RESUMO

Elaeidobius kamerunicus is the most efficient pollinator of oil palm. Wolbachia is an endosymbiotic bacteria associated with E. kamerunicus that has a potential to affect the fecundity and fitness of the E. kamerunicus. Despite their importance, no studies have been conducted to investigate its prevalence in E. kamerunicus. The objectives of this study were to detect and characterise Wolbachia in E. kamerunicus and determine the phylogenetic relationship of Wolbachia strains that infect E. kamerunicus by using three genetic markers namely Filamenting temperature-sensitive mutant Z (ftsZ), Chaperonin folding protein (groEL), and Citrate Synthase Coding Gene (gltA). DNA was extracted from 210 individuals of E. kamerunicus and the Wolbachia infections were detected using the wsp marker. The infected samples (n = 25, 11.9%) were then sequenced using ftsZ, gltA and groEL markers for strain characterization. In this study, a combination of four markers was used to construct the phylogeny of Wolbachia. Similar topologies were shown in all trees; Neighbour-Joining (NJ), Maximum Parsimony (MP), and Bayesian Inference (BI), which showed the mixing of individuals that harbor Wolbachia between populations. Interestingly, Wolbachia on E. kamerunicus was claded together with the species Drosophila simulans under supergroup B. This is the first report of Wolbachia infecting E. kamerunicus which is very valuable and significant as one of the parameters to evaluate the quality of the E. kamerunicus population for sustaining its function as a great pollinator for oil palm.

The oil palm pollinating weevil, Elaeidobius kamerunicus Faust (Coleoptera: Curculionidae) was first introduced into Malaysia from Cameroon as the main oil palm pollinator in 1981. Since then, oil palm pollination has improved and the need for assisted pollination has reduced. However, their pollinating behavior may be influenced by starvation, sexes and sources (wild-caught and lab-reared). A study to determine the effect of starvation level, sexes and sources of E. kamerunicus on their diurnal behavior was conducted utilizing two hours of direct observation between 1130 and 1330 h. The frequency and the time spent for several diurnal behaviors on oil palm flowers were recorded and evaluated. Starvation prevented the weevils from conducting the activities because they probably had to focus more on searching for food to gain energy to perform other behavior. Wild-caught and lab-reared E. kamerunicus differed in their size, while sexes of E. kamerunicus significantly affect the diurnal behavior. However, an in-depth study is needed to determine the relationship between the diurnal behavior of E. kamerunicus and oil palm production.

We studied the water quality of the riparian firefly sanctuary of Sungai Rembau, or Rembau River, in Negeri Sembilan, Malaysia, from January 2018 to November 2018 to determine the possible influence of the physico-chemical characteristics of the water on the firefly populations living within the sanctuary. We set up a total of five water quality sampling stations and 10 firefly sampling stations along the river. Dissolved oxygen (DO), temperature, pH and electrical conductivity (EC) were measured in situ, while chemical oxygen demand (COD), total suspended solids (TSS), biochemical oxygen demand (BOD) and ammonia-nitrogen (NH3-N) were analysed in the laboratory. Firefly samples were collected using a sweep net at both day and night for 1 min. Sungai Rembau was categorized as Class II on the Malaysian water quality index (WQI), which indicates slight pollution. Except for EC and DO, the water quality parameter values were not significantly different (p > 0.05) between the sampling stations. A total of 529 firefly individuals consisting of Pteroptyx tener (n = 525, 99.24%), P. malaccae (n = 3, 0.57%) and P. asymmetria (n = 1, 0.19%) were collected. There was significant correlation between firefly abundance and BOD (r = - 0.198, p < 0.05), NH3-N (r = - 0.150, p < 0.05) and pH (r = 0.408, p < 0.05). The results show that the firefly populations in Sungai Rembau are sensitive to organic compounds, which may be present in the form of pollutants from anthropogenic activities near their natural habitat.

Riparian areas hold vast number of flora and fauna with exceptional contributions to the ecosystem. A study was conducted in Sungai Sepetang, Sungai Rembau and Sungai Chukai to identify the insect community in a riparian zone of Peninsular Malaysia. Sampling was conducted in six consecutive months from December 2017 to May 2018 during both day and night using sweep nets. Twenty sampling stations (S1-S20) had been assembled along the riverbanks with an average distance of 200 m between each station. The 17,530 collected insects were from 11 orders and consisted of Diptera, Coleoptera, Hemiptera, Hymenoptera, Lepidoptera, Neuroptera, Orthoptera, Blattodea, Thysanoptera, Mantodea and Odonata. The three most abundant orders were Diptera (33.84%; 5933 individuals), Coleoptera (28.82%; 5053 individuals) and Hemiptera (25.62%: 4491 individuals). The collected insect community consisted of different guilds such as the scavenger, predator, herbivore, pollinator and parasitoid. Sungai Sepetang and Sungai Rembau were dominated by mangrove flora, Sonneratia caseolaris (Myrtales: Lythraceae), while Sungai Chukai was dominated by Barringtonia racemosa. There was a significant difference (p < 0.05) in the composition of insects between the three rivers though clustering analysis showed that the insect communities in Sungai Sepetang and Sungai Rembau were 100% similar compared to Sungai Chukai which consisted of a totally different community. There is a significant negative correlation between abundance of insects with salinity and wind speed at Sungai Chukai and Sungai Sepetang.
