternal virus-specific CTL played a role in the formation of developmental abnormalities and immunodeficient states in newborns.

Using 51Cr isotope label it was demonstrated that a very low per cent of syngeneic lymphocytes derived from healthy donors and inoculated in the blood stream of uninfected or influenza virus-infected pregnant mice is found in fetuses before delivery. Similar results were obtained after inoculation of virus-specific cytotoxic lymphocytes (CTL) into uninfected pregnant mice. After inoculation into the blood stream of infected pregnant mice of virus-specific CTL their migration into fetuses before delivery increases, being most marked in 25-30% of mice. Intravenous inoculation of excess CTL (10(6) cells) to infected pregnant mice resulted in rapid development of signs of slow influenza infection in the progeny with typical clinical picture and histopathological lesions in organs and tissues. Large doses (10(7)-10(8) cells) of CTL inoculated into the blood stream cause higher reduction and death of fetuses and increase the rate of stillbirths. The role of maternal virus-specific CTL in the pathogenesis of experimental congenital and especially slow influenza infection is discussed.

As a result of our studies, strain differences in the sensitivity of CBA and BALB/c mice to partially attenuated Francisella tularensis strain have been revealed. Relationship between the increased migration of lymphocytes to the liver and lymphoid organs and the intensive development of cell-mediated immunity reactions has been shown. An important role of local reactions (the skin at the site of the inoculation of F. tularessis + a regional lymph node) in the development of the pathological process and the formation of immunity to tularemia infection has been noted. A high level of resistance to F. tularensis strain used for inoculation in BALB/c mice seems to be greatly due to the fact that in these mice more intensive cell-mediated immunity reactions develop at the early stages of infection, than in CBA mice.

RESUMO

Lymph nodes implanted subcutaneously to syngeneic recipients were shown to regenerate after mass cell destruction. Regenerated lymphoid tissue has a resemblance to the cortical zone of intact lymph nodes. Microenvironment of regenerated lymphoid tissue provides homing of lymphocytes. However, migration of 51Cr-labelled lymphocytes to implants declined drastically, as compared to lymphocyte migration to intact lymph nodes. Attenuation of proliferative activity and the data of morphological analysis indicate a more prolonged retention of lymphocytes in implanted lymph nodes. The results obtained could be attributable to only partial recovery of sinus and vessel systems regulating the inflow and outflow of lymphocytes in lymph nodes.

A study was made of dynamics of the content of colony-forming units (CFU) in the spleen and the bone marrow of the nonirradiated F1 (CBA times C57BL) mice to which polysaccharide (PS) from typhoid bacilli was administered. A method of exogenous colony formation was used in this work. A single PS administration increased the CFU count in the bone marrow (2-2 1/2 times) and in the spleen (3 times). Repeated (6- and 9- time) administration of PS was not more effective than a single one. Apparently PS acted as an inductor which caused proliferation of the stem hemopoietic cells and maintained this process for a definite period of time automatically.
