RESUMO

Neurodegenerative diseases (ND) are characterized, especially, by the progressive loss of neurons, resulting in neuropsychomotor dysfunctions. Even with a high prevalence, NDs are treated with drugs that alleviate the symptoms of patients, but which develop adverse events and still do not inhibit the progression of the disease. Thus, within a new pharmacological perspective, this review aimed to verify the therapeutic potential of natural compounds of marine origin against ND. For this, an integrative review was carried out, according to the PRISMA methodology, which included steps such as: search, pre-selection and inclusion of articles. The results described revealed species such as Acaudina malpodioides, Holothuria scabra and Xylaria sp., which presented important evidence in relation to Alzheimer's, reducing the generation of ROS, presenting neuroprotective effects and reducing the concentration of Aß peptide. Regarding Parkinson's disease (PD), another example of ND, the bioactive compounds from Holothuria scabra and Xylaria sp., showed to be able to reduce the degeneration of dopaminergic neurons, reduce the deposition of alpha synuclein and reduce the formation of Mutant Huntingtin protein (Mhtt). The other marine compounds and bioactive substances are also described in this review. In conclusion, the evaluated studies indicate that compounds of marine origin emerge as a promising source of bioactive compounds, revealing an important therapeutic potential for the treatment of ND.

KEY MESSAGE: Weighted outperformed unweighted genomic prediction using an unbalanced dataset representative of a commercial breeding program. Moreover, the use of the two cycles preceding predictions as training set achieved optimal prediction ability. Predicting the performance of untested single-cross hybrids through genomic prediction (GP) is highly desirable to increase genetic gain. Here, we evaluate the predictive ability (PA) of novel genomic strategies to predict single-cross maize hybrids using an unbalanced historical dataset of a tropical breeding program. Field data comprised 949 single-cross hybrids evaluated from 2006 to 2013, representing eight breeding cycles. Hybrid genotypes were inferred based on their parents' genotypes (inbred lines) using single-nucleotide polymorphism markers obtained via genotyping-by-sequencing. GP analyses were fitted using genomic best linear unbiased prediction via a stage-wise approach, considering two distinct cross-validation schemes. Results highlight the importance of taking into account the uncertainty regarding the adjusted means at each step of a stage-wise analysis, due to the highly unbalanced data structure and the expected heterogeneity of variances across years and locations of a commercial breeding program. Further, an increase in the size of the training set was not always advantageous even in the same breeding program. The use of the two cycles preceding predictions achieved optimal PA of untested single-cross hybrids in a forward prediction scenario, which could be used to replace the first step of field screening. Finally, in addition to the practical and theoretical results applied to maize hybrid breeding programs, the stage-wise analysis performed in this study may be applied to any crop historical unbalanced data.

This paper presents results from an experiment designed to improve the understanding of the relationship between extreme breaking waves and their mechanical loading on heritage offshore lighthouses. The experiment, conducted at La Jument, an iconic French offshore lighthouse, featured several records of wave, current and structure accelerations acquired during severe storm conditions, with individual waves as high as 24 m. Data analysis focuses on a storm event marked by a strong peak in the horizontal accelerations measured inside La Jument. Thanks to stereo-video wave measurements synchronized to the acceleration record we were able to identify and describe the breaking wave responsible for this intense loading. Our observations suggest that this giant wave (19 m high) had a crest elevation high enough to directly hit the lighthouse tower, above the substructure. This paper reveals the potential for conducting ambitious field experiments from offshore lighthouses in order to collect valuable storm waves and wave loading observations. This offers a possible second service life for these heritage structures as in situ laboratories dedicated to the study of the coastal hydrodynamics and its interaction with marine structures. Background This study was performed to assess adhesion molecules in systemic lupus erythematosus (SLE). Methods This case-control study examined 126 SLE patients and 48 healthy individuals. Blood levels of six adhesion molecules, cortisol, nuclear autoantibody (ANA) and anti-double stranded DNA (anti-dsDNA) titers were measured, while disease activity was assessed using the SLE Disease Activity Index (SLEDAI) score. Results Platelet endothelial cell adhesion molecule 1 (PECAM-1), vascular cell adhesion molecule 1 (VCAM-1), E-selectin, P-selectin, and plasminogen activator inhibitor type-1 (PAI-1) were significantly higher in SLE patients than in controls. Binary logistic regression analysis showed that PECAM-1 and PAI-1 predicted SLE with a sensitivity of 86.5% and a specificity of 81.3%. ANA titers were significantly and positively associated with PECAM-1, VCAM-1, E-selectin, and PAI-1, whereas there were no associations between anti-dsDNA titers and adhesion molecules. Cortisol was negatively associated with PCAM-1 and ICAM-1. There were significant associations between metabolic syndrome (MetS) and E-selectin and PAI-1. 14.8% of the variance in the SLEDAI score was explained by the regression on PECAM-1 and MetS. Conclusions Our data show that adhesion molecules, especially PECAM-1, are significantly associated with SLE and disease activity, suggesting that they play a role in SLE pathophysiology. While MetS, ANA titers and cortisol levels modulate adhesion molecule levels, these associations do not explain the increased levels of adhesion molecules in SLE. Increased levels of adhesion molecules are new drug targets in SLE.

Background: This study tested the hypothesis that the birthweight paradox would not be observed when assessing the effect of maternal education on neonatal mortality in the presence of socioeconomic inequality in access to health care. Methods: Non-concurrent cohort study. Passive follow-up of live-born infants using probabilistic record linkage of birth and death records for Rio de Janeiro (2004-2010; n = 1 445 367). Maternal age, birthweight and neonatal death were evaluated according to maternal educational level strata (<4, 4-11 and ≥12 years of study). We estimated the association between maternal educational level and neonatal mortality using logistical regression models adjusted for maternal age and birthweight (<2500 g and ≥2500 g). Results: Neonatal mortality was 1.8 times higher in low educational level group compared with high educational level. We did not find birthweight-specific mortality curves crossing over in the stratum under 2500 g (birthweight paradox). The odds of a low birthweight child being born in facilities without neonatal intensive care units was about 70% higher in the group of low education when compared with mothers with high education. Conclusions: The absence of crossing birthweight-specific mortality curves may be a reason for concern about the severity of the disadvantages faced by low maternal education women.

The aim of this study was to evaluate changes in intraocular pressure (IOP), pupil size (PS), blood pressure (BP), heart rate (HR), and ECG variables (Pms wave PmV, PR interval, QRS complex, RMV wave and QT intervals) over time during the instillation of 0.5% timolol, 0.5% levobunolol and 0.5% apraclonidine in clinically normal dogs. Ten adult beagles were used. Baseline values were measured at 8a.m., 2p.m. and 8p.m., for three consecutive days. A waiting period of 10 days between the administrations of each drug was established. For 15 consecutive days, the drug being tested was instilled in one eye of each dog twice a day (7a.m. and 7p.m.). The parameters were evaluated at the aforementioned times on days 3, 6, 9, 12 and 15. Data were statistically compared using the Bonferroni test and one-way repeated measures analysis of variance (P<0.05). The Pearson test was used to evaluate any correlation between QT interval, HR and BP. The tested drugs did not find a decrease in IOP. A significant decreased in PS was observed in almost all dogs following levobunolol administration, relative to the control eye. A significant decrease in HR was observed on day 3 following levobunolol treatment, while apraclonidine induced an increase on day 15. Blood pressure was reduced in all measurement time points following apraclonidine treatment. A negative correlation between QT interval and HR was only observed in dogs treated with timolol. In conclusion, levobunolol was the only drug that induced significant alterations in PS. Apraclonidine was the only drug that induced systemic hypotension. Timolol was the only drug to that induced a negative correlation between QT and HR.(AU)


O objetivo deste estudo foi avaliar as mudanças na pressão intraocular (PIO), no diâmetro pupilar (DP), na pressão sanguínea (PS), na frequência cardíaca (FC) e nas variáveis eletrocardiográficas (onda Pms, PmV, intervalo PR, complexo QRS, onda RmV e intervalo QT), ao longo do tempo da instilação do timolol 0,5%, do levobunolol 0,5% e da apraclonidina 0,5% em cães clinicamente normais. Dez Beagles adultos compuseram o estudo. Valores basais foram mensurados às oito,, 14 e 20 horas, durante três dias consecutivos. Foi instituído um período de espera de 10 dias entre a administração de cada fármaco. Durante 15 dias consecutivos, um olho de cada animal recebeu uma gota de cada um deles, a intervalos de 12 horas (às sete e às 19 horas). Os parâmetros foram avaliados nos momentos acima referidos, nos dias três, seis, nove, 12 e 15. Os dados foram comparados estatisticamente empregando-se o teste de Bonferroni após análise de variância para medidas repetidas (P<0,05). Teste de Pearson foi utilizado para correlação entre o intervalo QT com a FC e a PS. Não se encontrou diminuição da PIO. Observou-se redução significativa do DP na quase totalidade dos animais que receberam levobunol, relativamente ao olho controle. Diminuição significativa da FC foi vista ao terceiro dia após a administração do levobunolol, enquanto apraclonidina induziu aumento no 15º dia. A pressão arterial foi reduzida em todos os momentos com a apraclonidina. Observou-se correlação negativa entre o intervalo QT e a FC apenas nos indivíduos tratados com o timolol. Em conclusão, levobunolol foi o único fármaco que induziu alterações significativas no DP. A apraclonidina foi o único fármaco que induziu hipotensão sistêmica significativa. O timolol foi o único a ensejar correlação negativa entre o intervalo QT e a FC.(AU)

Foram avaliadas as taxas de reconcepção de 98 novilhas primíparas (34 Guzerá - G, 32, 1/2 Guzerá x 1/2 Nelore - GN e 32,1/2 Red Angus x 1/2 Nelore - AN) com 14 meses de idade e peso médio de aproximadamente 249,65kg, criadas em pastos de Brachiaria brizantha cv Marandu. Na segunda estação de monta (EM), foram utilizadas apenas as 36 primíparas gestantes na primeira EM (três G, nove GN e 24 AN), com média de idade de 26 meses e peso corporal de 313,67±25,01kg, 336,50±45,84kg e 399,86±44,45kg, respectivamente, para as fêmeas dos grupos G, GN e AN. O grupo AN apresentou ganho médio diário (GMD) de 0,30±0,06 (kg/dia) e maiores taxas (58,33) de reconcepção (P<0,05), comprovando que a heterose resultante do cruzamento entre raças distintas com maior distância genética (Bos taurus taurus x Bos taurus indicus) proporciona maior desempenho produtivo e reprodutivo.


A total of 98 heifers of three genetic groups: 34 Guzerá (G), 32, 1/2 Guzerá x 1/2 Nelore (GN) and 32 1/2 Red Angus x 1/2 Nelore (AN), 14 month old and average body weight of 249,65kg, raised in Brachiaria brizantha cv Marandu were used to evaluate reconception rates. In the second breeding season (BS), only those 36 pregnant in the first BS (three G, nine GN and 24 AN), age 26 months and body weight of 313.67±25.01kg, 336.50±45.84kg and 399.86±44.45kg, respectively, G , GN and AN were evaluated. The highest reconception rate (58.33%, P<0.05) was registered for the AN group and showed that average daily gain of 0.30±0.06kg, showing that crosses between Bos taurus taurus x Bos taurus indicus leads to higher reproductive and productive performances.

We study the effects of next-nearest-neighbor (NNN) interactions on the dynamics of the one-dimensional spin-1/2 transverse Ising model in the high-temperature limit. We use exact diagonalization to obtain the time-dependent transverse correlation function and the corresponding spectral density for a tagged spin. Our results for chains of 13 spins with periodic boundary conditions produce results which are valid in the infinite-size limit. In general we find that the NNN coupling produces slower dynamics accompanied by an enhancement of the central mode behavior. Even in the case of a strong transverse field, if the NNN coupling is sufficiently large, then there is a crossover from collective mode to central mode behavior. We also obtain several recurrants for the continued fraction representation of the relaxation function.

We investigate the effects of the excitation power on the photoluminescence spectra of aqueous CdTe/CdS core-shell quantum dots. We have focused our efforts on nanoparticles that are drop-cast on a silicon nitride substrate and dried out. Under such conditions, the emission intensity of these nanocrystals decreases exponentially and the emission center wavelength shifts with the time under laser excitation, displaying a behavior that depends on the excitation power. In the low-power regime a blueshift occurs, which we attribute to photo-oxidation of the quantum dot core. The blueshift can be suppressed by performing the measurements in a nitrogen atmosphere. Under high-power excitation the nanoparticles thermally expand and aggregate, and a transition to a redshift regime is then observed in the photoluminescence spectra. No spectral changes are observed for nanocrystals dispersed in the solvent. Our results show a procedure that can be used to determine the optimal conditions for the use of a given set of colloidal quantum dots as light emitters for photonic crystal optical cavities.

OBJECTIVES: Dolutegravir is a second-generation integrase strand transfer inhibitor (InSTI) that has been recently approved by the FDA to treat antiretroviral therapy-naive as well as treatment-experienced HIV-infected individuals, including those already exposed to the first-generation InSTI. Despite having a different mutational profile, some cross-resistance mutations may influence its susceptibility. The aim of this study was to evaluate the impact of a raltegravir-containing salvage regimen on dolutegravir activity. PATIENTS AND METHODS: Blood samples of 92 HIV-infected individuals with virological failure (two or more viral loads >50 copies/mL after 6 months of treatment) using raltegravir with optimized background therapy were sequenced and evaluated according to the Stanford University HIV Drug Resistance Database algorithm. RESULTS: Among the 92 patients analysed, 32 (35%) showed resistance to dolutegravir, in most cases associated with the combination of Q148H/R/K with G140S/A mutations. At genotyping, patients with resistance to dolutegravir had viral load values closer to the highest previously documented viral load. CONCLUSIONS: Changes in viraemia during virological failure may indicate the evolution of raltegravir resistance and may predict the emergence of secondary mutations that are associated with a decrease in dolutegravir susceptibility. Early discontinuation of raltegravir from failing regimens might favour subsequent salvage with dolutegravir, but further studies are necessary to evaluate this issue.

Water quality alterations due to forest fires may considerably affect aquatic organisms and water resources. These impacts are cumulative as a result of pollutants mobilized from fires, chemicals used to fight fire, and postfire responses. Few studies have examined postfire transport into water resources of trace elements, including the polycyclic aromatic hydrocarbons (PAH), which are organic pollutants produced during combustion and are considered carcinogenic and harmful to humans. PAH are also known to adversely affect survival, growth, and reproduction of many aquatic species. This study assessed the effects of forest wildfires on groundwater from two mountain regions located in protected areas from north and central Portugal. Two campaigns to collect water samples were performed in order to measure PAH levels. Fifteen of 16 studied PAH were found in groundwater samples collected at burned areas, most of them at concentrations significantly higher than those found in control regions, indicating aquifer contamination. The total sum of PAH in burned areas ranged from 23.1to 95.1 ng/L with a median of 62.9 ng/L, which is one- to sixfold higher than the average level measured in controls (16.2 ng/L). In addition, in control samples, the levels of light PAH with two to four rings were at higher levels than heavy PAH with five or six rings, thus showing a different profile between control and burned sites. The contribution of wildfires to groundwater contamination by PAH was demonstrated, enabling a reliable assessment of the impacts on water quality and preparation of scientifically based decision criteria for postfire forest management practices.

Spatial structure in landscapes impacts population stability. Two linked components of stability have large consequences for persistence: first, statistical stability as the lack of temporal fluctuations; second, synchronisation as an aspect of dynamic stability, which erodes metapopulation rescue effects. Here, we determine the influence of river network structure on the stability of riverine metapopulations. We introduce an approach that converts river networks to metapopulation networks, and analytically show how fluctuation magnitude is influenced by interaction structure. We show that river metapopulation complexity (in terms of branching prevalence) has nonlinear dampening effects on population fluctuations, and can also buffer against synchronisation. We conclude by showing that river transects generally increase synchronisation, while the spatial scale of interaction has nonlinear effects on synchronised dynamics. Our results indicate that this dual stability - conferred by fluctuation and synchronisation dampening - emerges from interaction structure in rivers, and this may strongly influence the persistence of river metapopulations.

We study the reflectivity spectra of photonic crystal slab cavities using an extension of the scattering matrix method that allows treating finite sizes of the spot of the excitation beam. The details of the implementation of the method are presented and then we show that Fano resonances arise as a consequence of the electromagnetic interference between the discrete contribution of the fundamental cavity mode and the continuum contribution of the light scattered by the photonic crystal pattern. We control the asymmetry lineshape of the Fano resonance through the polarization of the incident field, which determines the relative phase between the two electromagnetic contributions to the interference. We analyse the electric field profile inside and outside of the crystal to help in the understanding of the dependence on polarization of the reflectivity lineshape. We also study with our implementation the dependence of the Fano resonances on the size of the incident radiation spot.

RESUMO

We find that a common mutation that increases angiotensin I-converting enzyme activity occurs with higher frequency in male patients suffering from refractory temporal lobe epilepsy. However, in their brains, the activity of the enzyme is downregulated. As an explanation, we surprisingly find that carbamazepine, commonly used to treat epilepsy, is an inhibitor of the enzyme, thus providing a direct link between epilepsy and the renin-angiotensin and kallikrein-kinin systems.

Estudou-se o quadro clínico do veneno decorrente do envenenamento por Tityus fasciolatus e sua ação refletida nos componentes hematológicos no modelo murino. Para o estudo do perfil hematológico, foram utilizados 54 camundongos Swiss CF1, machos, com 30g, distribuídos em três grupos (G) (n=18) inoculados via subcutânea com: 50µL de PBS (G1); 24µg de veneno de T. fasciolatus (G2) e 8µg de veneno de T. serrulatus (G3). Cada grupo foi subdividido em três subgrupos (n=6) de acordo com o momento da coleta de sangue que foi estipulada em uma, oito e 24h após a inoculação do veneno, e foram realizados o hemograma e a dosagem de proteínas totais e fracionadas. O veneno de T. fasciolatus na dose de 24µg causou piloereção, comportamento nociceptivo, secreção nasal e oral acentuada, dispneia, prurido na face e reflexos exacerbados. No exame hematológico, foram observadas policitemia relativa e leucocitose com linfocitose.


The aim of this study was to evaluate the clinical picture caused by Tityus fasciolatus and its action reflected in mouse blood profile. To study the blood profile, 54 Swiss CF1 mice, male, 30g, distributed into three groups (G) (n= 18) and inoculated subcutaneously with: 50µl PBS (G1), 24µg of venom of T. fasciolatus (G2) and 8µg venom T. serrulatus (G3) were used. Each group was subdivided into three subgroups (n= 6) according to the time of blood collection that was set at 1h, 8h and 24h after inoculation of the venom, and blood profile, total protein and fractionated were evaluated. The venom of T. fasciolatus at a dose of 24µg caused piloerection, behavior pain, nasal and oral sharp, dyspnea, rash on the face and exaggerated reflexes. In blood profile were observed relative polycythemia, leukocytosis with lymphocytosis.

RESUMO

Radiography (conventional and contrasted - canalography) and audioelectrophysiologic (impedance audiometry) tests were correlated with otitis media in 12 middle ear affected and 12 healthy dogs. When comparing both diagnostic methods, it was found that the reliability in the detection of otitis media in dogs was 100 percent for the first and 58.3-60.0 percent for the latter. Thus, it was possible to attest that the impedance audiometry was the most accurate method in the identification of otitis media in dogs when compared with radiographic assessments.

Foram avaliadas as técnicas radiográficas dentárias intra (TIB) e extrabucal (TEB) em 50 cães com doença periodontal, no intuito de padronizar os procedimentos de diagnóstico dessa síndrome. A TIB revelou que 16 animais não apresentaram lesões ósseas visíveis, enquanto a TEB apontou que 39 pacientes foram negativos para as mesmas lesões. Em resumo, a TIB foi mais eficaz na detecção de sinais radiográficos, especialmente as chamadas lesões finas, que a TEB, sendo a técnica de escolha na síndrome periodontal.


The intra (IOT) and extraoral (EOT) dental radiographic techniques in 50 dogs with periodontal disease were compared to standardize the most appropriate procedure for accurate diagnosis of abnormalities that occur in this syndrome. For IOT, 16 animals showed no visible radiographic lesions, whereas in EOT, 39 patients were negative for the same radiographic lesions. For incisor and canine teeth, IOT presented the highest sensitivity of lesion detection, while there was no significant difference for the premolar and molar teeth group. It is concluded that IOT is more effective in detecting radiographic signs (especially the so-called thin lesions) than the EOT and should therefore be the technique of choice in the periodontal syndrome.

1. Much of the current understanding of ecological systems is based on theory that does not explicitly take into account individual variation within natural populations. However, individuals may show substantial variation in resource use. This variation in turn may be translated into topological properties of networks that depict interactions among individuals and the food resources they consume (individual-resource networks). 2. Different models derived from optimal diet theory (ODT) predict highly distinct patterns of trophic interactions at the individual level that should translate into distinct network topologies. As a consequence, individual-resource networks can be useful tools in revealing the incidence of different patterns of resource use by individuals and suggesting their mechanistic basis. 3. In the present study, using data from several dietary studies, we assembled individual-resource networks of 10 vertebrate species, previously reported to show interindividual diet variation, and used a network-based approach to investigate their structure. 4. We found significant nestedness, but no modularity, in all empirical networks, indicating that (i) these populations are composed of both opportunistic and selective individuals and (ii) the diets of the latter are ordered as predictable subsets of the diets of the more opportunistic individuals. 5. Nested patterns are a common feature of species networks, and our results extend its generality to trophic interactions at the individual level. This pattern is consistent with a recently proposed ODT model, in which individuals show similar rank preferences but differ in their acceptance rate for alternative resources. Our findings therefore suggest a common mechanism underlying interindividual variation in resource use in disparate taxa.

Avaliou-se o hemograma de 12 cães adultos, saudáveis (14,2±5,4kg) após a inoculação de veneno do escorpião amarelo (Tityus serrulatus). Os animais foram distribuídos em dois grupos (G), com seis em cada: os do GI foram usados como controle e receberam 0,5mL de salina tamponada com fosfato (PBS) por via subcutânea (SC) na face medial da coxa esquerda (FMCE), e os do GII receberam veneno liofilizado do T. serrulatus (250µg/kg) diluído em PBS por via SC na FMCE. Foram realizadas colheitas de sangue com anticoagulante EDTA a 10 por cento antes da inoculação do veneno (T0) e após 2h, (T1), 6h (T2), 12h (T3), 24h (T4), 48h (T5) e 72h (T6), para contagem de eritrócitos, leucócitos e plaquetas em aparelho contador eletrônico e esfregaços sanguíneos para contagem diferencial de leucócitos. Houve aumento significativo (P<0,05) dos valores de eritrócitos, volume globular e hemoglobina 2h e 6h após o envenenamento, devido à contração esplênica decorrente da dor local causada pelo veneno e pela liberação de catecolaminas. Foi observada leucocitose por aumento significativo (P<0,05) de neutrófilos e linfócitos 2h e 6h após o envenenamento. Concluiu-se que o veneno de Tityus serrulatus na dose de 250µg/kg, é capaz de aumentar os valores do eritrograma e do leucograma dos cães, provavelmente devido à dor local, com liberação de catecolaminas.


The canine blood profile after scorpion envenomation was evaluated using 12 healthy mongrel male dogs (14.2±5.4kg) distributed in two groups, with six animals in each: group I (control group) and group II (venom group). The lyophilized yellow scorpion (Tityus serrulatus) venom (250µg/kg) diluted in 0.5mL phosphate buffered saline (PBS) was given to group II animals by subcutaneous injection, in the medial face of the left thigh. Group I animals received only 0.5mL of PBS, by subcutaneous injection, in the medial face of the left thigh. Blood samples were collected with EDTA before (T0) and 2 (T1), 6 (T2), 12 (T3), 24 (T4), 48 (T5), and 72h (T6) after envenomation. Significant increases (P<0.05) in erythrocytes counting, hematocrit and hemoglobin concentration, 2 and 6h after envenomation were observed. Leukocytosis with significant increases (P<0.05) of neutrophils and lymphocytes 2 and 6h after envenomation was found. Then, T. serrulatus venom may induce alterations in blood profile in dogs, probably due to spleen contraction evoked by pain and catecholamines releasing.
