RESUMO

Background: Unilateral absence of the pulmonary artery (UAPA) is a very rare congenital anomaly. OBJECTIVE: To analyze the diagnostic strategy applied to seven patients with UAPA who were examined and subsequently treated at the National Lung Hospital, Hanoi, Vietnam. METHODS: All seven patients, including three pediatric cases (1, 2, and 14 years old) and four adult cases (21, 26, 44, and 53 years old), had a history of recurrent pneumonia, and the clinical symptoms on admission included cough, progressive dyspnea, chest pain, and fatigue. The patients were initially examined clinically, followed by hematological testing, blood biochemistry testing, and chest X-ray radiology. The results suggested UAPA, so echocardiography and contrast-enhanced chest computed tomography (CT) were performed as soon as practical. RESULTS: The echocardiographic and CT imaging findings confirmed the suspected diagnosis of UAPA in all seven patients, which was accompanied by congenital heart disease in three patients. Three of the seven patients had mild and medium pulmonary hypertension. All seven patients were treated with drugs, which led to improvement in symptoms. CONCLUSION: Frontal chest X-ray provided the initial signs suggesting a diagnosis of UAPA. Subsequent echocardiography and contrast-enhanced chest CT were effective diagnostic tools for fast and accurate confirmation of UAPA.

BACKGROUND: We have followed a birth cohort from 2008 to 2009 near a dioxin-contaminated area of Da Nang, Vietnam, and investigated the effects of perinatal dioxin exposure on neurodevelopment from infancy to pre-school age. The present study aimed to investigate the effects of perinatal dioxin exposure on the learning abilities of the elementary-school children from the Da Nang birth cohort. METHODS: From 241 mother-infant pairs recruited at baseline (134 boys and 107 girls), 185 (76.8%) participated in a follow-up when the children were 8 years of age (108 boys and 77 girls). The children's perinatal dioxin exposure was estimated using the dioxin levels in their mothers' breast milk. The Colorado Learning Difficulties Questionnaire (CLDQ) was used to evaluate the children's learning difficulties. Math- and language-achievement scores were obtained using paper-based tests. Reading fluency was examined by having the children read passages in Vietnamese. RESULTS: In boys exposed to high levels of 2,3,7,8-tetrachlorodibenzodioxin (2,3,7,8-TetraCDD), CLDQ reading scores were significantly higher (worse), and language achievement scores were significantly lower. Boys exposed to high levels of 2,3,7,8-TetraCDD as well as high levels of the toxic equivalent (TEQ) of polychlorodibenzodioxins and polychlorodibenzofurans (PCDDs/Fs) had higher numbers of reading errors. Reading errors were higher and math achievement scores were lower with increasing concentrations of 1,2,3,4,7,8-HexaCDD and 1,2,3,4,6,7,8-HeptaCDD. In girls, no significant differences of any learning ability markers were found between high and low exposure groups to TEQ-PCDDs/Fs and these 3 congeners. CONCLUSIONS: Perinatal dioxin exposure may have adverse effects on the learning abilities of school children, especially boys.

BACKGROUND: Aneuploidy is a major cause of miscarriages and implantation failure. Preimplantation genetic testing for aneuploidy (PGT-A) by Next Generation Sequencing (NGS) is able to detect of the numeral and structural chromosomal abnormalities of embryos in vitro fertilization (IVF). AIM: This study was aimed to assess the relationship between maternal age and chromosomal abnormalities NGS technology. METHODS: A group of 603 human trophectoderm (TE) biopsied samples were tested by Veriseq kit of Illumina. The relation of marternal age and chromosomal abnormality of blastocyst embryo was evaluated. RESULTS: Among the 603 TE samples, 247 samples (42.73%) presented as chromosomal abnormalities. The abnormalities occurred to almost chromosomes, and the most popular aneuploidy observed is 22. Aneuploidy rate from 0.87% in chromosome 11 to 6.06% in chromosome 22. The rate of abnormal chromosome increased dramatically in group of mother's ages over 37 (54.17%) comparing to group of mother's ages less than 37 (38.05%) (p < 0.000). The Abnormal chromosome and maternal age has a positive correlation with r = 0.4783 (p<0.0001). CONCLUSION: These results showed high rate abnormal chromosome and correlated with advanced maternal age of blastocyst embryos.

BACKGROUND: Vorinostat (VRS), a histone deacetylases inhibitor, has significant cytotoxic potential in a large number of human cancer cell lines. OBJECTIVE: To clarify its promising anticancer potential and to improve its drawback related to physical properties and in vivo performance of VRS. METHODS: VRS was successfully incorporated into nanostructured lipid carriers (NLCs) by the hot microemulsion method using sonication following a homogenization technique. RESULTS: After the optimization process, VRS-loaded NLCs (VRS-NLCs) were obtained as ideal quality nanoparticles with a spherical shape, small size (∼150 nm), negative charge (∼-22 mV), and narrow size distribution. In addition, the high entrapment efficiency (∼99%) and sustained drug release profile were recorded. Cytotoxicity study in three different cell lines (A549, MCF-7, and SCC-7) demonstrated higher cytotoxicity of VRS-NLCs than free drug. Finally, the AUC of VRS (118.16 ± 17.35 µgh/mL) was enhanced ∼4.4 times compared with that of free drug (27.03 ± 3.25 µgh/mL). CONCLUSION: These results suggest the potential of NLCs as an oral delivery system for enhancement of cellular uptake, in vitro cytotoxicity in cancer cell lines and the oral bioavailability of VRS.

We investigated the pharmacokinetic profile of (R)- and (S)-zaltoprofen (ZPF) in rats using rapid and selective liquid chromatography with solid-phase extraction (SPE). The ZPF enantiomers were extracted from a small volume of plasma (0.2 mL) by means of SPE using cartridges and were analyzed on a Chiralcel OJ-H (4.6 mm × 150 mm, 5 µm) column with ultraviolet detection at 244 nm. The lower limit of quantification of the ZPF enantiomers in plasma was 0.1µg/mL. The validated method was successfully applied to chiral pharmacokinetic studies of oral administration of racemic ZPF to rats. (S)-ZPF showed significantly higher AUC, T(max), and C(max) and a longer half-life than (R)-ZPF, indicating higher bioavailability of the (S)-isomer. A total of 8 samples (about 12% of the total number of samples) were selected for incurred sample reanalysis (ISR). The % difference between the re-assay concentrations and the original concentrations were all less than 15% of their mean values and met the acceptance criteria for ISR.

RESUMO

For the purpose of reconstructive surgery training and research, we have developed a new skin flap model: canine lateral thoracic fasciocutaneous flap. Anatomical study found that the lateral thoracic arteries in dogs have similar anatomical characteristics to human's ones. Based on these vessels, if a skin flap was designed within the vessels territory (size 5 x 8 cm) it could survive completely, whereas, if designed beyond the vessels territory (size 5 x 14 cm) would result in partial necrosis of the flap. This fasciocutaneous flap model closely simulates the human surgery and could be valuable for training and research. Furthermore, this flap could be applied in the veterinary practice for reconstruction of canine forelimbs and cervical area.
