RESUMO

Ankyrin repeat-rich membrane spanning (ARMS) plays roles in neural development, neuropathies, and tumor formation. Such pleiotropic function of ARMS is often attributed to diverse ARMS-interacting molecules in different cell context. However, it might be achieved by ARMS' effect on global biological mediator like reactive oxygen species (ROS). We established ARMS-knockdown in melanoma cells (siARMS) and in Drosophila eyes (GMR>dARMS RNAi ) and challenged them with H2O2. Decreased ARMS in both systems compromises nuclear translocation of NF-κB and induces ROS, which in turn augments autophagy flux and confers susceptibility to H2O2-triggered autophagic cell death. Resuming NF-κB activity or reducing ROS by antioxidants in siARMS cells and GMR>dARMS RNAi fly decreases intracellular peroxides level concurrent with reduced autophagy and attenuated cell death. Conversely, blocking NF-κB activity in wild-type flies/melanoma enhances ROS and induces autophagy with cell death. We thus uncover intracellular ROS modulated by ARMS-NFκB signaling primes autophagy for autophagic cell death upon oxidative stress.

BACKGROUND: Work ability is an important component of occupational health assessments and reflects how a persons' physical and mental health affect their ability to perform their job. However, little is known about factors relating to the work ability status of flight attendants. The aim of this study was to investigate the physical, mental, and work-related factors that affect flight attendants' work ability.METHODS: A questionnaire-based cross-sectional study and simple random sampling was conducted with participants employed at a Taiwan-based airline for longer than 1 yr. Analysis of variance and Pearson correlation tests were carried out to analyze work ability according to the flight attendants' social demographics, physical and mental health, and work-related factors. Multiple regression analysis was used to predict the flight attendants' work abilities.RESULTS: A total of 472 flight attendants were recruited and the response rate was 78.67%. The work ability of the flight attendants ranged from 'moderate' to 'excellent' (WAI score, 34.1 ± 1.8 to 45.1 ± 1.5). In a regression analysis, work ability was positively associated with gender, age, and good eating habits; in contrast, insomnia and work-related burnout were negatively associated with work ability (R² = 32.4%).DISCUSSION: Insomnia, work-related burnout, and eating habits had a significant impact on flight attendants' work abilities. OBJECTIVES: To establish a practical method for assessing the general metabolic health conditions among different employee groups, this study utilized the total count of metabolic syndrome (MetS) elements as a parameter, and performed a retrospective analysis comparing changes of MetS component count (MSC) of 5 years among day-time work (DW) and day-andnight rotating shift work (RSW) employees. MATERIAL AND METHODS: The data of personal histories, physical examinations, blood tests, abdominal sonographic examinations and occupational records were collected from a cohort of workers in an electronics manufacturing company. We first defined the arithmetic mean value of MSC as MSC density (MSCD) for the employee group; then we compared the changes of MSCD over 5 years between DW and RSW workers. Occupational, personal and health records were analyzed for the 1077 workers with an initial mean age of 32.4 years (standard deviation (SD): 6.2 years), including 565 RSW workers (52%). RESULTS: The initial MSCDs were 1.26 and 1.12 (p = 0.06) for DW and RSW workers, respectively; after 5 years, the increments of MSCD for DW and RSW workers were 0.10 and 0.39, respectively (p < 0.01). By performing multivariate logistic regression analyses, and comparing with DW co-workers, final results indicated that the workers exposed to RSW have 1.7-fold increased risk of elevated MSCD (95% confidence interval (CI): 1.28-2.25, p < 0.01); and are 38% less likely (adjusted rate ratio (aRR) 0.62, 95% CI: 0.45-0.86, p < 0.01) to attain decreased MSCD. CONCLUSIONS: These observations demonstrate that changes of MSCD are significantly different between DW and RSW workers, and are increasingly associated with RSW exposure. In conclusion, MSCD can represent the general metabolic health conditions of a given employee group; MSC, MSCD and their transitional changes can be applied as simple and standardized tools for monitoring metabolic health risk profiles when managing employee health, at both the individual and company levels.

To evaluate the impact of day-and-night rotating shift work (RSW) on liver health, we performed a retrospective analysis of the association between long-term RSW exposure and the normalization of plasma alanine transaminase (ALT) levels over a five-year period. The data from physical examinations, blood tests, abdominal sonographic examinations, personal histories, and occupational records were collected from a cohort of workers in a semiconductor manufacturing company. The sample population was divided into three subgroups for analysis, according to self-reported shift work status over the five-year interval: persistent daytime workers, workers exposed intermittently to RSW (i-RSW), and workers exposed persistently to RSW (p-RSW). Records were analyzed for 1196 male workers with an initial mean age of 32.5 years (SD 6.0 years), of whom 821 (68.7%) were identified as rotating shift workers, including 374 i-RSW (31.3%) and 447 p-RSW workers (37.4%). At the beginning of the follow-up, 275 were found to have elevated ALT (e-ALT): 25.1% daytime workers, 23.0% i-RSW workers, and 21.3% p-RSW workers (p = 0.098). Of those with e-ALT at the beginning, 101 workers showed normalized serum ALT levels at the end of five-year follow-up: 40 (10.7%) of 375 daytime workers, 32 (8.6%) of 374 i-RSW workers, and 29 (6.5%) of 447 p-RSW workers (p = 0.016). Compared with the workers having persistent e-ALT at the end of follow-up, the workers normalized serum ALT levels had significantly lesser exposures to RSW during follow-up. By performing multivariate logistic regression analyses, and comparing with the persistent daytime co-workers, after controlling for confounding variables (age, occupational factors, educational levels, lifestyle factors, metabolic syndrome, hepatovirus infection, and fatty liver), analysis indicated that the workers exposed to p-RSW were 46% less likely (OR, 0.54; 95% CI, 0.30-0.95; p = 0.03) to attain normal ALT levels within a five-year interval. These observations demonstrate that persistent day-and-night RSW pose a vigorous obstacle to the normalization of e-ALT among workers with preexisting abnormal liver function. We suggest that workers and managers approach with caution the consideration of assigning or accepting long-term day-and-night RSW when an employee health screening shows evidence of abnormal liver function.

Vectorial optical arbitrary waveform generation is experimentally demonstrated by applying polarization line-by-line pulse shaping on a phase-modulated continuous laser frequency comb. Polarization shaped optical waveforms extending a 50-ps time window are successfully synthesized. Temporal Talbot effect is extended into the vectorial regime, where the distinct periodic temporal phases of the two orthogonally polarized pulse trains are exploited. In one example, we generate repetition-rate doubled circularly polarized pulses with alternating pulse-by-pulse handedness. In another example, complex instantaneous field polarizations are synthesized through the combination of line-by-line amplitude and temporal Talbot phase shaping. Our experimental results are measured through a dual-quadrature spectral interferometry system and are found in excellent agreements to the applied shaping controls.

According to the known effects of each ingredient, Gan-Lu-Yin (GLY), a traditional Chinese herbal formula, has the potential to be an antiangiogenic agent. The purpose of this study was to explore the putative effect of GLY on antiangiogenesis. An ethanol extract of GLY was tested on chicken chorioallantoic membrane (CAM) and human umbilical vein endothelial cells (HUVEC) to evaluate the effects of GLY extract on cell proliferation, migration, and tube formation. The results showed that treatment with 1.0 mg/mL of GLY extract could markedly reduce cell migration and in vitro tube formation of HUVEC, and 1.5 mg/mL of GLY extract was sufficient to inhibit proliferation of HUVEC. The expression level of vascular endothelial growth factor (VEGF) of HUVEC was significantly decreased by 1.5 and 2.0 mg/mL of GLY extract. In chicken CAM assay, all tested concentrations of GLY extract were found to reduce the capillary mesh on the CAM of fertilized eggs. The inhibitory effects of GLY extract (1 mg/mL) were also found on tumor cell-induced HUVEC proliferation and tube formation. These observations suggested that GLY extract has an inhibitory effect on angiogenesis, which in turn may prevent tumor growth, and its mechanism might be partially associated with blocking VEGF protein expression of HUVEC.
