RESUMO

The scope of this research was to determine the bioactive composition, antioxidant, binding, and anti-proliferative properties of red sweet paprika growing under artificial light. The amounts of carotenoids, chlorophyll, polyphenols, tannins, and flavonoids in red paprika (RP), cultivated in Korea, before and after light treatments under high pressure sodium (HPS) and lighting emitting plasma (LEP) lamps (RPControl, RPHPS, RPLEP), were analyzed in water (W) and ethanolic extracts (Et). Spectroscopic, radical scavenging assays, fluorescence and cytotoxicity measurements were applied. The results of this study showed that total chlorophyll and carotenes were the highest in RPHPS (10.50 ± 1.02 and 33.90 ± 3.26 µg/g dry weight (DW)). The strongest antioxidant capacity (µM TE/g DW) in a 2, 2'-azino-bis (3-ethyl-benzothiazoline-6-sulfonic acid) diammonium salt (ABTS(•+)) assay was in RPControlEt (24.34 ± 2.36), in a ferric-reducing/antioxidant power (FRAP) assay in RPHPSW (27.08 ± 2.4) and in a cupric reducing antioxidant (CUPRAC) in RPLEPW (70.99 ± 7.11). The paprika ethanolic extracts showed lower values in their bioactivity than the water ones. The binding and cytotoxicity abilities of extracted polyphenols correlated with their amounts. LEP treatment is better for plant growth characteristics than other conventional treatments. The investigated paprika samples can be used as a source of antioxidants.

Seeds and leaves of indigo (Polygonum tinctorium Ait.) plant were investigated and compared with another medicinal plant named prolipid for their properties such as chemical composition, antioxidant, and anticancer effects by Fourier transform infrared, three-dimensional fluorescence spectroscopy, and electrospray ionization-MS in negative mode. It was found that polyphenols, flavonoids, and flavanols were significantly higher in prolipid (P<0.05), following by indigo mature leaves, immature leaves, and seeds. Methanol extract of mature indigo leaves in comparison with the ethyl acetate extract showed higher inhibition of proliferation. The interaction between polyphenol extracts of indigo mature leaves and BSA showed that indigo has a strong ability, as other widely used medicinal plants, to quench the intrinsic fluorescence of BSA by forming complexes. In conclusion, indigo mature leaves were compared with prolipid. High content of bioactive compounds, antioxidant, fluorescence, and antiproliferative properties of indigo justifies the use of this plant as a medicinal plant and a new source of antioxidants.

The aim of this investigation was to determine the bioactive compounds in kiwifruit as an indication of quality after extraction using methanol and ethyl acetate. Using FTIR and three-dimensional fluorescence spectroscopy and electrospray ionization/MS, the contents of polyphenols, flavonoids, flavanols, and tannins, and the level of the antioxidant activity by 2, 2-azino-bis (3-ethyl-benzothiazoline-6-sulfonic acid) diammonium salt, 1, 1-diphenyl-2-picrylhydrazyl, ferric-reducing/ antioxidant power, and cupric reducing antioxidant capacity assays were determined and compared. It was found that the methanol extracts of kiwifruit showed significantly higher amounts of bioactive acetate extracts. The cultivar Bidan, in comparison compounds and antioxidant activities than the ethyl with the classic Hayward, showed significantly higher bioactivity. For the first time, Bidan organic kiwifruit was analyzed for its antioxidant activities and compared with the widely consumed Hayward organic based on its bioactive compounds and fluorescence properties. Relatively high content of bioactive compounds and positive antioxidant and antiproliferative properties of kiwifruit determined by the advanced analytical methods justify its use as a source of valuable antioxidants. The methods used are applicable for bioactivity determination, in general, for any food products.

Anthocyanin content and the activities of polyphenol oxidase (PPO), peroxidase (POD) and phenylalanine ammonia-lyase (PAL) and their relationships were determined in the leaves of six lettuce (Lactuca sativa L.) cultivars, exposed for 6 weeks to alternating three different day/night temperatures. Anthocyanin content was found to be highest at 20/13°C, followed by 25/20°C and 30/25°C, showing accumulation of anthocyanin at low temperatures. Activities of PPO and PAL were also found to be highest at low day/night temperatures, whereas the POD activity was decreased at low day/night temperatures. The most significant positive correlation existed between anthocyanin content and PPO activity (r² = 0.71). The results suggest that various day/night temperature regimes affect anthocyanin content and the activities of PPO, POD and PAL in lettuce.

The contents of two bioactive compounds (polyphenols and flavonoids) and their antioxidant and enzyme activities were determined in the leaves of six lettuce (Latuca sativa L.) cultivars subjected to 4 different day/night temperatures for 6 weeks. The total polyphenol and anthocyanin contents and the corresponding antioxidant activities were the highest at 13/10°C and 20/13°C, followed by 25/20°C and 30/25°C. The enzymatic activities of polyphenol oxidase (PPO) and phenylalanine ammonia-lyase (PAL) were also the highest at low day/night temperatures, but the peroxidase (POD) activity was decreased at low day/night temperatures and increased at high day/night temperatures. The most significant positive correlation existed between anthocyanin content and PPO activity, total polyphenols and their antioxidant activities. The results showed that at relatively low temperatures, lettuce plants have a high antioxidant and enzymatic status. These results provide additional information for the lettuce growers.

This study was conducted in order to compare the nutrient and chemical contents of two Korean teas: traditional Chungtaejeon tea (CTJ) with that of green tea (GT). Main bioactive compounds and the antioxidant activities using four radical scavenging assays (ABTS, CUPRAC, FRAP and DPPH) in methanol and acetone extracts of both teas were determined. It was found that the contents of vitamin C, amino acids and total nitrogen in CTJ were lower than that of GT (p < 0.05). Caffeine, reducing sugar and chlorophyll contents in CTJ were similar to GT. Catechin (C), epicatechin (EC), and epigallocatechin (EGC) contents were lower in CTJ than in green tea (p < 0.05), but gallocatechin (GC), epicatechin gallate (ECG), epigallocatechin gallate (EGCG) and catechin gallate (CG) showed no significant differences between CTJ and GT (p >/= 0.05). The contents of polyphenols, flavonoids, flavanols and tannins in CTJ methanol were 229.30 +/- 11.3 mg GAE/g dry weight (DW), 15.24 +/- 0.8 mg CE/g DW, 109.10 +/- 5.1 mg CE/g DW and 25.68 +/- 1.2 mg CE/g DW, respectively, and significantly higher than in acetone extracts (p < 0.05). Flavonoids (quercetin and kaempferol) were higher in GT than in CTJ and myricetin was higher in CTJ (p < 0.05). Threonine and aspartic acid was lower, and glutamic acid was higher in CTJ compared with GT (p < 0.05). Free amino acid content in CTJ and GT showed no significant difference. Potassium and magnesium in CTJ were lower compared to GT, but no significant difference was found for iron, manganese and calcium. Also, the level of the antioxidant activity by all four used assays was significantly higher in CTJ and in methanol was higher than in acetone extracts (p < 0.05). In conclusion, traditional fermented Korean tea Chungtaejeon contains high quantities of bioactive compounds and possesses high antioxidant activity. The contents of the bioactive compounds and the levels of antioxidant activities are significantly higher in methanol than in acetone extracts. 3-D fluorescence and FTIR- spectroscopy showed slight differences between the two investigated tea samples and can be used as additional tools for identification of polyphenols. Both studied teas can be recommended as a source of bioactive compounds.

Garlic and white and red varieties of onion were subjected to processing by a variety of culinary methods, and bioactive compounds then determined. For in vivo studies, 84 male Wistar rats were randomly divided into 14 diet groups, each of six rats, including two control groups (one with no supplementation and one with cholesterol supplementation only). During the 30-day trial, the basal diets of the other 12 groups were supplemented with 1% cholesterol and raw or processed vegetables. Both raw red onion and red onion subjected to blanching for 90 s hindered the rise in plasma lipids more than the other vegetables studied in the supplemented diets. The decrease in antioxidant activity compared to the cholesterol-supplemented control group was significantly less for the group fed with red onion subjected to blanching for 90 s. No histological changes were detected in the studied organs of rats that had been fed cholesterol. In conclusion, blanching for 90 s most fully preserved the bioactive compounds and antioxidant potentials, and hindered the rise in plasma lipid levels and the decrease in plasma antioxidant activity of rats fed cholesterol. Alkaline phosphatase levels correlated with classical atherosclerosis indices, and determination of alkaline phosphatase is suggested as an additional index in atherosclerosis testing.

This study was conducted to determine the antiproliferative activity of 24 Korean wild vegetables. The methanol extracts of these wild vegetables were used against lung, breast, colon and gastric cancer cells, and the results were assessed by MTT assay. It was found that at the extract concentration of 400 mgL(-1) 14 plants exercised antiproliferative activity over 80% against the lung cancer cells, one plant among six--against breast cancer cells, and two plants among six--against colon cancer cells, respectively. Eighteen wild vegetables had the hyperplasia inhibition activity against gastric cancer cells over 23.6% at all extract concentrations, however, only six plants had the antiproliferative activity over 80% in 600 mgL(-1). It was found that the extracts from Youngia sonchifolia, Synurus deltoides, Syneilesis palmata, and Cephalonoplos segetum, in concentration of 400 mgL(-1) inhibited the hyperplasia of lung cancer cells over 95% and Angelica gigas-both lung and colon cancer cells over 95%. In conclusion, the studied wild vegetables' methanol extracts possess dose dependent antiproliferative properties, based on their bioactive compounds, mainly polyphenols, but some of them as Hypericum ascyron against lung cancer are not effective and even course harm.

In traditional Korean medicine, lotus (Nelumbo nucifera Gaertn) roots have been used as an antidiabetic and an antiproliferative remedy. However, scientific publications on lotus properties are very limited. Therefore, it was decided to investigate the Korean white lotus cultivars in order to find out their bioactivity. It was found that all lotus cultivars (Inchisa, Muan, Garam, and Chungyang) possess high amounts of bioactive compounds: total phenols, between 7.95 +/- 0.8 and 4.21 +/- 0.3 mg of gallic acid equivalents (GAE)/g dry weight (DW); ascorbic acid, between 15.8 +/- 1.1 and 22.3 +/- 1.7 mg of ascorbic acid/g DW; and amino acids, between 15.05 +/- 0.82% and 16.62 +/- 0.90% DW. The highest contents of polyphenols (7.95 +/- 0.8 mg of GAE/g DW) and the highest levels of antioxidant [by 2,2-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) and 1,1-diphenyl-2-picrylhydrazyl assays, 54.27 +/- 6.1 and 21.98 +/- 2.5 microM Trolox equivalents/g DW, respectively] and antiproliferative activities on both human cancer cell lines (Calu-6 for human pulmonary carcinoma and SMU-601 for human gastric carcinoma, 59.75 +/- 3.99% and 71.21 +/- 2.79% cell viability, respectively) were found in the Chungyang cultivar. Fluorometry and Fourier transform infrared spectroscopy can be applied as rapid methods for determination of bioactive compounds, such as polyphenols. The correlation between the bioactive compounds and the antioxidant activity was high. In conclusion, all Korean white lotus cultivars are valuable medicinal foods, and in order to receive the best results a combination of lotus cultivars has to be consumed.

In the time of globalization many of the tropical fruits can be find at the markets of Europe and North America. Most customers are not familiar with the nutritional and proliferative values of these fruits. Therefore, a less known snake fruit was compared with better known kiwi fruit, using fluorometry, FT-IR spectroscopy, several radical scavenging and proliferative assays and statistical evaluation. It was found similarity between snake fruit (cultivar Sumalee) and kiwi fruit (cultivar Hayward) in the contents of polyphenols (8.15-7.91, mg GAE g(-1) DW), antioxidant values by DPPH (11.28-10.24, microMTE g(-1) DW), and antiproliferative activities on both human cancer cell lines (Calu-6 for human pulmonary carcinoma, and SMU-601 for human gastric carcinoma, 90.5-87.6 and 89.3-87.1%, cell survival, respectively). In conclusion, snake fruit cultivar Sumalee is comparable with kiwi fruit cultivar Hayward. Two fruits can be used as supplements to the normal diet. Consumption of a combination of both fruits could be recommended in order to receive the best results.

UNLABELLED: The aim of this investigation was to study the antioxidant and anticancer activities of young sprouts of some traditional Korean salad plants. Total phenolics, antioxidant and anticancer activities of the methanol extracts from young sprouts of 11 salad plants were determined. The highest amount of phenolics was found in methanol extracts of Euonymus alatus (235.7 mg kg(-1)), followed by Hypericum ascyron (197.1 mg kg(-1)), Zanthoxylum piperitum (194.1 mg kg(-1)) and Zanthoxylum schinifolium (142.5 mg kg(-1)). Methanol extracts of E. alatus, H. ascyron, and Z. piperitum at 63 mg kg(-1) exhibited the highest dose-depend DPPH radical scavenging activity by 91.2, 91.2 and 83.9%, respectively. According to the MTT results, the methanol extracts from Stellaria aquatica, Eleutherococcus sessilifolrus and Z. schinifolium showed the highest anticancer activities against Calu-6 (IC50<25.0 microg ml(-1)) and from S. aquatica-the highest anticancer activities against SNU-601 (153.3 microg ml(-1)), following by E. sessilifolrus (196.7 microg ml(-1)) and Amaranthus mangostanus (303.1 microg ml(-1)). Total phenolics were highly correlated with the DPPH, suggesting that they contribute to the antioxidant properties of the studied plants. IN CONCLUSION: young sprouts of Korean salad possess antioxidant and anticancer properties and could be used as a supplement to proper drugs.

The antioxidant activities of different durian cultivars at the same stage of ripening (Mon Thong, Chani, Kan Yao, Pung Manee and Kradum) were compared in order to choose the best as a supplement in the human diet. Total polyphenols (mg gallic acid equivalent/100 g fresh weight (FW)) and flavonoids (mg catechin equivalent (CE)/100 g FW) in Mon Thong (361.4+/-23.2 and 93.9+/-7.4) were significantly higher (P<0.05) than in Kradum (271.5+/-11.2 and 69.2+/-5.3) and Kan Yao (283.2+/-16.5 and 72.1+/-6.8). The free polyphenols and flavonoids showed lower results than the hydrolyzed ones. Anthocyanins (microg cyanidin-3-glucoside equivalent/100 g FW) and flavanols (microg CE/100 g FW) were significantly higher in Mon Thong (427.3+/-23.8 and 171.4+/-16.3) than in Kradum (320.2+/-12.1 and 128.6+/-9.7) and Kan Yao (335.3+/-14.1 and 134.4+/-11.7). Ultraviolet spectroscopy and high-performance liquid chromatography/diode array detection analyses showed that caffeic acid and quercetin were the dominant bioactive substances in Mon Thong cultivar. The antioxidant activity (microM trolox equivalent/100 g FW) of Mon Thong cultivar (260.8+/-20.2, 1,075.6+/-81.4 and 2,352.7+/-124.2) determined by ferric-reducing/antioxidant power (FRAP), cupric reducing antioxidant capacity (CUPRAC) and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) with Trolox equivalent antioxidant capacity (TEAC) assays was significantly higher (P<0.05) than in Kradum (197.4+/-8.9, 806.5+/-31.2 and 1,773.2+/-102.5) and in Kan Yao (204.7+/-9.7, 845.5+/-48.6 and 1,843.6+/-107.5). The correlation coefficients between polyphenols, flavonoids, flavanols and FRAP, CUPRAC and TEAC capacities were between 0.89 and 0.98. In extracted and separated by electrophoresis durian proteins, some differences were found in the sodium dodecyl sulfate-protein bands in the region of 16 and 68 kDa for Kradum, 45 kDa for Mon Thong and three bands for Kan Yao. Antioxidants and proteins can be used for characterization of the quality of durian cultivars. In conclusion, the bioactivity of durian cultivars Mon Thong, Chani and Pung Manee was high and the total polyphenols were the main contributors to the overall antioxidant capacity. The results of our investigation in vitro are comparable with other fruits that widely used in human diets. Therefore, Durian can be used as a supplement for nutritional and healthy purposes, especially Durian Mon Thong, Chani and Pung Manee.

The polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs) and organotins were analyzed in mussels Mytilus galloprovincialis from polluted and unpolluted sites from Mokpo Bay, Korea. The total PAH's concentrations (10(-3)mgkg(-1)) measured by GC-MS were in the range from 31+/-23 to 1+/-1. Among the eight PAHs the predominant ones were fluoranthene, phenanthrene and pyrene and accounted approximately 63% of the total PAHs. Among the four detected PCBs the highest content was of PCB 153, which accounted about 47% of the total PCBs. The main organotin compounds were dibutyltindichloride (DBT) and tributyltinchloride (TBT) and their composition was approximately 33% and 24%. PAHs, PCBs and organotins were found only in the mussels from polluted site. The antioxidant activity by ABTS [2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid)] test was higher in mussels from polluted than from unpolluted sites (P<0.05). It was found a correlation between the determined compounds (PAHs, PCBs and organotins) and the antioxidant activity of the mussel tissue from polluted site and the correlation coefficients were 0.96, 0.92 and 0.80, respectively. Such correlation can be explained by the properties of mussels. Since the mussel cell wall and tissues are hydrophobic, they can concentrate a number of hydrophobic pollutants like PAHs and PCBs from the marine environment by solubility rules. On the other hand, proteins are lipophilic compounds having antioxidant properties. Certain amino acid residues and thiol (-SH) groups, contained in proteins, respond to the ABTS antioxidant activity assay. Thus there may be a correlation between the total antioxidant activity of the organism and the PAH-PCB pollutants which were concentrated from its environment. The studied properties of mussels from polluted site can be used as an additional indicator of pollution.

The influence of diets supplemented with mussels, from polluted (MPoll) and non-polluted (MNPoll) areas, on some atherosclerosis indices in rats fed cholesterol (Chol) were studied. According to the results of our investigation in vitro, mussels from polluted areas had higher contents of proteins, metals and antioxidant compounds, mostly phenolics and higher antioxidant capacities. 28 male Wistar rats were divided into 4 groups of 7 and named Control, Chol, Chol/MPoll and Chol/MNPoll. The rats of the Control group received basal diet (BD) only, and the diets of the other 3 groups were supplemented with 1% of non-oxidized cholesterol (NOC), 1% of NOC and 5.6% of mussel dry matter (DM) from polluted and 1% of NOC and 5.6% of mussel DM from non-polluted areas for Chol, Chol/MPoll and Chol/MNPoll, respectively. The histology of the aorta and brain in rats fed cholesterol did not show any signs of atherosclerosis. Some differences were registered in the electrophoretic protein patterns of plasma in rats, with mussel-supplemented diets. In full plasma electrophoretic patterns of the Chol/MPoll diet group more proteins were detected than in both Chol and Control groups, and the differences were significant. In conclusion, in groups of rats fed cholesterol with mussels supplementation, a significant hindering in the rise of plasma lipid levels and also hindering in the decrease of plasma antioxidant activity were registered.

The aim of this investigation was to determine the content of total phenolics, antioxidant activity and cytotoxicity of methanol extracts from the aerial parts of 11 Korean medicinal salad plants. The highest total phenolic content of the methanol extracts was found in Aster scaber (17.1 mg 100 g(-1)), followed by Ixeris dentate (16.4 mg 100 g(-1)), Aster yomena (12.0 mg 100 g(-1)) and Sedum sarmentosum (9.1 mg 100 g(-1)) of FW. Methanol extracts of Ixeris dentate and Aster scaber at 50 microg mL(-1) exhibited the highest DPPH radical scavenging activity by 86.4 and 83.3%, respectively. It was registered a dose-dependent increase of DPPH free radical scavenging activity. Total phenolic content of the studied plant extracts was correlated with the DPPH radical scavenging activity. It was found by means of MTT assay, that cytotoxicity of the methanol extracts was the highest against HCT-116. Methanol extracts from Petasites japonicus (IC(50)<25.0 microg mL(-1)) showed the highest activity against HCT-116, following by Angelica gigas (34.75 microg mL(-1)), Erythronium japonicum (44.06 microg mL(-1)), and Aster scaber (54.87 microg mL(-1)). In conclusion, the studied salad plants have high total phenolics content and high antioxidant activity. These plants dose-dependently increased DPPH free radical scavenging activity. The total phenolics level was highly correlated with the free radical scavenging activity. Most of the studied salad plants have potent cytotoxicity activity. The results of this investigation suggest that the extracts of studied salad plants could be an addition to basic medicine for some diseases.
