RESUMO

Imidacloprid and thiacloprid, two neonicotinoid insecticides that are extensively used in urban areas, are potentially toxic to non-target aquatic organisms. In this study, the concentrations of imidacloprid and thiacloprid in surface runoff after rainfall were 20.79-43.77 ng/L and 25.13-63.84 ng/L, respectively, whereas the levels for the Licun River were 10.78-41.70 ng/L and 2.66-39.68 ng/L, respectively. The acute and chronic criteria for imidacloprid and thiacloprid are 0.865, 0.006, 0.83, and 0.012 µg/L, respectively. Tiered ecological risk assessments revealed the chronic ecological risks of these micropollutants to local aquatic species. There was a moderate chronic toxicity risk associated with imidacloprid and thiacloprid in the Licun River, and the joint probability curves showed a probability of chronic ecological risk to 5 % of the aquatic organisms at 68 %-97 %. The results provide evidence of urban surface runoff transporting micropollutants from surface into rivers and estuaries, highlighting the ecological risks to aquatic ecosystems.

RESUMO

Introduction: Metritis is a common postpartum disease in dairy cows. As a mast cell (MC) mediator, leukotriene B4 (LTB4) is the strongest phagocyte chemokine. It is important in inflammation for the recruitment of immune cells to resist infection. This study investigated the effect of LTB4 in metritis. Material and Methods: Twenty Holstein cows 3 to 6 years old and at 6 to 10 days postpartum were selected, ten of which with postpartum metritis were the experimental group, and the other ten of which as healthy cows were the control group. The levels of LTB4, substance P (SP) and vasoactive intestinal peptide (VIP) were measured by ELISA, the expression of LTB4 receptor 2 (BLT2), matrix metalloproteinase (MMP)-2 and MMP-9 mRNA was measured by qPCR, and collagens I and IV were detected by immunohistochemical staining. Results: Concentrations of SP and LTB4 were significantly higher, but those of VIP were significantly lower in the experimental group than those in the control group. The expression of BLT2, MMP-2 and MMP-9 mRNA was significantly higher in the experimental group than that in the control group. The expression of collagen Ⅰ and collagen Ⅳ was significantly lower in the experimental group than that in the control group. Conclusion: In metritis, SP promotes the activation of MC and the synthesis and release of LTB4. Leukotriene B4 chemotactic immune cells promote the high expression of collagenase, which accelerated the hydrolysis of collagen, while the inhibitory effect of VIP on MC was weakened. This may further aggravate the damage to uterine tissue.

RESUMO

The reactions of Ni(OAc)2 2H2O with Schiff base ligands 5-bromo-2-((cyclopentylimino)methyl)phenol (HL1) and 5-bromo-2-(((2-(isopropylamino)ethyl)imino)methyl)phenol (HL2) in methanol afforded two discrete trinuclear com-plexes [Ni3(L1)2(?2-?1:?1-OAc)2(DMF)2(BrSal)2] (1) and [Ni3(L2)2(?2-?1:?1-OAc)2(?2-?2:?1-OAc)2] (2), where BrSal is the monoanionic form of 4-bromosalicylaldehyde. The complexes were characterized by elemental analysis, IR and UV-Vis spectroscopy. The crystal structures of the complexes have been determined by X-ray crystallography. In both com-plexes, the nickel atoms are in octahedral coordination geometries. The L1 ligand coordinates to the nickel atoms through the phenolate O and imino N atoms, and the L2 ligand coordinates to the nickel atoms through the phenolate O, imino N and amino N atoms. The antimicrobial activities of the complexes were assayed.

RESUMO

Salmonella is a common zoonotic and foodborne pathogen that causes high morbidity and mortality in developing countries. In this study, we established and validated a polymerase spiral reaction (PSR) assay which targeted the conserved invasion gene (invA) of Salmonella by SYBR Green I indicator methods. Subsequently, assays for determination of the optimal conditions for optimal specificity and sensitivity of PSR were performed. We performed comprehensive evaluations using loop-mediated isothermal amplification (LAMP) and realtime PCR. A total number of 532 samples of daily food were analyzed by PSR. Twenty-seven bacterial strains were tested in the specificity assay, from which positive results were obtained only for 14-Salmonella strains. However, none of the 13 non-Salmonella strains was amplified. Similarly with LAMP and real-time PCR, the detection limit of the PSR assay was 50 CFU/ml. The PSR method was also successfully applied to evaluate the contamination with Salmonella in 532 samples of daily food, corroborating traditional culture method data. The novel PSR method is simple, sensitive, and rapid and provides new insights into the prevention and detection of foodborne diseases.

RESUMO

Canine distemper virus (CDV) is the causative agent of canine distemper (CD) that is a highly contagious, lethal, multisystemic viral disease of receptive carnivores. The prevalence of CDV is a major concern in susceptible animals. Presently, it is unclear whether intragenic recombination can contribute to gene mutations and segment reassortment in the virus. In this study, 25 full-length CDV genome sequences were subjected to phylogenetic and recombinational analyses. The results of phylogenetic analysis, intragenic recombination, and nucleotide selection pressure indicated that mutation and recombination occurred in the six individual genes segment (H, F, P, N, L, M) of the CDV genome. The analysis also revealed pronounced genetic diversity in the CDV genome according to the geographically distinct lineages (genotypes), namely Asia-1, Asia-2, Asia-3, Europe, America-1, and America-2. The six recombination events were detected using SimPlot and RDP programs. The analysis of selection pressure demonstrated that a majority of the nucleotides in the CDV individual gene were under negative selection. Collectively, these data suggested that homologous recombination acts as a key force driving the genetic diversity and evolution of canine distemper virus.

RESUMO

Computational prediction of compound-protein interactions (CPIs) is of great importance for drug design as the first step in in-silico screening. We previously proposed chemical genomics-based virtual screening (CGBVS), which predicts CPIs by using a support vector machine (SVM). However, the CGBVS has problems when training using more than a million datasets of CPIs since SVMs require an exponential increase in the calculation time and computer memory. To solve this problem, we propose the CGBVS-DNN, in which we use deep neural networks, a kind of deep learning technique, instead of the SVM. Deep learning does not require learning all input data at once because the network can be trained with small mini-batches. Experimental results show that the CGBVS-DNN outperformed the original CGBVS with a quarter million CPIs. Results of cross-validation show that the accuracy of the CGBVS-DNN reaches up to 98.2 % (σ<0.01) with 4 million CPIs.

RESUMO

We demonstrate a new and simple process to fabricate tungsten tips with good control of the tip profile. In this process, we use a commercial function generator without any electronic cutoff circuit or complex mechanical setup. The tip length can be varied from 160 µm to 10 mm, corresponding to an aspect ratio of 1.6-100. The radius of curvature of the tip apex can be controlled to a size <10 nm. Surface roughness and the taper angle can be controlled independently. Through control of the etching parameters, the tip length, the radius of curvature, surface roughness, and the taper angle can be controlled to suit different requirements of various applications. The possible etching mechanisms are also discussed.

RESUMO

We report a reliable method for preparing a pure Ir single-atom tip by thermal treatment in oxygen. The atomic structure of the tip apex and its ion emission characteristics are investigated with field ion microscopy. We have shown that the Ir single-atom tip can be a good field ion emitter, capable of emitting a variety of gas ion beams, such as He+, H2+, N2+, and O2+, with high brightness and stability. In addition, this tip can easily be maintained and regenerated in vacuum, ensuring it has sufficient lifetime for practical applications.

RESUMO

The title compound, C(15)H(12)BrClN(2)O(3), was obtained by the condensation reaction between 3-bromo-5-chloro-2-hydroxy-benzaldehyde and 2-methoxy-benzohydrazide. The mol-ecule is essentially planar, with a dihedral angle between the two benzene rings of 4.7 (2)°, and displays an E configuration about the C=N double bond. The mol-ecular conformation is stabilized by intramolecular O-H⋯N and N-H⋯O hydrogen bonds. In the crystal structure, mol-ecules are linked into zigzag chains running parallel to the c axis by inter-molecular C-H⋯O hydrogen bonds. The chains are further connected through aromatic π-π stacking inter-actions with centroid-centroid distances of 3.583 (4) Å.
