RESUMO

We examined the surface structure, binding conditions, electrochemical behavior, and thermal stability of self-assembled monolayers (SAMs) on Au(111) formed by N-(2-mercaptoethyl)heptanamide (MEHA) containing an amide group in an inner alkyl chain using scanning tunneling microscopy (STM), X-ray photoelectron spectroscopy (XPS), and cyclic voltammetry (CV) to understand the effects of an internal amide group as a function of deposition time. The STM study clearly showed that the structural transitions of MEHA SAMs on Au(111) occurred from the liquid phase to the formation of a closely packed and well-ordered ß-phase via a loosely packed α-phase as an intermediate phase, depending on the deposition time. XPS measurements showed that the relative peak intensities of chemisorbed sulfur against Au 4f for MEHA SAMs formed after deposition for 1 min, 10 min, and 1 h were calculated to be 0.0022, 0.0068, and 0.0070, respectively. Based on the STM and XPS results, it is expected that the formation of a well-ordered ß-phase is due to an increased adsorption of chemisorbed sulfur and the structural rearrangement of molecular backbones to maximize lateral interactions resulting from a longer deposition period of 1 h. CV measurements showed a significant difference in the electrochemical behavior of MEHA and decanethiol (DT) SAMs as a result of the presence of an internal amide group in the MEHA SAMs. Herein, we report the first high-resolution STM image of well-ordered MEHA SAMs on Au(111) with a (3 × 2√3) superlattice (ß-phase). We also found that amide-containing MEHA SAMs were thermally much more stable than DT SAMs due to the formation of internal hydrogen networks in MEHA SAMs. Our molecular-scale STM results provide new insight into the growth process, surface structure, and thermal stability of amide-containing alkanethiols on Au(111).

RESUMO

Background/Aims: The management of hepatic hydrothorax (HH) remains a challenging clinical scenario with suboptimal options. We investigated the effect and safety of pigtail catheter drainage compared to intermittent thoracentesis. Methods: This multicenter, retrospective study included 164 cirrhotic patients with recurrent pleural effusion from March 2012 to June 2017. Patients with neoplasms, cardiopulmonary disease, and infectious conditions were excluded. We compared the clinical outcomes of pigtail catheter drainage versus thoracentesis for variables including complications related to procedures, overall survival, and re-admission rates. Results: A total of 164 patients were divided into pigtail catheter (n = 115) and thoracentesis (n = 49) groups. During the follow-up period of 6.93 months after discharge, 98 patients died (pigtail; n = 47 vs. thoracentesis; n = 51). The overall survival (p = 0.61) and 30-day mortality (p = 0.77) rates were similar between the pigtail catheter and thoracentesis groups. Only MELD scores were associated with overall survival (adjusted HR, 1.08; p < 0.01) in patients with HH. Spontaneous pleurodesis occurred in 59 patients (51.3%) in the pigtail catheter group. Re-admission rates did not differ between the pigtail catheter and thoracentesis groups (13.2% vs 19.6% p = 0.7). A total of five complications occurred, including four total cases of bleeding (one patient in the pigtail catheter group and three in the thoracentesis group) and one case of empyema in the pigtail catheter group. Conclusions: Pigtail catheter drainage is not inferior to that of intermittent thoracentesis for the management of HH, proving it may be an effective and safe clinical option.

RESUMO

To probe the effects of deposition temperature on the formation and structural order of self-assembled monolayers (SAMs) on Au(111) prepared by vapor deposition of 2-(2-methoxyethoxy)ethanethiol (CH3O(CH2)2O(CH2)2SH, EG2) for 24 h, we examined the surface structure and electrochemical behavior of the resulting EG2 SAMs using scanning tunneling microscopy (STM) and cyclic voltammetry (CV). STM observations clearly revealed that EG2 SAMs vapor-deposited on Au(111) at 298 K were composed of a disordered phase on the entire Au surface, whereas those formed at 323 K showed improved structural order, showing a mixed phase of ordered and disordered phases. Moreover, at 348 K, uniform and highly ordered EG2 SAMs on Au(111) were formed with a (2 × 3√3) packing structure. CV measurements showed sharp reductive desorption (RD) peaks at -0.818, -0.861, and -0.880 V for EG2 SAM-modified Au electrodes formed at 298, 323, and 348 K, respectively. More negative potential shifts of RD peaks with increasing deposition temperature are attributed to an increase in van der Waals interactions between EG2 molecular backbones resulting from the improved structural quality of EG2 SAMs. Our results obtained herein provide new insights into the formation and thermally driven structural order of oligo(ethylene glycol)-terminated SAMs vapor-deposited on Au(111).

RESUMO

BACKGROUND: Multiple sclerosis is characterised by inflammation, oligodendrocyte loss and axonal demyelination and shows an additional impact on astrocytes, and their polarization. Although a certain degree of spontaneous myelin repair can be observed, disease progression, and aging impair regeneration efforts highlighting the need to better understand glial cell dynamics to establish specific regenerative treatments. METHODS: Applying a chronic demyelination model, we here analysed demyelination and remyelination related effects on astrocytes and stem cell niches and studied the consequences of medrysone application on myelin repair, and astrocyte polarization. FINDINGS: Medrysone induced recovery of mature oligodendrocytes, myelin expression and node formation. In addition, C3d/S100a10 co-expression in astrocytes was enhanced. Moreover, Timp1 expression in C3d positive astrocytes revealed another astrocytic phenotype with a myelination promoting character. INTERPRETATION: Based on these findings, specific astrocyte subpopulations are suggested to act in a myelin regenerative way and manner the regulation of which can be positively modulated by this corticosteroid. RESUMO

OBJECTIVE: Aquatic therapy is a significant intervention method for both patients and healthy individuals. However, in clinical practice, quantitative measurements are rarely applied in aquatic therapy due to the disadvantages of submerging expensive instruments in water. In this study, we used readily available smartphones and armbands to measure leg segments and joint angles during aquatic gait and evaluated the reliability of these measurements. METHODS: Waterproof smartphones were strapped to the trunk, thighs, and shanks of 19 healthy young adults using armbands. The angles of the trunk, thigh, and shank segments were measured during aquatic gait. The measurements were repeated 1 day later. The data were analyzed to obtain the angles of the hip and knee joints. RESULTS: Measurement repeatability, calculated using the intraclass correlation coefficient (ICC), was the highest for the shank segment range of motion (ROM) (first 46.79° ± 5.50°, second 50.12° ± 9.98°, ICC = 0.78). There was high agreement in trunk segment ROM (first 6.36° ± 1.42°, second 4.29° ± 1.83°, ICC = 0.73), thigh segment ROM (first 33.49° ± 5.18°, second 37.31° ± 8.70°, ICC = 0.62), and knee joint ROM (first 52.43° ± 11.26°, second 62.19° ± 16.65°, ICC = 0.68) and fair agreement in hip joint ROM (first 34.60°±4.71°, second 37.80° ± 7.84°, ICC = 0.59). CONCLUSIONS: Smartphones can be used to reliably measure leg segments and joint angles during aquatic gait, providing a simpler method for obtaining these measurements and enabling the wider use of aquatic motion analysis in clinical practice and research.

RESUMO

Laser-induced breakdown spectroscopy (LIBS) was explored as a method of monitoring oxygen (O) concentration in electrolytic reduction salt of pyroprocessing. Simulated salt samples were fabricated, and each sample was put in a transparent and sealed vial filled with argon gas. An neodymium-doped yttrium aluminum garnet (Nd:YAG) laser pulse was applied to the sample through the vial surface, and the optical emission spectrum was measured. O(I) 777.2 nm lines were clearly identified in the spectrum of a sample containing Li2O, and the intensity of the O peak and the intensity ratio of O and lithium (Li) peaks, in which Li was used as the normalization, increased linearly as the O concentration in the salt sample was increased. The limit of detection and root mean square error were calculated for the cases of O peak area, O peak height, peak area ratio of O-Li, and the peak height ratio of O-Li, and all the cases could indicate that the O concentration in the electrolytic reduction salt was out of normal range. Our result shows that LIBS has the possibility to be used as a method for monitoring of O in electrolytic reduction salt.

RESUMO

INTRODUCTION: Dynamic balance devices are costly and uncommon, requiring dedicated space for use. Thus, studies often utilize smartphones to measure dynamic balance by attaching them to body parts, as they are inexpensive, small, and readily available. However, little consideration has been given to determine which body part would yield the most reliable measurements. METHOD: ː A total of 25 healthy college students consented to stand on a balance pad on their dominant leg for 30 s. The test and retests were performed at a one-week interval. RESULTS: ː Agreement between the test and retest values was moderate for those obtained from the sternum and waist (0.50 ≤ ICC ≤ 0.74) and was good for those obtained from the shin (0.75 ≤ ICC ≤ 0.89). There were significant moderate positive correlations between the test and retest results for all the attachment sites (0.40 < r < 0.69, p < 0.05). DISCUSSION: Based on a review of related literature and the experiment's results, this study determined the pros and cons of using a smartphone as a measuring tool and the various measurement locations in the body. Despite the smartphone being a low-cost measuring tool, greater care must be taken to ensure the validity of the interpreted data. CONCLUSION: ː Although quantifying balance is necessary for evidence-based medicine, clinics seldom use existing modern devices due to their costliness. This study's results suggest that a smartphone, which is a cheaper alternative, produces the most reliable results when attached to the shin.

RESUMO

Accurate measurement of polyp size during colonoscopy is crucial. The usefulness of cap-assisted colonoscopy and external grid application on monitor (gCAP) was evaluated for polyp size measurement in this 3-year, single-center, single-blind, randomized trial. Using the endoscopic forceps width as reference, the discrepancy percent (DP), error rate (ER), and measurement time were compared between gCAP and visual estimation (VE) after randomization. ER was calculated within a 20% and 33% limit. From the 111 patients, 280 polyps were measured. The mean polyp sizes were 4.0 ± 1.7 mm and 4.2 ± 1.8 mm with gCAP and VE, respectively (p = 0.368). Compared with that by the forceps method, DP was significantly lower in the gCAP group than in the VE group. Moreover, ER was significantly lower in the gCAP group within its preset limit. The measurement time was 4 s longer in the gCAP group than in the VE group (8.2 ± 4.8 s vs. 4.2 ± 1.5 s; p < 0.001). However, the forceps method lasted 28 s longer than the others. On subgroup analysis by size, gCAP was more accurate for polyp size ≥ 5 mm. The gCAP method was more accurate for polyp size measurement than VE, especially for polyps ≥ 5 mm, and was more convenient than the forceps method.

RESUMO

Growth processes and electrochemical behaviors of 4-fluorobenzenethiol (4-FBT) self-assembled monolayers (SAMs) on Au(111) prepared by vapor deposition at 323 K were examined using scanning tunneling microscopy (STM) and cyclic voltammetry (CV). STM imaging revealed that 4-FBT SAMs at the initial growth stage (deposition for 1 min) were mainly composed of bright molecular aggregates and liquid-like disordered phase. After longer deposition for 3 min, 4-FBT SAMs had three distinct surface features: a few molecular aggregates, small ordered domains, and disordered phase. These small ordered domains with sizes ranging from 5 to 10 nm had a (4× âˆš3)R30° packing structure. As deposition time increased to 24 h, long-range ordered domains larger than 40 nm were formed on Au(111) surfaces. From this STM study, we demonstrate that phase transitions of 4-FBT SAMs on Au(111) occur from molecular aggregates to large ordered domains via formation of small ordered domains as deposition time increases. CV measurements showed reductive desorption peaks for 4-FBT SAMs in the range of -638~-648 mV regardless of SAM morphology, suggesting that S-Au binding strength of 4-FBT SAMs on Au electrodes is a dominant factor for electrochemical stability.

RESUMO

The metabotropic glutamate receptor 7 (mGlu7) is a class C G protein-coupled receptor that modulates excitatory neurotransmitter release at the presynaptic active zone. Although post-translational modification of cellular proteins with ubiquitin is a key molecular mechanism governing protein degradation and function, mGlu7 ubiquitination and its functional consequences have not been elucidated yet. Here, we report that Nedd4 ubiquitin E3 ligase and ß-arrestins regulate ubiquitination of mGlu7 in heterologous cells and rat neurons. Upon agonist stimulation, ß-arrestins recruit Nedd4 to mGlu7 and facilitate Nedd4-mediated ubiquitination of mGlu7. Nedd4 and ß-arrestins regulate constitutive and agonist-induced endocytosis of mGlu7 and are required for mGlu7-dependent MAPK signaling in neurons. In addition, Nedd4-mediated ubiquitination results in the degradation of mGlu7 by both the ubiquitin-proteasome system and the lysosomal degradation pathway. These findings provide a model in which Nedd4 and ß-arrestin act together as a complex to regulate mGlu7 surface expression and function at presynaptic terminals.

RESUMO

Displacement processes of pre-adsorbed 1-admanetanethiol (ADT) self-assembled monolayers (SAMs) on Au(111) by 1-hexanethiol (HT) at room temperature were investigated by scanning tunneling microscopy (STM) and cyclic voltammetry (CV). Molecular-scale STM imaging clearly revealed that phase transitions from the (7 × 7) phase for ADT SAMs to the c(4 × 2) phase for HT SAMs via intermediate phase including bright aggregated islands and disordered phase. Moreover, it was found that ADT SAMs were completely displaced by HT molecules with a short hexyl chain within an hour. The CV measurements showed that cathodic peaks for SAM-modified Au(111) electrodes as a function of displacement time were varied with the structural change of displaced SAMs. Our results provide new insight into understanding the displacement processes of ADT SAMs on Au(111) by HT.

RESUMO

Solvent effect on the formation and electrochemical behavior of octaneselenolate (C8Se) self-assembled monolayers (SAMs) on Au(111) derived from the adsorption of octaneselenocyanate (C8Se-CN) molecules in various solvents at 363 K for 1 h was examined by scanning tunneling microscopy (STM) and cyclic voltammetry (CV). STM imaging clearly revealed that the formation and structure of C8Se SAMs were markedly influenced by the polarity of solvent. C8Se SAMs formed in octane were composed of liquid-like disordered phase. In contrast, C8Se SAMs formed in DMF had ordered domains but not a uniform surface, whereas C8Se SAMs formed in ethanol had uniform surface and highly ordered domains with a (2 × 2 √13) structure. It was found that the structural quality of C8Se SAMs increased with increasing solvent polarity in the order of ethanol > DMF > octane. CV measurements also showed that blocking efficiency of C8Se SAMs for electrode reaction increased with increasing the structural quality of SAMs.

RESUMO

Displacement processes of pre-covered cyclohexanethiol (CHT) self-assembled monolayers (SAMs) by 4-fluorobenzenethiol (4-FBT) on Au(111) were examined as a function of displacement time by scanning tunneling microscopy (STM) and water static contact angle (CA) measurements. STM imaging revealed that the adsorption of 4-FBT on Au(111) in a 1 mM ethanol solution at room temperature for 24 h generated disordered SAMs, whereas well-ordered 4-FBT SAMs with a (4√6 × âˆš3)R5° packing structure were formed over the entire Au(111) surfaces via the displacement of pre-covered CHT SAMs by 4-FBT molecules. The CA measurements also showed that CA values increase with increasing displacement time, reflecting that the displacement reaction took place and the resulting SAMs had greater hydrophobicity compared with CHT SAMs. In this study, we found that the displacement technique using CHT SAMs as a molecular template is very useful in obtaining 4-FBT SAMs with a high degree of structural order and large ordered domains.

RESUMO

OBJECTIVES: This study evaluated and validated the reliability of smartphones as measuring equipment for the dorsi-plantar flexion of ankle joint position sense (JPS) ability. METHODS: The subjects were 20 healthy young students in their 20s. We confirmed the concurrent validity by comparison with existing electrogoniometer data. The reliability of the smartphone was confirmed using the test-retest method. RESULTS: In the case of dorsiflexion, there was no significant difference between the smartphone and electrogoniometer groups (p > 0.05). Regarding the correlation, it was significantly high (r = 0.65, p < 0.05), and ICC(3,1) was good (ICC(3,1) = 0.79). For the case of plantar flexion, there was no significant difference between the smartphone and electrogoniometer groups (p > 0.05), the correlation was significantly high (r = 0.69, p < 0.05), and the ICC(3,1) was very good (ICC(3,1) = 0.82). In the case of dorsiflexion, there was no significant difference between test and retest (p > 0.05), the correlation was intermediate (r = 0.59, p < 0.05), and the ICC(3,1) value was good (ICC(3,1) = 0.74). For plantar flexion, there was no significant difference between test and retest (p > 0.05), the correlation was significantly high (r = 0.63, p < 0.05), and the ICC(3,1) was good (ICC(3,1) = 0.76). CONCLUSIONS: The results showed that smartphones provide high validity and reliability as measurement equipment for JPS of dorsi-plantar flexion of the ankle. Finally, the study also considers that smartphone-based JPS measuring methods may replace the traditional and expensive methods that are currently being used for the same purpose.

RESUMO

[Purpose] The purpose of this study was to investigate the perceived treatment times and emotional reactions under different light colors in the treatment room. [Subjects and Methods] Subjects in this study were 20 healthy young students in their 20s. Under each lighting condition (blue, red, white, and yellow) differentiated by color, each subject laid on a therapeutic bed and underwent ultrasound therapy. Subjects were instructed to press a stopwatch every 1 minute, for a total of 5 times, after therapy started according to their perception of time while the stopwatch's time indicator was blocked. After the experiments, self-administered questionnaires were given to subjects to measure their emotional reactions. [Results] In terms of K-POMS scores, the mood states of depression-dejection, anger-hostility, and confusion-bewilderment were higher scores for blue and red lights compared to yellow light. The mood state of vigor-activity were higher scores for yellow and white lights compared to blue and red lights. [Conclusion] Therefore, it is important to take necessary measures to prevent the negative effects that blue and red light-based therapy can have on patient mood.

RESUMO

[Purpose] This study's purpose was to identify abduction angles of the shoulder joint that can provide effective infraspinatus muscle exercises while minimizing the muscle activity of the posterior deltoid muscle. [Subjects and Methods] The subjects of this study were 15 healthy young students in their 20s. Subjects adopted a standing position and performed shoulder external abduction while pushing their hands against the wall. The current research was undertaken to compare the activity of the infraspinatus muscle and the posterior deltoid muscle at abduction angles of 45, 90 and 135° of the shoulder joint during closed kinetic chain exercises. [Results] A activity of the infraspinatus muscle showed no statistically significant differences. The activities of posterior deltoid muscle were greater at the angle of 45° than at 90 and 135°. [Conclusion] These results indicate that to strengthen the infraspinatus, shoulder external rotation exercises at abduction angles of the shoulder joint greater than 45° are more effective.

RESUMO

[Purpose] Evaluation of the reliability of smartphones as measuring equipment for dynamic balance ability was the goal of this study. [Subjects and Methods] Subjects were 30 healthy young students in their 20s. The first and second rounds of measurements were taken at a one-day interval to confirm test-retest reliability. The subjects stood on the footboard of the Biodex Balance System. Balance was measured using a smart phone. [Results] Acceleration rates corresponding to subjects with open eyes were 2.7 ± 2.2 (first measurement) and 3.3 ± 1.5 (second measurement), and the interclass correlation coefficient ICC (1,1) was 0.8. Acceleration rates corresponding to subjects with closed eyes were 4.1 ± 2.4 (first measurement) and 4.5 ± 1.8 (second measurement), and the ICC (1,1) was 0.9. Gyroscope rates corresponding to subjects with open eyes were 1.7 ± 1.2 (first measurement) and 2.3 ± 1.5 (second measurement), and the ICC (1,1) was 0.7. Gyroscope rates corresponding to subjects with closed eyes were 6.7 ± 2.4 (first measurement) and 6.6 ± 2.3 (second measurement), and the ICC (1,1) was 0.6. [Conclusion] The results of this study suggest that smartphones have sufficient potential as measuring equipment for dynamic balance ability.

RESUMO

[Purpose] This study was intended to examine the most effective feedback method for contracting the musculus transversus abdominis muscle by using basic training, a pressure biofeedback unit, and real-time ultrasonographic imaging during abdominal hollowing exercise training. [Subjects and Methods] The subjects in this study were 30 healthy young students in their twenties. Thirty subjects were divided randomly and equally into the basic training, the pressure biofeedback unit, and real-time ultrasonographic imaging groups. All of the subjects received abdominal hollowing exercise training for 15 minutes. The subjects in the pressure biofeedback unit group were trained by using a pressure biofeedback unit. Those in the real-time ultrasonographic imaging group received training with monitoring of possible contraction of their musculus transversus abdominis muscles on ultrasonography. [Results] In all the three groups, the musculus transversus abdominis became significantly thicker, but more significantly in the real-time ultrasonographic imaging group than in the basic training group. [Conclusion] The feedback method using real-time ultrasonographic imaging may be more effective in thickening the musculus transversus abdominis muscle during exercise than the traditional feedback method with manual contact only. However, it is insufficient in terms of overall qualitative improvement of exercise outcome.

RESUMO

[Purpose] The purpose of this study was to examine the effects of ankle exercises on balance ability when using shoe height insoles. [Subjects and Methods] Thirty adults in their 20s, who were students, were randomly divided into an experimental group and a control group (n=15/group). Individuals in the experimental group wore height-adjustable silicon insoles measuring 3, 5, and 7 cm three times per week for four weeks. The ankle exercise program consisted of stretching exercises, resistance training, and a balance exercise. [Results] There was a significant improvement in the balance ability of the experimental group for all height insoles (3, 5, and 7 cm) following the exercise program, as compared with before the exercise program. There was no significant improvement in the balance ability of the control group for all height insoles following the exercise program as compared with before the exercise program. The experimental group exhibited a significant improvement in balance ability following the exercise program for all the height insoles as compared with the control group. [Conclusion] According to the results of this study, active ankle exercises are helpful in preventing ankle joint damage, as they improve balance ability.

RESUMO

The deposit of polyubiquitinated aggregates has been implicated in the pathophysiology of Parkinson's disease (PD), and growing evidence indicates that selective autophagy plays a critical role in the clearance of ubiquitin-positive protein aggregates by autophagosomes. The selective autophagic receptor p62/SQSTM-1, which associates directly with both ubiquitin and LC3, transports ubiquitin conjugates to autophagosomes for degradation. Leucine-rich repeat kinase 2 (LRRK2), a PD-associated protein kinase, is tightly controlled by autophagy-lysosome degradation as well as by the ubiquitin-proteasome pathway. However, little is known about the degradation of ubiquitinated LRRK2 via selective autophagy. In the present study, we found that p62/SQSTM-1 physically interacts with LRRK2 as a selective autophagic receptor. The overexpression of p62 leads to the robust degradation of LRRK2 through the autophagy-lysosome pathway. In addition, LRRK2 indirectly regulates Ser351 and Ser403 phosphorylation of p62. Of particular interest, the interaction between phosphorylated p62 and Keap1 is reduced by LRRK2 overexpression. Therefore, we propose that the interplay between LRRK2 and p62 may contribute to the pathophysiological function and homeostasis of LRRK2 protein.
