RESUMO

Glioma, a common malignant tumor of the central nervous system, has high invasiveness. The objective of the present study was to identify genes playing an important role in the development of glioma and to reveal their potential research value. Conjoint analysis on the GSE16011 dataset in the Gene Expression Omnibus (GEO) database and the 'Messenger RNA Expression Microarray of Diffuse Gliomas and Controls' dataset and 'RNA sequencing of Diffuse Gliomas' dataset in the Chinese Glioma Genome Atlas (CGGA) database is carried out in the study. The weighted correlation network analysis (WGCNA) was used to carry out co-expression analysis on the GSE16011. Finally, 24 genes significantly related to grade and prognosis were obtained. In addition, there is no report about CACNG2, JPH3, TUBB6 (tubulin ß 6 class V), NRSN1, FAM19A2, NALCN, CDH18, GNAL on glioma.

Arctigenin (ARC-G) is the main active ingredient extracted from Great Burdock Achene, with extensive pharmacological effects. In addition, ARC-G has been suggested to show excellent efficacy on inflammatory disease. This study aimed to defined that the function of Arctigenin attenuates inflammation in dextran sulfate sodium (DSS)-induced acute colitis, to determine its possible mechanism. Mice was induced by giving 2.0% DSS in the drinking water for DSS-induced acute colitis. Mice of acute colitis were injected intraperitoneally with 20 mg/kg per day of Arctigenin for 7 days. MPO activity levels were measured using MPO activity kits. Western Blot Analysis was used to determine the protein expression. Arctigenin prevents colitis and attenuates inflammation in DSS-induced acute colitis. Arctigenin suppressed NLRP3 inflammasome by SIRT1 in DSS-induced acute colitis. In THP-1 cell by LPS model, Arctigenin suppressed NLRP3, caspase-1 and IL-1ß protein expression by SIRT1. Si-NLRP3 increases the effects of Arctigenin on inflammation in THP-1 cell by LPS model. Si-SIRT1 decreases the effects of Arctigenin on inflammation in THP-1 cell by LPS model. INF39, NLRP3 inhibitor also increased the effects of Arctigenin on inflammation in DSS-induced acute colitis. SIRT1 inhibitor also decreases the effects of Arctigenin on inflammation in DSS-induced acute colitis. Taken together our results demonstrated that Arctigenin attenuates inflammation in DSS-induced acute colitis through suppression of NLRP3 inflammasome by SIRT1.

BACKGROUND: Wuqinxi is a traditional medicinal exercise that is widely practiced in China now. Because of its obvious medical rehabilitation, Wuqinxi has been used in the physical education for more than 1.2 million people in at least 24 Chinese Medicine university campuses in China for many years. This investigation aimed to evaluate whether Wuqinxi has the positive effect on physical improvements for female college students. METHODS: Infrared scanners were used for real-time monitoring of body calorie dynamics; the electromyography (EMG) was used to detect the iEMG on biceps, brachioradialis, quadriceps, and gastrocnemius; beside, the physical health elements, heart rate and cardiopulmonary function were also taken within the scope of our investigation and records. RESULTS: Wuqinxi exercise can improve the body function through making the abdominal muscles, back muscles and limbs strength exercise more effectively; Wuqinxi exercise had also made the athletes better control their muscles to have a good way to contraction and keeping balance; Moreover, the performances of speed of 800 m run, setting flexion, set-ups and grip strength had a comprehensive promotion for each of the participants including long-time practitioners and short-term practitioners. CONCLUSIONS: Therefore, the new gymnastics derived from ancient Chinese Wuqinxi exercise can improve the physical health of female college students so that it can be used as part of the development of higher education's health quality in the future.

Wogonoside, the main effective constituent of traditional Chinese medicine Scutellaria, belongs to the glucuronide family, with various functions, including detoxification, anti-inflammation and nourishing gallbladder, lowering blood pressure, diuresis and anti-allergic reactions. However, the effects of wogonoside on human colon cancer cells remain unclear. The present study aimed to investigate the anticancer effect of wogonoside on human colon cancer cells in vitro and its anticancer mechanisms. The results demonstrated that wogonoside significantly inhibited cell growth, induced apoptosis and mitochondrial-mediated autophagy of colon cancer cells. Furthermore, the results revealed that wogonoside significantly increased caspase-3 and caspase-9 expression levels, induced apoptosis regulator Bax/Bcl-2 and microtubule-associated protein 1A/1B-light chain 3 protein expression, suppressed the phosphatidylinositol 3 kinase (PI3K)/RAC-α serine/threonine-protein kinase (Akt)/mechanistic target of rapamycin (mTOR)/p70 S6 kinase (p70S6K) signaling pathway and induced p62 protein expression in colon cancer cells. In conclusion, these results demonstrated that wogonoside inhibits cell growth and induces mitochondrial mediated autophagy-related apoptosis in human colon cancer cells through modulation of the PI3K/Akt/mTOR/p70S6K signaling pathway.
