Present study aimed to investigate the effect of adding antioxidants, cysteine and ascorbic acid on the levels of glutamic oxaloacetic transaminase (GOT), glutamic-pyruvate (GPT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH) and γ-glutamyl transpeptidase (GGT) enzymes of post-thawed stallion sperm. Ten ejaculates were collected each from four healthy stallions and cryopreserved using HF-20 freezing extender containing either 0 mg/ml cysteine or ascorbic acid, 0.5 mg/ml cysteine and 0.5 mg/ml ascorbic acid. All samples in freezing extender containing cysteine or ascorbic acid or none of them were assessed for sperm motility, viability, plasma membrane integrity, morphology and enzymes concentration. The ALP, LDH and GGT were significantly higher in 0-group compared with cysteine and ascorbic acid groups. The sperm motility of frozen-thawed semen with 0-group was significantly better compared with cysteine and ascorbic acid groups. The variation on viability, sperm membrane integrity and morphology were insignificant between all treated groups. Therefore, these enzymes were reduced when using antioxidants in the freezing extender. Results of the present study suggest that concentration of ALP, LDH and GGT enzymes could be used as parameters for prediction of frozen-thawed stallion semen.

OBJECTIVE To characterize signalment, clinical signs, reproductive history, surgical management, and outcomes of beef cattle undergoing cesarean section because of dystocia at a veterinary teaching hospital. DESIGN Retrospective case series with nested cohort study. ANIMALS 173 beef cattle admitted to a veterinary teaching hospital from 2001 through 2010 that underwent cesarean section because of dystocia. PROCEDURES Medical records were reviewed and information collected on cattle signalment; reproductive history; cause of dystocia; anesthetic protocol; surgical management; number, sex, and body weight of calves delivered (alive or dead); perioperative treatment; duration of hospitalization; and discharge status. A questionnaire regarding postoperative fertility was mailed to all owners, and owners who did not respond were contacted via telephone. RESULTS Overall mortality rate for calves was high, with 37.6% (62/165) of calves delivered dead or dying ≤ 24 hours after cesarean section. Mortality rate was higher for female versus male calves and for calves from dams with signs of labor for ≥ 3 hours versus < 3 hours before hospital admission. Overall mortality rate for dams was low, with only 10 of 161 (6.2%) dams failing to survive for ≥ 21 days after hospital discharge. Postoperative fertility rate was acceptable, with 75% (44/59) of dams that were rebred after cesarean section giving birth to ≥ 1 live calf. CONCLUSIONS AND CLINICAL RELEVANCE Cesarean section was a clinically useful method for resolving dystocia in beef cattle, providing a high dam survival rate and an acceptable postoperative fertility rate. Beef cattle producers should seek veterinary assistance whenever clinical signs of dystocia are noticed, preferably within 6 hours after onset of parturition.

Calcium homeostatic mechanisms are challenged in periparturient multiparous dairy cattle due to the rapid transport of large amounts of calcium into the mammary gland associated with colostrogenesis, resulting in decreased plasma total calcium concentration ([Ca]). An unresolved issue is the timing of the decrease in plasma [Ca] relative to the time of parturition, with the consensus view being that plasma [Ca] does not decrease until after parturition. The objective of this study, therefore, was to characterize the change in plasma [Ca] over time in periparturient dairy cattle. Plasma and mid-stream urine samples were collected daily starting 3 d before calving from 104 periparturient Holstein-Friesian dairy cows in a herd fed an acidogenic total mixed ration during the late dry period. Mixed-models ANOVA and linear and multivariable regression analyses were conducted. Plasma [Ca] decreased in periparturient multiparous cattle (n = 70) but not in primiparous cattle (n = 34). Compared with mean values approximately 72 h before parturition ([Ca] = 2.32 mmol/L), mean plasma [Ca] in multiparous cattle first decreased at 9 h before parturition (2.13 mmol/L) and remained decreased for up to 48 h after parturition, with the lowest mean value (1.87 mmol/L) occurring at 28 h after parturition. Mean 24-h urine Ca excretion was calculated to decrease by 3.5 to 3.8 g in periparturient multiparous cattle. Regression analysis indicated that plasma [Ca] in the 12-h period before and 24-h period after parturition was strongly and negatively associated with age but was also negatively associated with milk production indices. We conclude that plasma [Ca] was decreased at least 9 h before parturition in multiparous dairy cattle fed an acidogenic diet in late gestation, and that calcium homeostasis was disrupted for 2 to 3 d around parturition.

BACKGROUND: The Precision Xtra electrochemical meter is widely used to measure blood ß-hydroxybutyrate concentration (BHBb) in dairy cattle. The meter uses an algorithm optimized for human blood that assumes the HCT in cattle is the same as in people, and that intra-erythrocyte ß-hydroxybutyrate (BHBe) and plasma ß-hydroxybutyrate (BHBp) concentration are equivalent. OBJECTIVES: The first objective was to characterize the analytic performance of the meter for measuring BHBb and BHBp in dairy cattle. The second objective was to characterize the influence of HCT and sample temperature on BHBp concentration measured by the meter. METHODS: Blood and plasma samples were obtained from 106 periparturient Holstein cattle and 15 lactating Holstein cows with experimentally induced electrolyte and acid-base imbalances. Meter performance was evaluated using Deming regression and Bland-Altman plots. Multivariable linear regression was used to determine the effect of HCT and sample temperature on BHBb and BHBp concentration, respectively. RESULTS: The meter was linear up to BHB = 3.0 mmol/L as measured by the reference method, equivalent to meter values for BHBb > 4.5 mmol/L and BHBp > 5.2 mmol/L. An increase in HCT resulted in higher BHBb concentration. This result was partially explained by BHBe being much lower than BHBp. Changes in sample temperature caused a linear change in measured BHBp whenever BHBp > 3.0 mmol/L. CONCLUSIONS: Meter accuracy was markedly dependent on the BHBe-to-BHBp ratio and consequently the HCT. Therefore, the algorithm used by the meter should be revised when applied to bovine blood for improved accuracy.

The objective of this study was to determine the clinical utility of measuring calf front hoof circumference, maternal intrapelvic area, and selected morphometric values in predicting dystocia in dairy cattle. An observational study using a convenience sample of 103 late-gestation Holstein-Friesian heifers and cows was performed. Intrapelvic height and width of the dam were measured using a pelvimeter, and the intrapelvic area was calculated. Calf front hoof circumference and birth weight were also measured. Data were analyzed using Spearman's correlation coefficient (rs), Mann-Whitney U test, and binary or ordered logistic regression; P < 0.05 was significant. The calving difficulty score (1-5) was greater in heifers (median, 3.0) than in cows (median, 1.0). Median intrapelvic area immediately before parturition was smaller in heifers (268 cm(2)) than in cows (332 cm(2)), whereas front hoof circumference and birth weight of the calf were similar in both groups. The calving difficulty score was positively associated with calf birth weight in heifers (rs = 0.39) and cows (rs = 0.24). Binary logistic regression using both dam and calf data indicated that the ratio of front hoof circumference of the calf to the maternal intrapelvic area provided the best predictor of dystocia (calving difficulty score = 4 or 5), with sensitivity = 0.50 and specificity = 0.93 at the optimal cutpoint for the ratio (>0.068 cm/cm(2)). Determining the ratio of calf front hoof circumference to maternal intrapelvic area has clinical utility in predicting the calving difficulty score in Holstein-Friesian cattle.
