RESUMO

Oligomeric proanthocyanidins (OPCs) are found mostly in red grape seeds. Many publications have reported that OPCs possess an excellent anti-oxidant effects. Since it could against cellular damage from reactive oxygen species (ROS) led to reduce the risk of chronic disease and cancers. We carried out this study on the Thai OPCs to evaluate the mutagenicity/ anti-mutagenicity and anti-oxidative DNA damage effects in TK6 cells by micronucleus (MN) and comet assays. In the MN assay, OPCs-treatment of TK6 cells at concentrations ranging from 10-200 ?g/ml (4 and 24 h) did not cause micronucleus induction over the negative control group but revealed a significant reduction the micronucleus frequencies against the known mutagen (mitomycin C). In the comet assay, OPCs-treated TK6 cells at concentrations of 100, 250, 500, and 1,000 ?g/ml could inhibit DNA damage induced by H(2)O(2) as indicated by 18.7, 36.4, 30.6, and 60.1%, respectively. Our results suggest that OPCs possess the anti-mutagenic and anti-oxidative DNA damage effects in TK6 cells under the conditions of this assay.

Cleistanthin A (CleinA) and cleistanthoside A (CleisA) isolated from plant Phyllanthus taxodiifolius Beille have previously shown potent anticancer effects. To promote their medicinal benefits, CleisA was modified to cleistanthoside A tetraacetate (CleisTA) and evaluated for genotoxic and anti-mutagenic properties in comparison with CleinA. Both compounds showed no significant mutagenic activity to S. typhimulium bacteria and no cytotoxic effect to normal mammalian cells. The non genotoxic effect of CleinA was further confirmed by un-alteration of cytokinesis-block proliferation index (CBPI) and micronucleus (MN) frequency assays in Chinese hamster lung fibroblast (V79) cells, and of CleisTA was confirmed by un-changes of human peripheral blood lymphocytes (HPBL) chromosomal structure assay. Moreover, the metabolic form of CleinA efficiently demonstrated cytostasis effect to V79 cell and prevented mutagen induced Salmonella TA98 and TA100 reversion, whereas both metabolic and non-metabolic forms of CleisTA reduced HPBL mitotic index (%M.I) in a concentration-dependent relationship. The results support CleinA and CleisTA as the new lead compounds for anti-cancer drug development.

Piperine is a major pungent substance and active component of black pepper (Piper nigrum Linn.) and long pepper (Piper longum Linn.). Both plants are used worldwide as household spices and condiments. They are also used as important ingredients in folklore medicine in many Asian countries. Therefore, it is of interest to study antimutagenic effects of piperine. In this study, its influence on chromosomes was investigated in rat bone marrow cells. Male Wistar rats were orally administered piperine at the doses of 100, 400 and 800 mg/kg body weight for 24 hours then challenged with cyclophosphamide at a dose of 50 mg/kg body weight by intraperitoneal injection. Twenty-four hours thereafter, all animals were sacrificed and bone marrow samples were collected for chromosomal analysis. The results demonstrated that piperine at a dose of 100 mg/kg body weight gave a statistically significant reduction in cyclophosphamide-induced chromosomal aberrations. In conclusion, piperine may have antimutagenic potential. The underlying molecular mechanisms now require attention.
