RESUMO

Neutrophils play a pivotal role in innate immunity by releasing ROS through respiratory bursts to neutralize various pathogenic factors. However, excessive ROS release can cause tissue damage. Adenosine is an endogenous anti-inflammatory molecule inhibiting respiratory burst to protect the host. Adenosine aptamers with antibody-like properties and good stability are expected to act as adenosine antagonists with functional modulation capability. This study compares the effects of adenosine and its aptamer on the respiratory bursts of salivary polymorphonuclear leukocytes and circulating polymorphonuclear leukocytes using a programmable stopped-flow injection approach, ensuring rapid and efficient analysis while maintaining the neutrophils' viability. The results show that primed salivary polymorphonuclear leukocytes exhibit specificities that differ from circulating polymorphonuclear leukocytes. Adenosine aptamer can function as an inhibitory antagonist that distinguishes between physiologically controlled and excessive priming of neutrophils, showing potential application prospects in immunotherapy.

This study aimed to analyze the independent risk factors for predicting preeclampsia severity and explore its underlying mechanism. Clinical data of patients with preeclampsia were collected from the Medical Information Mart for Intensive Care (MIMIC)-IV database. Univariate and multivariate analyses were employed to assess the significant factors associated with preeclampsia severity. Additionally, we performed multivariate logistic regression analysis and mediation analysis to investigate the potential regulatory path. Based on inclusion and exclusion criteria, 731 participants were enrolled: severe preeclampsia (n = 381) and mild to moderate preeclampsia (n = 350). Age, white blood cells (WBC), platelet, creatinine, albumin, uric acid, aspartate aminotransferase, alanine aminotransferase, international normalized ratio, and prothrombin time were significantly related to preeclampsia severity. Besides, hospital length of stay was significantly higher in the severe group. Notably, age and uric acid were independent predictors for preeclampsia severity. Further, WBC and creatinine were significantly associated with uric acid. Finally, the mediation analysis showed that uric acid was a mediator of the relationship between WBC and preeclampsia severity. In conclusion, WBC might affect preeclampsia severity and progression via the mediation of uric acid. This study might provide novel insight into preventing preeclampsia development.

RATIONALE: Ovarian vein thrombosis (OVT) is a rare yet potentially life-threatening condition associated with thromboembolic events. Group B Streptococcus (GBS) is a type of ß-hemolytic Gram-positive bacterium known for asymptomatic colonization in the lower genital and gastrointestinal tracts. Here we reported a 35-year-old multiparous woman with gestational diabetes who suffered from placental abruption, stillbirth, OVT, septic shock, and renal failure due to severe GBS infection. PATIENT CONCERNS: A 35-year-old woman with gestational diabetes presented with acute and sustained lower abdominal cramping, vaginal bleeding, and fever at 35 gestational weeks. DIAGNOSES: Based on preoperative ultrasound and intraoperative findings, the patient was diagnosed with placental abruption, intrauterine fetal demise, and right OVT. GBS was cultured from the amniotic fluid obtained during cesarean section. INTERVENTIONS: The patient underwent a right adnexectomy during a cesarean section and received intravenous antibiotics. Subsequently, an ultrasound-guided uterine curettage was performed due to recurrent fever. OUTCOMES: After a prolonged course of intravenous antibiotics for over a month, the patient recovered and was discharged from the hospital. LESSONS: This case underscores the need for early initiation of anticoagulant protocols in cases of OVT, particularly when GBS infection is identified as a predisposing factor. Further research and awareness are warranted to better understand the relationship between GBS infection and OVT and to optimize management strategies in such cases.

Activated polymorphonuclear neutrophils play an important role in host defense system against invading pathogens via releasing reactive oxygen species through respiratory bursts. Nowadays, neutrophil activation process has been found to be modeled as a two-stages continuum from dormant to primed, and to fully activated. Although the differences of salivary and circulatory neutrophils have been investigated in various experimental designs, priming state of salivary polymorphonuclear neutrophils with respiratory burst has not been comprehensively studied. Here, for the first time so far, a programmable flow injection analysis based on Stop in Flow Cell operation mode has been applied to batch analyzing comparative study of neutrophil activation. The high-sensitivity luminol-dependent chemiluminescence assay has been employed with just 330 µL of cell suspension consumption per sample, and the process has been conducted rapidly and efficiently within 15 min to keep the neutrophils viability. ROS from either unstimulated or substimulated or stimulated salivary polymorphonuclear neutrophils, in comparison with those from autologous circulatory polymorphonuclear neutrophils, have been determined, together with further monitoring the impact of extracellular and intracellular calcium ion (Ca2+) on neutrophils ROS generation. Conclusively, the priming state of salivary polymorphonuclear neutrophils during activation has been testified from multiple aspects.

OBJECTIVE: To explore the clinical curative effects of preoperative neoadjuvant chemotherapy (NACT) on locally advanced cervical cancer. METHODS: A total of 62 patients of stage Ib2-IIb cervical cancer received neoadjuvant chemotherapy of paclitaxel plus cisplatin for 2 - 3 courses. The clinical curative effects were evaluated according to the changes of lesion size, intraoperative conditions and postoperative pathological reactions. RESULTS: The overall response rate was 90.32% (56/62) and the complete response rate 30.65% (19/62). The tumor volumes decreased after NACT. The differences were significant (P < 0.05). After NACT, 56 patients undergoing radical hysterectomy recovered smoothly. The surgical resection rate was 90.32%. Chemotherapeutic reactions of cancerous tissue and a large number of infiltrated lymphocytes were seen in 50 cases. Lymph nodes were positive in 3 cases. There were parametrial invasion (n = 2) and vascular tumor emboli (n = 2). CONCLUSION: Preoperative neoadjuvant chemotherapy is efficacious in cervical cancer. The parametrium becomes softer and the tumor staging decreases.

OBJECTIVE: To investigate the difference of anticytomegaloviral activity of three kinds of traditional Chinese medicines which are the injections of Jinye Baidu, Radix Isatidis and Indigowoa in vitro. METHOD: The inhibitory activity of three traditional Chinese medicines against human cytomeglovirus (HCMV AD169) infected human embryo lung fibroblasts (HELF) was observed by cytopathic effect method (CPE) and MTT method in vitro. According their value of A, anticytomegaloviral activity has evaluated. RESULT: Experimental study in vitro showed that the 50% toxicity dose (TD50) of Jinye Detoxifying, Radix Isatidis root and Indigowoa were 20, 10.23, 20.23 g x L(-1) respectively; the 50% inhibitory concertration (IC50) were 5.65, 3, 5.71 g x L(-1) respectively; the therapeutic index (TI) were 3.54, 3.41 and 3.54 respectively. It suggested that three traditional Chinese medicines had anticytomeglovirus activity and their effect increased with their concentration. CONCLUSION: Three traditional Chinese medicines of the parenteral solution of Jinye Detoxifying, Radix Isatidis root and Indigowoa have antiviral activity when they are diluted in 1:200. They are safe and valuable drug for inhibiting cytomeglovirus infection.
