RESUMO

A novel molecularly imprinted polymer coated stir bar was prepared using 2-chlorophenol (2-CP) as template molecule and used for selective extraction of phenolic compounds including chlorophenols (CPs) directly from seawater samples. Various parameters affecting extraction have been evaluated using one variable-at-a-time and Box-Behnken design. A three-level Box-Behnken experimental design by six factors, which combined the response surface methodology, was used to optimize the selective extraction of the CPs. Six independent variables, including the absorption and desorption times (ranging from 15 to 25 min), stirring speed (ranging from 550 to 650 rpm), pH of solution (ranging from 4.0 to 6.0), amount of NaCl (ranging from 0.30 to 0.40 mol/L) and temperature of extraction process (ranging from 40 to 50°C) were coded as A, B, C, D, E and F at three various levels (-1, 0 and 1). Under the optimum conditions, the detection limits calculated were 0.17, 0.33 and 0.38 µg/L, respectively, for 2-CP, 2,4-dichlorophenol and 2,4,6-trichlorophenol. The linear ranges of method were 1.0-100.0 µg/L for all CPs studied. This method was employed for the direct extraction and determination of three CPs in seawater samples.

AIM: Schmallenberg virus (SBV) is a newly emerging virus in Simbu group that 1st time is reported in 2011 in Germany and now spread to Europe. The clinical signs of infection to this virus are fever, loss of appetite, reduced milk yield and in some cases, diarrhea and in pregnant animals congenital malformations in calves, lambs, and kid goats. MATERIALS AND METHODS: In this study for a serologic survey of SBV, blood samples from 200 horse in different rural areas of the northern and northeast of Iran with the high equine population collected and were analyzed using an indirect ELISA test. RESULTS: Based on our results 5% (n=10) of total 200 samples were positive for SBV antibody and 2% (n=4) was doubtful and 93% (n=186) was negative. There were no significant differences between age and sex and breed properties (p>0.05). CONCLUSION: This study demonstrated the presence of antibodies against the SBV on horse populations in Iran. The high population and activity of Culicoides biting midges and their proper living conditions, especially the areas of temperate and humid environmental conditions, are the possible causes of arboviruses related diseases seen in this country.

Soil samples at two depths were collected and analyzed to determine the concentrations of 16 polycyclic aromatic hydrocarbons (PAHs), organic carbon, and soil pH. The Σ16PAHs were 0.13 to 3.92 mg kg(-1) at depth 1 and 0.21 to 50.32 mg kg(-1)at depth 2. The averages of the PAH compounds indicate that the area is contaminated with oil, and this pollution was greater at depth 2. Interpolation maps showed that the southern region, especially at depth 2, has been contaminated more by anthropogenic activity. The diagnostic ratios indicate several sources of pollution of the agricultural soil. A comparison of average PAHs and standard values revealed that higher molecular weight compounds in the topsoil (InP and BghiP) and subsoil (BaA, BkF, BaP, DBA, and BghiP) exceed standard values for farmland. The pH interpolation map for both depths showed that most of the area has alkaline soil from long-term irrigation with untreated urban wastewater.

The application of a granular activated carbon-sequencing batch biofilm reactor (GAC-SBBR) for treatment of wastewater containing 1,000 mg/L Acid Red 18 (AR18) was investigated in this research. The treatment system consisted of a sequencing batch reactor equipped with moving GAC as biofilm support. Each treatment cycle consisted of two successive anaerobic (14 h) and aerobic (8 h) reaction phases. Removal of more than 91% chemical oxygen demand (COD) and 97% AR18 was achieved in this study. Investigation of dye decolorization kinetics showed that the dye removal was stimulated by the adsorption capacity of the GAC at the beginning of the anaerobic phase and then progressed following a first-order reaction. Based on COD analysis results, at least 77.8% of the dye total metabolites were mineralized during the applied treatment system. High-performance liquid chromatography analysis revealed that more than 97% of 1-naphthyalamine-4-sulfonate as one of the main sulfonated aromatic constituents of AR18 was removed during the aerobic reaction phase. According to the scanning electron microscopic analysis, the microbial biofilms grew in most cavities and pores of the GAC, but not on the external surfaces of the GAC.

BACKGROUND: Respiratory diseases cause a considerable amount of morbidity and mortality in the world. Pulmonary function tests are important measures for the diagnosis and management of respiratory disorders. Workers in tile and ceramic industry are exposed to high amounts of respiratory pollutants. OBJECTIVE: To identify the changes in spirometric parameters in a 2-year period among tile and ceramic workers in Yazd and compare it with a control group. METHODS: The study was conducted in 5 tile and ceramic factories selected by cluster sampling between 2009 and 2011 in Yazd, southeastern Iran. Demographic data and spirometric parameters of participants were recorded. RESULTS: Spirometric parameters were significantly reduced during the 2 years. The largest decrease was observed in FVC (≈500 mL) in ball-mill and grinding after 2 years. Decrease in all spirometric parameters was significantly higher in industrial workers than office workers. CONCLUSION: Respiratory exposure in tile and ceramic industry can significantly affect pulmonary function tests.

Two integrated anaerobic/aerobic fixed-bed sequencing batch biofilm reactor (FB-SBBR) were operated to evaluate decolorization and biodegradation of azo dye Acid Red 18 (AR18). Volcanic pumice stones and a type of plastic media made of polyethylene were used as packing media in FB-SBBR1 and FB-SBBR2, respectively. Decolorization of AR18 in both reactors followed first-order kinetic with respect to dye concentration. More than 63.7% and 71.3% of anaerobically formed 1-naphthylamine-4-sulfonate (1N-4S), as one of the main sulfonated aromatic constituents of AR18 was removed during the aerobic reaction phase in FB-SBBR1 and FB-SBBR2, respectively. Based on statistical analysis, performance of FB-SBBR2 in terms of COD removal as well as biodegradation of 1N-4S was significantly higher than that of FB-SBBR1. Spherical and rod shaped bacteria were the dominant species of bacteria in the biofilm grown on the pumice stones surfaces, while, the biofilm grown on surfaces of the polyethylene media had a fluffy structure.

The application of aerobic moving bed biofilm process as post-treatment of anaerobically degraded azo dye Acid Red 18 was investigated in this study. The main objective of this work was to enhance removal of anaerobically formed the dye aromatic metabolites. Three separate sequential treatment systems were operated with different initial dye concentrations of 100, 500 and 1000 mg/L. Each treatment system consisted of an anaerobic sequencing batch reactor (An-SBR) followed by an aerobic moving bed sequencing batch biofilm reactor (MB-SBBR). Up to 98% of the dye decolorization and more than 80% of the COD removal occurred anaerobically. The obtained results suggested no significant difference in COD removal as well as the dye decolorization efficiency using three An-SBRs receiving different initial dye concentrations. Monitoring the dye metabolites through HPLC suggested that more than 80% of anaerobically formed 1-naphthylamine-4-sulfonate was completely removed in the aerobic biofilm reactors. Based on COD analysis results, at least 65-72% of the dye total metabolites were mineralized during the applied treatment systems. According to the measured biofilm mass and also based on respiration-inhibition test results, increasing the initial dye concentration inhibited the growth and final mass of the attached-growth biofilm in MB-SBBRs.

In visceral leishmaniasis (VL), the development of protective immunity is associated with expansion of leishmania-specific T-cell responses. Because of the essential role of CD28 in the effectiveness of T-cell activation, this study was carried out to investigate the CD28 gene polymorphism and plasma levels of soluble (s) CD28 molecule in Iranian patients with VL. Plasma concentrations of CD28 in 88 patients with VL, 132 individual with subclinical leishmaniasis, and 100 seronegative healthy controls were measured by enzyme-linked immunosorbent assay. Genotyping of CD28 gene polymorphism was performed by polymerase chain reaction based allotyping method using allele-specific primers for C or T at intron 3 position +17 in three groups. The frequency of CC genotype was significantly higher in subclinical VL patients (42.4%) than active VL group (27.3%) and healthy controls (16%) (P<0.001). Also, the frequency of allele C among subclinical VL group (57.6%) was significantly higher than active VL (40.9%) and control groups (34%) (p=0.003). No significant differences were observed between the plasma levels of sCD28 in three groups. Our findings suggest that the CD28 gene may have significant role in the protection of active VL in the Iranian population.

Since their introduction, poly(ethylene glycol)-phospholipid (PEG-PL) conjugates have found many applications in design and engineering of nanosized delivery systems for controlled delivery of pharmaceuticals especially to non-macrophage targets. However, there are reports of idiosyncratic reactions to certain PEG-PL engineered nanomedicines in both experimental animals and man. These reactions are classified as pseudoallergy and may be associated with cardiopulmonary disturbance and other related symptoms of anaphylaxis. Recent studies suggest that complement activation may be a contributing, but not a rate limiting factor, in eliciting hypersensitivity reactions to such nanomedicines in sensitive individuals. This is rather surprising since PEGylated structures are generally assumed to suppress protein adsorption and blood opsonization events including complement. Here, we examine the molecular basis of complement activation by PEG-PL engineered nanomedicines and carbon nanotubes and discuss the challenges ahead.

We reviewed the clinical manifestations and outcome of 20 cases of neurobrocellosis out of 1375 patients with brucellosis admitted to the infectious diseases ward of a tertiary hospital in Hamedan, Islamic Republic of Iran. Of the 20 cases, 12 had meningitis (acute and subacute), 4 had meningoencephalitis, 2 had myelopathy, 1 had polyradiculopathy and 1 had meningovascular complications manifested by stroke. The most prevalent symptoms were headache (75%), vomiting and fever (50%) and unconsciousness (35%). Among 20 neurobrucellosis patients, 1 died and the other 19 recovered; 14 fully recovered and 5 patients had residual neurological deficits. In endemic areas, the disease should be ruled out in all patients who develop unexplained neurological symptoms.

An analytical cross-sectional study determined the serogroups and serotypes of Vibrio cholerae, and their antibiotic resistance rates, in the 2005 cholera epidemic in Hamadan. All 190 patients with positive stool cultures had V. cholerae serogroup O1, biotype El Tor and serotype Inaba positive. Of 60 cases selected randomly for antibiogram testing, sensitivity to norfloxacin, ciprofloxacin, kanamycin, amikacin, tetracycline and doxycycline was 97%, 92%, 88%, 85%, 77% and 67% respectively. Resistance to furazolidone, trimethoprim-sulfamethoxazole and erythromycin was 100%, 98% and 62% respectively. Comparison with the results of the 1998 epidemic suggests a worrying increase in the resistance of V. cholerae to erythromycin, doxycycline and ciprofloxacin.

BACKGROUND: The aim of this study was to investigate the importance of somatostatin receptor subtype 2 (SSTR2) expression for 111In-labelled diethylenetriamine-pentaacetic acid (DTPA)-D-Phe1-octreotide binding and uptake of 111In in neuroendocrine tumours. METHODS: 111In activity concentrations in surgical biopsies from neuroendocrine tumours (midgut carcinoid and medullary thyroid carcinoma), breast carcinoma and blood were determined 1-8 days after intravenous injection of 111In-labelled DTPA-D-Phe1-octreotide (140-350 MBq). The ratio of 111In activity concentrations between tumour tissue and blood (T/B value) was calculated. The expression of SSTR2 messenger RNA (mRNA) in tumour biopsies was quantitated by ribonuclease protection assay and SSTR2 protein was localized by immunocytochemistry. RESULTS: T/B values were highest for tumour biopsies from midgut carcinoids (mean 160 (range 4-1200); n = 65) followed by medullary thyroid carcinoma (mean 38 (range 2-350); n = 88) and breast carcinoma (mean 18 (range 4-41); n = 4). The expression of SSTR2 mRNA (relative to the NCI-H69 cell line) was highest in tumour biopsies from midgut carcinoids (mean 2.5 (range 0.83-6.0); n = 40) followed by medullary thyroid carcinoma (mean 1.3 (range 0.20-6.0); n = 7) and breast carcinoma (mean 0.66 (range 0.29-1.0); n = 9). In tumour biopsies SSTR2 protein was localized exclusively to tumour cells. CONCLUSION: Midgut carcinoid tumours showed a much higher level of SSTR2 expression than medullary thyroid carcinoma in accordance with superior tumour imaging by octreotide scintigraphy. The high SSTR2 mRNA values and T/B values observed in midgut carcinoid tumours were positively correlated.

Somatostatin sst2(a) receptor was observed, by immunofluorescence, in ependymal cells and in tanycytes of the wall of the ventricle and the hypothalamic recess of the male rat median eminence. Strong immunoreactivity for the receptor protein was observed in lateral tanycytes (alpha-type) while a moderate signal was seen in medial tanycytes (beta-type). In high magnification the immunoreactive material, of moderate intensity, had a coarse granular appearance. Only few of the alpha-tanycytes also displayed immunoreactive GFAP. The apical portion of the ependymal cells as well as of tanycytes contained immunoreactive S-100 (alphabeta). Since rather high levels of somatostatin are demonstrated to occur in the cerebrospinal fluid of the third ventricle, it is suggested that somatostatin via the sst2(a) receptor may regulate the physiology of tanycytes.

UNLABELLED: The purpose of this study was to evaluate the potential for therapy of thyroid tumors using the radiolabeled somatostatin (SS) analog octreotide. METHODS: Concentrations of 111In activity in human thyroid tumors and normal thyroid tissue and blood samples were determined 1-15 d after intravenous injection of 111In-diethylenetriaminepentaacetic acid-Phe1-octreotide. The results were compared with SS receptor (sstr subtype profile (by Northern blot analysis) and the relative expression of the second subtype, sstr2 (by ribonuclease protection assay, RPA). The true tumor volumes in lymph node metastases from 1 patient were estimated. In total, tissues from 68 patients were included in the study. RESULTS: The highest tumor-to-blood ratio (T/B) for medullary thyroid carcinoma (MTC) was 360; for follicular adenoma (FA), 190; for Hurthle cell adenoma (HCA), 140; and for Hurthle cell carcinoma (HCC) and papillary carcinoma (PC), 70. The corresponding value was 7-18 for normal thyroid tissue, with higher values for colloid goiter (8-48) and thyroiditis (7-120). A high T/B was related to a large fraction of tumor cells in lymph node metastases. T/Bs were higher for the tumor samples with expression of sstr2 at Northern blot analysis than for those without. All thyroid tumor types regularly expressed sstr1, sstr3, sstr4, and sstr5. sstr2 was expressed in most MTC tumors but was not detected in FA or PC and was irregularly expressed in HCA and HCC. However, RPA analysis detected sstr2 in all tumors studied. CONCLUSION: Despite the lack of sstr2 at Northern blot analysis in most of the thyroid tumors studied, high T/Bs were in general found when compared with corresponding values for normal thyroid tissue. The sometimes extremely high ratios are promising and indicate a possibility of using radiolabeled octreotide for radiation therapy of sstr-positive tumors in the future.
