RESUMO

BACKGROUND: The Fat mass and obesity-associated gene (FTO) was the first gene reliably associated with body mass index in genome-wide association studies on a population level. At present, the genetic variations within the FTO gene are still the common variants that have the largest influence on body mass index. METHODS: In the current study, we amplified the entire FTO gene, in total 412 Kbp, in over 200 long-range PCR fragments from each individual, from 524 severely obese and 527 lean Swedish children, and sequenced the products as two DNA pools using massive parallel sequencing (SOLiD). RESULTS: The sequencing achieved very high coverage (median 18 000 reads) and we detected and estimated allele frequencies for 705 single nucleotide polymorphisms (SNPs) (19 novel) and 40 indels (24 novel) using a sophisticated statistical approach to remove false-positive SNPs. We identified 19 obesity-associated SNPs within intron one of the FTO gene, and validated our findings with genotyping. Ten of the validated obesity-associated SNPs have a stronger obesity association (P<0.007) than the commonly studied rs9939609 SNP (P<0.012). CONCLUSIONS: This study provides a comprehensive obesity-associated variation map of FTO, identifies novel lead SNPs and evaluates putative causative variants. We conclude that intron one is the only region within the FTO gene associated with obesity, and finally, we establish next generation sequencing of pooled DNA as a powerful method to investigate genetic association with complex diseases and traits.

RESUMO

Persons with psychiatric disabilities often lead an isolated life, and their living conditions are usually not as good as those of other people. There are few studies that describe these individuals' own opinions of what can contribute to increased health. The aim of this study was therefore to describe the importance of health for persons with psychiatric disabilities. The study group comprised 12 persons, living in the community in west of Sweden, who were interviewed during 2005. The data were analysed by means of qualitative content analysis. The findings revealed five categories: (1) help and support in daily life; (2) being part of a social context; (3) having a meaningful daily life; (4) using resources in the right way; and (5) experiencing well-being. The result showed that interventions that promote health need to be clarified and strengthened by means of coordinated efforts from the healthcare services and the local community. In addition, persons with psychiatric disabilities should be invited and encouraged to participate in society as well as being listened to and having their needs addressed.

RESUMO

The structure of iron pentacarbonyl, Fe(CO)(5), was reinvestigated by gas-phase electron diffraction using an experimental rotational constant available from the literature as a constraint on the structural parameters. The study utilized a B3LYP/6-311+G(d) ab initio quadratic force field, scaled to fit observed infrared wavenumbers, from which were calculated corrections for the effects of vibrational averaging on distances and certain other quantities useful for the structural analysis. The results confirm that the equatorial Fe-C bonds are longer than the axial ones, an important difference with the structure in the crystal where the equatorial Fe-C bonds are the shorter. Some distance (r(g)/A) and vibrational amplitude (l(alpha)/A) parameter values with estimated 2sigma uncertainties based on assumption of D(3h) symmetry are [r(Fe-C)] = 1.829(2), r(Fe-C)(eq) - r(Fe-C)(ax) = 0.032(20), [r(C=O)] = 1.146(2), r(C=O)(eq) - r(C=O)(ax) = 0.006(27), r(Fe-C)(ax) = 1.810(16), r(Fe-C)(eq) = 1.842(11), r(C=O)(ax) = 1.142(23), r(C=O)(eq) = 1.149(16), l(Fe-C)(ax) = l(Fe-C)(eq) = 0.047(5), and l(C=O)(ax) = l(C=O)(eq) = 0.036(3).

We report an extended version of our normal coordinate program ASYM40, which may be used to transform Cartesian force constants from ab initio calculations to a force field in nonredundant internal (symmetry) coordinates. When experimental data are available, scale factors for the theoretical force field may then be optimized by least-squares refinement. The alternative of refining an empirical force field to fit a wide variety of data, as with the previous version ASYM20, has been retained. We compare the results of least-squares refinement of the full harmonic force field with least-squares refinement of only the scale factors for an SCF calculated force field and conclude that the latter approach may be useful for large molecules where more sophisticated calculations are impractical. The refinement of scale factors for a theoretical force field is also useful when there are only limited spectroscopic data. The program will accept ab initio calculated force fields from any program that presents Cartesian force constants as output. The program is available through Quantum Chemistry Program Exchange. RESUMO

Electron diffraction patterns of the fullerene C(60) in the gaseous state have been obtained by volatilizing it from a newly designed oven-nozzle at 730 degrees C. The many peaks of the experimental radial distribution curve calculated from the scattered intensity are completely consistent with icosahedral symmetry for the free molecule. On the basis of this symmetry assumption, least-squares refinement of a model incorporating all possible interatomic distances led to the values r(g)(C(1)-C(2)) = 1.458(6) angstroms (A) for the thermal average bond length within the five-member ring (that is, for the bond fusing five- and six-member rings) and r(g)(C(1)-C(6)) = 1.401(10) A for that connecting five-member rings (the bond fusing six-member rings). The weighted average of the two bond lengths and the difference between them are the values 1.439(2) A and 0.057(6) A, respectively. The diameter of the icosahedral sphere is 7.113(10) A. The uncertainties in parentheses are estimated 2sigma values.
