The composition of melanoidins in dried apple that affects quality remains unclear. The composition and structure of melanoidins in dried apple slices by hot air drying (HAAM), instant controlled pressure drop drying (DICAM), and vacuum freeze-drying (FDAM) were investigated. It showed that the melanoidins were highly heterogeneous mixtures with a light-yellow color and blue-green fluorescence, belonging to polysaccharide-type melanoidins. Specifically, HAAM had a large molecular weight (929.5 g/mol) and wide molecular weight distribution, with more double-bond conjugated systems. DICAM (610.9 g/mol), possessing the strongest fluorescence intensity, was mainly composed of compounds with fewer π-conjugated structures and more electron-donating groups. As a control, the low level of Maillard reaction in FDAM resulted in the formation of the smallest molecular weight (458.6 g/mol) with weak fluorescence intensity. Moreover, 10 compounds were tentatively identified in apple melanoidins. This study provides the foundation for the future functional preparation of apple melanoidins.

Browning commonly appeared in apple processing, which varied in different apple varieties. Present work investigated the metabolomics of four varieties apple of Yataka, Gala, Sansa, and Fuji, which possessed different browning characteristics and related enzymes. Sansa as browning insensitive apple variety, exhibited the least chroma change with the lowest PPO activity and the highest SOD activity among the four apple varieties. Browning inhibition pretreatment increased the activity of SOD and PAL and decreased PPO and POD activity. In addition, metabolomic variances among the four apple varieties (FC), their browning pulp (BR) and browning inhibition pulp (CM) were compared. And the key metabolites were in-depth analyzed to match the relevant KEGG pathways and speculated metabolic networks. There were 487, 644, and 494 significant differential metabolites detected in FC, BR and CM, which were consisted of lipids, benzenoids, phenylpropanoids, organheterocyclic compounds, organic acids, nucleosides, accounting for 23 %, 11 %, 15 %, 16 %, 11 % of the total metabolites. The differential metabolites were matched with 39, 49, and 36 KEGG pathways in FC, BR, and CM, respectively, in which other secondary metabolites biosynthesis metabolism was the most significant in FC, lipid metabolism was the most significant in BR and CM, and energy metabolism was markedly annotated in CM. Notably, Sansa displayed the highest number of differential metabolites in both its BR (484) and CM (342). The BR of Sansa was characterized by flavonoid biosynthesis, while the other three apple varieties were associated with α-linolenic acid metabolism. Furthermore, in browning sensitive apple varieties, the flavonoid and phenylpropanoid biosynthesis pathway was significantly activated by browning inhibition pretreatment. Phenolic compounds, lipids, sugars, organic acids, nucleotides, and adenosine were regulated differently in the four apple varieties, potentially serving as key regulatory sites. Overall, this work provides novel insight for browning prevention in different apple varieties.

Domestic wastewater is one of the major carbon sources that cannot be ignored by human society. Against the background of carbon peaking & carbon neutrality (Double Carbon) goals, the continuous urbanization has put heavy pressure on urban drainage systems. Nevertheless, the complex subjective and objective conditions of drainage systems restrict the field monitoring, measurement, and analysis of drainage systems, which has become a great obstacle to the study of carbon emissions from drainage system. In this paper, 3389 sensor terminals of Internet of Things (IoT) are used to build a field monitoring IoT for urban domestic wastewater methane (CH4) carbon emission, with 21 main districts of Chongqing Municipality in China as the study area. Incorporating Fick's law of diffusion, this field monitoring IoT derives a measurement model for methane carbon emissions based on measured concentrations, and solves the problems of long-term and stable monitoring and measurement of methane gas in complex underground environment. With GIS spatio-temporal analysis used to analyze the spatial and temporal evolution patterns of carbon emissions from septic tanks in drainage systems, it successfully reveals the spatial and temporal distribution of methane carbon emissions from drainage systems in different seasons, as well as the relationship between methane carbon emissions from drainage systems and the latitude of direct sunlight. Applying the DTW method, it quantifies the stability of methane monitoring in drainage systems and evaluates the effects of Sampling Frequency (SF) and Number of Devices Terminal (NDT) on the stability of methane monitoring. Consequently, an intelligent management system for carbon emissions from urban domestic wastewater is constructed on the base of IoT, which integrates methane monitoring, measurement and analysis in septic tanks of drainage systems.

Due to the cardiotoxicity of doxorubicin (DOX), its clinical application is limited. Lipid peroxidation caused by excessive ferrous iron is believed to be a key molecular mechanism of DOX-induced cardiomyopathy (DIC). Dexrazoxane (DXZ), an iron chelator, is the only drug approved by the FDA for reducing DIC, but it has many side effects and cannot be used as a preventive drug in clinical practice. Single-nucleus RNA sequencing (snRNA-seq) analysis identified myocardial and epithelial cells that are susceptible to DOX-induced ferroptosis. The glutathione peroxidase 4 (GPX4) activator selenomethione (SeMet) significantly reduced polyunsaturated fatty acids (PUFAs) and oxidized lipid levels in vitro. Consistently, SeMet significantly decreased DOX-induced lipid peroxidation in H9C2 cells and mortality in C57BL/6 mice compared to DXZ, ferrostatin-1, and normal saline. SeMet can effectively reduce serum markers of cardiac injury in C57BL/6 mice and breast cancer patients. Depletion of the GPX4 gene in C57BL/6 mice resulted in an increase in polyunsaturated fatty acid (PUFA) levels and eliminated the protective effect of SeMet against DIC. Notably, SeMet exerted antitumor effects on breast cancer models with DOX while providing cardiac protection for the same animal without detectable toxicities. These findings suggest that pharmacological activation of GPX4 is a valuable and promising strategy for preventing the cardiotoxicity of doxorubicin.

Background: The relationship between leisure sedentary behavior (LSB) and periodontitis risk remains unclear in terms of causality and the potential mediating effects of intermediate factors. Materials and methods: Using the aggregate data of several large-scale genetic association studies from participants of European descent, we conducted a univariate, two-step, and multivariate Mendelian random (MR) analysis to infer the overall effect of LSB on periodontitis, and quantified the intermediary proportion of intermediary traits such as smoking. Results: Our findings indicated that per 1-SD increase (1.87 h) in leisure screen time (LST), there was a 23 % increase in the risk of periodontitis. [odds ratios (95 % CI) = 1.23 (1.04-1.44), p = 0.013]. Smoking was found to partially mediate the overall causal effect of LST on periodontitis, with a mediation rate of 20.7 % (95 % CI: 4.9%-35.5 %). Multivariate MR analysis demonstrated that the causal effect of LST on periodontitis was weakened when adjusting for smoking, resulting in an odds ratio of 1.19 (95 % CI: 1.01-1.39, p = 0.049) for each 1 standard deviation increase in exposure. Conclusion: The study provides evidence of a potential causal relationship between LSB characterized by LST and periodontitis, thereby further supporting the notion that reducing LSB is beneficial for health. Furthermore, it confirms the role of smoking as a mediator in this process, suggesting that inhibiting smoking behavior among individuals with long-term LSB may serve as a strategy to mitigate the risk of periodontitis.

Maillard reaction browning is one of the quality deterioration in dried fruit products, but how pectin affects Maillard reaction in the fruit drying and storage process is not clear. This study aimed at investigating the mechanism of pectin variation impact on the browning of Maillard reaction by using simulated system (l-lysine, d-fructose and pectin) in thermal (60 °C and 90 °C for 8 h) and storage (37 °C for 14 days) process. Results showed that apple pectin (AP) and sugar beet pectin (SP) significantly enhanced the browning index (BI) of the Maillard reaction system by 0.01 to 134.51 in the thermal and storage processes, respectively, which were methylation degree of pectin-dependent. The pectin depolymerization product participated Maillard reaction by reacting with l-lysine, and increasing the 5-hydroxymethyl furfural (5-HMF) content (1.25-11.41-fold) and Abs420nm (0.01-0.09). It also produced a new product (m/z 225.1245), which finally increased browning level of the system.

The application of carbon-based nanomaterials (CBNMs) in plant science and agriculture is a very recent development. Many studies have been conducted to understand the interactions between CBNMs and plant responses, but how fullerol regulates wheat subjected to drought stress is still unclear. In this study, seeds of two wheat cultivars (CW131 and BM1) were pre-treated with different concentrations of fullerol to investigate seed germination and drought tolerance. Our results indicate that the application of fullerol at certain concentrations (25-200 mg L-1) significantly promoted seed germination in two wheat cultivars under drought stress; the most significant effective concentration was 50 mg L-1, which increased the final germination percentage by 13.7% and 9.7% compared to drought stress alone, respectively. Wheat plants exposed to drought stress induced a significant decrease in plant height and root growth, while reactive oxygen species (ROS) and malondialdehyde (MDA) contents increased significantly. Interestingly, wheat seedlings of both cultivars grown from 50 and 100 mg L-1 fullerol-treated seeds were promoted in seedling growth under water stress, which was associated with lower ROS and MDA contents, as well as higher antioxidant enzyme activities. In addition, modern cultivars (CW131) had better drought adaptation than old cultivars (BM1) did, while the effect of fullerol on wheat had no significant difference between the two cultivars. The study demonstrated the possibility of improving seed germination, seedling growth and antioxidant enzyme activities by using appropriate concentrations of fullerol under drought stress. The results are significant for understanding the application of fullerol in agriculture under stressful conditions.

Browning of some processed fruit products was affected not only by polyphenol oxidation but also by cell wall polysaccharides (pectin). The study was performed to understand the mechanism of browning in the pectin system. The catechin/chlorogenic acid oxidation system in three pectins significantly enhanced their browning during thermal storage with pectin structure- and concentration-dependent. Particularly, the structural and physicochemical properties of pectin were examined to determine its effects on the kinetics of polyphenol oxidation and the stability of free polyphenols. Moreover, pectin impacted the fluorescence characteristics of polyphenols by cross-linking with the aromatic ring of polyphenols. In turn, the interaction between polyphenols and pectin impacted the chemical bond vibration of pectin, thereby affecting its optical features and browning. The correlation analysis revealed that the monosaccharide composition, Ratio 1, Ratio 2, Ratio 3, methyl esterification, ζ-potential, and polydispersity index of pectin were significantly correlated with the browning of the pectin-polyphenol oxidation system.

Osteomyelitis is commonly developed via hematogenous spreading or direct inoculation of bacteria from orthopedics trauma. Pathogens-induced bone destruction impedes the penetration of antibiotics to the infection site, and the severe inflammation further compromises the traditional treatment outcome. In this work, vancomycin-loaded oligochitosan nanoparticles (Van-NPs) with antibacterial, antibiofilm, antioxidant as well as bone regenerative properties are prepared using sodium tripolyphosphate (TPP) as a crosslinker, and employed for the treatment of osteomyelitis. Van-NPs exhibit strong interactions with dissociative S. aureus and biofilms due to the positive zeta potential, the additional effect between vancomycin (Van) and oligochitosan (OCS) further contributes to an enhanced antibacterial and antibiofilm outcome. The in vitro osteogenic differentiation of rBMSCs is facilitated by the antioxidant ability of Van-NPs and the TPP-induced activation of ERK1/2 and p38 signaling pathways. Moreover, the combination of Van-NPs with PLGA-PEG-PLGA gel (Gel/Van-NPs) achieves successful localized treatment of osteomyelitis in terms of enhanced bacteria elimination, inflammatory modulation, and accelerated bone regeneration. Therefore, Gel/Van-NPs may serve as a promising biomaterial for the optimal treatment of osteomyelitis.

The field measurement of wind-induced response is of great significance to the wind resistance design of high-rise buildings, in particular torsional responses measured from high-rise buildings under typhoons. The measured high-rise building, with a height of 108 m, has 32 stories and is supported by giant trusses with four massive columns. Acceleration responses along translational and torsional directions were monitored synchronously and continuously during the passage of Typhoon Sarika on 18 October 2016. The wind speed and wind direction at the height of 115 m, the translational accelerations on a total of six floors and the angular accelerations on a total of four floors were recorded. The time and frequency domain characteristics of translational acceleration and torsional angular accelerations were analyzed. The amplitude-dependent translational and torsional modal frequencies of the measured building were identified by NExT-ERA, SSI, and RDT methods. The full-scale study is expected to provide useful information on the wind-resistant design of high-rise buildings in typhoon-prone regions.

The chip is the core component of the integrated circuit. Degradation and failure of chip solder joints can directly lead to function loss of the integrated circuit. In order to establish the degradation model of chip solder joints under coupled stress, this paper takes quad flat package (QFP) chip solder joints as the study object. First, solder joint degradation data and failure samples were obtained through fatigue tests under coupled stress. Three types of micro failure modes of solder joints were obtained by scanning electron microscope (SEM) analysis and finite element model (FEM) simulation results. Second, the characterization of degradation data was obtained by the principal component of Mahalanobis distance (PCMD) algorithm. It is found that solder joint degradation is divided into three stages: strain accumulation stage, crack propagation stage, and failure stage. Later, Coffin-Manson model and Paris model were modified based on the PCMD health index and strain simulation. The function relationship between strain accumulation time, crack propagation time, and strain was determined, respectively. Solder joint degradation models at different degradation stage were established. Finally, through strain simulation, the models can predict the strain accumulation time and failure time effectively under each failure mode, and their prediction accuracy is above 85%.

Up-conversion ß-NaYF4:Yb(3+),Tm(3+)/NaYF4 core-shell nanoparticles (NYF NPs) with a high luminous intensity in the visible light region were synthesized by a hydrothermal reaction process. Photocurrent densities of the mesoscopic perovskite solar cells fabricated by incorporating up-conversion NYF NPs into the electron transporting layer are effectively enhanced. The effects of the thicknesses of the electron transporting layer and the weight ratio of up-conversion NYF NPs/TiO2 on the power conversion efficiency (PCE) of the as-fabricated devices were also investigated. The results indicate that the PCE of the optimized device achieves 16.9%, which is 20% higher than that of the device without introducing NYF NPs, and the steady-state PCE of the as-fabricated devices is close to its transient-state PCE. The up-conversion effect of NYF NPs is conducive to higher device performance rather than the nanoparticles as scattering centers to increase possible light absorption of the perovskite film or the electronic effect of the NaYF4 shell surface. These results can be further confirmed by finite-difference time-domain simulation. Photoluminescence results suggest that the multiphonon-assistance can accelerate the nonradiative recombination process at a lower temperature. Incorporating NYF NPs into the electron transporting layer opens a new approach to a promising family of electron transporting materials for mesoscopic perovskite solar cells.

A convenient metal-free method for the synthesis of 3-aryl isoindolinones via TfOH catalyzed aromatic C-H functionalization of electron-rich arenes with 2-formylbenzonitriles is developed. This process provided a new efficient strategy for the synthesis of isoindolinone derivatives in good to high yields and regioselectivities by forming two bonds.

RESUMO

A convenient one-pot reaction was conducted by mixing bromoacetophenone, a functionalized α,ß-unsaturated ketone and potassium hydroxide in tetrahydrofuran at room temperature, ammonium acetate was added and heated to reflux, resulting in four chemical bonds forming from easily accessible substrates. This process provides a flexible and rapid synthetic route for the construction of polysubstituted benzofuro[2,3-c]pyridines in moderate to good yields.

A honeycomb-like regular nanoporous pattern built in the photosensitive organic-inorganic hybrid film was fabricated by an UV soft nanoimprint technique. Polydimethylsiloxane (PDMS) soft mold was firstly replicated from an anodic aluminum oxide (AAO) template obtained by using a two-step anodization method. Scanning electron microscopy images show that the AAO template has a regular honeycomb-like nanoporous structure, while the PDMS soft mold has a relief structure of nanopillar arrays. Photosensitive TiO2-contained organic-inorganic hybrid films, which were prepared by combining a low temperature sol-gel process with a spin-coating technique, were used as the imprinted layer. Thus, a honeycomb-like regular nanoporous pattern built in the hybrid film can be easily obtained by imprinting the PDMS soft mold into the photosensitive hybrid film under an UV-irradiation. The as-fabricated organic-inorganic regular nonporous arrays have potential applications in two-dimensional photonic crystal.

OBJECTIVE: To establish the HPLC fingerprint analysis method for the quality control of acetoacetate extraction of Polygonum orientale. METHODS: Agilent C18 (4.6 mm x 250 mm, 5 microm); DAD detector; Mobile phase: methyl alcohol-water gradient elution at a flow rate of 1.0 mL/min, and at a column temperature of 30 degrees C. RESULTS: 26 peaks were selected as the common fingerprint peaks. The relative standard deviations for relative retention values were less than 3% in the precision and repeated test. CONCLUSION: The repetition, stability and precision of the method is good and it can be used to control the quality of Polygonum orientale.
