RESUMO

Application of manure, biosolids, and recycled wastewater to croplands could be a potential pathway through which trace organic compounds (TOrCs) may be taken up by food crops. We present a framework to prepare a short list of TOrCs for detailed risk assessment and evaluation in terms of bioaccumulation. The framework was modified from Lipinski's method to predict drug permeability based on four critical properties: (i) molecular weight (MW); (ii) lipophilicity (expressed as log, the octanol-water partition coefficient); (iii) H-bond acceptors; and (iv) H-bond donors. The literature shows that the compounds with MW ranging from 200 to 500 can readily diffuse through mammalian membranes, the uptake of compounds with log >5 is hindered, and an excessive number of H-bond donors and H-bond acceptors reduces the permeability across a mammalian membrane bilayer. In general, mammalian and plant membranes are similar in structure and functions. Based on these four properties, we developed the "Rule of 3," which states that greater absorption and higher permeability of a TOrC is likely when its log is <3, its MW is <300, H-bond donors are <3, and H-bond acceptors are <6. Applicability of the framework was tested with published data, which showed that uptake and bioaccumulation of TOrCs in plants decreased in the order: Rule of 3 > Rule of 3 to 5 (log between 3 and 5, MW between 300 and 500, H-bond acceptors between 3 and 6, and H-bond donors between 3 and 5) > Rule of 5 (log >5, MW >500, H-bond acceptors >10, and H-bond donors <5). We conclude that TOrCs following the "Rule of 3" could be prioritized for detailed risk assessment involving dietary exposure.

A 17 kJ transportable plasma focus (PF) device with flexible transmission lines is developed and is characterized. Six custom made capacitors are used for the capacitor bank (CB). The common high voltage plate of the CB is fixed to a centrally triggered spark gap switch. The output of the switch is coupled to the PF head through forty-eight 5 m long RG213 cables. The CB has a quarter time-period of 4 µs and an estimated current of 506 kA is delivered to the PF device at 17 kJ (60 µF, 24 kV) energy. The average neutron yield measured using silver activation detector in the radial direction is (7.1 ± 1.4) × 10(8) neutrons/shot over 4π sr at 5 mbar optimum D2 pressure. The average neutron yield is more in the axial direction with an anisotropy factor of 1.33 ± 0.18. The average neutron energies estimated in the axial as well as in the radial directions are (2.90 ± 0.20) MeV and (2.58 ± 0.20) MeV, respectively. The flexibility of the PF head makes it useful for many applications where the source orientation and the location are important factors. The influence of electromagnetic interferences from the CB as well as from the spark gap on applications area can be avoided by putting a suitable barrier between the bank and the PF head.

Development of a palm top plasma focus device generating (5.2 ± 0.8) × 10(4) neutrons∕pulse into 4π steradians with a pulse width of 15 ± 3 ns is reported for the first time. The weight of the system is less than 1.5 kg. The system comprises a compact capacitor bank, a triggered open air spark gap switch, and a sealed type miniature plasma focus tube. The setup is around 14 cm in diameter and 12.5 cm in length. The energy driver for the unit is a capacitor bank of four cylindrical commercially available electrolytic capacitors. Each capacitor is of 2 µF capacity, 4.5 cm in diameter, and 9.8 cm in length. The cost of each capacitor is less than US$ 10. The internal diameter and the effective length of the plasma focus unit are 2.9 cm and 5 cm, respectively. A DC to DC converter power supply powered by two rechargeable batteries charges the capacitor bank to the desired voltage and also provides a trigger pulse of -15 kV to the spark gap. The maximum energy of operation of the device is 100 J (8 µF, 5 kV, 59 kA) with deuterium gas filling pressure of 3 mbar. The neutrons have also been produced at energy as low as 36 J (3 kV) of operation. The neutron diagnostics are carried out with a bank of (3)He detectors and with a plastic scintillator detector. The device is portable, reusable, and can be operated for multiple shots with a single gas filling.

The durability and restorable properties of superhydrophobicity are a most important issue essential for utilization of superhydrophobic materials in industrial and domestic fields. In this work, we have focused on the sol-gel dip coating synthesis, durability, and restoring properties of a superhydrophobic surface composed of spherical silica particles with low energy free surface successfully achieved on the surface of glass substrates. The water contact angle (WCA) of the as-prepared superhydrophobic coatings reached as high as 170±1° (WCA=170±1°) and the sliding angle was 3±1° (WSA=3±1°). The behavior of wettability on superhydrophobic coating surface under various external disturbances including outdoor environments, heat, and mild acidic condition is investigated. It is shown in particular that degradation and repairing of surface methyl groups between two such surfaces leads to a fully restorable coating surface by the surface modification with trimethylchlorosilane (TMCS) at room temperature. The coating also holds good thermal stability; it holds superhydrophobicity up to 550 °C and gets degrade into superhydrophilicity above 600 °C without deforming surface morphology. The results confirm the durability of the silica coating under different external disturbances and sol-gel dip coating method and restoring property provide the best solution to fabricate superhydrophobic silica coating surface with extended durability at low cost.

Development and operation of a portable and compact pulsed neutron source based on sealed-type plasma focus (PF) device are reported. The unit is the smallest sealed-type neutron producing PF device. The effective volume of the PF unit is 33 cm(3) only. A compact size single capacitor (4 µF) is used as the energy driver. A battery based power supply unit is used for charging the capacitor and triggering the spark gap. The PF unit is operated at 10 kV (200 J) and at a deuterium gas filling pressure of 8 mb. The device is operated over a time span of 200 days and the neutron emissions have been observed for 200 shots without changing the gas in between the shots. The maximum yield of this device is 7.8 × 10(4) neutrons/pulse. Beyond 200 shots the yield is below the threshold (1050 neutrons/pulse) of our (3)He detector. The neutron energy is evaluated using time of flight technique and the value is (2.49 ± 0.27) MeV. The measured neutron pulse width is (24 ± 5) ns. Multishot and long duration operations envisage the potentiality of such portable device for repetitive mode of operation.

The surface free energy of a solid determines its surface and interfacial behavior in processes like wetting and adhesion which is crucial for silica aerogels in case of organic liquid absorption and transportation of chemicals at nano-scale for biotechnological applications. Here, we have demonstrated that the surface free energy of aerogels can be tuned in wide range from 5.5892 to 0.3073 mJ/m(2) by modifying their surface using TMCS and HMDZ silylating reagents. The alcogels were prepared by two step acid-base catalyzed process where the molar ratio of precursors Tetraethoxysilane (TEOS):Methanol (MeOH):Oxalic acid:NH(4)OH:NH(4)F was kept at optimal value of 1:2.7:0.18×10(-4):0.02:0.22×10(-3), respectively. To modify gel surfaces, TMCS and HMDZ concentration have been varied from 3% to 12% and such alcogels were dried at ambient pressure. It is observed from FTIR for aerogels that increase in concentration of silylating reagent resulted increase in hydrophobicity. This leads to increase in contact angle for water from 123° to 155° but leads to decrease in surface free energy from 5.5892 to 0.3073 mJ/m(2). As there is not direct method, we have used Neumann's equation of state to estimate surface energy of aerogels.

The wetting of solid surfaces by water droplets is ubiquitous in our daily lives as well as in industrial processes. In the present research work, water repellent porous silica films are prepared on glass substrate at room temperature by sol-gel process. The coating sol was prepared by keeping the molar ratio of methyltriethoxysilane (MTES), methanol (MeOH), water (H(2)O) constant at 1:12.90:4.74, respectively, with 2M NH(4)OH throughout the experiments and the molar ratio (M) of MTES/Ph-TMS was varied from 0 to 0.22. A simple dip coating technique is adopted to coat silica films on the glass substrates. The static water contact angle as high as 164° and water sliding angle as low as 4° was obtained for silica film prepared from M=0.22. The surface morphological studies of the prepared silica film showed the porous structure with pore sizes typically ranging from 200nm to 1.3µm. The superhydrophobic silica films prepared from M=0.22 retained their superhydrophobicity up to a temperature of 285°C and above this temperature the films became superhydrophilic. The porous and water repellent silica films are prepared by proper alteration of the Ph-TMS in the coating solution. The prepared silica films were characterized by surface profilometer, scanning electron microscopy (SEM), transmission electron microscopy (TEM), Fourier Transform Infrared (FT-IR) spectroscopy, humidity tests, chemical aging tests, static and dynamic water contact angle measurements.

An electrically triggered rail gap switch has been designed over a commercially available copper clad fiberglass sheet commonly used in making printed circuit boards for applications requiring compact design and direct integration to parallel plate transmission lines. Switch performance has been investigated in terms of its inductance, jitter, and gap closing time. With an electrode separation of 9.0 mm, it has been found to have an inductance of 6 nH, gap closing time of 5 ns, and jitter of about 4-10 ns measured at 95% of self-breakdown voltage. An application of this switch has been demonstrated as an electrically exploding foil accelerator developed over the same board and velocities up to 1.6 km/s have been achieved on Kapton flyers with diameter of 3.0 mm and thickness of 125 microm using a compact 1 microF capacitor bank.

Two low energy (1.6 and 8 kJ) portable electrically exploding foil accelerators are developed for moderately high pressure shock studies at small laboratory scale. Projectile velocities up to 4.0 km/s have been measured on Kapton flyers of thickness 125 microm and diameter 8 mm, using an in-house developed Fabry-Perot velocimeter. An asymmetric tilt of typically few milliradians has been measured in flyers using fiber optic technique. High pressure impact experiments have been carried out on tantalum, and aluminum targets up to pressures of 27 and 18 GPa, respectively. Peak particle velocities at the target-glass interface as measured by Fabry-Perot velocimeter have been found in good agreement with the reported equation of state data. A one-dimensional hydrodynamic code based on realistic models of equation of state and electrical resistivity has been developed to numerically simulate the flyer velocity profiles. The developed numerical scheme is validated against experimental and simulation data reported in literature on such systems. Numerically computed flyer velocity profiles and final flyer velocities have been found in close agreement with the previously reported experimental results with a significant improvement over reported magnetohydrodynamic simulations. Numerical modeling of low energy systems reported here predicts flyer velocity profiles higher than experimental values, indicating possibility of further improvement to achieve higher shock pressures.

Excessive loading of sediment and nutrients to rivers is a major problem in many parts of the United States. In this study, we tested the non-parametric Seasonal Kendall (SEAKEN) trend model and the parametric USGS Quality of Water trend program (QWTREND) to quantify trends in water quality of the Minnesota River at Fort Snelling from 1976 to 2003. Both methods indicated decreasing trends in flow-adjusted concentrations of total suspended solids (TSS), total phosphorus (TP), and orthophosphorus (OP) and a generally increasing trend in flow-adjusted nitrate plus nitrite-nitrogen (NO(3)-N) concentration. The SEAKEN results were strongly influenced by the length of the record as well as extreme years (dry or wet) earlier in the record. The QWTREND results, though influenced somewhat by the same factors, were more stable. The magnitudes of trends between the two methods were somewhat different and appeared to be associated with conceptual differences between the flow-adjustment processes used and with data processing methods. The decreasing trends in TSS, TP, and OP concentrations are likely related to conservation measures implemented in the basin. However, dilution effects from wet climate or additional tile drainage cannot be ruled out. The increasing trend in NO(3)-N concentrations was likely due to increased drainage in the basin. Since the Minnesota River is the main source of sediments to the Mississippi River, this study also addressed the rapid filling of Lake Pepin on the Mississippi River and found the likely cause to be increased flow due to recent wet climate in the region.

Water quality concerns, including greater potential for nutrient transport to surface waters resulting in eutrophication and nutrient leaching to ground water, exist when agricultural or food processing industry wastes and by-products are land applied. Plot- and field-scale studies were conducted to evaluate the effects of sugar beet by-products on NO3-N and P losses and biochemical oxygen demand (BOD) in runoff and NO3-N concentrations in percolating waters. In the runoff plot study, treatments in the first year included two rates (224 and 448 Mg ha(-1) fresh weight) of pulp and spoiled beets and a nonfertilized control. In the second year, no by-products were applied on the treated plots, the control treatment was fertilized with N fertilizer, and an additional treatment was added as a nonfertilized control in buffer areas. Wheat (Triticum aestivum L.) was grown in the year of by-product application and sugar beet (Beta vulgaris L.) in the following year. In the percolation field study, the treatments were the control, pulp (224 Mg ha(-)(1)), and spoiled beets (224 Mg ha(-1)). Results from the runoff plot showed that both by-products caused immobilization of soil inorganic N and thus reduced NO3-N losses in runoff and soil waters during the first growing season. There was some risk of NO3-N exceeding the drinking water limit of 10 mg L(-1), especially between the period of wheat harvest and soil freezing in fall when pulp was applied at 448 Mg ha(-1). The field-scale study showed that by-product application at 224 Mg ha(-1) did not result in increased ground water NO3-N concentrations. Application of spoiled beets at both rates caused significantly higher BODs in runoff in the first year of application. The concentrations of total and soluble reactive P (SRP) were also higher from both rates of spoiled beet application and from the higher application rate of pulp during the 2-yr study period. These high BODs and total P and SRP concentrations in runoff waters from land application of sugar beet by-product suggest that application rates should not be higher than 224 Mg ha(-1). Best management practices that prevent runoff from entering surface waters directly from these fields are warranted.

Land application of food processing wastes has become an acceptable practice because of the nutrient value of the wastes and potential cost savings in their disposal. Spoiled beets and pulp are among the main by-products generated by the sugar beet (Beta vulgaris L.) processing industry. Farmers commonly land apply these by-products at rates >224 Mg ha(-1) on a fresh weight basis. However, information on nutrient release in soils treated with these by-products and their subsequent impacts on crop yield is lacking. Field studies were conducted to determine the effects of sugar beet by-product application on N release and crop yields over two growing seasons. Treatments in the first year were two rates (224 and 448 Mg ha(-1) fresh weight) of pulp and spoiled beets and a nonfertilized control. In the second year after by-product application, the control treatment was fertilized with N fertilizer and an additional treatment was added as a nonfertilized control in buffer areas. Wheat (Triticum aestivum L.) was grown in the year of by-product application and sugar beet in the subsequent year. By-product treatments caused a significant reduction in wheat grain yield compared with the control. This was due to a decline in N availability as a result of immobilization. Based on microplots receiving 15N labeled beets, wheat took up <1% of spoiled beet-N (approximately 4.7 kg ha(-1)) during the year of by-product application. In the second cropping year, sugar beet root yields were significantly higher in the fertilized control and by-product treatments than the nonfertilized control. The lack of significant difference in sugar beet yield between the fertilized control and by-product treatments was likely due to the greater availability of N in the second year. Labeled 15N data also showed that the sugar beet crop recovered a 17% of sugar beet-N, an equivalent of 86 kg N ha(-1), during the second cropping year. There was no difference in sugar beet root yield, N uptake, or soil N mineralization during the sugar beet cropping season between the pulp and the spoiled beet treatments at comparable rates of application.

First principles total energy calculations on hcp, ω (a three atom simple hexagonal), ß (bcc) and fcc phases of osmium have been performed as a function of hydrostatic compression employing the FP-LAPW method. The comparison of total energies of these phases up to a maximum compression V/V(0) = 0.58 (pressure∼700 GPa) shows that the hcp structure remains stable up to this compression. The 300 K isotherm is determined after adding finite temperature thermal contributions to the total energy calculated as a function of volume at 0 K. From the theoretically determined isotherm, we have derived the shock Hugoniot of this metal and determined the shock parameters C(0) and s to be 4.48 km s(-1) and 1.32, respectively. Employing the theoretically calculated Gruneisen parameter in the differential form of the Lindemann melting rule, we have determined the variation of melting point of the osmium with pressure. The theoretically derived melting curve and the temperature rise along the Hugoniot predict the shock melting of osmium at ∼447 GPa with a corresponding temperature of ∼9203 K.

Manure management is a major concern in livestock production systems. Although historically the primary concerns have been nutrients and pathogens, manure is also a source of emerging contaminants, such as antibiotics, to the environment. There is a growing concern that antibiotics in manure are reaching surface and ground waters and contributing to the development and spread of antibiotic resistance in the environment. One such pathway is through leaching and runoff from manure stockpiles. In this study, we quantified chlortetracycline, monensin, and tylosin losses in runoff from beef manure stockpiles during two separate but consecutive experiments representing different weather conditions (i.e., temperature and precipitation amount and form). Concentrations of chlortetracycline, monensin, and tylosin in runoff were positively correlated with initial concentrations of antibiotics in manure. The highest concentrations of chlortetracycline, monensin, and tylosin in runoff were 210, 3175, and 2544 microg L(-1), respectively. Relative antibiotic losses were primarily a function of water losses. In the experiment that had higher runoff water losses, antibiotic losses ranged from 1.2 to 1.8% of total extractable antibiotics in manure. In the experiment with lower runoff water losses, antibiotic losses varied from 0.2 to 0.6% of the total extractable antibiotics in manure. Manure analysis over time suggests that in situ degradation is an important mechanism for antibiotic losses. Degradation losses during manure stockpiling may exceed cumulative losses from runoff events. Storing manure in protected (i.e., covered) facilities could reduce the risk of aquatic contamination associated with manure stockpiling and other outdoor manure management practices.

RESUMO

Providencia spp. are Gram negative bacteria, belonging to the family Enterobacteriaceae and are responsible for gastrointestinal disorders both in humans and animals. We report on the isolation of multi-drug resistant Providencia spp. (n=4) from animal manure. The isolates were found to be resistant to clindamycin and to tetracycline, as well as to the macrolide and sulphonamide groups of antibiotics.

Phosphorus-immobilizing amendments can be useful in minimizing P leaching from high P soils that may be irrigated with wastewater. This study tested the P-binding ability of various amendment materials in a laboratory incubation experiment and then tested the best amendment in a field setup using drainage lysimeters. The laboratory experiment involved incubating 100-g samples of soil (72 mg kg(-1) water-extractable phosphorus, WEP) with various amendments at different rates for 63 d at field moisture capacity and 25 degrees C. The amendments tested were alum [Al2SO4)3.14H2O], ferric chloride (FeCl3), calcium carbonate (CaCO3), water treatment residual (WTR), and sugarbeet lime (SBL). Ferric chloride and alum at rates of 1.5 and 3.9 g kg(-1), respectively, were the most effective amendments that decreased WEP to 20 mg kg(-1), below which leaching has previously been shown to be low. Alum (1.3 kg m(-2)), which is less sensitive to redox conditions, was subsequently tested under field conditions, where it reduced WEP concentration in the 0- to 0.15-m layer from 119 mg kg(-1) on Day 0 to 36.1 mg kg(-1) (85% decrease) on Day 41. Lysimeter breakthrough tests using tertiary-treated potato-processing wastewater (mean total phosphorus [TP] = 3.4 mg L(-1)) showed that alum application reduced leachate TP and soluble reactive phosphorus (SRP) concentrations by 27 and 25%, respectively. These results indicate that alum application may be an effective strategy to immobilize P in high P coarse-textured soils. The relatively smaller decreases in TP and SRP in the leachate compared to WEP suggest some of the P may be coming from depths below 0.2 m. Thus, to achieve higher P sequestration, deeper incorporation of the alum may be necessary.

Land application of wastewater has become an important disposal option for food-processing plants operating year-round. However, there are concerns about nutrient leaching from winter wastewater application on frozen soils. In this study, P and N leaching were compared between nongrowing season application of tertiary-treated wastewater plus growing season application of partially treated wastewater (NGS) vs. growing season application of partially treated wastewater (GS) containing high levels of soil P. As required by the Minnesota Pollution Control Agency (MPCA), the wastewater applied to the NGS fields during October through March was treated such that it contained < or =6 mg L(-1) total phosphorus (TP), < or =10 mg L(-1) NO3-N, and < or =20 mg L(-1) total Kjeldahl nitrogen (TKN). The only regulation for wastewater application during the growing season (April through September) was that cumulatively it did not exceed the agronomic N requirements of the crop in any sprayfield. Application of tertiary-treated wastewater during the nongrowing season plus partially treated wastewater during the growing season did not significantly increase NO3-N leaching compared with growing season application of nonregulated wastewater. However, median TP concentration in leachate was significantly higher from the NGS (3.56 mg L(-1)) than from the GS sprayfields (0.52 mg L(-1)) or nonirrigated sites (0.52 mg L(-1)). Median TP leaching loss was also significantly higher from the NGS sprayfields (57 kg ha(-1)) than from the GS (7.4 kg ha(-1)) or control sites (6.9 kg ha(-1)). This was mainly due to higher hydraulic loading from winter wastewater application and limited or no crop P uptake during winter. Results from this study indicate that winter application of even low P potato-processing wastewater to high P soils can accelerate P leaching. We conclude that the regulation of winter wastewater application on frozen soils should be based on wastewater P concentration and permissible loading. We also recommend that winter irrigation should take soil P saturation into consideration.

There is some concern that antibiotic residues in land-applied manure may promote the emergence of antibiotic resistant bacteria in the environment. The goal of this study was to determine whether or not soil bound antibiotics are still active against bacteria. The procedure involved sorbing various amounts of tetracycline or tylosin on two different textured soils (Webster clay loam [fine-loamy, mixed, superactive, mesic Typic Endoaquolls] and Hubbard loamy sand [sandy, mixed, frigid Entic Hapludolls]), incubating these soils with three different bacterial cultures (an antibiotic resistant strain of Salmonella sp. [Salmonella(R)], an antibiotic sensitive strain of Salmonella sp. [Salmonella(S)], and Escherichia coli ATCC 25922), and then enumerating the number of colony forming units relative to the control. Incubation was done under both static and dynamic conditions. Soil-adsorbed antibiotics were found to retain their antimicrobial properties since both antibiotics inhibited the growth of all three bacterial species. Averaged over all other factors, soil adsorbed antimicrobial activity was higher for Hubbard loamy sand than Webster clay loam, most likely due to higher affinity (higher clay content) of the Webster soil for antibiotics. Similarly, there was a greater decline in bacterial growth with tetracycline than tylsoin, likely due to greater amounts of soil-adsorbed tetracycline and also due to lower minimum inhibitory concentration of most bacteria for tetracycline than tylosin. The antimicrobial effect of tetracycline was also greater under dynamic than static growth conditions, possibly because agitation under dynamic growth conditions helped increase tetracycline desorption and/or increase contact between soil adsorbed tetracycline and bacteria. We conclude that even though antibiotics are tightly adsorbed by clay particles, they are still biologically active and may influence the selection of antibiotic resistant bacteria in the terrestrial environment.

Land application of wastewater presents potential for ground water pollution if not properly managed. In situ breakthrough tests were conducted using potato (Solanum tuberosum L.)-processing wastewater and a Br tracer to characterize P leaching in seasonally frozen sandy outwash soils. In the first test, P and Br breakthrough were measured in a 7-m deep well following wastewater [2.94 mg L(-1) total P (TP); 280 mg L(-1) Br] application at the site that had 13.1 mg water-extractable P (WEP) kg(-1)and 94.4 mg Bray-1 P kg(-1). Bromide was detected in the well after approximately 0.4 pore volumes, but there was no P break-through after 7 pore volumes. In the second breakthrough test, wastewater containing 3.6 mg L(-1) TP and 259 mg L(-1) Br was applied on 1.5-m deep lysimeters at low (0.8 mg WEP kg(-1); 12.1 mg Bray-1 P kg(-1)) and high soil test P sites (104 mg WEP kg(-1); 585 mg Bray-1 P kg(-1)). Leachate TP concentration during the test remained constant (0.04 mg L(-1)) at the low P sites but increased from approximately 3.5 to 5.6 mg L(-1) at the high P sites. These results indicate no P leaching in low P soils, but leaching in high P soils, thus suggesting that most of the P leached at the high P sites was mainly due to desorption and dissolution of weakly adsorbed P from prior P applications. This was consistent with P transport simulations using the convective-dispersive equation. We conclude that P concentration in land-applied wastewater should be regulated based on soil test-P level plus wastewater P loading.
