RESUMO

In the present study, methanolysis of poplar biomass was conducted for the selective transformation of hemicellulose and lignin, which leads to methyl glycosides (mainly C5 glycosides) and lignin fragments in the liquefied products that can be separated according to their difference in hydrophilicity. The distribution of methyl glycosides and delignification was dependent on the presence of acid catalysts and reaction temperatures. The obtained lignin fraction was separated into solid lignin fragments and liquid lignin oil according to their molecular weight distribution. Subsequently, directional conversion of methyl C5 glycosides into methyl levulinate was performed with dimethoxymethane/methanol as the cosolvent. A yield of 12-30% of methyl levulinate yield (based on the methyl glycoside) was achieved under these conditions. The remaining cellulose-rich substrate showed enhanced susceptibility to enzymatic hydrolysis, resulting in a yield of glucose of above 70%. Overall, the described strategy shows practical implications for the effective valorization of biomass.

RESUMO

Tripyrrole molecules have received renewed attention due to reports of numerous biological activities, including antifungal, antibacterial, antiprotozoal, antimalarial, immunosuppressive, and anticancer activities. In a screen of bacterial strains with known toxicities to termites, a red pigment-producing strain, HDZK-BYSB107, was isolated from Chamaecyparis lawsoniana, which grows in Oregon, USA. Strain HDZK-BYSB107 was identified as Serratia marcescens subsp. lawsoniana. The red pigment was identified as prodigiosin using ultraviolet absorption, LC-MS, and 1H-NMR spectroscopy. The bacterial prodigiosin had an inhibitory effect on both Gram-negative and Gram-positive bacteria. The main objective of this study was to explore the anticancer activities and mechanism of strain HDZK-BYSB107 prodigiosin by using human choriocarcinoma (JEG3) and prostate cancer cell lines (PC3) in vitro and JEG3 and PC3 tumor-bearing nude mice in vivo. In vitro anticancer activities showed that the bacterial prodigiosin induced apoptosis in JEG3 cells. In vivo anticancer activities indicated that the prodigiosin significantly inhibited the growth of JEG3 and PC3 cells, and the inhibitory activity was dose and time dependent. The anticancer efficacy of the bacterial prodigiosin on JEG3 and PC3 cells, JEG3 and PC3 tumor exhibited a correlation with the down regulation of the inhibitor of IAP family, including XIAP, cIAP-1 and cIAP-2, and the activation of caspase-9 and caspase-3 accompanied by proteolytic degradation of poly (ADP-ribose)-polymerase. The expressions of P53 and Bax/Bcl-2 in JEG3 and PC3 cells were significantly higher than in untreated groups. Our results indicated that the bacterial prodigiosin extracted from C. lawsoniana is a promising molecule due to its potential for therapeutic applications.

RESUMO

Development of new drugs is one of the solutions to fight against the existing antimicrobial resistance threat. Cinnamaldehyde-amino acid Schiff base compounds, are newly discovered compounds that exhibit good antibacterial activity against gram-positive and gram-negative bacteria. Quantitative structure⁻activity relationship (QSAR) methodology was applied to explore the correlation between antibacterial activity and compound structures. The two best QSAR models showed R² = 0.9354, F = 57.96, and s² = 0.0020 against Escherichia coli, and R² = 0.8946, F = 33.94, and s² = 0.0043 against Staphylococcus aureus. The model analysis showed that the antibacterial activity of cinnamaldehyde compounds was significantly affected by the polarity parameter/square distance and the minimum atomic state energy for an H atom. According to the best QSAR model, the screening, synthesis, and antibacterial activity of three cinnamaldehyde-amino acid Schiff compounds were reported. The experiment value of antibacterial activity demonstrated that the new compounds possessed excellent antibacterial activity that was comparable to that of ciprofloxacin.

RESUMO

Cinnamaldehyde amino acid Schiff base (CAAS) is a new class of safe, bioactive compounds which could be developed as potential antifungal agents for fungal infections. To design new cinnamaldehyde amino acid Schiff base compounds with high bioactivity, the quantitative structure-activity relationships (QSARs) for CAAS compounds against Aspergillus niger (A. niger) and Penicillium citrinum (P. citrinum) were analysed. The QSAR models (R2 = 0.9346 for A. niger, R2 = 0.9590 for P. citrinum,) were constructed and validated. The models indicated that the molecular polarity and the Max atomic orbital electronic population had a significant effect on antifungal activity. Based on the best QSAR models, two new compounds were designed and synthesized. Antifungal activity tests proved that both of them have great bioactivity against the selected fungi.

RESUMO

Homogeneous modification of cellulose with succinic anhydride was performed using tetrabutylammonium acetate (TBAA)/dimethyl sulfoxide (DMSO) mixed solvent. The molar ratio of succinic anhydride (SA) to free hydroxyl groups in the anhydroglucose units (AGU), TBAA dosage, reaction temperature, and reaction time were investigated. The highest degree of substitution (DS) value of 1.191 was obtained in a 10 wt% TBAA/DMSO mixed solvent at 60 °C for 60 min, and the molar ratio of SA/AGU was 6/1. The molar ratio of SA/AGU and the TBAA dosage showed a significant influence on the reaction. The succinoylated cellulose was characterized by ATR-FTIR, TGA, XRD, solid state CP/MAS 13C NMR spectroscopy (CP/MAS 13C NMR), and SEM. Moreover, the modified cellulose was applied for the adsorption of Cu2+ and Cd2+, and both the DS values of modified cellulose and pH of the heavy metal ion solutions affected the adsorption capacity of succinylated cellulose. The highest capacity for Cu2+ and Cd2+ adsorption was 42.05 mg/g and 49.0 mg/g, respectively.

RESUMO

Cellulose nanofibers were successfully isolated from bamboo using microwave liquefaction combined with chemical treatment and ultrasonic nanofibrillation processes. The microwave liquefaction could eliminate almost all the lignin in bamboo, resulting in high cellulose content residues within 7min, and the cellulose enriched residues could be readily purified by subsequent chemical treatments with lower chemical charging and quickly. The results of wet chemistry analyses, SEM images, and FTIR and X-ray spectra indicated the combination of microwave liquefaction and chemical treatment was significantly efficient in removing non-cellulosic compounds. Ultrasonication was used to separate the nanofibrils from the purified residues to extract nanofibers. The TEM images confirmed the presence of elementary fibrils, nano-sized fibril bundles, and aggregated fibril bundles. As evidenced by the TGA analysis, cellulose nanofibers isolated by this novel technique had high thermal stability indicating that the isolated nanofibers could possibly be applied as reinforcing elements in biomaterials.

RESUMO

Termites are responsible for an estimated US$1 billion annually in property damage, repairs, pest control, and prevention. There is an urgent need of finding a better alternative way to control and prevent termites. Port-Orford-Cedar (POC) has been known to have significant levels of natural durability and termiticidal activities due to its extractive contents. In this study, 25 endophytes including 22 fungal and 3 bacterial strains were isolated from the POC. Four strains, namely, HDZK-BYF21, HDZK-BYF1, HDZK-BYF2, and HDZK-BYB11, were chosen to test their termiticidal activities. The fermentation broth of strain HDZK-BYF21 displayed the potent antimicrobial and termiticidal activities. Morphological examination and 18 S rDNA sequence analysis demonstrated that strain HDZK-BYF21 belonged to the genus Aspergillus. This finding indicates the existence of an interesting chemical symbiosis between an endophytic fungus and its host. This is also the first report on endophytes isolated from the POC that may have potential termiticidal activities. Endophytes with termiticidal activities can be grown in bioreactor to provide an inexhaustible supply of bioactive compounds and thus can be exploited commercially.

RESUMO

Bagasse flour (BF) was liquefied using bi-component polyhydric alcohol (PA) as a solvent and phosphoric acid as a catalyst in a microwave reactor. The effect of BF to solvent ratio and reaction temperatures on the liquefaction extent and characteristics of liquefied products were evaluated. The results revealed that almost 75% of the raw bagasse was converted into liquid products within 9 min at 150 °C with a BF to solvent ratio of 1/4. The hydroxyl and acid values of the liquefied bagasse (LB) varied with the liquefied conditions. High reaction temperature combining with low BF to solvent ratio resulted in a low hydroxyl number for the LB. The molecular weight and polydispersity of the LB from reactions of 150 °C was lower compared to that from 125 °C. Rigid polyurethane (PU) foams were prepared from LB and methylene diphenyl diisocyanate (MDI), and the structural, mechanical and thermal properties of the PU foam were evaluated. The PU foams prepared using the LB from high reaction temperature showed better physical and mechanical performance in comparison to those from low reaction temperature. The amount of PA in the LB has the ability of increasing thermal stability of LB-PU foams. The results in this study may provide fundamental information on integrated utilizations of sugarcane bagasse via microwave liquefaction process.

RESUMO

Chromated copper arsenate (CCA)-treated wood was liquefied with polyethylene glycol/glycerin and sulfuric acid. After liquefaction, most CCA metals (98% As, 92% Cr, and 83% Cu) were removed from liquefied CCA-treated wood by precipitation with calcium hydroxide. The original CCA-treated wood and liquefied CCA-treated wood sludge were fractionated by a modified Community Bureau of Reference (BCR) sequential extraction procedure. The purpose of the BCR-sequential extraction used in this study was to examine the availability of CCA metals in treated wood for reuse. Both As and Cr had a slightly higher concentration in the sludge sample than in original CCA-treated wood. The sequential extraction showed that As and Cr were principally existed in an oxidizable fraction (As, 67%; Cr, 88%) in original CCA-treated wood. Only 1% of both As and Cr were extracted by hot nitric acid with the last extraction step. The distribution of As and Cr changed markedly in liquefied CCA-treated wood sludge. The amount of As in the exchangeable/acid extractable fraction increased from 16% to 85% while the amount of Cr increased from 3% to 54%. Only about 3% of As was present in the oxidizable fraction. However, there was still about 34% of Cr in the same fraction. Based on these results from sequential extraction procedures, it can be concluded that the accessibilities of CCA metals increase markedly by the liquefaction-precipitation process.

RESUMO

Near infrared spectroscopy is widely used as a quantitative method, and the main multivariate techniques consist of regression methods used to build prediction models, however, the accuracy of analysis results will be affected by many factors. In the present paper, the influence of different sample roughness on the mathematical model of NIR quantitative analysis of wood density was studied. The result of experiments showed that if the roughness of predicted samples was consistent with that of calibrated samples, the result was good, otherwise the error would be much higher. The roughness-mixed model was more flexible and adaptable to different sample roughness. The prediction ability of the roughness-mixed model was much better than that of the single-roughness model.

RESUMO

Rapid prediction of annual ring density of Paulownia elongate standing trees using near infrared spectroscopy was studied. It was non-destructive to collect the samples for trees, that is, the wood cores 5 mm in diameter were unthreaded at the breast height of standing trees instead of fallen trees. Then the spectra data were collected by autoscan method of NIR. The annual ring density was determined by mercury immersion. And the models were made and analyzed by the partial least square (PLS) and full cross validation in the 350-2 500 nm wavelength range. The results showed that high coefficients were obtained between the annual ring and the NIR fitted data. The correlation coefficient of prediction model was 0.88 and 0.91 in the middle diameter and bigger diameter, respectively. Moreover, high coefficients of correlation were also obtained between annual ring density laboratory-determined and the NIR fitted data in the middle diameter of Paulownia elongate standing trees, the correlation coefficient of calibration model and prediction model were 0.90 and 0.83, and the standard errors of calibration (SEC) and standard errors of prediction(SEP) were 0.012 and 0.016, respectively. The method can simply, rapidly and non-destructively estimate the annual ring density of the Paulownia elongate standing trees close to the cutting age.
