RESUMO

Ganoderma is a genus of biomedical fungus that is used in the development of numerous health products throughout the world. The Lower Volta River Basin of Ghana is an undulating land surface covered by extensive vegetation and water bodies and is rich in polypore mushrooms resembling various members of the Ganoderma genus. Despite the extensive biopharmaceutical benefits of Ganoderma spp., the isolates from the Lower Volta River Basin have not been properly characterized, thus limiting their use in the development of biotechnological products. In this study, Ganoderma spp. collected from the Lower Volta River Basin were genetically analyzed using the nuclear ribosomal sequences, the internal transcribed spacer 2 (ITS 2), the complete internal transcribed spacer (ITS), and the nuclear large subunit (nLSU). Blastn search and sequence analysis revealed that the sample we coded as Ganoderma LVRB-2 belongs to G. mbrekobenum, whereas Ganoderma LVRB-1, Ganoderma LVRB-14, and Ganoderma LVRB-16 belong to the species G. enigmaticum. Our analysis further demonstrates that Ganoderma LVRB-17 belongs to the species G. resinaceum. Thus, the five samples collected in the present study were positioned in three different distinct groups, namely G. mbrekobenum, G. enigmaticum, and G. resinaceum. The current data may serve as reference points for future studies.

In this work, we sought to determine the differences and/or similarities in the metabolite composition of the mycelial biomass of three ganoderma isolates (Ganoderma LVRB-1, Ganoderma LVRB-9 and Ganoderma LVRB-17) from the Lower Volta River Basin of Ghana. The cultured mycelial mass of the three isolates were subjected to DNA sequencing. BLASTn searches of the internal transcribed spacer. (ITS) sequences of the isolates were conducted in the GenBank and the data obtained subjected to ITS phylogenetic analysis. Thereafter, extracts of the cultured mycelial biomass of the three isolates were subjected to untargeted ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS)-based metabolomic analysis. A cursory examination of the total ion chromatograms of the isolates gave evidence of the differential levels of the metabolites present. Further analysis of the metabolomic data using multivariate analysis better captured these marked differences in terms of the presence and/or levels of the metabolites. Finally, four lanostane triterpenoids, namely ganoderic acid C6, ganoderenic acid A, Ganoderenic acid D and ganoderic acid G, together with two annotated compounds (ganoderic acids K and AM1) were detected in the mycelia biomass of the three ganoderma isolates from the Lower Volta River Basin of Ghana. The results provide the first ever metabolomic data on the chemical constituents of the mycelial biomass of ganoderma isolates from the Lower Volta River Basin of Ghana.

RESUMO

The pale brittle stem mushroom, Psathyrella candolleana, a species new to Iraq, is described from the sub-arid region of Aljazira (Iraq). Both classical taxonomy and DNA analyses confirm the identification of the fungus strain (RM-0861) as P. candolleana, a species that belongs to the family Psatherellaceae known to possess medicinal properties. Being a saprophyte, this fungus is cultivatable in laboratory conditions and therefore shows potential for production and use as a medicinal mushroom in human and veterinary health.

RESUMO

The importance and prospect of growing mushrooms through utilization of low-cost, technologically simple methods for developing Third World countries has recently been outlined. Three different species from genus Pleurotus (P. djamor, P. pulmonarius, and P. sajor-caju) and one from genus Hypsizygus (H. ulmarius) were studied. Whole stalk wheat straw, shredded wheat straw, and ground maize cob (Zea mays) were used as the substrates. Wheat straw is the post-harvest stalk of Triticum aestivum. Biological efficiency (BE), growth dynamics, and photographs are provided for each cultivated strain, on different substrates, and substrate treatment comparisons are made. From several experiments conducted with various technologically simple methods of cultivation, it was found that all above mentioned species can be successfully cultivated at the village level in any country, be it highly developed or the poorest country on earth.

In the present investigation 16 species of Phellinus and 6 species of Hymenochaete (collected as an adulterant) were screened for sesquiterpenoids and triterpenoids. A total of 11 spots were detected in both sesquiterpenoids and triterpenoids chromatograms. The terpenoids were not identified. The spots obtained for Phellinus were compared with the spots of Hymenochaete. It was revealed from the SAB and D values that almost half of the species of Phellinus have about 45% to 57% dissimilarity in case of sesquiterpenoids and 50% to 60% dissimilarity with the triterpenoids. However, since both the taxa are from the same family, the spots that were common may be characteristic of the family. Such a study may be used to reveal the adulteration in medicinal Phellinus samples.

Immune enhancement through the use of natural products is a potentially valuable therapeutic modality in HIV-infected people, especially those who are not good candidates for aggressive ARV therapy. One such immune enhancement, a medicinal mushroom product from the United States, is Immune Assist 24/7. In this study the effect of Immune Assist 24/7, which is a naturally derived immune-modulating and antiviral agent, on CD4+ T-lymphocyte counts was evaluated in 8 HIV-infected patients at the Sunyani Regional Hospital (Ghana). The subjects were administered three tablets of 800 mg Immune Assist 24/7 once daily (2.4 g/day) and peripheral blood samples were drawn at baseline, day 30, and day 60, and the CD4+ count measured. The study revealed that Immune Assist 24/7, used as a sole therapeutic agent without additional ARV drugs, significantly increased CD4+ T-lymphocyte populations in all of the patients. In one patient, the CD4+ T-lymphocyte count went from 4 at the baseline, to 170 cells in 60 days, representing an increase of more than 4000%. In another patient, the CD4+ count went from 88 to 470 cells within the same period. Even in the patients with the highest CD4+ counts of around 800, there was a significant elevation in the CD4+ count noted. This study did not deal with the effect of Immune Assist 24/7 on other immune parameters such as CD3+ T-lymphocyte count, natural killer cells count, or viral load among HIV-infected patients. These initial results are promising, and indicate the potential value of further evaluating the effects of Immune Assist 24/7 on other immune parameters and viral load among HIV patients, administered either as a sole therapeutic agent, as an adjuvant with standard ARV therapy, or in comparison with standard ARV therapy alone.

The morphological and cultural characteristics of vegetative mycelia of 29 Tibetan strains of medicinal caterpillar fungus Ophiocordyceps sinensis (= Cordyceps sinensis) were studied. Data on mycelial growth of the above-mentioned fungi strains on different types of nutrients, the macro- and micromorphological description of colonies grown on different agar media, and anamorph stage identification are provided. It was shown that strains of O. sinensis demonstrated moderately slow growth on selected nutrients compared with other ascomycetous fungi. The highest growth rate value from all analyzed strains is O. sinenis N14-2.7 mm/day was completed with a mycelial run on potato-dextrose agar (pH = 6.0) in 15 d. Most of the examined strains preferred Sabouraun’s dextrose agar; some of the strains preferred potato-dextrose agar as the medium for optimal development. The least favorable nutrient for all strains was Czapek solution agar. Analyses of morphological and microstructural peculiarities on different types of nutrients were conducted and detailed descriptions and illustrations were provided. Based on macro- and micromorphological characteristics, the investigated strains were identified as Hirsutella sinensis and Tolypocladium sinensis species, which were identified as the anamorphs of Ophiocordyceps sinensis.
