RESUMO

Despite of all the developments in DNA microarray technology, there is not sufficient knowledge about protein abundance or their function in processes such as proteolysis, phosphorylation. Therefore, there is a significant need for direct detection and quantification of proteins, especially in processes such as proteomics, drug design and disease prediction. The present work introduce the new generation of polymeric substrate based on polyaniline and, polylactic acid, which it was used for impedimetric sensor in detection of proteins in particular for bovine serum albumin (BSA). In this copolymerization, the polylactic acid-block-polyaniline copolymer (PLA-b-PANI) was synthesized to attach polylactic acid and polyaniline using epichlorohydrin as a coupling agent. The structure of synthesized compounds in all steps, were confirmed by FT-IR and, 1H-NMR. The thermal properties and, morphology were analyzed by DSC, TGA, and, SEM. Also the electrochemical characteristics of fabricated PLA-b-PANI electrode were investigated by Electrochemical Impedance Spectroscopy (EIS) and Cyclic Voltammetry (CV). The results demonstrated that morphology of the PLA-b-PANI is sphere shape nanoparticles with dimension less than 100 nanometer diameters and, reasonable thermal properties. PLA-b-PANI was used to modify a screen-printed carbon electrode (SPCE) to fabricate a BSA impedimetric sensor. In order to increase the performance of the proposed impedimetric sensor, optimization of incubation time, pH and amount of PLA-b-PANI were investigated. The results show that the impedimetric sensor has the highest response when the electrode surface is covered with 5 microliters of PLA-b-PANI, and is incubated in BSA solution with pH 6.5 for 5 min. Impedimetric results showed that the PLA-b-PANI has excellent properties in reducing the charge transfer resistance and increasing the electron charge transfer rate. The final impedimetric sensor exhibited good repeatability, reproducibility, and chemical stability within the linear concentration range of 0.1-20 µg L-1 of BSA, and a detection limit of 0.05 µg L-1.

It is highly desired and yet challenging to develop eco-friendly cation exchange membranes with a combination of good mechanical, electrochemical, and biocompatible properties with a rational economic efficiency for given applications. In this study, new biocompatible double network (DN) hydrogels were prepared based on a blend of modified K-Carrageenan (KC) and polyvinyl alcohol (PVA). Acrylic acid (AA)-grafted KC (KC-g-(PAA)) and (AA-co-tertbutyl acrylate (TBA))-grafted KC (KC-g-P(AA-co-TBA)) were synthesized through an in situ free radical copolymerization. The grafted copolymers were blended with PVA and mixed with ZrOCl2/KCl and glutaraldehyde (Glu) as the physical and chemical cross-linkers, respectively to produce KC-g-P(AA)/PVA and KC-g-P(AA-co-TBA)/PVA DN hydrogels. The membranes were prepared by a solution casting method. Various techniques were carried out to compare the structural, thermal, mechanical, flammability, and electrochemical properties of the membranes with those of the cross-linked KC, PVA, and KC/PVA membranes. The KC-g-P(AA-co-TBA)/PVA DN membrane showed more desirable properties as the cation exchange membrane with water uptake of 70.7 %, ion exchange capacity of 0.47 meq H+ /g, the ionic conductivity of 1.99 × 10-2 S/cm2, and elongation at break of 71.8 %. The prepared biopolymer membrane is very cost-effective and self-extinguished with admissible conductivity for electrochemical applications.

The autism is an abnormality in the neuronal advance which starts before age 3 recognized by defective behaviors. This study aimed to make quercetin-loaded nanophytosomes (QNP) on behavioral deficits, cerebellar oxidative stress and apoptosis in an autistic-like model caused by maternal separation (MS). The newborn rats are randomly categorized into seven groups, including control, positive control, disease, and diseases treated with quercetin (10 and 40 mg/kg) and QNP (10 and 40 mg/kg). Pups exposed to MS for 3 h per day from postnatal days (PND) 1-9 showed behavioral impairment in adult rats compared to control group. The oral administration of quercetin and QNP was constantly started after the lactation period (21 postnatal days) for three weeks. Autistic-like behaviors, antioxidant parameters, and Nrf2, Bax/Bcl-2, and Caspase-3 expressions were surveyed in the cerebellum. Quercetin (40 mg/kg) treated improved some behavioral disorders. Also, the improvement of oxidative stress parameters, Nrf2 and apoptotic factors gene expression was observed in the cerebellum of quercetin (40 mg/kg) treated (p < 0.01). QNP treatment (10 and 40 mg/kg) significantly ameliorated anxiety-like behaviors, line crossing, and grooming index (p < 0.001), lipid peroxidation (p < 0.001), and increased catalase (CAT) (p < 0.001), superoxide dismutase (SOD) (p < 0.001), glutathione peroxidase (GPx) (p < 0.001) activity, and glutathione (GSH) levels (p < 0.05). Moreover, QNP significantly reduced Caspase-3 and Bax expression (p < 0.001), but increased Bcl-2, and Nrf2 expressions (p < 0.001). These findings indicated that QNP due to its high bioavailability was more effective than quercetin can be reduced autistic-like behavior, oxidative and apoptotic damages in the model of MS rats.

This study reports the successful synthesis of a ZnS@GO@Pani polymeric nanocomposite (NC) via chemical polymerization. The product was used for simultaneous photocatalytic degradation-adsorption of malachite green (MG), a carcinogenic and widely used dye. The physicochemical properties of the prepared NC were characterized by various techniques. Morphological and XRD results confirmed the fine size of ZnS nanoparticles (NPs) with an approximate mean size of 5 nm, uniformly distributed within the polymeric matrix. For comparative purposes, photocatalytic dye degradation-adsorption of this nanohybrid was explored both in the dark and under natural light. It was observed that 0.1 g of the ternary NC in MG aqueous solution (20 ppm) leads to dye adsorption within 15 minutes with an efficiency of 70% under dark conditions. Also, MG removal efficiency of up to 90% was achieved in 15 minutes under natural light owing to integrated photocatalytic degradation-adsorption mechanisms. Adsorption isotherm studies were performed considering Langmuir, Freundlich, Temkin, and Dubinin-Radushkevich (D-R) models. The results showed that the Freundlich isotherm with R 2 = 0.988 is well consistent with the experimental data. Integrated photocatalytic degradation-adsorption kinetics were modeled with pseudo-first-order (PFO) and pseudo-second-order (PSO) models where PSO with R 2 = 0.999 best fitted the data, implying the predominant role of chemical adsorption in the dye removal process. Antibacterial tests revealed superior antibacterial activity of the prepared ZnS@GO@Pani NC against both Gram-negative and Gram-positive bacteria, demonstrating the remarkable synergistic effect of ZnS NPs embedded in the GO@Pani matrix. Accordingly, the prepared NC could be regarded as a promising candidate for wastewater treatment applications. The leaching and regeneration studies also confirmed that the prepared NC is a non-toxic dye removal agent with good reusability.

As a serious neurodegenerative disorder, the prevalence of Parkinson's disease is predicted to dramatically increase in the coming decades. Despite the development of numerous drugs for its treatment, oral administration of levodopa has remained the simplest and most effective pharmacological approach in the management of Parkinson's disease. In this research, the levodopa-imprinted hydrogel was synthesized by reverse emulsion polymerization in the presence of levodopa followed by modification with polypyrrole. The antioxidant activity of amphiphilic non-levodopa-imprinted hydrogel was studied by 2,2-Diphenyl-1-picrylhydrazyl active radicals, which indicated 100% efficiency in the applied amount. Amphiphilic non-levodopa-imprinted hydrogel cytotoxicity was evaluated by MTT assay, which confirmed no significant toxicity after 24 and 48 h even at high concentrations. Moreover, in vitro releasing property of the levodopa-imprinted hydrogel was studied in the pH range of 4 to 7.4, which reached 60 and 80% within 160 h, respectively.

The present study aimed to develop novel hesperetin-loaded on multiple wall carbon nanotubes (Hst-MWCNTs) to resolve the restricted bioavailability of hesperetin (Hst) and to enhance its preventive effect on cerebral ischemia-reperfusion (I/R). The physicochemical characteristics of Hst-MWCNTs were evaluated by Fourier-transform infrared spectra (FT-IR) and field emission scanning electron microscopy (FE-SEM). Forty male Wistar rats were randomly divided into five groups (control, I/R, MWCNTs, Hst, and Hst-MWCNTs). Hst, MWCNTs and Hst-MWCNTs (15 mg/kg orally) were pretreated for 14 days, and then I/R was induced by bilateral common carotid artery occlusion (BCCAO). Learning and memory deficits were evaluated using the novel object recognition test (NORT). The percentage of infarct size, catalase (CAT), superoxide dismutase (SOD), glutathione reductase (GRx), glutathione peroxidase (GPx) activities, malondialdehyde (MDA), and glutathione (GSH) levels was evaluated. Caspase-3 and Bcl-2 expressions were detected by qRT-PCR and Western blot analysis. Compared to the I/R group, Hst-MWCNTs considerably reduced learning and memory deficits, infarct size, and MDA levels. CAT, SOD, GRx, GPx activities and GSH levels were significantly increased in the Hst-MWCNTs group than in the I/R group. Additionally, Hst-MWCNTs significantly reduced the Caspase-3 expression but increased the Bcl-2 expression. All these results indicated that MWCNTs could be used as a promising novel carrier to enhance the oral bioavailability of Hst and to treat cerebral I/R injury.

Certainly water pollution control will play a key role in environmental health. Therefore, researchers are attempting to reduce the ecological effects of water pollution by creating restrictions in the use of chemicals or water reuse. Among all the available techniques, adsorption method attracted much attention due to the higher efficiency, cost-effectiveness and simplicity. So, in this work, novel biodegradable hydrogel based on carboxymethyl cellulose (CMC) and polyaniline (PANI) was introduced to remove toxic dyes from wastewater. The synthesis process was accomplished in two steps: free radical polymerization of acrylic acid (AA) on the CMC (0.25 g) in the presence of ammonium per sulfate (APS) as radical initiator (0.2 g) and N,N-methylene-bis-acrylamide (MBA) as crosslinker (0.1 g) at 70 °C, and after 30 min, growing PANI chains on the synthesized CMC-PAA hydrogel by radical polymerization of aniline (1.0 mL) in acidic condition (20 mL of hydrochloric acid 1.0 M) to form macro-porous conductive hydrogel (CMC-PAA-PANI). The resulted hydrogel showed high antibacterial activity and excellent biodegradability by natural soil microorganisms with decomposition to 91.7 %. Also, the final hydrogel exhibited reasonable conductivity and pH sensitivity properties.

Chitosan-based polymeric nanocarrier has been utilized as a novel drug delivery device in recent years due to its prominent role in the treatment of central nervous system disorders. The aim of this study was to investigate the anti-oxidative and anti-inflammatory effects of silymarin-loaded chitosan nanoparticles (SM-CS-NPs) on rat model of global cerebral ischemia/reperfusion (I/R). All rats were randomly distributed into four groups: Control, I/R, SM and SM-CS-NPs. Oral administration of SM and SM-CS-NPs was started 14 days prior to bilateral common carotid artery occlusion (BCCAO). Depressive-like behaviors, infarct volume, some oxidative stress markers and inflammatory factors were assessed after induction of I/R. SM-CS-NPs pretreatment significantly ameliorated depressive-like behaviors and infarct volume after I/R. SM-CS-NPs also significantly decreased the levels of malondialdehyde (MDA), and expression of interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α), and significantly increased the activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GRx), and glutathione (GSH) levels in I/R brain. The current study demonstrated that SM-CS-NPs pretreatment effectively prevents oxidative and inflammatory damage in the brain caused by I/R, and it can be considered as a useful pretreatment to attenuate the negative effects of I/R.

RESUMO

The survey on the characteristic data presented here, are related to the study entitled "Transparent chitosan based nanobiocomposite hydrogel: Synthesis, thermophysical characterization, cell adhesion and viability assay" [1]. Scanning electron microscopy images, evidence for structural confirmation and more description about biological assay are presented. The thermophysical characteristic including Differential scanning calorimetery and thermogravimetery analysis are described. Swelling kinetic parameters for the prepared hydrogel were calculated and showed that Schott's equation is well suited for explaining the swelling behavior of this transparent hydrogel.

This study designed to explore the characteristic features of the novel prepared hydrogel. This transparent nanocomposite hydrogel was formulated with employing environmental friendly biopolymer, "chitosan". To increase the hydrophilicity of chitosan, it was quaternized with triethyl amine. Also by incorporating click protocol, the triazole rings were inserted in the structure. After decoration with appropriate chemicals using efficient methods, functionalized chitosan and the corresponding hydrogel were investigated by Fourier transform infrared (FT-IR), thermal gravimetric analysis (TGA), differential scanning calorimetric (DSC) and dynamic-mechanical thermal analysis (DMTA). Swelling behavior of the synthesized hydrogel was assayed in both room temperature and 37 °C. Moreover, swelling kinetics were appraised and found that the experimental data fit the Schott's equation. To study the cell adhesion and proliferation, MTT assay was performed and the SEM images of 24, 48 and 72 h of direct cell culture on the surface of the scaffold were obtained. Morphological features of cultured cells were confirmed with Giemsa staining. The results displayed the potential capability of the synthesized scaffold for being used in bioapplications.

The evaluation on the characteristic dataset and figures presented here, are related to our latest research data entitled "Fabrication of chitosan based magnetic nanocomposite by click reaction strategy; evaluation of nanometric and Cytotoxic characteristics" [1]. FTIR, Vibrating Sample Magnetometer (VSM) measurements, Xray diffraction (XRD) information and the resulted figures for structural confirmation of the prepared chitosan based nanocomposite are presented in this article. The morphological changes of the Fibroblast, Saos, MCF7 and Hela cell lines after treatment with the mention compound were displayed. The additional adsorption data for the synthesized nanobiocomposite were also demonstrated with graphs.

Efficient procedure for preparation of the novel magnetic nanobiocomposite (MNbC) was successfully established by employing click chemistry protocol. The resulted structure exhibited valuable characteristics from biological viewpoints. The prepared materials and obtained nanobiocomposite were characterized by means of nuclear magnetic resonance (NMR), fourier transform infrared (FT-IR), thermal gravimetric analysis (TGA) and differential scanning calorimetry (DSC). To assay the antiproliferative effects of the MNbC; as cancerous cells, Hela, MCF-7 and Saos were evaluated by means of MTT assay and compared with normal fibroblast cell line. Promising antiproliferative effects were observed on both epitheloid and mesenchymal cancer cell lines depending on MNbC concentration and time. To detect the apoptosis-associated changes of cell membranes during the process of apoptosis, Acridine Orange/Ethidium Bromide (AO/EB) dual staining fluorescent technique was incorporated. The studies in this article including all of these specifications represent convincing key findings that make MNbC as an appreciable candidate for further bio-applications.
