RESUMO

Autoantibodies against apolipoprotein A-I (ApoA-I) are associated with cardiovascular disease risks. We aimed to examine the 4-hydroxy-2-nonenal (HNE) modification of ApoA-I in coronary artery disease (CAD) and evaluate the potential risk of autoantibodies against their unmodified and HNE-modified peptides. We assessed plasma levels of ApoA-I, HNE-protein adducts, and autoantibodies against unmodified and HNE-peptide adducts, and significant correlations and odds ratios (ORs) were examined. Two novel CAD-specific HNE-peptide adducts, ApoA-I251-262 and ApoA-I70-83, were identified. Notably, immunoglobulin G (IgG) anti-ApoA-I251-262 HNE, IgM anti-ApoA-I70-83 HNE, IgG anti-ApoA-I251-262, IgG anti-ApoA-I70-83, and HNE-protein adducts were significantly correlated with triglycerides, creatinine, or high-density lipoprotein in CAD with various degrees of stenosis (<30% or >70%). The HNE-protein adduct (OR = 2.208-fold, p = 0.020) and IgM anti-ApoA-I251-262 HNE (2.046-fold, p = 0.035) showed an increased risk of progression from >30% stenosis in CAD. HNE-protein adducts and IgM anti-ApoA-I251-262 HNE may increase the severity of CAD at high and low levels, respectively.

Afforestation is a promising nature-based climate solution for mitigating climate change, but it is subject to a complex web of biophysical, cost-benefit, market, and policy processes. Although its biophysical feasibility has been established, the cost, market, and policy constraints that affect climate change mitigation through afforestation are still unclear. Here, we estimate such cost, market, and policy constraints on the basis of biophysical feasibility. Our findings reveal that implementation costs are a more relevant constraint than opportunity costs on mitigating climate change through afforestation. The China Certified Emission Reduction market currently provides only a 0.308 % incentive for climate change mitigation through afforestation, due to market access constraints. The current market prices of China Certified Emission Reduction, China Carbon Emissions Trading Exchange, and Nature Based Carbon Offset in Voluntary Carbon Market constrain 88.15 %, 87.95 %, and 85.75 % of CO2 removal actions through afforestation, compared to the carbon price scenario (US$62.97 tCO2-1) of the EU Emissions Trading System. Moreover, land policy under the scenarios of prohibiting conversion of cultivated land to forest and forest restoration in degraded areas exhibit 8.87-29.59 % and 65.16-74.10 % constraints, respectively, on mitigating climate change through afforestation compared to land-use freedom conversion scenarios from 2020 to 2060. Thus, enhancing the incentive price of CO2 removal, addressing the market access barrier, strengthening cooperation between global carbon markets, and exploring carbon-neutral and food multi-oriented land policies can be valuable sources of mitigation efforts over the next 40 years.

BACKGROUND: Previous studies show that using 12-lead electrocardiogram (ECG) or 24-h ECG monitor for the detection of cardiac arrhythmia events in patients with stroke or syncope is ineffective. HYPOTHESIS: The 14-day continuous ECG patch has higher detection rates of arrhythmias compared with conventional 24-h ECG monitoring in patients with ischemic stroke or syncope. METHODS: This cross-sectional study of patients with newly diagnosed ischemic stroke or syncope received a 24-h ECG monitoring and 14-day continuous cardiac monitoring patch and the arrhythmia events were measured. RESULTS: This study enrolled 83 patients with ischemic stroke or syncope. The detection rate of composite cardiac arrhythmias was significantly higher for the 14-day ECG patch than 24-h Holter monitor (69.9% vs. 21.7%, p = .006). In patients with ischemic stroke, the detection rates of cardiac arrhythmias were 63.4% for supraventricular tachycardia (SVT), 7% for ventricular tachycardia (VT), 5.6% for atrial fibrillation (AF), 4.2% for atrioventricular block (AVB), and 1.4% for pause by 14-day ECG patch, respectively. The significant difference in arrhythmic detection rates were found for SVT (45.8%), AF (6%), pause (1.2%), AVB (2.4%), and VT (9.6%) by 14-day ECG patch but not by 24-h Holter monitor in patients with ischemic stroke or syncope. CONCLUSIONS: A 14-day ECG patch can be used on patients with ischemic stroke or syncope for the early detection of AF or other cardiac arrhythmia events. The patch can be helpful for physicians in planning medical or mechanical interventions of patients with ischemic stroke and occult AF.

Current therapies for hepatitis B virus (HBV) infection can slow disease progression but cannot cure the infection, as it is difficult to eliminate or permanently silence HBV covalently closed circular DNA (cccDNA). The interaction between host factors and cccDNA is essential for their formation, stability, and transcriptional activity. Here, we focused on the regulatory role of the host factor ENPP1 and its interacting transcription factor LMNB1 in HBV replication and transcription to better understand the network of host factors that regulate HBV, which may facilitate the development of new antiviral drugs. Overexpression of ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1) in Huh7 cells decreased HBV pregenomic RNA (pgRNA) and hepatitis B core antigen (HBcAg) expression levels, whereas knockdown of ENPP1 increased them. A series of HBV promoter and mutant plasmids were constructed, and a luciferase reporter assay showed that overexpression of ENPP1 caused inhibition of the HBV promoter and its mutants. A DNA pull-down assay showed that lamin B1 (LMNB1), but not ENPP1, interacts directly with the HBV enhancer II/ basic core promoter (EnhII/BCP). ZDOCK and PyMOL software were used to predict the interaction of ENPP1 with LMNB1. Overexpression of LMNB1 inhibited the activity of the HBV promoter and its mutant. The acetylation levels at the amino acids 111K, 261K, and 483K of LMNB1 were reduced compared to the control, and an LMNB1 acetylation mutant containing 111R, 261Q, 261R, 483Q, and 483R showed increased promoter activity. In summary, ENPP1 together with LMNB1 increased the acetylation level at 111K and 261K, and LMNB1 inhibited the activity of HBV promoter and downregulated the expression of pregenomic RNA and HBcAg. Our follow-up studies will investigate the expression, clinical significance, and relevance of ENPP1 and LMNB1 in HBV patient tissues, explore the effect of LMNB1 on post-transcriptional progression, and examine whether ENPP1 can reduce cccDNA levels in the nucleus.

The situations are complex and variant in the three stages of "carbon emission peak", "rapid reduction of carbon emission" and "deep decarbonization for carbon neutrality" in China's carbon neutralization roadmap. Forest carbon sequestration is an important means to achieve the goal of carbon neutralization in China. Its intertemporal allocation is a vital way to balance industrial emission reduction and forest carbon sequestration, reduce the cost of carbon neutrality, and gradually achieve the goal of carbon neutrality based on optimal cost. Based on the cost optimization allocation theory, we simulated the cost change process of three stages of carbon neutralization in China by quoting the theory of marginal carbon sequestration cost and combining with the existing domestic marginal abatement cost theory. The results showed that annual forest carbon sequestrations with the optimal cost in China was 20 million t, 775 million t and 1.982 billion t respectively in the three stages of "carbon emission peak", "rapid reduction of carbon emission" and "deep decarbonization for carbon neutrality", accounting for 1.8%, 17.5%, and 37.6% of the total emission reduction in each period. Compared with the way relying only on industrial emission reduction, forest carbon sequestration under the optimal cost design reduced the total cost by 48, 79136, and 909253 million US$ in the three stages of carbon neutralization, respectively. Due to the limited cost advantage of forest carbon sequestration, industrial emission reduction should be emphasized in the "carbon emission peak" stage. In the "rapid reduction of carbon emissions" stage, the cost advantage of forest carbon sequestration will be increasingly prominent. In the stage of "deep decarbonization for carbon neutrality", it is necessary to fully exploit the cost advantage of forest carbon sequestration to achieve the goal of "zero carbon" to avoid the risk of high costs, especially for industries with high decarbonization cost or that will never be completely decarbonized. The optimal cost design for forest carbon sequestration can save 988.437 billion US $ in carbon-neutral costs.

PURPOSE: To explore how perceptions of fall risk influence decisions to undertake activities in people with Parkinson's disease who have fallen, along with their care-partners. MATERIALS AND METHODS: This qualitative study used semi-structured interviews to collect data from eight people with moderate to severe Parkinson's Disease and freezing of gait (including those with cognitive impairments) and their care-partners. An inductive approach to thematic analysis was used to analyse the data. RESULTS: Four main, interconnected themes emerged, and a framework was developed to illustrate these connections. Weighing up the risks and benefits reflected the constant tension between assessing the likelihood of falling and potential benefit of any activity; Being fearful heightened the perceived risk resulting in a tendency to avoid or modify activities; Desire to lead a normal life heightened the perceived benefit of any activity, leading towards risk taking behaviour; It's a part of Parkinson's provided the context in which the decision-making process took place, with the reality of a progressive disorder influencing choices and contributing to a belief that falls were inevitable. CONCLUSIONS: There is a complex interaction between perceptions of fall risk and behaviour. An understanding of these interactions will assist therapists to tailor individualised fall prevention interventions.IMPLICATIONS FOR REHABILITATIONPeople are constantly weighing up the risks and benefits of activities while balancing fear of falling and a desire to lead a normal life in the context of Parkinson's disease.The resulting activity choices vary along a continuum from avoiding activities to taking risks, with a common approach to minimise the risk and then embrace the activity.Therapists can help people with Parkinson's and their care-partners to determine what level of risk is acceptable for them when balancing risk with quality of life.Understanding how people decide if they will undertake an activity that poses a risk of falling will allow therapists and patients to co-design fall prevention and management interventions based on the patient's values and priorities.

Machine learning (ML) algorithms have been applied to predicting coronary artery disease (CAD). Our purpose was to utilize autoantibody isotypes against four different unmodified and malondialdehyde (MDA)-modified peptides among Taiwanese with CAD and healthy controls (HCs) for CAD prediction. In this study, levels of MDA, MDA-modified protein (MDA-protein) adducts, and autoantibody isotypes against unmodified peptides and MDA-modified peptides were measured with enzyme-linked immunosorbent assay (ELISA). To improve the performance of ML, we used decision tree (DT), random forest (RF), and support vector machine (SVM) coupled with five-fold cross validation and parameters optimization. Levels of plasma MDA and MDA-protein adducts were higher in CAD patients than in HCs. IgM anti-IGKC76-99 MDA and IgM anti-A1AT284-298 MDA decreased the most in patients with CAD compared to HCs. In the experimental results of CAD prediction, the decision tree classifier achieved an area under the curve (AUC) of 0.81; the random forest classifier achieved an AUC of 0.94; the support vector machine achieved an AUC of 0.65 for differentiating between CAD patients with stenosis rates of 70% and HCs. In this study, we demonstrated that autoantibody isotypes imported into machine learning algorithms can lead to accurate models for clinical use.

Ecological capital is an important fulcrum for the construction of ecological civilization. Measuring the spatial and temporal changes and influencing factors of ecological capital efficiency can help understand the current status of ecological capital efficiency and improve the level of ecological civilization to achieve green development. We used the super-efficient DEA model and Malmquist index to measure the spatial and temporal changes of ecological capital efficiency in Northeast China and analyzed the influencing factors. The results showed that the ecological capital efficiency in Northeast China was generally good, showing a trend of rising first and then decreasing. The ecological capital efficiency of Heilongjiang was high, showing U-shaped development, while that in Jilin and Liaoning provinces was relatively low. Results from the Malmquist index analysis showed that technological progress was the main driving force for the improvement of ecological capital efficiency. The results of Tobit regression analysis showed that the environment scale effect and the demographic effect variables had a significant positive impact on the efficiency of ecological capital. The low efficiency of science and technology investment led to negative correlation between environmental technology effect variables and ecological capital efficiency. To improve the ecological capital efficiency in Northeast China, on the one hand, we should accelerate the adjustment of industrial structure and develop ecological economy, on the other hand, we should increase the input efficiency of science and technology funds and rely on technological progress to realize green development of economy in Northeast China.
