RESUMO

As a global emerging contaminant, microplastics (MPs) in water or soil can accumulate in vegetables, making them easily ingested through the diet. With excellent and tunable optical properties, carbon dots (CDs) are highly advantageous for tracing the entry process of MPs. Originally, long-wavelength CDs were synthesized from leaf-derived extracts, and fluorescent submicrometer plastics (CDs-MPs) with clean surfaces and concentrated particle sizes were obtained by soap-free microemulsion polymerization. The concentration of CDs-MPs exhibits a significant linear relationship with long-wavelength fluorescence intensity (λEx/λEm: 415/676 nm). Soybean sprouts (SBS), as an important type of food, are susceptible to contamination of MPs due to their soft epidermis and rapidly growing biomass. The results showed that CDs-MPs could be embedded into the cortex of SBS and enter the plant with cell division and elongation, leading to an increase in pore size on the cell wall surface. After entering the root system, CDs-MPs will pass through the Casparian strip and migrate in the vessels. Then, CDs-MPs enter the leaves through vascular bundles, and the distribution and size of epicuticular wax on leaves have changed. Furthermore, SBS showed resistant growth and increased levels of oxidative response when exposed to MPs/CDs-MPs. It is the first study to demonstrate the application of leaf-derived CDs in the prevention of MPs pollution by revealing the migration behavior of submicrometre plastics in SBS.

RESUMO

Limited research has been undertaken regarding the homogeneity of CoCrPtB alloy billets. A CoCrPtB alloy was processed through casting and vacuum hot pressing. This investigation delved into the interconnection between the secondary dendrite arm spacing (SDAS) in the as-hot-pressed samples and their corresponding attributes, specifically Vickers hardness and magnetic properties. Systematic sampling was conducted on the cross-sectional layer and longitudinal surface. Upon examination of the cross-sectional layer proximate to the uppermost region of the hot casting, a discernible parabolic trend was observed for the SDAS that exhibited a gradual increment from the peripheral regions toward the central area along the width. Simultaneously, the fraction of the dendrite phase displayed a consistent linear decline, attaining its peak value at the central portion of the billet. Conversely, on the longitudinal surface, SDAS and the fraction of the dendrite phase remained fairly uniform within the same column sampling regions. However, a notable divergence was identified in the central section, characterized by an augmented SDAS and diminished dendrite phase content. This inherent microstructural inhomogeneity within the CoCrPtB alloy engendered discernible disparities in material properties.

RESUMO

With the progress of science and technology, artificial intelligence is widely used in various disciplines and has produced amazing results. The research of the target detection algorithm has significantly improved the performance and role of unmanned aerial vehicles (UAVs), and plays an irreplaceable role in preventing forest fires, evacuating crowded people, surveying and rescuing explorers. At this stage, the target detection algorithm deployed in UAVs has been applied to production and life, but making the detection accuracy higher and better adaptability is still the motivation for researchers to continue to study. In aerial images, due to the high shooting height, small size, low resolution and few features, it is difficult to be detected by conventional target detection algorithms. In this paper, the UN-YOLOv5s algorithm can solve the difficult problem of small target detection excellently. The more accurate small target detection (MASD) mechanism is used to greatly improve the detection accuracy of small and medium targets, The multi-scale feature fusion (MCF) path is combined to fuse the semantic information and location information of the image to improve the expression ability of the novel model. The new convolution SimAM residual (CSR) module is introduced to make the network more stable and focused. On the VisDrone dataset, the mean average precision (mAP) of UAV necessity you only look once v5s(UN-YOLOv5s) is 8.4% higher than that of the original algorithm. Compared with the same version, YOLOv5l, the mAP is increased by 2.2%, and the Giga Floating-point Operations Per Second (GFLOPs) is reduced by 65.3%. Compared with the same series of YOLOv3, the mAP is increased by 1.8%, and GFLOPs is reduced by 75.8%. Compared with the same series of YOLOv8s, the detection accuracy of the mAP is improved by 1.1%.

RESUMO

In recent years, highway accidents occur frequently, the main reason is that there is always foreign body invasion on the highway, which makes people unable to respond to emergencies in time. In order to reduce the occurrence of highway incidents, an object detection algorithm for highway intrusion was proposed in this paper. Firstly, a new feature extraction module was proposed to better preserve the main information. Secondly, a new feature fusion method was proposed to improve the accuracy of object detection. Finally, a lightweight method was proposed to reduce the computational complexity. We compare the algorithm in this paper with existing algorithms, the experimental results showed that: On the Visdrone dataset (small size targets), (a) the CS-YOLO was 3.6% more accurate than the YOLO v8. (b) The CS-YOLO was 1.2% more accurate than the YOLO v8 on the Tinypersons dataset (minimal size targets). (c) CS-YOLO was 1.4% more accurate than YOLO v8 on VOC2007 data set (normal size).

RESUMO

In this study, pure copper's hot deformation behavior was studied through isothermal compression tests at deformation temperatures of 350~750 °C with strain rates of 0.01~5 s-1 on a Gleeble-3500 isothermal simulator. Metallographic observation and microhardness measurement were carried out of the hot compressed specimens. By analyzing the true stress-strain curves of pure copper under various deformation conditions during the hot deformation process, the constitutive equation was established based on the strain-compensated Arrhenius model. On the basis of the dynamic material model proposed by Prasad, the hot-processing maps were acquired under different strains. Meanwhile, the effect of deformation temperature and strain rate on the microstructure characteristics was studied by observing the hot-compressed microstructure. The results demonstrate that the flow stress of pure copper has positive strain rate sensitivity and negative temperature correlation. The average hardness value of pure copper has no obvious change trend with the strain rate. The flow stress can be predicted with excellent accuracy via the Arrhenius model based on strain compensation. The suitable deforming process parameters for pure copper were determined to be at a deformation temperature range of 700~750 °C and strain rate range of 0.1~1 s-1.

RESUMO

This study aimed to explore how kinesin family member 2C (KIF2C) influences the progression of non-small cell lung cancer (NSCLC). The levels of KIF2C and microRNA-186-3p (miR-186-3p) were examined by quantitative real-time polymerase chain reaction (qRT-PCR). Through the utilization of cell counting kit-8 (CCK-8) assay, colony formation assay, wound closure assay, and Transwell assay, NSCLC cell proliferation, migration, and invasion were identified, respectively. NSCLC cell apoptosis was assessed using the TUNEL assay and flow cytometry (FCM) assay. Luciferase reporter analysis was used to investigate the relationship between KIF2C and miR-186-3p. Western blot assays were conducted to investigate the influence of KIF2C on the AKT-GSK3ß-ß-catenin pathway. The results showed that KIF2C was up-regulated in NSCLC cells, which predicted poor prognosis. KIF2C overexpression promoted the proliferation, migration, and invasion of NSCLC cells as well as inhibited NSCLC cell apoptosis. KIF2C was as a key target of miR-186-3p. High expression of KIF2C, meanwhile, increased the levels of ß-catenin, p-GSK-3ß and phosphorylated protein kinase B (p-AKT). KIF2C downregulation and miR-186-3p upregulation reversed these outcomes. As an oncogenic factor, KIF2C is negatively regulated by miR-186-3p and participates in the progression of NSCLC through the AKT-GSK3ß-ß-catenin pathway.

RESUMO

Acute myeloid leukemia (AML) is widely considered an immunoresponsive malignancy. However, potential association between glycolysis-immune related genes and AML patients' prognosis has been seldom studied. AML-related data was downloaded from TCGA and GEO databases. We grouped patients according to Glycolysis status, Immune Score and combination analysis, basing on which overlapped differentially expressed genes (DEGs) were identified. The Risk Score model was then established. The results showed that totally 142 overlapped genes were probably correlated with glycolysis-immunity in AML patients, among which 6 optimal genes were screened to construct Risk Score. High Risk Score was an independent poor prognostic factor for AML. In conclusion, we established a relatively reliable prognostic signature of AML based on glycolysis-immunity related genes, including METTL7B, HTR7, ITGAX, TNNI2, SIX3 and PURG.

RESUMO

Intercropping the arsenic (As) hyperaccumulator Pteris vittata with fruit trees can safely yield peaches in As-polluted orchards in South China. However, the soil As remediation effects and the related mechanisms of P. vittata intercropped with peach trees with additives in the north temperate zone have rarely been reported. A field experiment was conducted to systematically study the intercropping of peach (Amygdalus persica) with P. vittata with three additives [calcium magnesium phosphate (CMP), ammonium dihydrogen phosphate (ADP), and Stevia rebaudiana Bertoni residue (SR)] in a typical As-contaminated peach orchard surrounding a historical gold mine in Pinggu County, Beijing City. The results showed that compared with monoculture (PM) and intercropping without addition (LP), the remediation efficiency of P. vittata intercropping was significantly increased by 100.9 % (CMP) to 293.5 % (ADP). CMP and ADP mainly compete with available As (A-As) adsorbed to the surface of Fe-Al oxide through PO43-, while SR might activate A-As by enhancing dissolved organic carbon (DOC) in P. vittata rhizospheres. The photosynthetic rates (Gs) of intercropped P. vittata were significantly positively correlated with pinna As. The intercropping mode applied with the three additives did not obviously affect fruit quality, and the net profit of the intercropping mode (ADP) reached 415,800 yuan·ha-1·a-1. The As content in peaches was lower than the national standard in the intercropping systems. Comprehensive analysis showed that A. persica intercropped with P. vittata applied with ADP is better than other treatments in improving risk reduction and agricultural sustainability. In this study, a theoretical and practical basis is provided for the safe utilization and remediation of As-contaminated orchard soil in the north temperate zone.

RESUMO

The influence of environmental factors on Cd accumulation by Hylotelephium spectabile and its physiological mechanisms are unclear. A field trial was conducted to investigate the effects of nitrogen, soil moisture, and light regulation on plant growth, Cd absorption and translocation, and the photosynthetic characteristics of two H. spectabile populations (LN with high Cd accumulation capacity and HB1 with relatively low Cd accumulation capacity). The results showed that Cd accumulation in LN was 59.6% higher than that in HB1 which may partly be explained by the inherent high transpiration rate of LN, especially at the terminal stage. In addition, the photosynthetic rate of LN responded more positively to nitrogen than HB1, which further amplified its advantages on plant growth and Cd accumulation. Moderate drought significantly stimulated root growth of LN, indicating that LN possesses stronger resistance to drought. Shade inhibited Cd distribution, rather than directly affecting Cd concentrations in H. spectabile. The combined stress of shade and drought had a synergistic effect on Cd translocation in H. spectabile. Moreover, LN achieved 17.3%∼444.5% higher transpiration levels than HB1 under environmental stress, which ensured a more efficient Cd transport capacity of LN. Therefore, the investigation of photosynthetic characteristics further revealed the physiological mechanism by which LN accumulated Cd superior to HB1 under environmental stress and responded more positively to nitrogen nutrition.

RESUMO

Intercropping a Cd-accumulator with economically valuable crops is common in slightly or moderately Cd-polluted farmland soils. A field experiment was conducted to evaluate the effects of water-soluble chitosan (WSC) on the growth and Cd uptake of the Cd-accumulator Hylotelephium spectabile and soybean (Glycine max) during a co-cultivation in Cd-contaminated agricultural soil (WSC, 0 and 10 g·m-2). The results indicated that soybean yields were highest in response to the intercropping and WSC treatment. The results from the field trials generally showed that intercropping and WSC treatments significantly decreased Cd concentrations in inedible parts of soybean by 42.9-72.1% (except for stems), in the meantime, increased 95.8%-334.6% in shoot and root tissues of H. spectabile compared with the control (p < 0.05). The data revealed that Cd uptake was highest for H. spectabile during the intercropping and WSC treatment. The application of WSC in the intercropping system significantly increased the uptake of Cd by H. spectabile, but not by soybean. The findings of this study suggest that combining an intercropping system with a WSC treatment may be better for remediating Cd-contaminated soils than other methods involving the growth of a single hyperaccumulator.

This paper clearly focused on the accumulation and uptake of Cd in the system of intercropping of Cd-accumulator (Hylotelephium spectable) and soybean (Glycine max) grown in Cd-polluted farmland soils supplied with water-soluble chitosan (WSC) under field conditions. Some studies mainly focused on active agent to promote remediation efficiency of (hyper) accumulators. This study indicated that combining the intercropping system with WSC may be better for remediating Cd-contaminated soils than the methods involving a single hyperaccumulator.

RESUMO

The controllable design of red-emitting carbon dots and further exploration of their application in the trace determination of environmental pollutants remains a tremendous challenge. Herein, the novel strategy for red fluorescent carbon dots (R-CDs) with a higher quantum yield of 58.9% was proposed by doping small-molecule urea into the bio-dye of resazurin for the first time, which can retain the luminophore of precursors and exhibit exceptional optical, advantageous reversibility and outstanding photostability. Importantly, the R-CDs exhibit a remarkable fluorescence reduction towards tetracyclines (TCs) accompanied by a noticeable color change of R-CDs solution from red to yellow, which can realize the trace detection of TCs at strelatively low levels, including tetracycline (TC), oxytetracycline (OTC), and chlortetracycline (CTC). The linear range of TC, CTC, and OTC are 3-40 µM, 4-50 µM, and 2-50 µM, and the corresponding detection limits are 38.5 nM, 64.6 nM, and 45.4 nM, respectively (S/N = 3). Furthermore, the R-CDs demonstrate sensitivity to the physiological pH in the linear range of 4.0-5.0 and 5.0-6.2 with a pKa of 5.61. As a multifunctional fluorescent sensor, R-CDs can provide a new perspective for the preparation of long-wavelength CDs, and further realize the trace determination of environmental pollutants.

RESUMO

Lung adenocarcinoma is a malignancy with a low overall survival and a poor prognosis. Studies have shown that lung adenocarcinoma progression relates to locus-specific/global changes in histone modifications. To explore the relationship between histone modification and gene expression changes, we focused on 11 histone modifications and quantitatively analyzed their influences on gene expression. We found that, among the studied histone modifications, H3K79me2 displayed the greatest impact on gene expression regulation. Based on the Shannon entropy, 867 genes with differential H3K79me2 levels during tumorigenesis were identified. Enrichment analyses showed that these genes were involved in 16 common cancer pathways and 11 tumors and were target-regulated by trans-regulatory elements, such as Tp53 and WT1. Then, an open-source computational framework was presented (https://github.com/zlq-imu/Identification-of-potential-LUND-driver-genes). Twelve potential driver genes were extracted from the genes with differential H3K79me2 levels during tumorigenesis. The expression levels of these potential driver genes were significantly increased/decreased in tumor cells, as assayed by RT-qPCR. A risk score model comprising these driver genes was further constructed, and this model was strongly negatively associated with the overall survival of patients in different datasets. The proportional hazards assumption and outlier test indicated that this model could robustly distinguish patients with different survival rates. Immune analyses and responses to immunotherapeutic and chemotherapeutic agents showed that patients in the high and low-risk groups may have distinct tendencies for clinical selection. Finally, the regions with clear H3K79me2 signal changes on these driver genes were accurately identified. Our research may offer potential molecular biomarkers for lung adenocarcinoma treatment.

RESUMO

Field experiments were conducted to investigate the effects of two foliar fertilizers, water-soluble chitosan (WSC) and Na2SiO3 (Si), on the accumulation of Pb by a low-Pb accumulator Brassica napus cultivar (QY-1) grown at two mildly Pb-contaminated farmland sites surrounding working smelters in Jiyuan city, Henan province, China. Regardless of the frequency of the fertilizer treatments, the foliar application of WSC (0.01%) or Si (0.15%) significantly increased the QY-1 biomass and decreased the grain Pb concentrations. Compared with the control treatment, spraying plants once with WSC or Si during the flowering period achieved the best effect in the two soils with different pollution, which may be because inhibiting the accumulation of Pb in grains by decreasing the husk-to-grain transfer coefficient. Thus, the foliar application of WSC or Si combined with the cultivation of a low-Pb accumulator is a promising approach for optimizing the utility of Pb-contaminated farmland affected by atmospheric deposition.

RESUMO

In order to explore the distribution characteristics of heavy metal contamination of farmland soil surrounding Luancheng town, Shijiazhuang City, Henan province, the concentrations of As, Cd, Cr, Cu, Hg, Ni, Pb, and Zn in the surface soil and crops were determined and assessed. The principal components were also analyzed for source apportionment. The heavy metal concentrations in crops were further detected, and the non-carcinogenic health risks in the study area were evaluated using the probabilistic risk assessment method, as to provide a theoretical basis for the treatment, prevention, and control of heavy metal pollution in farmland soil in Luancheng. According to the results, ω(Cd), ω(Cr), ω(Cu), ω(Pb), and ω(Zn) in the soils were 0.06-1.08, 22.14-473.47, 12.83-150.74, 10.75-577.72, and 62.23-652.78 mg·kg-1, which exceeded the standard with over-standard rates reaching 1.83%, 1.22%, 0.61%, 0.61%, and 1.22%, respectively. Further, Cd and Pb were transported into crops, in which Cd concentrations exceeded the standard in some corn samples, and Cd and Pb concentrations exceeded the standard in some wheat samples. The total non-carcinogenic health risks (TTHQ) to the human body caused by the consumption of heavy metals in corn grown in the study area were all less than 1, with no obvious negative effects, and TTHQ was higher than 1 in wheat, increasing the likelihood of negative impacts on the human body. With the influence of the distribution of pollution-related enterprises in the industrial zone, heavy metal concentrations were higher in the south, west, and middle directions of the study area. Among them, the study area soil was slightly contaminated by Cd (Level 1). Cd and Hg had a slight potential ecological risk (Level 2), whereas other heavy metals had low potential ecological risk (Level 1). In general, most of the surface cultivated soil was not obviously polluted by heavy metals in the study area. According to the PMF results and survey, we speculated that soil heavy metals mainly came from soil parent material (52.05%), artificial pollution sources (historical sewage irrigation and industrial manufacture) (32.98%), and atmospheric deposition (14.97%). To summarize, the study area should be divided into a priority protection category and safe utilization category. The input of pollution sources should be strictly controlled for the priority protection category, and alternative planting, rotating, and fallow should be implemented for the safe utilization category to reduce the risk of standard-exceeding agricultural products.

RESUMO

Pot and field trials were conducted to investigate Cd uptake and phytoremediation efficiency of two Brassica napus cultivars (QY-1 and SYH) with applied water-soluble chitosan (WSC, Pot: 0, 2% and 4%; Field: 0 and 10 g·m-2) grown in Cd-contaminated soils. The results from the pot and field trials generally showed that WSC treatments significantly increased Cd concentrations in shoot and root tissues by 33.77-159.71% (except for SYH/JY) and 7.42-168.71% of two B. napus cultivars compared with the control (p < 0.05). The uptake of Cd by shoots of SYH was obviously higher than by shoots of QY-1 treated with WSC under pot and field conditions, which was 1.54-2.22 times than that of QY-1 (p < 0.05). The results indicated that 2% WSC treatment significantly increased the water-soluble and acid extractable Cd in rhizosphere soils of both B. napus cultivars. Furthermore, Cd concentrations in the oils of two B. napus cultivars with applied WSC (10 g·m-2) grown under field conditions were not significantly different from commercial rapeseed oils. Rapeseed oil of B. napus is not only an edible oil with high nutritional value, but it can also be converted into biomass diesel that can be used as a substitute for petroleum diesel.

RESUMO

Hyperaccumulator-cash crop intercropping remediation is a research hotspot for heavy metal contaminated farmland, but few studies evaluated its feasibility based on practice. Field experiments and survey statistics were conducted to obtain parameters of Pteris vittata-Citrus reticulata/Zea mays intercropping, and potential of intercropping remediation was evaluated based on remediation efficiency and economic benefits. The results showed that intercropping hyperaccumulator with cash crop (especially herbs) had a certain negative effect on remediation efficiency because of the influence on planting density and harvest times of hyperaccumulator; while trees could partly alleviate this effect. Until achieving the predetermined target, the net remediation cost of P. vittata-Z. mays was 18.2 $/g As, followed by P. vittata monoculture (13.3 $/g) and P. vittata-C. reticulata (8.6 $/g). Based on the proposed evaluation model, nealy half of the P. vittata intercropping modes had low economic benefits, insufficient to compensate the cost of sacrificing remediation efficiency. Based on the data from two soil remediation projects, when net income of cash crops intercropped with As-hyperaccumulators exceeded 5865/1607 $/hm2 (herbs/trees), the economic benefit of intercropping will be relatively obvious. Therefore, cash crops should be considered from three aspects: planting conditions, spatial allocation and economic benefits. Novelty statement: This work analyzed remediation efficiency and economic benefits of intercropping remediation. An economic benefit evaluation model was established to evaluate intercropping remediation modes. The selection principle and net income threshold of cash crops in intercropping was put forward for the first time.[Figure: see text]HighlightsThe selection principle of cash crops in intercropping remediation was put forward. An evaluation model of P. vittata intercropping remediation was established. The net cost of extracting 1.0 g of soil As in each remediation mode was proposed.Net income of herb/tree intercropped with P. vittata should exceed 5865/1607 $/hm2.

RESUMO

BACKGROUND: The dysregulation of LncRNAs is related to the malignant progression of many cancers. OBJECTIVE: The study aimed to investigate the expression and the biological role of LncSNHG3 in hepatocellular carcinoma (HCC). METHODS: The TCGA data of the LncSNHG3 in HCC were analyzed. The expression in HCC cell lines was detected by qRT-PCR. Proliferation, migration, and invasion of HepG2 and Huh7 were examined by cell counting kit-8, colony formation, transwell assays, and wound healing assays. At the same time, the interactions among LncSNHG3, miR-152-3p, and JAK1 were confirmed by dual-luciferase reporter assay, RNA immunoprecipitation, subcellular distribution. Xenograft tumor-bearing mice models were used to measure the effect of LncSNHG3 on the growth of HCC in vivo. The apoptosis and epithelial mesenchymal transition (EMT)-associated proteins were checked by WB and IHC. RESULTS: LncSNHG3 was overexpressed in HCC tissues and cell lines. In addition, it is correlated with the tumor stage and survival time of HCC patients. Down-regulated LncSNHG3 could significantly suppress the EMT progression of HCC in vivo and in vitro. LncSNHG3 could promote the JAK1 expression by sponging miR-152-3p. CONCLUSIONS: LncSNHG3 acted as an oncogene and promoted the EMT procession in HCC by binding miR-152-3p and promoting JAK1 expression. Predictably, LncSNHG3 was used as a potential marker and will be used as a novel therapy target for HCC in the future.

RESUMO

Compared with phytoextraction, growing suitable crops may be a more profitable and practical approach for managing contaminated farmland, especially when there are multiple pollutants. In this 5-year field study, the phytoaccumulator Hylotelephium spectabile, the high-biomass species amaranth (Amaranthus hypochondriacus), and a winter rapeseed/maize rotation crop were cultivated on farmland contaminated with cadmium (Cd) and lead (Pb). Over 4 consecutive years, the annual Cd uptake and extraction efficiency of H. spectabile was 117.6 g hm-2 and 2.36%, respectively. The Cd extraction efficiency of amaranth was equivalent to that of H. spectabile because of its high biomass, and it extracted more Pb (660-2210 g hm-2) from the soil than did H. spectabile. However, neither of these species was able to remediate contaminated farmland rapidly and inexpensively, even with enhancing strategies such as variety screening and the addition of fertilizers and a chelating agent. A safe utilization approach to cultivate rapeseed instead of wheat significantly reduced the carcinogenic and noncarcinogenic risks. The concentrations of heavy metals in rapeseed oil were below the limits specified in the Chinese national food standard, and the heavy metal concentrations in the byproducts (rapeseed meal and straw) were below the limits specified in Chinese national standards for organic fertilizer and feed. The cost of safe utilization was one-quarter that of phytoextraction, and the net economic benefit was 33.5%-123.5% higher than that of wheat crops. Therefore, the rapeseed/maize rotation is a profitable and feasible approach for the safe utilization of Cd- and Pb-contaminated farmland on the northern plains of China.

RESUMO

The acquisition of cancer stem-like properties is believed to be responsible for cancer metastasis and therapeutic resistance in cervical cancer (CC). CC tissues display a high expression level of hexokinase 2 (HK2), which is critical for the proliferation and migration of CC cells. However, little is known about the functional role of HK2 in the maintenance of cancer stem cell-like ability and cisplatin resistance of CC cells. Here, we showed that the expression of HK2 is significantly elevated in CC tissues, and high HK2 expression correlates with poor prognosis. HK2 overexpression (or knockdown) can promote (or inhibit) the sphere-forming ability and cisplatin resistance in CC cells. In addition, HK2-overexpressing CC cells show enhanced expression of cancer stem cell-associated genes (including SOX2 and OCT4) and drug resistance-related gene MDR1. The expression of HK2 is mediated by miR-145, miR-148a, and miR-497 in CC cells. Overexpression of miR-148a is sufficient to reduce sphere formation and cisplatin resistance in CC cells. Our results elucidate a novel mechanism through which miR-148a regulates CC stem cell-like properties and chemoresistance by interfering with the oncogene HK2, providing the first evidence that dysregulation of the miR-148a/HK2 signaling plays a critical role in the maintenance of sphere formation and cisplatin resistance of CC cells. Our findings may guide future studies on therapeutic strategies that reverse cisplatin resistance by targeting this pathway.

RESUMO

Cervical cancer is one of the most common gynecological cancers. Cisplatin resistance remains a major hurdle in the successful treatment of cervical cancer. Aberrant expression of long non-coding RNAs (lncRNAs) and microRNAs (miRNAs) are implicated in cisplatin resistance. However, the regulatory functions of lncRNAs and miRNAs in cervical cancer cisplatin resistance and the underlying mechanisms are still elusive. Our qRT-PCR assays verified that miR-206 levels were down-regulated in cisplatin-resistant cervical cancer cells. The introduction of miR-206 sensitized cisplatin-resistant cervical cancer cells to cisplatin. Our qRT-PCR and luciferase reporter assays showed that Cyclin D2 (CCND2) was the direct target for miR-206 in cervical cancer cells. The cisplatin-resistant cervical cancer cells expressed higher CCND2 expression than the parental cells, whereas inhibition of CCND2 could sensitize the resistant cells to cisplatin treatment. Furthermore, we demonstrated that lncRNA OTUD6B-AS1 was up-regulated in cisplatin-resistant cervical cancer cells, and knocking down OTUD6B-AS1 expression induced re-acquirement of chemosensitivity to cisplatin in cervical cancer cells. We also showed that OTUD6B-AS1 up-regulated the expression of CCND2 by sponging miR-206. Low miR-206 and high OTUD6B-AS1 expression were associated with significantly poorer overall survival. Taken together, these results suggest that OTUD6B-AS1-mediated down-regulation of miR-206 increases CCND2 expression, leading to cisplatin resistance. Modulation of these molecules may be a therapeutic approach for cisplatin-resistant cervical cancer.
