OBJECTIVE: Coronavirus disease 2019 (COVID-19) spread around the world in 2020. Abnormal pulmonary function and residual CT abnormalities were observed in COVID-19 patients during recovery. Appropriate rehabilitation training is around the corner. The correlation between spirometric impairment and residual CT abnormality remains largely unknown. PATIENTS AND METHODS: A cross-sectional study conducted on the pulmonary function of 101 convalescent COVID-19 patients before discharge. Multivariate analysis was used to establish a scoring system to evaluate the spirometric abnormality based on residual chest CT. RESULTS: Lung consolidation area >25% and severe-type COVID-19 were two independent risk factors for severe pulmonary dysfunction. Besides, a scoring system was established. People scoring more than 12 points have more chances (17 times) to get severe pulmonary function impairment before discharge. CONCLUSIONS: For the first time, a chest CT characteristics-based grading system was suggested to predict the pulmonary dysfunction of COVID-19 patients during convalescence in this study. This study may provide suggestions for pulmonary rehabilitation.

OBJECTIVE: We aimed to compare and analyze the long-term efficacy and safety between kelp micro gelation (KMG) and gelfoam particles in partial splenic embolization (PSE). PATIENTS AND METHODS: This study retrospectively included 65 liver cirrhosis patients with comorbidity of hypersplenism who were admitted in Nanfang Hospital from July 2008 to May 2012. Among the included patients, 33 patients were in KMG-PSE group, and 32 cases were included in Gelfoam-PSE group. According to the Child-Pugh criteria, all cases were divided into grade A, B, and C, respectively. All eligible subjects received CT or MR examination and laboratory examination. RESULTS: Our results showed that both KMG and gelfoam particles could substantially improve the short-term efficacy of thrombocytopenia leukopenia. However, the efficacy of KMG is superior to that of gelfoam. Due to the characteristic of KMG as a permanent agent, KMG may result in an obvious pain in the spleen after PSE, especially in patients with megalosplenia. KMG was more expensive than gelfoam particles. The complication rate in patents with great embolization was much higher than that with less embolization. CONCLUSIONS: The efficacy of KMG is superior to that of gelfoam in both short-term and long-term. To effectively control the occurrence of severe complication, the embolization should be controlled less than 70% regardless of the embolization agents.

Transition metal dichalcogenides exhibit an excellent catalytic activity for hydrogen evolution reaction and continue to attract many studies. Some heterojunctions are also used to enhance the catalytic activity. In this work, the TiO2 nanotubes/MoSe2 and Si-doped TiO2 NTs/MoSe2 hybrids were synthesized. The MoSe2 in the hybrids exhibited nanosheet morphology and metallic (1T) structure with high conductivity and catalytic activity for hydrogen evolution reaction. Moreover, MoSe2 decoration provided high light absorbance for the hybrids and highly efficient interface-induced effect at the heterojunction. Furthermore, the Si-doping increased specific surface area and hydrophilicity. Thus, the Si-doped TiO2 NTs/MoSe2 hybrids showed remarkably greater photocatalytic and electrocatalytic activities than the nanotubes.

The transition metal dichagenides and their metallic 1T structure are attracting contemporary attentions for applications in high-performance devices because their peculiar optical and electrical properties. The single and few layers 1T structure is generally obtained by mechanical or chemical exfoliation. This work presents facile one-step synthesis of 2H-1T MoS2:Cu/reduced graphene oxide nanosheets. The experiment results indicated that the MoS2 and MoS2:Cu prepared by simple chemical solution reaction possessed 2H-1T structures. The reduced graphene oxide (rGO) incorporation further induced the phase transition from 2H-MoS2 to 1T-MoS2 and morphology transition from granular/nanosheet to more nanosheet. The 2H-1T structure and 2H → 1T phase transition, together with the Cu doping and interface effect between the MoS2 and rGO, remarkably enhanced the conduction and photoconduction of the nanostructures. Thus, Cu doping and rGO incorporation obviously enhanced the catalytic activity and its stability, making the MoS2:Cu/rGO nanosheet a most active and stable catalyst for hydrogen evolution. This work clearly indicates that the 1T-MoS2 nanosheets with high catalytic activity for hydrogen evolution can be easily obtained by the facile low temperature chemical method and induction of rGO.

OBJECTIVE: We investigated the interactions among the dysregulated genes in hepatocellular carcinoma (HCC) and identified the hub genes in the protein-protein interaction (PPI) network. Also, we also investigated the regulative effect of miR-379 on the IGF1/IGF1R signaling pathway and chemoresistance in HCC. MATERIALS AND METHODS: Raw data of a microarray that compared transcriptional gene profile between 3-paired HCC tissue samples and adjacent normal tissues were downloaded from Expression Atlas (E-GEOD-33006). The raw data was reanalyzed to identify the significantly dysregulated genes, which were further used for PPI network and KEGG pathway analysis. The regulative effect of miR-379 on IGF1R expression was studied by dual luciferase assay and Western blotting. The functional role of miR-379 in chemosensitivity of HCC cells was studied by drug sensitivity and flow cytometric assay. RESULTS: IGF1 is a hub gene that is mostly upregulated in HCC and it is an important player in the p53 signaling pathway. Knockdown of IGF1R significantly decreased IC50 of 5-FU, paclitaxel (PTX) and Doxorubicin (DOX) in Huh7 and HepG2 cells. MiR-379 could directly bind to the 3'UTR of IGF1R and suppress IGF1R expression. MiR-379 overexpression sensitized Huh7 and HepG2 cells to 5-FU, PTX and DOX and also enhanced DOX-induced cell apoptosis. CONCLUSIONS: IGF1 is a hub gene in HCC and is also one of the most upregulated genes in HCC tissues compared to normal tissues. It is involved in the p53 signaling pathway regulation. MiR-379 can sensitize HCC cells to chemotherapeutic reagents via targeting IGF1R and suppressing its expression.

BACKGROUND: Non-pharmacological interventions are effective neonatal pain reduction strategies. We aimed to study the effects of non-nutritive sucking (NNS) and swaddling on infants' behavioural and physiological parameters during shallow or deep heel stick procedures. METHOD: In this prospective, multi-centred, randomized controlled clinical trial, we enrolled 671 newborns. The infants undergoing shallow or deep heel stick procedures were randomized into four groups: oral sucrose (routine care, group S), oral sucrose combined with NNS (group NS), oral sucrose combined with swaddling (group SS) and oral sucrose combined with NNS and swaddling (group NSS). The behavioural responses were evaluated by the Revised Neonatal Facial Coding System and the physiological signals were monitored by electrocardiogram monitors. RESULTS: A significant synergistic analgesic effect was observed between the NS and SS groups in both the shallow (F = 5.952, p = 0.015) and deep heel stick (F = 7.452, p = 0.007) procedure. NSS group exhibited the lowest pain score. For the deep heel stick procedure, the NS group had a significantly lower increase in heart rate (HR)% and decrease in SPO2 % than the S group (F = 17.540, p = 0.000, F = 10.472, p = 0.001), while this difference was not observed in the shallow heel stick procedure. No difference was found between the S and SS groups, in terms of different physiological parameters. CONCLUSION: Non-nutritive sucking and swaddling had synergistic effects on pain relief when used with oral sucrose. For the deep heel stick procedure, oral sucrose combined with NNS and swaddling provided the best pain relief effect. For the shallow heel stick procedure, addition of NNS and swaddling did not improve the effects.

Membranous nephropathy (MN), characterized by the presence of diffuse thickening of the glomerular basement membrane and subepithelial in situ immune complex disposition, is the most common cause of idiopathic nephrotic syndrome in adults, with an incidence of 5-10 per million per year. A number of studies have confirmed the relevance of several experimental insights to the pathogenesis of human MN, but the specific biomarkers of MN have not been fully elucidated. As a result, our knowledge of the alterations in histone methylation in MN is unclear. We used chromatin immunoprecipitation followed by high-throughput sequencing (ChIP-seq) to analyze the variations in a methylated histone (H3K9me3) in peripheral blood mononuclear cells from 10 MN patients and 10 healthy subjects. There were 108 genes with significantly different expression in the MN patients compared with the normal controls. In MN patients, significantly increased activity was seen in 75 H3K9me3 genes, and decreased activity was seen in 33, compared with healthy subjects. Five positive genes, DiGeorge syndrome critical region gene 6 (DGCR6), sorting nexin 16 (SNX16), contactin 4 (CNTN4), baculoviral IAP repeat containing 3 (BIRC3), and baculoviral IAP repeat containing 2 (BIRC2), were selected and quantified. There were alterations of H3K9me3 in MN patients. These may be candidates to help explain pathogenesis in MN patients. Such novel findings show that H3K9me3 may be a potential biomarker or promising target for epigenetic-based MN therapies.

The aim of this study was to observe the dynamic expression of osteopontin, type I collagen, and osteocalcin genes during the differentiation of bone marrow mesenchymal stem cells (BMSCs) into osteoblasts and confirm whether BMSCs can differentiate and mature into osteoblasts. A healthy, 2-month-old Altay tailed sheep was obtained, and 10 mL bone marrow was extracted from the posterior superior iliac spine of the sheep via puncture. Sheep BMSCs were extracted using a whole bone marrow culture method and cultured for osteogenic induction. Total RNA from the BMSCs was extracted, and the gene expression levels of osteopontin, type I collagen, and osteocalcin at the 1st, 2nd, 3rd, and 4th passages of uninduced and induced BMSCs were detected using reverse transcription-polymerase chain reaction. The surface antigen CD44 was also detected in the BMSCs. After induction by culturing in osteoinductive medium, BMSCs in the 1st, 2nd, 3rd, and 4th passages displayed stage-specific expression of osteopontin, type I collagen, and osteocalcin genes. The positive expression rate of BMSC-specific antigen CD44 in the 4th passage of osteogenic-induced BMSCs was 99.8%. In vitro-cultured sheep BMSCs in osteogenic culture medium gradually differentiated into osteoblasts, which stage-specifically expressed osteoblast-specific genes such as osteopontin, type I collagen, and osteocalcin at the 4th passage after induction. At this passage, these cells had osteoblast functions.

It has been generally acknowledged that the diagnosis, treatment and prognosis evaluation of leukemia largely rely on an adequate identification of genetic abnormalities. A systemic analysis of genetic aberrations was performed in a cohort of 1346 patients with newly diagnosed acute lymphoblastic leukemia (ALL) in China. The pediatric patients had higher incidence of hyperdiploidy and t(12;21) (p13;q22)/ETV6-RUNX1 than adults (P<0.0001); in contrast, the occurrence of Ph and Ik6 variant of IKZF1 gene was much more frequent in adult patients (all P<0.0001). In B-ALL, the existence of Ik6 and that of BCR-ABL were statistically correlated (P<0.0001). In comparison with Western cohorts, the incidence of t(9;22) (q34;q11)/BCR-ABL (14.60%) in B-ALL and HOX11 expression in T-ALL (25.24%) seemed to be much higher in our group, while the incidence of t(12;21) (p13;q22)/ETV6-RUNX1 (15.34%) seemed to be lower in Chinese pediatric patients. The occurrence of hyperdiploidy was much lower either in pediatric (10.61% vs 20-38%) or adult patients (2.36% vs 6.77-12%) in our study than in Western reports. In addition, the frequencies of HOX11L2 in adult patients were much higher in our cohort than in Western countries (20.69% vs 4-11%). In general, it seems that Chinese ALL patients bear more adverse prognostic factors than their Western counterparts do.

Readily available N-acylbenzotriazoles 2a-q efficiently acylate aqueous ammonia and primary and secondary amines to give primary, secondary, and tertiary amides in good to excellent yields. The wide applicability of the procedure is illustrated by the preparation of (i) alpha-hydroxyamides from alpha-hydroxy acids and of (ii) perfluoroalkylated amides.
