RESUMO

Little is known about the transition mechanisms that govern early lymphoid lineage progenitors from common lymphoid progenitors (CLPs). Pellino2 (PELI2) is a newly discovered E3 ubiquitin ligase, which plays important roles in inflammation and immune system. However, the physiological and molecular roles of PELI2 in the differentiation of immune cells are largely unknown. Here, by using a conditional knockout mouse model, we demonstrated that PELI2 is required for the early B-cell development and stressed hematopoiesis. PELI2 interacted with and stabilized PU.1 via K63- polyubiquitination to regulate IL-7R expression. The defects of B cell development induced by PELI2 deletion were restored by overexpression of PU.1. Similarly, PELI2 promoted TCF3 protein stability via K63- polyubiquitination to regulate IL-7R expression, which is required for the proliferation of B-cell precursor acute lymphoblastic leukemia (BCP-ALL) cells. These results underscore the significance of PELI2 in both normal B lymphopoiesis and malignant B-cell acute lymphoblastic leukemia via the regulation of IL-7R expression, providing a potential therapeutic approach for BCP-ALL.

Chronic myeloid leukemia (CML) is a hematopoietic malignancy driven by the fusion gene BCR: ABL1. Drug resistance to tyrosine kinase inhibitors (TKIs) due to BCR: ABL1 mutation and residual leukemia stem cells (LSCs) remain major challenges for CML treatment. Here, we revealed the requirement of VDR in the progression of CML, in which VDR was upregulated by BCR: ABL1, accounting for its high expression. Interestingly, VDR knockdown inhibited the CML cell proliferation driven by BCR: ABL1 regardless of its mutations with resistance to TKIs. Mechanistically, VDR transcriptionally regulated DDIT4 expression, and the inhibition of DDIT4 triggered DNA damage-induced senescence via p53 signaling activation in CML cells. Furthermore, VDR deficiency was sufficient to not only ameliorate the disease burden and progression in primary CML mice but also reduce the self-renewal of CML-LSCs. Together, our study demonstrated that targeting VDR is a promising strategy to overcome TKI resistance and eradicate leukemia stem cells in CML.

We examined gender and grade differences in the relationship between students' perceived competence, their enjoyment of physical education (PE), and their PA persistence on the frequency of their physical activity (PA). We also used structural equation modeling to assess the direct, indirect, and total effects of perceived competence and PE enjoyment on PA frequency through the mediator of PA persistence. Participants were 223 middle school students (115 boys, 108 girls) in grades 7 and 8. We found that, regardless of grade level, girls had lower perceived competence and PE enjoyment than boys. Both perceived competence and PE enjoyment had significant direct and positive connections to persistence, but they had no significant indirect effects on PA frequency through the mediator of persistence. These findings highlight the need for physical educators to be aware of gender differences in perceived competence and PE enjoyment, and the important roles these factors have in enhancing students' PA participation.

Our aim in the study was to examine age and gender differences in achievement goal orientations and their impact on self-reported persistence toward physical activities of middle school, high school, and college students. A total of 1254 students from four middle schools, two high schools, and one university participated in this study. Multiple regression analyses revealed that age, as a continuous variable, had a significant and positive effect on students' mastery-approach goals, performance-approach goals, mastery-avoidance goals, and their persistence toward physical activities. In contrast, the endorsement of performance-avoidance goals was negatively related to students' age. However, no significant gender differences or age-by-gender interactions were found for the four achievement goals. These findings suggest that future investigators should consider age (vs. grade) when examining maturational differences in young people's achievement goal orientation in physical activity settings.

This study aimed to provide evidence of validity and reliability for the Perceived Mattering Questionnaire-Physical Education (PMQ-PE) developed by Richards et al. (2017) for the Chinese physical education (PE) teachers. The PMQ-PE consisted of two factors (i.e., PE matters and PE teacher matters) with four items in each, measuring four dimensions (attention, importance, dependence, and ego-extension). PMQ-PE in Chinese (PMQ-PEC) was validated among 1,278 elementary and secondary school PE teachers in China, of whom 59.0% were secondary PE teachers and 70.2% were males. Confirmatory factor analysis (CFA) with the entire sample found a poor model fit. Then exploratory factor analysis (EFA) using half of the sample indicated there was only one factor in PMQ-PEC. CFA of the other half of the sample suggested a one-factor model with the elimination of three unfitted items showed a better fitness to the data. Cronbach's alpha value was also acceptable. The final version of PMQ-PEC included five items with one factor that demonstrated acceptable validity and reliability and was deemed acceptable among Chinese PE teachers after the above modifications were made.

In the present study, we examined the longitudinal stability and change in mastery-approach goals for running as offered in regularly scheduled physical education (PE) and athletics classes in the United States. There were five waves of data collection from 806 students (431 boys; 375 girls) who were tracked from fourth to eighth grade while participating in running activities in PE or athletics classes. We assessed the participants' mastery-approach goals using four items on a 5-point scale. We found acceptable longitudinal construct validity, measurement invariance, and scale reliability for the scores of mastery-approach goals, and we found these goals to have moderate stability across this 5-year period. Latent growth modeling revealed a linear decline in mean mastery-approach goal scores over the five school years. We concluded that the assessment of mastery-approach goals over time was both valid and reliable and that these running mastery-approach goals were stable in most respects but became less intense over time.

D, D-carboxypeptidase DacA plays an important role in the synthesis and stabilization of Escherichia coli cell wall peptidoglycan. The production level of extracellular recombinant proteins in E. coli can be enhanced by high D, D-carboxypeptidase activity. Construction of expression systems under optimal promoters is one of the main strategies to realize high protein production in E. coli. In this study, the promoter PdacA-3 from DacA on the genome of E. coli BL21 (DE3) was verified to be efficient for recombinant green fluorescent protein using the plasmid mutant pET28a-PdacA with PdacA-3. Meanwhile, the promoter PdacA-3 was engineered to increase the production level of proteins via inserting one or two Shine-Dalgarno (SD) sequences between the promoter PdacA-3 and the target genes. The expression level of dacA on the genome was increased by the improved transcription of the engineered promoters (especially after inserting one additional SD sequence). The engineered promoters increased cell membrane permeabilities to significantly enhance the secretion production of extracellular recombinant proteins in E. coli. Among them, the extracellular recombinant amylase activities in E. coli BL21::1SD-pET28a-amyK and E. coli BL21::2SD-pET28a-amyK were increased by 2.0- and 1.6-fold that of the control (E. coli BL21-pET28a-amyK), respectively. Promoter engineering also affected the morphology and growth of the E. coli mutants. It was indicated that the engineered promoters enhanced the expression of dacA on the genome to disturb the synthesis and structural stability of cell wall peptidoglycans.

PURPOSE: The study aimed to test the overall changes of health-related fitness (HRF) in minority Chinese college students and to examine HRF differences in gender, race, and year in college. Method: Participants (n = 1320) were minority college students with more than two-thirds females (ie 76.1%), and Hui, Tibetan, and Mongolia consisted of 13.8%, 13.8%, and 11.2%, respectively. Student HRF was tracked for four years. Data were analyzed using multilevel latent growth curve modeling. Results: Muscular strength and endurance were the weakest component in minority college students' HRF, while body mass index was within the category of "excellent". Males outperformed female on all components of HRF. Conclusions: It is suggested that interventions concerning minority females' HRF and muscular strength and endurance for both genders be constructed and tested.

Acute myeloid leukemia (AML) is a highly heterogeneous disease featured by the clonal accumulation of immature myeloid cells. Zinc finger E­box binding homeobox 2 (ZEB2)­antisense RNA 1 (AS1) has been verified to participate in the progression of several types of cancer, including AML. However, the potential mechanisms of ZEB2­AS1 in AML have not yet been fully elucidated. The present study aimed to elucidate the role and regulatory mechanisms of ZEB2­AS1 in AML. The expression of ZEB2­AS1, microRNA­122­5p (miRNA/miR­122­5p) and polo­like kinase 1 (PLK1) was detected by reverse transcription­quantitative polymerase chain reaction (RT­qPCR) in AML tissues or cells. Cell proliferation and apoptosis were examined by methyl thiazolyl tetrazolium (MTT) assay and apoptosis assay, respectively. The protein levels were examined by western blot analysis. The targeted sequence between miR­122­5p and ZEB2­AS1 or PLK1 was predicted using an online database and verified by dual­luciferase reporter assay. A mouse tumor xenograft model was established to confirm the effects of ZEB2­AS1 on tumor growth in vivo. The results revealed that the expression levels of ZEB2­AS1 and PLK1 were upregulated, while those of miR­122­5p were downregulated in AML tissues and cells. The knockdown of ZEB2­AS1 inhibited proliferation and induced apoptosis in vitro, and inhibited tumor growth in vivo. By experimental verification, ZEB2­AS1 was found to negatively regulate miR­122­5p expression and PLK1 was found to be a target gene of miR­122­5p. Furthermore, ZEB2­AS1 was verified to regulate the expression of PLK1 by sponging miR­122­5p in AML cells. On the whole, the findings of the present study demonstrate that ZEB2­AS1 promotes cell proliferation and inhibits apoptosis, at least partly by targeting PLK1 mediated by miR­122­5p in AML cells.

The sewage sludge flocculated with ferrous sulfate (SFS) was prepared by one-step pyrolysis to obtain magnetic Fe-containing carbon. Results showed that only a small amount of FexOy as well as extremely weak magnetism were observed at pyrolysis temperatures of less than 500 °C. SFS tended to exhibit intensive agglomeration, leading to the drastic increase of the crystalline-phase particle size at high pyrolysis temperature. The optimal pyrolysis temperature is 700 °C, corresponding to the production of some sulfides, an optimal content of FexOy, and a suitable BET surface. Hg0 removal efficiency of SFS700 (SFS pyrolyzed at 700 °C) reached 80.7% at the reaction temperature of 125 °C. The presence of O2 and low concentration of SO2 enhanced the Hg0 removal, while the H2O vapor and high SO2 concentration inhibited it. Meanwhile, good resistance for the adsorbent to moderate concentrations of SO2 and H2O was observed. Moreover, the good magnetism performance is conducive to the recovery and utilization of the SFS700 in flue gas. Therefore, SFS can be used for Hg0 removal without any chemical modification after undergoing one-step pyrolysis and this study has guiding significance for the resource utilization and engineering practices.

Iris sibirica biomass (ISBM) used for cobalt (Co) pollution remediation was prepared by one-step pyrolysis and employed to remove elemental mercury (Hg0) from flue gas. Results showed that the ISBM pyrolyzed at 700 °C (ISBM700) exhibited good Hg0 removal performance (about 86%) at 150 °C. The existence of NO and O2 facilitated the removal of Hg0, while SO2 and water vapor inhibited it. Characterization analysis (including N2 adsorption-desorption, X-ray diffraction, scanning electron microscopy, X-ray photoelectron spectroscopy, and Fourier transform infrared spectrometry) showed that ISBM700 has a relatively higher specific surface area, a quantity of lattice oxygen derived from well-dispersed amorphous-phase CoO x , and abundant oxygen functional groups. A Mars-Maessen mechanism is thought to be involved in the Hg0 removal process. The adsorbed Hg0 could be oxidized to HgO by the surface oxygen species derived from CoO x , and then, the consumed surface oxygen species can be replenished by O2. Therefore, the Co-contaminated I. sibirica biomass (CCIB) from phytoremediation could be utilized for Hg0 removal after being pyrolyzed instead of any chemical modification.

This study aimed to systematically review previous studies on the reliability and concurrent validity of the Global Physical Activity Questionnaire (GPAQ). A systematic literature search was conducted (n = 26) using the online EBSCOHost databases, PubMed, Web of Science, and Google Scholar up to September 2019. A previously developed coding sheet was used to collect the data. The Modified Quality Assessment Tool for Observational Cohort and Cross-Sectional Studies was employed to assess risk of bias and study quality. It was found that GPAQ was primarily revalidated in adult populations in Asian and European countries. The sample size ranged from 43 to 2657 with a wide age range (i.e., 15-79 years old). Different populations yielded inconsistent results concerning the reliability and validity of the GPAQ. Short term (i.e., one- to two-week interval) and long-term (i.e., two- to three-month apart) test-retest reliability was good to very good. The concurrent validity using accelerometers, pedometers, and physical activity (PA) log was poor to fair. The GPAQ data and accelerometer/pedometer/PA log data were not compared using the same measurements in some validation studies. Studies with more rigorous research designs are needed before any conclusions concerning the concurrent validity of GPAQ can be reached.

PURPOSE: The relationships among students' self-efficacy, 2 x 2 achievement goals (mastery-approach [MAp], mastery-avoidance [MAv], performance-approach [PAp], and performance-avoidance goals), and achievement performance remain largely unanswered. We tested a model of the mediating role of self-efficacy on the relationship between 2 x 2 achievement goals and cardiorespiratory fitness METHOD: A sample of 276 middle school students (115 boys and 161 girls; 91 sixth graders, 87 seventh graders, and 98 eighth graders), aged 12 to 15 years (Mage = 13.34, SD = 0.96), responded to the Achievement Goals Questionnaire (Conroy, Elliot, & Hofer, 2003) and Self-Efficacy Questionnaire (Gao, Newton, & Carson, 2008) referenced to the fitness test. Their cardiorespiratory fitness was assessed via the Progressive Aerobic Cardiorespiratory Endurance Run (PACER) 3 days later. RESULTS: Structural equation modeling demonstrated an acceptable model fit to the data, Chi2 (2, N = 105) = 1.66. Self-efficacy had a statistically significant direct effect on the PACER after controlling for the effects of the achievement goals (gamma(self-efficacy))-PACER = .21). MAv and PAp also had direct effects on PACER performance (gammaMAv-PACER = -.24, and gammaPAp-PACER = .24, respectively). MAp failed to exert direct effect on the PACER. However, the indirect effect of MAp on the PACER via self-efficacy was small although it was statistically significant. Additionally, the indirect effects of MAv and PAp on PACER through self-efficacy were not significant. CONCLUSIONS: Students' self-efficacy fully mediated the effect of MAp on fitness performance, as well as partially mediated the effects of MAv and PAp on cardiorespiratory fitness performance. Study implications are provided for educators and practitioners.

OBJECTIVE: To investigate the prevalence of anti-hepatitis E virus (HEV) and genotypes of hepatitis E virus in 8 species of animals including swine, cattle, sheep, horse, donkey, dog, chicken and duck in the suburb of Beijing. METHODS: Serum samples were collected from the 8 species of animals, and fecal samples of younger swine were collected from 2 stock farms. Anti-HEV was detected by Double Antigen Sandwich Assay. HEV RNA from fecal samples was detected by a reverse transcription nested polymerase chain reaction (RT-nPCR). Parts of the PCR products were cloned and sequenced. The swine HEV sequences were analyzed genetically. RESULTS: The positive rates of anti-HEV in serum specimens of swine, cattle, horse, donkey, sheep, dog, duck and chicken were 80.43% (481/598), 15.02% (52/346), 14.29% (40/280), 0 (0/26), 9.88% (33/334), 0 (0/21), 3.03% (7/231) and 2.53% (8/316), respectively. The anti-HEV prevalence of adult swine (>/= 6 months) and younger swine ( < / = 3 months) were 87.86% (369/420) and 62.92% (112/178) respectively. 74 of 111 (66.67%) pig faces were positive for HEV RNA. Sequence analysis on these positive samples showed that there were 6 groups of HEV designated as bjsw1, bjsw2, bjsw3, bjsw4, bjsw5 and bjsw6. The 6 strains of HEV shared 94.5% - 99.6% sequence identity of partial HEV ORF2 nucleotide with each other. The identities of HEV ORF2 nucleotide sequences between the 6 strains and genotype 1, 2, 3 and 4 were 75.6% - 78.6%, 75.6% - 76.2%, 77.1% - 80.7% and 83.7% - 94.5%, respectively. The sequence identity between the 6 strains and human HEV genotype 4d was 90.0% - 94.5%. CONCLUSION: HEV infection was seen in swine, cattle, horse, sheep, duck and chicken in the suburbs of Beijing. The anti-HEV positive rate appeared the highest in swine and the lowest in dog and donkey. The six strains of HEV isolated from younger swine belonged to genotype 4d.

The purpose of the study was to examine Hispanic and other minority ninth-grade students' health-related fitness (HRF) knowledge, using FitSmart, a standardized test for high school students. The test consisted of 50 weighted multiple choice items, measuring six subcontent components. Means and standard deviations of the overall scores and the subcontent scores were computed. The findings suggested that students demonstrated mastery of one subcontent component but failed to meet passing values for all other subcontents and overall HRF knowledge. Female students outscored their male counterparts, but there was no difference for ethnicity. More efforts are needed to ensure that Hispanic and other minority high school students master fundamental HRF knowledge.

RESUMO

The authors reviewed studies on college students' physical activity (PA) behaviors and found that previous research on this topic focused on describing college students' PA patterns and their determinants. Researchers reported that about 40% to 50% of college students are physically inactive. More important, health and PA professionals in higher education have not been able to effectively increase students' PA behaviors. Interventions to promote students' PA are still at an early stage and have only produced moderate effects. The authors found primary problems with the current research on this topic to be 3-fold: (1) college students' PA has been seriously neglected as a research topic, (2) there is a lack of multiple-level approaches (ie, personal, psychosocial, and environmental levels) for examining PA behaviors in the college student population, and (3) measures of PA are subjective and inconsistent, which makes comparisons of PA patterns among different samples very difficult or impossible.

The present study examined relationships among variables drawn from achievement goal theory and the expectancy-value model of achievement choice as well as mean level changes of these variables over time in elementary physical education. Participants (N = 207) completed questionnaires over a 2-year period: once while in the second and fourth grades and again when they were in the third and fifth grades. Results indicated that achievement goals, expectancy-related beliefs, and subjective task values were related to one another and were predictive of children's intention for future participation in physical education. Children's subjective task values of physical education decreased over time. Children in Cohort 1 (across second to third grades) generally had stronger motivation for learning in physical education than children in Cohort 2 (across fourth to fifth grades). Findings suggest the importance of integrating achievement goal theory and the expectancy-value model of achievement choice in understanding student motivation.

This study examined children's motivation in elementary physical education within an expectancy-value model developed by Eccles and her colleagues. Four hundred fourteen students in second and fourth grades completed questionnaires assessing their expectancy-related beliefs, subjective task values, and intention for future participation in physical education. Results indicated that expectancy-related beliefs and subjective task values were clearly distinguishable from one another across physical education and throwing. The two constructs were related to each other positively. Children's intention for future participation in physical education was positively associated with their subjective task values and/or expectancy-related beliefs. Younger children had higher motivation for learning in physical education than older children. Gender differences emerged and the findings provided empirical evidence supporting the validity of the expectancy-value model in elementary physical education.
