RESUMO

BACKGROUND: Small cell lung cancer (SCLC) is an aggressive tumor with a poor prognosis. Human IFN-λ1 (IL-29), belonging to the type III IFN family, captured increasing attention recently due to its crucial role in developing tumors. Recent studies have revealed that the recombinant Newcastle Disease Virus (NDV) expressing human IFN-λ1 (rL-hIFN-λ1) plays a critical role in the development of tumors. However, the role of rL-hIFN-λ1 in SCLC is still unknown. METHODS: We determined the concentration of the virus intervention, followed by successfully infection in virus. We also investigated the effects of rL-hIFN-λ1 on endoplasmic reticulum stress (ERS), apoptosis and autophagy in H446 cells, and explored the interaction among the three. RESULTS: We found that the ERS, autophagy and apoptosis related proteins were significantly upregulated after infected with rL-hIFN-λ1 or NDV. In addition, both 4-phenylbutyric acid (4-PBA) or 3-Methyladenine (3-MA) could downregulate the expression of related proteins which increased by rL-hIFN-λ1. Furthermore, we found that both B-cell lymphoma-2 (BCL-2) knockdown or Rapamycin (Rapa) could increase ERS, autophagy and apoptosis. CONCLUSIONS: Our findings suggest that rL-hIFN-λ1 can induce ERS, autophagy and apoptosis in SCLC H446 cells, particularly, autophagy plays an important role during this process. Furthermore, rL-hIFN-λ1 might provide a potential biological treatment target for lung cancer treatment.

RESUMO

Evaluation of prosthetic valve function is a challenging task. The clinician has to employ multiple parameters to quantify dysfunction (if present), the results of which can be mutually discrepant. This results from heterogeneity in the design of the valves themselves, implantation techniques, and both intra- and interpatient hemodynamic variability. Specifically, the location and angle of valve implantation can have a profound impact on its flow characteristics that can lead to symptoms despite satisfactory mechanical function. The authors present the case of inverted implantation of a prosthesis designed for the aortic position in the mitral annulus and resultant mitral stenosis. What follows is an examination of how the flow characteristics, such as pressure recovery, energy loss, and vortex formation, create a gradient that could not be explained by valve size alone.

OBJECTIVES: To assess the change in 3-dimensional (3D) echocardiography-derived right ventricular volumes before and after pulmonary thromboendarterectomy (PTE) and to evaluate the correlation of these variables with right heart catheterization-calculated pulmonary vascular resistance (PVR). SETTING: Single university hospitals. PARTICIPANTS: Patients undergoing elective PTE surgery between November 2016 and February 2018. METHODS: All patients received a pulmonary artery catheter and arterial line, and transesophageal echocardiographic monitoring was performed. Transesophageal echocardiographic monitoring before surgery (pre-PTE) and postsurgery (post-PTE) included comprehensive 2D examinations and 3D right ventricular data set acquisition for offline volumetric analysis. Right ventricular fractional area of change (RVFAC) was measured from a right ventricular-focused midesophageal 4-chamber view. TomTec-Arena 4D RV-Function 2.0 offline software (TomTec Imaging Systems GmbH, Unterschlessheim, Germany) was used to measure right ventricular end diastolic volume (RVEDV), right ventricular end systolic volume (RVESV), and right ventricular ejection fraction (RVEF). Paired t tests were used to evaluate for differences before and after surgery, and echocardiographic variables versus PVR were analyzed with linear regression. RESULTS: Forty patients were scheduled for elective PTE surgery; 35 patients had complete hemodynamic profiles and echocardiographic data sets and were included in the evaluation. Mean pulmonary artery pressure decreased from 40 ± 11 to 28 ± 7 mmHg, and PVR decreased from 708 ± 432 to 285 ± 136 dynes*s/cm5 after PTE. RVEDV decreased from 106 ± 43 to 79 ± 35 cm3 (p < 0.001), and RVESV decreased from 77 ± 36 to 59 ± 31 cm3 (p < 0.001). A statistically significant change was not identified in RVEF or RVFAC post-PTE compared with pre-PTE values. All volumetric analyses and RVFAC correlated poorly with PVR (pre-PTE RVEDV correlation to PVR [R2 = 0.004]; post-PTE RVEDV correlation to PVR [R2 = 0.024]). CONCLUSION: Even though RVEDV and RVESV displayed a statistically significant change after PTE, this study did not identify a correlation between those variables and PVR. In addition, markers of right ventricular systolic function (eg, RVFAC and RVEF) did not correlate with PVR. Therefore, the authors conclude that even though these echocardiographic measurements quantified a statistically significant change after PVR reduction, they cannot be reliably used as a surrogate marker of success immediately after PTE.

RESUMO

The microwave-assisted fast co-pyrolysis of chlorella and tire with additive under N2 and CO2 atmospheres were investigated. The pyrolysis profiles, yields of three-phase, the chemical composition of liquid and the ultimate analyses of solid residues were gained. With the tire ratio increasing, all the characters had the changes. The finial temperature had a wave change. The yield of liquid decreased and the chemical composition obtained in liquid of oxygenates compounds decreased, while hydrocarbon compounds increased, among which aromatic hydrocarbons had the highest content. The yield of solid increased, the HHV had a wave change and the values of H/C decreased. Under CO2 atmosphere, the final temperatures were lower with 70% and 100% chlorella ratios, the yield difference of liquid reached the minimal with 70% and 30% chlorella ratios. According to the quantity and quality of liquid and solid, and the former results, 50% percentage of tire was the suitable ratio.

In this study, the influences of ultrasonic pretreatment factors (frequency, power, treatment time) on blends of municipal solid waste (MSW) and paper sludge (PS) with additive (MgO) was explored, through orthogonal experiments design. The optimum operating condition wanted to be acquired. However, for the ultimate (H/C) and ash analysis after pretreatment, solid residue mass and oxygenates compounds contents in products, the influences of factors were in different results. With adding PS unceasingly, the contents of hydrocarbon compounds decreased. And the ultrasonic pretreatment had the obvious influence with high PS percentage. Longer treatment time resulted to the lower content of oxygenates compounds. After adding MgO, the residue mass reduced, which meant MgO had the catalytic action, and the oxygenates compounds content reduced only with 100 kHz, which had the sonochemical effect.

RESUMO

Obtaining high stereoselectivity in glycosylation reactions is often challenging in the absence of neighboring group participation. In this study, we demonstrate that activation of glycosyl trichloroacetimidate donors with immobilized perchloric acid on silica (HClO(4)-SiO(2)) provides higher alpha-selectivity than trimethylsilyl triflate (TMSOTf) for reactions that do not involve neighboring group participation.
