RESUMO

Patellofemoral pain syndrome (PFPS) is a common injury among runners, and it is thought that abnormal lower extremity biomechanics contribute to its development. However, the relationship between biomechanical changes after a marathon and PFPS injury remains limited. This study aims to investigate whether differences in knee and hip kinematics and lower extremity muscle activities exist in recreational runners before and after a marathon. Additionally, it aims to explore the relationship between these biomechanical changes and the development of PFPS injury. 12 recreational runners participated in the study. Kinematics and muscle activities of the lower extremity were recorded during walking (5 km/h) and running (10 km/h) tasks within 24 hours before and within 5 hours after a marathon. After the marathon, there was a significant decrease in peak knee flexion (walking: p = 0.006; running: p = 0.006) and an increase in peak hip internal rotation (walking: p = 0.026; running: p = 0.015) during the stance phase of both walking and running compared to before the marathon. The study demonstrates a decrease in knee flexion and an increase in hip internal rotation during the stance phase of gait tasks after completing a marathon, which may increase the risk of developing PFPS injury.

Suppressor of cytokine signalling (SOCS) 1/2/3/4 are involved in the occurrence and progression of multiple malignancies; however, their prognostic and developmental value in patients with glioblastoma (GBM) remains unclear. The present study used TCGA, ONCOMINE, SangerBox3.0, UALCAN, TIMER2.0, GENEMANIA, TISDB, The Human Protein Atlas (HPA) and other databases to analyse the expression profile, clinical value and prognosis of SOCS1/2/3/4 in GBM, and to explore the potential development mechanism of action of SOCS1/2/3/4 in GBM. The majority of analyses showed that SOCS1/2/3/4 transcription and translation levels in GBM tissues were significantly higher than those in normal tissues. qRT-PCR, western blotting (WB) and immunohistochemical staining were used to verify that SOCS3 was expressed at higher mRNA and protein levels in GBM than in normal tissues or cells. High SOCS1/2/3/4 mRNA expression was associated with poor prognosis in patients with GBM, especially SOCS3. SOCS1/2/3/4 were highly contraindicated, which had few mutations, and were not associated with clinical prognosis. Furthermore, SOCS1/2/3/4 were associated with the infiltration of specific immune cell types. In addition, SOCS3 may affect the prognosis of patients with GBM through JAK/STAT signalling pathway. Analysis of the GBM-specific protein interaction (PPI) network showed that SOCS1/2/3/4 were involved in multiple potential carcinogenic mechanisms of GBM. In addition, colony formation, Transwell, wound healing and western blotting assays revealed that inhibition of SOCS3 decreased the proliferation, migration and invasion of GBM cells. In conclusion, the present study elucidated the expression profile and prognostic value of SOCS1/2/3/4 in GBM, which may provide potential prognostic biomarkers and therapeutic targets for GBM, especially SOCS3.

Fluoride (F-) is a common trace component in groundwater, and long-term exposure to high-F- groundwater is harmful to human health. Fluoride concentrations that exceed the World Health Organization guideline for drinking water (1.5 mg/L) have been detected in thermal and non-thermal groundwater from Southeast China, where the causes of the high fluoride occurrence are lack of study. To study the formation and migration mechanism of high fluoride groundwater from Southeast China, we carried out a systemic sampling of water samples in the surrounding area of Heyuan deep fault zone and Zijin-Boluo fault zone, then a comprehensive discussion including water hydrogeochemical, stable isotope composition, as well as hydrogeology analysis was conducted. Fluoride concentrations in geothermal and non-thermal water samples range from 1.11 to 22.76 mg/L and 0.04-8.3 mg/L, respectively. High temperature, alkaline conditions, and the depleted Ca2+ by reverse cation exchange and calcite precipitation would promote the release of fluoride from host rock to geothermal water. The availability of Ca-bearing and F-bearing minerals in host rock causes significant differences in fluoride concentrations between carbonate reservoir and granite reservoir. Hydrochemical diagrams reveal that the composition of groundwater is affected by mixing and that fractures act as the mixing channels in our study area. The addition of cold groundwater limits the fluoride concentrations by lowing temperature and increasing Ca2+ levels of geothermal water. Additionally, the relationship between F- and SiO2 indicates that geothermal water promotes the fluoride enrichment in cold groundwater, especially in confined aquifers which are more susceptible to geothermal water. The direct input of geothermal fluoride and secondary enrichment caused by alkaline condition contribute to the formation of high F- concentrations (7.2-8.3 mg/L) in confined groundwater. Our findings highlight that the natural evolution of geothermal systems in fault zone can result in the formation of geogenic contaminated groundwater.

BACKGROUND: Measurement of knee laxity after anterior cruciate ligament (ACL) injury is crucial for appropriate treatment and rehabilitation decision-making. This study examined the potential of a new digital arthrometer (Ligs, Innomotion, Shanghai, China) to quantify anterior tibial translation (ATT) in patients with ACL injuries and in healthy subjects. METHODS: A total of 60 participants included 30 subjects with single-leg ACL injuries and 30 healthy subjects included as controls. The lower leg was immobilized. The thruster is positioned posterior to the lower leg and parallel to the tibial tuberosity in the sagittal plane. The load is applied vertically to the tibia under a dynamic load of 0-150 N, with continuous displacement recorded. The intrarater and interrater reliability will be examined. ATT and side-to-side differences (SSD) between the control and ACL injury groups were compared. Receiver operating characteristic (ROC) curves were analyzed, and the area under the curve (AUC) was calculated to determine the diagnostic accuracy of the Ligs. RESULTS: The interrater ICC was 0.909 and the intrarater ICC was 0.943. Significant differences in the SSD were observed between the control and ACL injury groups (for all P < 0.05), with the largest effect size (ES = 1.12) at 80 N. When comparing ATT at different loads between injured and healthy sides in the ACL injury group, displacement was statistically significant at different loads. At a load of 150 N, the AUC was the maximum (0.857) and the sensitivity and specificity were 0.87 and 0.73, respectively. CONCLUSIONS: A digital arthrometer can be used as a quantitative instrument to quantify knee laxity. Quantitative measurement of ATT and SSD under controlled loading can be an objective and effective tool for clinical practice.

Machine vision is being employed in defect detection, size measurement, pattern recognition, image fusion, target tracking and 3D reconstruction. Traditional cancer detection methods are dominated by manual detection, which wastes time and manpower, and heavily relies on the pathologists' skill and work experience. Therefore, these manual detection approaches are not convenient for the inheritance of domain knowledge, and are not suitable for the rapid development of medical care in the future. The emergence of machine vision can iteratively update and learn the domain knowledge of cancer cell pathology detection to achieve automated, high-precision, and consistent detection. Consequently, this paper reviews the use of machine vision to detect cancer cells in histopathology images, as well as the benefits and drawbacks of various detection approaches. First, we review the application of image preprocessing and image segmentation in histopathology for the detection of cancer cells, and compare the benefits and drawbacks of different algorithms. Secondly, for the characteristics of histopathological cancer cell images, the research progress of shape, color and texture features and other methods is mainly reviewed. Furthermore, for the classification methods of histopathological cancer cell images, the benefits and drawbacks of traditional machine vision approaches and deep learning methods are compared and analyzed. Finally, the above research is discussed and forecasted, with the expected future development tendency serving as a guide for future research.

INTRODUCTION: Biomimic electrospun matrix derived from silk fibroin nanofiber solution was recently prepared in our group. The feasibility of the matrix as mucosa repair scaffold was evaluated in a rat model in the present study. METHODS: Full thickness wound was established on the buccal mucosa of male Wister rats via microscopic oral surgery. 80 rats were randomly assigned into 4 groups: (1) silk fibroin matrix, (2) commercial cowhide acellular dermal matrix (Heal-All), (3) commercial acellular dermal matrix of human skin (RENOV), and (4) vaseline gauze, respectively. RESULTS: The silk fibroin matrix showed similar repair performance compared to the commercial acellular dermal matrices, implying promising applications in mucosa regeneration. More importantly, the silk fibroin matrix showed better wound healing ability, improved wound shrinkage inhibition, and reduced local immunological incompatibility. CONCLUSIONS: The silk fibroin scaffold performed satisfied in scar tissue inhibition and epidermis regeneration. Taking into account its improved mechanical properties, the biomimic electrospun silk matrix could become a promising substitute of acellular dermal matrix in clinical applications.
