RESUMO

Phthorimaea absoluta (Meyrick) is an invasive pest that has caused damage to tomatoes and other crops in China since 2017. Pest control is mainly based on chemical methods that pose significant threats to food safety and environmental and ecological security. Light-induced control, a green prevention and control technology, has gained attention recently. However, current light-trapping technology is non-specific, attracting targeted pests alongside natural enemies and non-target organisms. In this study, we characterized the phototactic behavior of tomato leaf miners for the development a specific light-trapping technology for pest control. In situ hybridization revealed opsin expression throughout the body. Furthermore, we investigated the tropism of pests (wild T. absoluta, Toxoptera graminum, and Bemisia tabaci) and natural enemies (Nesidiocoris tenuis and Trichogramma pintoi) using a wavelength-lamp tropism experiment. We found that 365 ± 5 nm light could accurately trap wild P. absoluta without trapping natural enemies and other insects. Finally, we analyzed the phototactic behavior of the mutant strains LW2(-/-) and BL(-/-). LW2 and BL mutants showed significant differences in phototactic behavior. The LW2(-/-) strain was attracted to light at 390 ± 5 nm and the BL(-/-) strain was unresponsive to any light. Our findings will help to develop specific light-trapping technology for controlling tomato leaf miners, providing a basis for understanding pest population dynamics and protecting crops against natural enemies.

Spodoptera exempta, known as the black armyworm, has been extensively documented as an invasive agricultural pest prevalent across various crop planting regions globally. However, the potential geographical distribution and the threat it poses to host crops of remains unknown at present. Therefore, we used an optimized MaxEnt model based on 841 occurrence records and 19 bioclimatic variables to predict the potential suitable areas of S. exempta under current and future climatic conditions, and the overlap with wheat, rice, and maize planting areas was assessed. The optimized model was highly reliable in predicting potential suitable areas for this pest. The results showed that high-risk distribution areas for S. exempta were mainly in developing countries, including Latin America, central South America, central Africa, and southern Asia. Moreover, for the three major global food crops, S. exempta posed the greatest risk to maize planting areas (510.78 × 104 km2), followed by rice and wheat planting areas. Under future climate scenarios, global warming will limit the distribution of S. exempta. Overall, S. exempta had the strongest effect on global maize production areas and the least on global wheat planting areas. Our study offers a scientific basis for global prevention of S. exempta and protection of agricultural crops.

Objective: Pancreatic carcinoma is characterised by high aggressiveness and a bleak prognosis; optimising related treatment decisions depends on the availability of reliable prognostic markers. This study was designed to compare various blood biomarkers, such as neutrophil/lymphocyte ratio (NLR), lymphocyte/monocyte ratio (LMR), platelet/lymphocyte ratio (PLR), C-reactive protein (CRP), albumin (Alb), plasma fibrinogen (PF), and CRP/Alb in patients with pancreatic carcinoma. Methods: Our study retrospectively reviewed 250 patients with pancreatic carcinoma diagnosed between July 2007 and December 2018. The Cutoff Finder application was used to calculate the optimal values of CRP/Alb and PF. The Chi-square test or Fisher's exact test was used to analyse the correlation of CRP/Alb and PF with other clinicopathological factors. Conducting univariate and multivariate analyses allowed further survival analysis of these prognostic factors. Results: Multivariate analysis revealed that, in a cohort of 232 patients with pancreatic ductal adenocarcinoma (PDAC), the PF level exhibited statistical significance for overall survival (hazard ratio (HR) = 0.464; p = 0.023); however, this correlation was not found in the entire group of 250 patients with pancreatic carcinoma. Contrastingly, the CRP/Alb ratio was demonstrated statistical significance in both the entire pancreatic carcinoma cohort (HR = 0.471; p = 0.026) and the PDAC subgroup (HR = 0.484; p = 0.034). CRP/Alb and PF demonstrated a positive association (r=0.489, p<0.001) as indicated by Spearman's rank correlation analysis. Additionally, in 232 PDAC patients, the combination of the CRP/Alb ratio and PF had synergistic effects on prognosis when compared with either the CRP/Alb ratio or the PF concentration alone. Conclusion: PF concentration is a convenient, rapid, and noninvasive biomarker, and its combination with the CRP/Alb ratio could significantly enhance the accuracy of prognosis prediction in pancreatic carcinoma patients, especially those with the most common histological subtype of PDAC.

The Mediterranean fruit fly, Ceratitis capitata (Wiedemann), which is native to tropical Africa, has invaded more than 100 countries and constitutes a risk to the citrus sector. Studying its potential geographical distribution (PGD) in the context of global climate change is important for prevention and control efforts worldwide. Therefore, we used the CLIMEX model to project and assess the risk of global invasion by C. capitata under current (1981-2010) and future (2040-2059) climates. In the prevailing climatic conditions, the area of PGD for C. capitata was approximately 664.8 × 105 km2 and was concentrated in South America, southern Africa, southern North America, eastern Asia, and southern Europe. Under future climate conditions, the area of PGD for C. capitata is projected to decrease to approximately 544.1 × 105 km2 and shift to higher latitudes. Cold stress was shown to affect distribution at high latitudes, and heat stress was the main factor affecting distribution under current and future climates. According to the predicted results, countries with highly suitable habitats for C. capitata that have not yet been invaded, such as China, Myanmar, and Vietnam, must strengthen quarantine measures to prevent the introduction of this pest.

In insects, vision is crucial in finding host plants, but its role in nocturnal insects is largely unknown. Vision involves responses to specific spectra of photon wavelengths and opsins plays an important role in this process. Long-wavelength sensitive opsin (LW opsin) and blue-sensitive opsin (BL opsin) are main visual opsin proteins and play important in behavior regulation.We used CRISPR/Cas9 technology to mutate the long-wavelength-sensitive and blue wavelength-sensitive genes and explored the role of vision in the nocturnal invasive pest Tuta absoluta. Light wave experiments revealed that LW2(-/-) and BL(-/-) mutants showed abnormal wavelength tropism. Both LW2 and BL mutations affected the preference of T. absoluta for the green environment. Mutations in LW2 and BL are necessary to inhibit visual attraction. The elimination of LW2 and BL affected the preference of leaf moths for green plants, and mutations in both induced a preference in moths for white plants. Behavioral changes resulting from LW2(-/-) and BL(-/-) mutants were not affected by sense of smell, further supporting the regulatory role of vision in insect behavior. To the best of our knowledge, this is the first study to reveal that vision, not smell, plays an important role in the host-seeking behavior of nocturnal insects at night, of which LW2 and BL opsins are key regulatory factors. These study findings will drive the development of the "vision-ecology" theory.

Invasive alien pests (IAPs) pose a major threat to global agriculture and food production. When multiple IAPs coexist in the same habitat and use the same resources, the economic loss to local agricultural production increases. Many species of the Diabrotica genus, such as Diabrotica barberi, Diabrotica undecimpunctata, and Diabrotica virgifera, originating from the USA and Mexico, seriously damaged maize production in North America and Europe. However, the potential geographic distributions (PGDs) and degree of ecological niche overlap among the three Diabrotica beetles remain unclear; thus, the potential coexistence zone is unknown. Based on environmental and species occurrence data, we used an ensemble model (EM) to predict the PGDs and overlapping PGD of the three Diabrotica beetles. The n-dimensional hypervolumes concept was used to explore the degree of niche overlap among the three species. The EM showed better reliability than the individual models. According to the EM results, the PGDs and overlapping PGD of the three Diabrotica beetles were mainly distributed in North America, Europe, and Asia. Under the current scenario, D. virgifera has the largest PGD ranges (1615 × 104 km2). In the future, the PGD of this species will expand further and reach a maximum under the SSP5-8.5 scenario in the 2050s (2499 × 104 km2). Diabrotica virgifera showed the highest potential for invasion under the current and future global warming scenarios. Among the three studied species, the degree of ecological niche overlap was the highest for D. undecimpunctata and D. virgifera, with the highest similarity in the PGD patterns and maximum coexistence range. Under global warming, the PGDs of the three Diabrotica beetles are expected to expand to high latitudes. Identifying the PGDs of the three Diabrotica beetles provides an important reference for quarantine authorities in countries at risk of invasion worldwide to develop specific preventive measures against pests.

Microorganisms can affect host reproduction, defense, and immunity through sexual or opportunistic transmission; however, there are few studies on insect reproductive organs and intestinal bacterial communities and their effects on mating. Tuta absoluta is a worldwide quarantine pest that seriously threatens the production of Solanaceae crops, and the microbial community within tomato leafminers remains unclear. In this study, 16s rRNA sequencing was used to analyze bacterial communities related to the reproductive organs and intestinal tracts of tomato leafminers (the sample accession numbers are from CNS0856533 to CNS0856577). Different bacterial communities were found in the reproductive organs and intestinal tracts of females and males. Community ecological analysis revealed three potential signs of bacterial sexual transmission: (1) Mating increased the similarity between male and female sex organs and intestinal communities. (2) The bacteria carried by mated individuals were found in unmated individuals of the opposite sex but not in unmated individuals of the same sex. (3) The bacteria carried by unmated individuals were lost after mating. In addition, the abundances of bacterial communities carried by eggs were significantly higher than those of adult worms. Our results confirm that mating leads to the transfer of bacterial communities in the reproductive organs and gut of tomato leafminers, and suggest that this community strongly influences the reproductive process.

Pine wilt disease (PWD), caused by the pine wood nematode (PWN, Bursaphelenchus xylophilus), a destructive, invasive forest pathogen, poses a serious threat to global pine forest ecosystems. The global invasion of PWN has been described based on three successive phases, introduction, establishment, and dispersal. Risk assessments of the three successive PWN invasion phases can assist in targeted management efforts. Here, we present a risk assessment framework to evaluate the introduction, establishment, and dispersal risks of PWD in China using network analysis, species distribution models, and niche concepts. We found that >88 % of PWN inspection records were from the United States, South Korea, Japan, Germany, and Mexico, and 94 % of interception records were primarily from the Jiangsu, Shanghai, Shandong, Tianjin, and Zhejiang ports. Based on the nearly current climate, the areas of PWN overlap with its host Pinus species were primarily distributed in southern, eastern, Yangtze River Basin, central, and northeastern China regions. Areas of PWN overlap with its insect vector Monochamus alternatus were primarily distributed in southern, eastern, Yangtze River Basin, central, and northeastern China regions, and those of PWN overlap with the insect vector Monochamus saltuarius were primarily distributed in eastern and northeastern China. The niche between PWN and the insect vector M. alternatus was the most similar (0.68), followed by that between PWN and the insect vector M. saltuarius (0.47). Climate change will increase the suitable probabilities of PWN and its two insect vectors occurring at high latitudes, further increasing their threat to hosts in northeastern China. This risk assessment framework for PWD could be influential in preventing the entry of the PWN and mitigating their establishment and dispersal risks in China. Our study provides substantial clues for developing a framework to improve the risk assessment and surveillance of biological invasions worldwide.

BACKGROUND: We invented Endoscopic Ruler, a new endoscopic device to measure the size of varices in patients with cirrhosis and portal hypertension. AIM: To assess the feasibility and safety of Endoscopic Ruler, and evaluate the agreement on identifying large oesophageal varices (OV) between Endoscopic Ruler and the endoscopists, as well as the interobserver agreement on diagnosing large OV using Endoscopic Ruler. METHODS: We prospectively and consecutively enrolled patients with cirrhosis from 11 hospitals, all of whom got esophagogastroduodenoscopy (EGD) with Endoscopic Ruler. The primary study outcome was a successful measurement of the size of varices using Endoscopic Ruler. The secondary outcomes included adverse events, operation time, the agreement of identifying large OV between the objective measurement of Endoscopic Ruler and the empirical reading of endoscopists, together with the interobserver agreement on diagnosing large OV by Endoscopic Ruler. RESULTS: From November 2020 to April 2022, a total of 120 eligible patients with cirrhosis were recruited and all of them underwent EGD examinations with Endoscopic Ruler successfully without any adverse event. The median operation time of Endoscopic Ruler was 3.00 min [interquartile range (IQR): 3.00 min]. The kappa value between Endoscopic Ruler and the endoscopists while detecting large OV was 0.52, demonstrating a moderate agreement. The kappa value for diagnosing large OV using Endoscopic Ruler among the six independent observers was 0.77, demonstrating a substantial agreement. CONCLUSION: The data demonstrates that Endoscopic Ruler is feasible and safe for measuring the size of varices in patients with cirrhosis and portal hypertension. Endoscopic Ruler is potential to promote the clinical practice of the two-grade classification system of OV.

The southern armyworm (Spodoptera eridania), a polyphagous crop pest native to tropical America, has been found in Africa (2016) and India (2019), causing defoliation and damage to the reproductive structures of cassava, soybean, and tomato. The damage caused by this pest to crop systems has raised concerns regarding its potential risks. Therefore, we predicted the potential geographical distribution of S. eridania under climate change conditions using 19 bioclimatic variables based on an optimized MaxEnt model. The results showed that annual precipitation (bio12), mean temperature of the warmest quarter (bio10), and precipitation of the driest month (bio14) were important bioclimatic variables influencing the potential distribution. The prediction showed that the suitable habitat area was approximately 3426.43 × 104 km2, mainly concentrated in southern North America, South America, western Europe, central Africa, southern Asia, and eastern Oceania. In response to global climate change, suitable habitats for S. eridania will expand and shift to higher latitudes in the future, especially under the SSP5-8.5 scenario. Because of the current devastating effects on crop production, countries without S. eridania invasion, such as the European Union, Southeast Asian countries, and Australia, need to strengthen phytosanitary measures at border ports to prevent the introduction of this pest.

Invasive crop pests (ICPs) are a major cause of crop losses and adversely affect global food security. Diuraphis noxia Kurdjumov is a significant ICP that feeds on the sap of crops, reducing crop yield and quality. Although estimating the geographical distribution patterns of D. noxia under climate change is critical for its management and global food security, such information remains unclear. Based on 533 global occurrence records and 9 bioclimatic variables, an optimized MaxEnt model was used to predict the potential global geographical distribution of D. noxia. The results showed that Bio1, Bio2, Bio7, and Bio12 were significant bioclimatic variables that influenced the potential geographical distribution of D. noxia. Under current climatic conditions, D. noxia was mainly distributed in west-central Asia, most of Europe, central North America, southern South America, southern and northern Africa, and southern Oceania. Under the SSP 1-2.6, SSP 2-4.5, and SSP 5-8.5 scenarios for the 2030s and 2050s, the potential suitable areas increased, and the centroid migrated to higher latitudes. The early warning of D. noxia in northwestern Asia, western Europe, and North America should be attended to further. Our results provide a theoretical basis for early monitoring and warning of D. noxia worldwide.

BACKGROUND: Geographic differences exist in the antibiotic resistance patterns of Helicobacter pylori. Personalized treatment regimens based on local or individual resistance data are essential. We evaluated the current status of H. pylori resistance in Ningxia, analyzed resistance-related factors, and assessed the concordance of phenotypic and genotypic resistance. METHODS: Strains were isolated from the gastric mucosa of patients infected with H. pylori in Ningxia and relevant clinical information was collected. Phenotypic antibiotic susceptibility assays (Kirby-Bauer disk diffusion) and antibiotic resistance gene detection (Sanger sequencing) were performed. RESULTS: We isolated 1955 H. pylori strains. The resistance rates of H. pylori to amoxicillin, levofloxacin, clarithromycin, and metronidazole were 0.9%, 42.4%, 40.4%, and 94.2%, respectively. Only five tetracycline-resistant and one furazolidone-resistant strain were identified. Overall, 3.3% of the strains were sensitive to all six antibiotics. Multidrug-resistant strains accounted for 22.9%, of which less than 20% were from Wuzhong. Strains isolated from women and patients with nonulcerative disease had higher rates of resistance to levofloxacin and clarithromycin. Higher rates of resistance to metronidazole, levofloxacin, and clarithromycin were observed in the older age group than in the younger age group. The kappa coefficients of phenotypic resistance and genotypic resistance for levofloxacin and clarithromycin were 0.830 and 0.809, respectively, whereas the remaining antibiotics showed poor agreement. CONCLUSION: H. pylori antibiotic resistance is severe in Ningxia. Therefore, furazolidone, amoxicillin, and tetracycline are better choices for the empirical therapy of H. pylori infection in this region. Host sex, age, and the presence of ulcerative diseases may affect antibiotic resistance of the bacteria. Personalized therapy based on genetic testing for levofloxacin and clarithromycin resistance may be a future direction for the eradication therapy of H. pylori infection in Ningxia.

Hymenopteran parasitoids generally show a haplo-diploid sex determination system. Haploid males are produced from unfertilized eggs, whereas diploid females develop from fertilized eggs (arrhenotokous). In some cases, diploid females develop from unfertilized eggs (thelytokous). Diglyphus wani (Hymenoptera: Eulophidae) is a biological control agent for agromyzid leafminers and have arrhenotokous and thelytokous strains. However, the morphological characteristics of two strains of D. wani are so similar that it is difficult to accurately distinguish them based on morphology. Here, a rapid molecular identification method was developed based on the mitochondrial gene cytochrome c oxidase I (COI) and one-step multiplex PCR. Two primer combinations, PC1 (Ar-F1/Th-F1/WR2) and PC2 (Ar-F1/Th-F4/WR2), were designed and repeatedly screened to distinguish two strains simultaneously, of which two special forward primers Th-F1/Th-F4 were used for the thelytokous strain and one special forward primer Ar-F1 was used for the arrhenotokous strain. In addition, a common reverse primer, WR2, was used for both strains. The PC1 and PC2 PCR assays were effective in distinguishing the two strains at different developmental stages and field colonies. This method provides a reliable, highly sensitive, and cost-effective tool for the rapid identification of the two strains of D. wani.

BACKGROUND: Both arrhenotoky (sexual reproduction of females and asexual reproduction of males) and thelytoky (asexual reproduction of females) occur within the order Hymenoptera. The existence of both thelytokous and arrhenotokous strains within one species provides an opportunity to compare the biocontrol efficiency between two reproductive modes. The parasitoid Neochrysocharis formosa (Westwood) (Hymenoptera: Eulophidae) has thelytokous and arrhenotokous strains with sympatric distributions. This parasitoid is used to control invasive leafminers through feeding, stinging, and parasitization. To compare the biocontrol efficiency of the two strains, we analyzed life tables and host-killing parameters of these two strains reared on the leafminer Liriomyza sativae Blanchard using the age-stage, two-sex life table and the CONSUME-MSChart software. RESULTS: Our results showed that the intrinsic rate of increase (r), finite rate of increase (λ), and net reproduction rate (R0 ) of the thelytokous strain were significantly higher than those of the arrhenotokous strain. The thelytokous females also performed better than the arrhenotokous females for the net host-feeding rate, net host-stinging rate, and net host-killing rate, but not the finite parasitism rate. Conclusively, the finite host-killing rate of the thelytokous strain (0.8720 ± 0.0516) was significantly higher than that of the arrhenotokous strain (0.5914 ± 0.0832). CONCLUSION: We concluded that thelytokous N. formosa is a better candidate as a biocontrol agent than arrhenotokous N. formosa to control leafminers. Our results shed light on how to choose a better biocontrol agent for integrated pest management (IPM) based on biological control, especially for co-occurring thelytokous and arrhenotokous parasitoids. © 2022 Society of Chemical Industry.

Historical polycyclic aromatic hydrocarbon (PAH) pollution was explored through the sedimentary records of three lakes: Huguangyan Maar Lake (HGY) in South China, Mayinghai Lake (MYH) in North China, and Sihailongwan Lake (SHLW) in Northeast China. In these three lakes, the PAH concentrations in sediments are still rising, showing the different trend to lakes in developed countries. PAH pollution in South China occurred from 1850, much earlier than the increases since 1980 observed in North and Northeast China. The temporal trends of PAH concentrations in lake sediments are highly correlated with local economic development. Spatially, although the region where HGY is located has the highest gross domestic product, higher fluxes of PAHs were found in MYH sediments, indicating that atmospheric PAH pollution in North China might be more serious, and that PAH pollution is not fully correlated with economic development. Source analysis suggested that the PAHs in lake sediments are mainly derived from oil leaks, coal and biomass combustion, vehicle emissions, and diagenesis. Positive matrix factorization (PMF) model revealed that the contribution of vehicle emissions and coal combustion to PAHs has increased significantly in the past 40 years. Benzo(a)pyrene equivalent (BaPE) in the surface sediments of MYH and SHLW were similar and higher than in HGY. In HGY, vehicle emissions posed the highest toxic risk, followed by coal combustion. However, in MYH, the toxicity risk of vehicle emissions was close to that of coal and biomass combustion due to the highly developed coal industry in Shanxi Province. In SHLW, the contribution of fossil fuel combustion to BaPE was significantly higher than that of biomass combustion. This study provides important information for understanding PAH pollution affected by anthropogenic activities in the Anthropocene and provides a scientific basis for formulating PAH pollution control strategies.

Ecological niche is a key concept that links species distributions. Ecological niche shifts are expected to affect the potential invasive risk of alien species. Rapistrum rugosum is an invasive agricultural weed in many countries. Wild populations of R. rugosum have been recorded in China, representing a great threat to the regional crops. Based on distribution records from different regions and relevant environmental variables, the present study predicted the potential distribution and estimated the invasive risk of R. rugosum in China. Ecological niche shifts strongly affected the potential invasive risk of R. rugosum in China. The two most important variables were annual temperature range (Bio7) and mean temperature of the coldest quarter (Bio11). The total suitable habitat for the species covered an area of 287.53 × 104km2 and was mainly distributed in Southwest, Southeast, and Central China. Australia, Canada, Brazil, the United States, and Argentina accounted for over 90% of the inspection records of R. rugosum from Chinese entry ports during 2015-2018. The intercepted R. rugosum was frequently mixed in Glycine max (L.) Merr., Hordeum vulgare L., linseed, Triticum aestivum L., and Sorghum bicolor (L.) Moench. Moreover, 80% interceptions were recorded from Tianjin, Guangdong, Nanjing, and Chengdu customs. Climatic conditions do not limit the establishment capability of R. rugosum in China. Our results provide a theoretical reference for the development of monitoring and control measures for this invasive weed.

The historical atmospheric heavy metal pollution of southern China over the past 200 years was explored by analyzing radiometric dating, heavy metals, and Pb isotopes from a sediment core in Huguangyan Maar Lake. Zn, Cd, Sb, Tl, and Pb in the lake are closely related to anthropogenic activities, while Cr and Ni are mainly derived from the weathering of basalt surrounding the lake. Atmospheric Zn, Cd, Sb, and Tl increased rapidly after 1980, consistent with the local industrial development. The increase of atmospheric Pb in southern China occurred earlier than in other regions of China, with the increase after 1850. War and the use of leaded gasoline were the main causes for the rapid increase in atmospheric Pb during 1910-1950. From 1950 to 2000, the input of Pb from anthropogenic activities decreased gradually due to the stable social environment. After 2000, atmospheric Pb continued to rise due to continued industrial development. The three-end-member model of Pb isotopes indicates that coal combustion is the main source of current atmospheric Pb. The proportion of Pb derived from vehicle exhaust emissions reached a peak in the 1960s, then gradually decreased and further reduced with the ban on leaded gasoline after 2000. These results are important in identifying the sources of atmospheric heavy metal pollution and in formulating pollution control strategies.

In Hymenoptera species, the reproductive mode is usually arrhenotoky, where haploid males arise from unfertilized eggs and diploid females from fertilized eggs. In addition, a few species reproduce by thelytoky, where diploid females arise from unfertilized eggs. Diploid females can be derived through various cytological mechanisms in thelytokous Hymenoptera species. Hitherto, these mechanisms were revealed mainly in endosymbiont-induced thelytokous Hymenoptera species. In contrast, thelytokous Hymenoptera species in which a reproductive manipulator has not been verified or several common endosymbionts have been excluded were paid less attention in their cytological mechanisms, for instance, Diglyphus wani (Hymenoptera: Eulophidae). Here, we investigated the cytological mechanism of D. wani using cytological methods and genetic markers. Our observations indicated that the diploid karyotypes of two strains of D. wani consist of four pairs of relatively large metacentric chromosomes and one pair of short submetacentric chromosomes (2n = 10). The arrhenotokous strains could complete normal meiosis, whereas the thelytokous strain lacked meiosis and did not expulse any polar bodies. This reproductive type of lacking meiosis is classified as apomictic thelytoky. Moreover, a total of 636 microsatellite sequences were obtained from thelytokous D. wani, dominated by dinucleotide repeats. Genetic markers results showed all three generations of offspring from thelytokous strain maintained the same genotype as their parents. Our results revealed that D. wani is the first eulophid parasitoid wasp in Hymenoptera whose thelytoky was not induced by bacteria to form an apomictic thelytoky. These findings provide a baseline for future inner molecular genetic studies of ameiotic thelytoky.

Diglyphus species are ecologically and economically important on agromyzid leafminers. In 2018, a thelytokous species, Diglyphuswani Liu, Zhu & Yefremova, was firstly reported and described. Subsequently, the arrhenotokous D.wani were discovered in Yunnan and Guizhou Provinces of China. We compared the morphological characteristics of thelytokous and arrhenotokous strains. However, the females of two strains had a strongly similar morphology and showed subtle differences in fore- and hind-wings. The difference was that forewing of arrhenotokous female was with denser setae overall, showing that costal cell with 2 ~ 4 rows of setae on dorsal surface and the setae of basal cell with 15 ~ 21 hairs and forewing of thelytokous female was with two rows of setae on dorsal surface and basal cell with 10 ~ 15 hairs generally. The setation beneath the marginal vein of the hind-wing of arrhenotokous female is denser than the same area of thelytokous female. To explore the genetic divergence between thelytokous and arrhenotokous strains of D.wani, the mitochondrial and nuclear gene were applied and sequenced. The polygenic analyses revealed that two strains can be distinguished by COI, ITS1 and ITS2. The mean sequence divergence between the two strains was 0.052, 0.010 and 0.007, respectively. Nevertheless, the 28S gene was unfeasible due to its containing a sharing haplotype between different strains. The two strains of D.wani are dominant parasitoids against agromyzid leafminers and such effective discernible foundation provides future in-depth studies on biological characteristics, along with insight into field application of two strains of D.wani.
