RESUMO

The effects of two typical heavy metal ions[Cu(Ⅱ) and Ni(Ⅱ)] and humic acid on ANAMMOX nitrogen removal (SAA) were studied through batch experiments, and the kinetic model was analyzed. At the same time, the effects of humic acid-heavy metal on ANAMMOX nitrogen removal were discussed. The results showed that ANAMMOX was promoted when ρ[Cu(Ⅱ)] and ρ[Ni(Ⅱ)] were 3 mg·L-1, and SAA was increased by 8.64% and 7.78%, respectively; ANAMMOX was inhibited when the ρ[Cu(Ⅱ)] and ρ[Ni(Ⅱ)] were 20 mg·L-1 and 5 mg·L-1, respectively, and the inhibition effect was more significant with the increase in heavy metal ion concentration. The index fitting showed that the IC50 of Cu(Ⅱ) and Ni(Ⅱ) on ANAMMOX were 29.67 mg·L-1 and 28.75 mg·L-1, respectively. SAA was increased by 7.37% when the ρ(humic acid) was 1 mg·L-1, and the inhibition rate reached 36.80% when the humic acid concentration was 40 mg·L-1. The linear fitting showed that the IC50 of humic acid on ANAMMOX was 58.36 mg·L-1. The modified Michaelis-Menten model could better describe the inhibitory kinetic behavior of heavy metals and humic acid on ANAMMOX. The model fitting showed that the complete inhibition concentrations (I*) of Cu(Ⅱ), Ni(Ⅱ), and humic acid on ANAMMOX were 49.59, 74.46, and 84.27 mg·L-1, respectively. An appropriate amount of humic acid was beneficial to improve the inhibition of heavy metals on ANAMMOX bacteria activity, and excessive humic acid would cause inhibition on ANAMMOX bacteria again.

Objective: To explore the changes in the excitatory/inhibitory (E/I) balance of pyramidal neurons in prefrontal cortex and hippocampus in mice with anxiety disorder induced by chronic unpredictable mild stress (CUMS). Methods: Twenty-four C57/BL6 male mice were randomly divided into control group (CTRL) and model group (CUMS), with 12 mice in each group. The mice in CUMS group were subjected to 21 days of stress, including restraint for 1 h, reversed day/night cycle for 24 h, forced warm water bath for 5 min, water/food deprivation for 24 h, housing in wet sawdust for 18 h, shaking the cage for 30 min, noise for 1 h, and social stress for 10 min. CTRL group mice were fed normally. Anxiety-related behavioral tests and whole-cell recording tests were performed after modeling. Results: Compared with CTRL group, the time of spent in the central arena of CUMS group was reduced significantly in open field test (P<0.01), the time and number of entering the open arms were decreased significantly in elevated plus maze test (P<0.01), and the time of staying in the closed arms was increased significantly in CUMS group (P<0.01). The sEPSC frequency, capacitance and E/I ratio of dlPFC, mPFC and vCA1 pyramidal neurons of mice in CUMS group were increased significantly (P<0.01), while sEPSC amplitude, sIPSC frequency, amplitude and capacitance were not significantly changed (P>0.05). The frequency, amplitude, capacitance and E/I ratio of sEPSC and sIPSC of dCA1 pyramidal neurons were not significantly changed (P>0.05). Conclusion: The anxiety-like behavior of CUMS-induced mice may be the result of the participation of multiple brain regions, which is mainly related to the increase of the excitability of pyramidal neurons in dlPFC, mPFC and vCA1 brain regions, but seems to have little relationship with dCA1 brain regions.

Transrectal ultrasound prostate biopsy is the most commonly used method for the diagnosis of prostate cancer. During the operation, the doctor needs to manually adjust the ultrasound probe for repeated adjustments, which is difficult to ensure the efficiency, accuracy, and safety of the operation. This paper presents a passive posture adjusting mechanism of transrectal ultrasound probe. The overall mechanism has 7 degrees of freedom, consisting of a position adjustment module, a posture adjustment module, and an ultrasonic probe rotation and feed module. In order to achieve the centering function, the posture adjustment module is designed based on the double parallelogram. Centering performance is verified based on SimMechanics, and remote center point error of physical prototypes is evaluated. The maximum error of the azimuth remote center point motion and the maximum error of the remote center point motion of the ultrasonic probe are 4 mm and 3.4 mm, respectively, which are less than the anus that can withstand 6 mm. Meanwhile, the analysis of measurement error shows that the random error correlation is weak in different directions, the systematic error confidence intervals of azimuth and elevation angle are less than 2.5 mm, and the maximum relative fixed point error and the maximum relative standard error are 14.73% and 14.98%, respectively. The simulation and testing results have shown the effectiveness and reliability of the propose mechanism.

The development of bilirubin adsorbents with high adsorption efficiencies towards albumin-bonded bilirubin is still a considerable challenge. In this work, a three-dimensionally porous graphene (3D-pGR) has been fabricated through a simple carbon dioxide (CO2) activation of thermally exfoliated graphite oxide (EGO). Intriguingly, the resultant 3D-pGR material showed hierarchically micro-meso-macroporous structure, high specific surface area of up to 843m2g-1, and large pore volume as high as 2.71cm3g-1. Besides, the large planar π-configuration structure of 3D-pGR made it possible to compete effectively with albumin for bilirubin binding. Taking advantages of these fantastic characteristics, the 3D-pGR was demonstrated to be extraordinarily efficient for bilirubin removal from a bovine serum albumin (BSA)-rich solution. Under optimized conditions, the maximum adsorption capacity of 3D-pGR for BSA-bonded bilirubin was up to 126.1mgg-1, which is not only significantly higher than the adsorption capacities of currently available adsorbents towards albumin-bonded bilirubin, but also superior to those of many reported adsorbents towards free bilirubin. In addition, the hemolysis assay of 3D-pGR indicated that this material had negligible hemolysis effect. Findings from this study may open up important new possibilities for removal of protein-bonded toxins.

5-Hydroxymethylcytosine (5hmC), a modified form of cytosine that is considered the sixth nucleobase in DNA, has been detected in mammals and is believed to play an important role in gene regulation. In this study, 5hmC modification was detected in rice by employing a dot-blot assay, and its levels was further quantified in DNA from different rice tissues using liquid chromatography-multistage mass spectrometry (LC-MS/MS/MS). The results showed large intertissue variation in 5hmC levels. The genome-wide profiles of 5hmC modification in three different rice cultivars were also obtained using a sensitive chemical labelling followed by a next-generation sequencing method. Thousands of 5hmC peaks were identified, and a comparison of the distributions of 5hmC among different rice cultivars revealed the specificity and conservation of 5hmC modification. The identified 5hmC peaks were significantly enriched in heterochromatin regions, and mainly located in transposable elements (TEs), especially around retrotransposons. The correlation analysis of 5hmC and gene expression data revealed a close association between 5hmC and silent TEs. These findings provide a resource for plant DNA 5hmC epigenetic studies and expand our knowledge of 5hmC modification.

OBJECTIVE: To investigate therapeutic effects of external fixator combined with artificial skin for the treatment of lower limb open fractures. METHODS: From January 2009 to February 2011,56 patients with lower limb open fractures were treated with external fixator and artificial skin following debridement. There were 42 males and 14 females, with a mean age of 43.6 years (ranged, 18 to 68 years). Wound surface or cavities were filled with artificial skin with embedded drainage tubes connected to vacuum bottle. The wounds were closed with secondary suturing, or free flap, or flap transfer at 5-7 days after primary operation. The therapeutic effects were evaluated by observing the wound healing, wound bacterial culture, healing time, recovery of limb function. RESULTS: The wounds of 56 patients all healed. Fifty-three patients had bone primary union, and 3 patients had delayed union. The average union time was 5.8 months. There was pin-tract infection in 3 cases and superficial infection in 1 case. According to Ovadia evaluation criteria,45 patients got an excellent results, 9 good and 2 fair. CONCLUSION: Artificial skin combined with external fixator is a simple and effective treatment method for Gustilo III type severe open fractures of legs, which is effective to provide rapid fixation, seal wounds, and shorten secondary healing time of wounds.

BACKGROUND: VHZ is a VH1-like (member Z) dual specific protein phosphatase encoded by DUSP23 gene. Some of the dual specific protein phosphatases (DSPs) play an important role in cell cycle control and have shown to be associated with carcinogenesis. Here, the expression of VHZ associated with cell growth and human cancers was investigated. RESULTS: We generated a mouse monoclonal antibody (mAb clone#209) and rabbit polyclonal antibodies (rAb) against VHZ. We performed cell proliferation assay to learn how VHZ is associated with cell cycle by retroviral transduction to express VHZ, VHZ(C95S), and control vector in MCF-7 cells. Overexpression of VHZ [but not VHZ(C95S)] in MCF-7 cells promoted cell proliferation compared to control cells. shRNA-mediated knockdown of VHZ in MCF-7 cells showed that reduction of VHZ resulted in increased G1 but decreased S phase cell populations. Using indirect immunofluorescence, we showed that both exogenous and endogenous VHZ protein was localized at the centrosome in addition to its cytoplasmic distribution. Furthermore, using immunohistochemistry, we revealed that VHZ protein was overexpressed either in enlarged centrosomes (VHZ-centrosomal-stain) of some invasive ductal carcinomas (IDC) Stage I (8/65 cases) or in entire cytoplasm (VHZ-cytosol-stain) of invasive epithelia of some IDC Stage II/III (11/47 cases) of breast cancers examined. More importantly, upregulation of VHZ protein is also associated with numerous types of human cancer, in particular breast cancer. VHZ mAb may be useful as a reagent in clinical diagnosis for assessing VHZ positive tumors. CONCLUSIONS: We generated a VHZ-specific mAb to reveal that VHZ has a novel subcellular localization, namely the centrosome. VHZ is able to facilitate G1/S cell cycle transition in a PTP activity-dependent manner. The upregulation of its protein levels in primary human cancers supports the clinical relevance of the protein in cancers.

OBJECTIVE: To study the effect of Extractum trametes robiniophila murr on cardiac allograft rejection in mice. METHODS: All abdominal heterotopic heart transplantation models were divided into three groups as follows: (A) Extractum trametes robiniophila murr group. (B) Rejection group. (C) Isograft group. In each group, mean survival times (MST) of transplanted hearts and their pathologic histological changes at postoperative fifth day were observed. With fluoroimmunoassay, granzyme B and CD8(+) expressions were examined. RESULTS: The MST of heart allografts in group A were (6.38 +/- 0.69) d, significantly shorter than that of group B [(8.31 +/- 0.59) d] (P < 0.01). In group A, acute rejection was present in advance; transplanted hearts were seriously damaged into acute rejection pathological grade 3, and CD8(+) T lymphocytes infiltrated diffusely and the expression of granzyme B increased significantly as compared with other groups. CONCLUSIONS: Exclusive application of Extractum trametes robiniophila murr can promote the acute rejection of graft in early phase of postoperation, and the mechanism may be the promoted proliferation and infiltration of CD8(+) T lymphocytes and the increased expression of granzyme B.
