RESUMO

@#Objective Focusing on four types acute myeloid leukemia ( AML) fusion oncogenes,so as to explore the network difference with time series expression data and further identify important genes in networks. Methods Gene network difference analysis was conducted while focusing on the global attributes of the union network. The CompNet neighborhood similarity index ( CNSI) was adopted to assess network similarity.“fast-greedy”algorithm was used to detect communities based on the union network,and further identify hub genes. Results The CNSI value between NUP98-HOXA9-3 d and NUP98-HOXA9-8 d was 0. 73,while AML1-ETO-6 h and PML-RARA-6 h was 0.25. We identified ten AML associated genes and sev- en of them ( TNF,VEGFA,EP300,EGF,CD44,PTGS2,SMAD3) were reported in the literature. Conclu- sions The network difference analysis revealed the pattern and heterogeneity of AML gene expression change across different time points,and further provided target genes for efficient treatment of AML with different types of fusion oncogenes.

RESUMO

Objective The Rothman-Keller model was used to establish a predictive model for the risk of birth defects on the basis of evidence-based medicine, which so as to provide the basis for pertinent interventions in China. Methods First, the odds ratio (OR) value of risk factors for birth defects was obtained by evaluating the literature of meta-analysis, and the risk score table of the Rothman-Keller model was constructed. Then the simulation data was used to build the model, the risk boundary value of risk prediction, and finally the actual data to was used for verification. Results The main risk factors for 20 birth defects were collected through 17 articles. In the actual data of Shanxi Province, the actual incidence rate of high-risk populations screened by Rothman-Keller model was 10.9%, and it was statistically different from other groups ( 2 =147.58,P<0.001). In addition, the rothman-keller model identified all patients with a family history of birth defects as high-risk. Conclusions Through the meta-analysis literature on birth defects in China, the study find the main risk factors and construct a risk prediction model. It can be used to predict the risk of birth defects and help screen high-risk groups. At the same time, it provides ideas for predicting the risk of other diseases.

RESUMO

Objective To observe the clinical efficacy of Recipe for Clearing Heat and Removing Dampness combined with Butyl Flufenamate Ointment for the treatment of facial seborrheic dermatitis. Methods A total of 92 patients with facial seborrheic dermatitis were randomly divided into treatment group and control group,46 cases in each group. The treatment group was treated with oral use of Recipe for Clearing Heat and Removing Dampness combined with external use of Butyl Flufenamate Ointment, and the control group was only treated with external use of Butyl Flufenamate Ointment. The treatment for the two groups covered 4 weeks. Before treatment and on treatment day 7, 14 and 28, scores of clinical symptoms, related skin barrier function indexes,and the serum level of cathepsin S(Cat S)in the two groups were observed. Results(1)On treatment day 14,28,the total effective rate in the treatment group was 84.8%, 89.1%,and that in the control group was 65.2%,71.7% respectively,the difference being significant(P < 0.05).(2)On treatment day 7,14,28, the scores of clinical symptoms in the two groups were obviously decreased(P < 0.05 compared with those before treatment),and the decrease in the treatment group was superior to that in the control group (P < 0.05).(3) On treatment day 7,14,28,facial sebum secretin,skin water content,and transepidermal water loss(TEWL) of the treatment group were improved to various degrees compared with those of the control group,the differences being significnat(P < 0.05 or P < 0.01).(4)On treatment day 28,serum Cat S of the two groups was markedly decreased(P < 0.01 compared with that before treatment),and the decrease in the treatment group was superior to that in the control group (P < 0.01). Conclusion Recipe for Clearing Heat and Removing Dampness combined with Butyl Flufenamate Ointment exerts certain effect for the treatment of facial seborrheic dermatitis.

RESUMO

<p><b>OBJECTIVE</b>To study the effects of Tongxinluo on the expressions of matrix metalloproteinase-3 (MMP-3), MMP-9 and peroxisome proliferators-activated receptor gamma (PPARgamma) in atherosclerotic rabbits and explore the mechanism of its anti-atherosclerotic effect.</p><p><b>METHODS</b>Twenty-four rabbits were randomized equally into control group, atherosclerotic model group (fed with high-fat diet for 14 weeks) and Tongxinluo group. The expressions of MMP-3, 9 and PPARgamma in the 3 groups were observed by means of immunohistochemistry.</p><p><b>RESULTS</b>The expressions of MMP-3, 9 and PPARgamma in the model group and Tongxinluo group were significantly higher than those in the control group (P<0.01). After high-fat diet feeding for 14 weeks, Tongxinluo group showed significantly lower expressions of MMP-3 and 9 but higher expression of PPARgamma than the model group (P<0.01).</p><p><b>CONCLUSION</b>Tongxinluo can inhibit the expression of MMP-3 and 9 and increase the expression of PPARgamma, which might be the mechanism of its anti-atherosclerotic effect.</p>

OBJECTIVE: This case-control study was carried out to assess the alteration of endothelium-dependent arterial dilation before and after subtotal thyroidectomy in subjects with hyperthyroidism. PATIENTS AND METHODS: The study subjects included 12 patients with hyperthyroidism and 39 apparently healthy individuals. We performed a subtotal thyroidectomy on the hyperthyroid patients. The endothelium-dependent arterial dilation was determined with a high-resolution ultrasound method in each patient at the hyperthyroid stage before treatment (stage H), the euthyroid stage induced immediately before surgery (stage E), and the transient hypothyroid stage 1 or 2 months after surgery (stage L). RESULTS: The flow-mediated arterial dilation decreased significantly from H to E and from E to L (P < 0.001). As compared with H, baseline blood flow decreased markedly at stages E and L (P < 0.001). The flow-mediated arterial dilation and baseline blood flow in the control subjects were very close to those at stage E of the hyperthyroid patients. The absolute change in the flow-mediated arterial dilation showed significant negative correlation with the changes in TSH (r =-0.86, P < 0.001), lipoprotein (a) [Lp(a)] (r =-0.77, P < 0.001) and low density lipoprotein (LDL) (r =-0.79, P < 0.001), and significant positive correlation with changes in fT3 (r =+0.88, P < 0.001). The absolute change in the baseline blood flow showed significant positive correlation with the change in fT3 (r =+0.85, P < 0.001) and significant negative correlation with the change in TSH (r =-0.63, P < 0.01). CONCLUSION: The endothelium-dependent arterial dilation increases significantly in untreated hyperthyroid patients, and decreases markedly after a subtotal thyroidectomy. Therefore, we conclude that the endothelium is more responsive to reactive hyperaemia in the hyperthyroid than the euthyroid state.

A cross-sectional survey with multiple-stage and random sampling was performed among middle and elderly aged permanent inhabitants in Wuhan area.The prevalences of metabolic syndrome,diabetes mellitus, impaired glucose tolerance,hypertension and obesity were 12.2%,11.8%,10.3%,31.9% and 48.0% respectively.

RESUMO

During oral glucose tolerance test(OGTT),endothelium-dependent vasodilation(EDD)at different time points in impaired glucose tolerance(IGT)group was lower than that in normal control group.EDD at 60 and 120 min in IGT + vitamin C group was higher than that in IGT group(all P<0.05).There was a negative relationship between blood glucose level and EDD during OGTT in IGT patients.
