Sunflower is one of the four major oil crops in the world. 'Zaoaidatou' (ZADT), the main variety of oil sunflower in the northwest of China, has a short growth cycle, high yield, and high resistance to abiotic stress. However, the ability to tolerate adervesity is limited. Therefore, in this study, we used the retention line of backbone parent ZADT as material to establish its tissue culture and genetic transformation system for new variety cultivating to enhance resistance and yields by molecular breeding. The combination of 0.05 mg/L IAA and 2 mg/L KT in MS was more suitable for direct induction of adventitious buds with cotyledon nodes and the addition of 0.9 mg/L IBA to MS was for adventitious rooting. On this basis, an efficient Agrobacterium tumefaciens-mediated genetic transformation system for ZADT was developed by the screening of kanamycin and optimization of transformation conditions. The rate of positive seedlings reached 8.0%, as determined by polymerase chain reaction (PCR), under the condition of 45 mg/L kanamycin, bacterial density of OD600 0.8, infection time of 30 min, and co-cultivation of three days. These efficient regeneration and genetic transformation platforms are very useful for accelerating the molecular breeding process on sunflower.

The aim of this study was to investigate the impact of water and nitrogen regulation on the characteristics of water and fertilizer demands and the yield, quality, and efficiencies of the water and nitrogen utilization of peanuts cultivated under mulched drip irrigation in a desert-oasis region. The experiment, conducted in Urumqi, Xinjiang, centered on elucidating the response mechanisms governing peanut growth, yield, quality, water consumption patterns, and fertilizer characteristics during the reproductive period under the influence of water and nitrogen regulation. In the field experiments, three irrigation levels were implemented, denoted as W1 (irrigation water quota of 22.5 mm), W2 (irrigation water quota of 30 mm), and W3 (irrigation water quota of 37.5 mm). Additionally, two nitrogen application levels, labeled N1 (nitrogen application rate of 77.5 kg·ha-1) and N2 (a nitrogen application rate of 110 kg·ha-1), were applied, resulting in seven treatments. A control treatment (CK), which involved no nitrogen application, was also included in the experimental design. The results indicate a direct correlation between the increment in the irrigation quota and increases in farmland water-related parameters, including water consumption, daily water consumption intensity, and water consumption percentage. The nitrogen harvest index (NHI) demonstrated a higher value in the absence of nitrogen application compared to the treatment with elevated nitrogen levels. The application of nitrogen resulted in an elevation in both nitrogen accumulation and nitrogen absorption efficiency within pods and plants. When subjected to identical nitrogen application conditions, irrigation proved to be advantageous in enhancing water-use efficiency (WUE), nitrogen partial factor productivity (NPFP), and the yield of peanut pods. The contribution rate of water to pod yield and WUE exceeded that of nitrogen, while the contribution rate of nitrogen to nitrogen-use efficiency (NUE) was higher. The total water consumption for achieving a high yield and enhanced water- and nitrogen-use efficiencies in peanuts cultivated under drip irrigation with film mulching was approximately 402.57 mm. Taking into account yield, quality, and water- and nitrogen-used efficiencies, the use of an irrigation quota of 37.5 mm, an irrigation cycle of 10-15 days, and a nitrogen application rate of 110 kg·ha-1 can be regarded as an appropriate water and nitrogen management approach for peanut cultivation under mulched drip irrigation in Xinjiang.

The WRKY family is an important family of transcription factors in plant development and stress response. Currently, there are few reports on the WRKY gene family in safflower (Carthamus tinctorius L.). In this study, a total of 82 CtWRKY genes were identified from the safflower genome and could be classified into 3 major groups and 5 subgroups based on their structural and phylogenetic characteristics. The results of gene structure, conserved domain and motif analyses indicated that CtWRKYs within the same subfamily maintained a consistent exon/intron organization and composition. Chromosomal localization and gene duplication analysis results showed that CtWRKYs were randomly localized on 12 chromosomes and that fragment duplication and purification selection may have played an important role in the evolution of the WRKY gene family in safflower. Promoter cis-acting element analysis revealed that the CtWRKYs contain many abiotic stress response elements and hormone response elements. Transcriptome data and qRT-PCR analyses revealed that the expression of CtWRKYs showed tissue specificity and a strong response to drought stress. Notably, the expression level of the CtWRKY55 gene rapidly increased more than eightfold under drought treatment and rehydration, indicating that it may be a key gene in response to drought stress. These results provide useful insights for investigating the regulatory function of the CtWRKY gene in safflower growth and development, as well as identifying key genes for future molecular breeding programmes.

The optimization of irrigation and fertilization indexes for peanuts with drip irrigation is urgently needed in Xinjiang. A field experiment was conducted during the 2021 peanut growing season at Urumqi, Xinjiang, in Northwestern China, to evaluate the effects of different water and nitrogen treatments on the growth, yield, and water and nitrogen utilization of peanuts. In field experiments, we set up three irrigation levels (irrigation water quotas of 22.5, 30, and 37.5 mm, respectively, for W1, W2, and W3), two nitrogen application levels (77.5 and 110 kg·ha-1, recorded as N1 and N2), and a control treatment (W2N0) that did not include the application of nitrogen. The results showed that nitrogen application enhanced the growth, physiological indexes, yield, and water use efficiency of the W1, W2, and W3 treatments when the irrigation volume remained the same. In comparison with no nitrogen application (W2N0), the peanut growth, physiological indexes, yield, and water use efficiency improved with increasing irrigation amounts in the N1 and N2 treatments. With an increase in the irrigation volume, the water use efficiency grew; the W3N2 treatment had the highest water use efficiency, which was 1.32 kg·m-3. The total water consumption and reproductive-stage water consumption of the peanuts in all treatments increased with the irrigation volume, and a high yield was achieved at 402.57 mm, which was 5.2974 Mg·ha-1. In the W1, W2, and W3 treatments, the nitrogen partial factor productivity significantly decreased as the nitrogen application increased, with the nitrogen partial factor productivity in the W3N1 treatment being the highest, at 60.61 kg·kg-1. A comprehensive evaluation based on principal component analysis assigned W3N2 the higher score. These findings suggest that irrigation water quotas of 37.5 mm should be coupled with 110 kg·ha-1 nitrogen applications for peanuts using drip irrigation in mulch film in Xinjiang.

Salt and drought are the major abiotic stress factors plaguing plant growth, development and crop yields. Certain abiotic-stress tolerant plants have developed special mechanisms for adapting to adverse environments in the long process of evolution. Elucidating the molecular mechanisms by which they can exert resistance to abiotic stresses is beneficial for breeding new cultivars to guide agricultural production. Halostachys caspica, a perennial halophyte belonging to Halostachys in Amaranthaceae, is extremely tolerant to harsh environments, which is commonly grown in the saline-alkali arid desert area of Northwest, China. However, the molecular mechanism of stress tolerance is unclear. Nuclear Factor Y-A (NFYA) is a transcription factor that regulates the expression of downstream genes in plant response to adverse environments. It has also been reported that some members of the NFYA family are the main targets of miR169 in plants. In this study, we mainly focused on exploring the functions and preliminary mechanism of the miR169b/NFYA1 module from H. caspica to abiotic stress. The main results showed that RLM-RACE technology validated that HcNFYA1 was targeted by HcmiR169b, qRT-PCR revealed that HcmiR169b was repressed and HcNFYA1 was induced in the H. caspica branches under various abiotic stress as well ABA treatment and Arabidopsis stable transformation platform with molecular methods was applied to elucidate that the HcmiR169b/HcNFYA1 module conferred the salt and drought tolerance to plants by enhancing ABA synthesis and ABA signal transduction pathways, maintaining ROS homeostasis and the stability of cell membrane. HcNFYA1 is expected to be a candidate gene to improve plant resistance to salt and drought stresses.

Wide uses of azo dyes produce a great risk of high residuals of carcinogenic aromatic amines, and hence it is important to rapidly analyze these carcinogenic compounds in the textile products to guarantee product safety. In the present work, a surface enhanced Raman spectroscopic (SERS) method was developed for rapid detection of carcinogenic aromatic amines in textiles. In this method, target aromatic amines are extracted from textiles, and then gold nanoparticles are added to the organic extractant, which assemble into closely packed Au array at liquid interface in situ. Finally, fingerprint SERS signals of the target aromatic amines are detected on the generated Au array on the basis of strong chemical interaction between the aromatic amines and the Au surface. The proposed method provided good reproducibility with a relative standard deviation of 3.5% for ten parallel tests of benzidine. It was applied to analyze 70 textile products. To strengthen the spectroscopic data processing, a cluster analysis model was established with 50 samples to automatically identify the spectra based on the good signal reproducibility. The other 20 samples were used as test sets to validate this model. It was found that all the positive samples were successfully identified with false positive rate of 20%. With the addition of the Artificial Intelligence step, the reliability of the discriminant results can be ensured.

We report a new method to determine the orientation of individual nitrogen-vacancy (NV) centers in a bulk diamond and use them to realize a calibration-free vector magnetometer with nanoscale resolution. Optical vortex beam is used for optical excitation and scanning the NV center in a [111]-oriented diamond. The scanning fluorescence patterns of NV center with different orientations are completely different. Thus, the orientation information on each NV center in the lattice can be known directly without any calibration process. Further, we use three differently oriented NV centers to form a magnetometer and reconstruct the complete vector information on the magnetic field based on the optically detected magnetic resonance(ODMR) technique. Compared with previous schemes to realize vector magnetometry using an NV center, our method is much more efficient and is easily applied in other NV-based quantum sensing applications.
