RESUMO

The objective of this study was to evaluate potency and timing of trenbolone acetate (TBA) administration on live performance and carcass characteristics of beef × dairy steers. A total of 6,895 beef × dairy steers [initial body weight (BW) = 157 ±â€…5.2 kg] were allotted into 30 pens, with pen as the experimental unit. Each pen was randomly assigned one of three implant treatments: 1) Revalor-IS (IS) at d 0, IS at d 80, and Revalor-XS (XS) at d 160 (IS/IS/XS); 2) Ralgro at d 0, IS at d 80, and XS at d 160 (Ral/IS/XS); or 3) Encore at d 0 and XS at d 160 (Enc/XS). Steers were blocked by arrival date, each pen was terminally sorted in three ways at 257 ±â€…22 days on feed and harvested at 329 ±â€…25 days on feed. For live and carcass outcomes, fixed effect of implant treatment and random effect of block was evaluated. Data are reported on a deads and removals out basis. Removals, morbidity, and mortality were similar (P ≥ 0.45). Steers administered TBA prior to d 160 were 5.8 kg heavier (P = 0.03) than Enc/XS steers at d 160. Final BW was not different (P = 0.78). Early administration of a TBA-containing implant resulted in an increased prevalence of bullers [2.40%, 5.18%, 6.86% (for Enc/XS, Ral/IS/XS, and IS/IS/XS) respectively; P < 0.01]. Dry matter intake (DMI) was 2.3% greater (P < 0.01) in steers administered Enc/XS compared to IS/IS/XS; however, DMI as a percentage of BW, average daily gain, and feed efficiency were not different (P ≥ 0.12). Dressing percentage, hot carcass weight, heavy carcass occurrence, Longissimus muscle area, and 12th rib fat thickness were similar among all steers (P ≥ 0.28). Marbling score tended to be greatest for Enc/XS and Ral/IS/XS (P = 0.09). Enc/XS graded a greater proportion of USDA Prime and fewer USDA Select carcasses than IS/IS/XS (P < 0.05). Enc/XS and Ral/IS/XS tended (P = 0.09) to have more USDA Yield Grade (YG) 1 carcasses. While delayed administration or decreased total potency of TBA-containing implants may decrease buller incidence and improve Quality Grade, few differences were observed in live or carcass outcomes.

STUDY DESIGN: Prospective, international, multicenter, single-arm, post-market study. OBJECTIVE: The aim of this study was to assess long-term safety and effectiveness of spinal cord stimulation using a passive recharge burst stimulation design for chronic intractable pain in the trunk and/or limbs. Herein we present 24-month outcomes from the TRIUMPH study (NCT03082261). SUMMARY OF BACKGROUND DATA: Passive recharge burst spinal cord stimulation (B-SCS) uniquely mimics neuronal burst firing patterns in the nervous system and has been shown to modulate the affective and attentional components of pain processing. METHODS: After a successful trial period, subjects received a permanent SCS implant and returned for follow-up at 6, 12, 18, and 24 months. RESULTS: Significant improvements in physical, mental, and emotional functioning observed after 6 months of treatment were maintained at 2 years. Pain catastrophizing scale (PCS) scores dropped below the population norm. Health-related quality of life on EQ-5D improved across all domains and the mean index score was within one standard deviation of norm. Pain reduction (on NRS) was statistically significant (P < 0.001) at all timepoints. Patient reported pain relief, a stated percentage of improvement in pain, was consistent at all timepoints at 60%. Patients reported significant improvements across all measures including activity levels and impact of pain on daily life. At 24 months, 84% of subjects were satisfied and 90% would recommend the procedure. Subjects decreased their chronic pain medication intake for all categories; 38% reduced psychotropic and muscle relaxants, 46% reduced analgesic, anti-convulsant and NSAIDs, and 48% reduced opioid medication. Adverse events occurred at low rates without unanticipated events. CONCLUSION: Early positive results with B-SCS were maintained long term. Evidence across multiple assessment tools show that B-SCS can alleviate pain intensity, psychological distress, and improve physical function and health-related quality of life.Level of Evidence: 3.

OBJECTIVE: Burst spinal cord stimulation (B-SCS) has been shown to reduce neuronal firing in the anterior cingulate cortex through selective modulation of the medial pain pathway tract. This pain pathway communicates the affective component of pain processing. The purpose of this study was to assess the effect of B-SCS on psychosocial functioning and its influence on pain and quality of life. MATERIALS AND METHODS: Eligible patients with chronic, intractable pain of the trunk, and/or lower limbs were enrolled. After a successful trial period, subjects received a permanent implant and returned for follow-up at 6- and 12-months. RESULTS: In total, 269 patients were enrolled at 22 centers. Trial success rate was 90%. Significant improvements in pain, physical, mental, and emotional functioning were observed from baseline to the 6- and 12-month follow-up (p < 0.001). Overall, patients had improved quality of life, became more active, and the negative impact of pain on daily life was decreasing. At one year, 81% of subjects were satisfied or very satisfied with their therapy. Subjects showing significant improvements on mental health outcomes reported enhanced pain relief and quality of life scores compared with subjects with continued impaired mental health at follow-up. At one year, 89% of subjects who were taking opioids at baseline decreased or stayed at the same level of opioid use; 19% stopped taking any opioids. No unanticipated adverse events have been reported. CONCLUSIONS: One-year outcomes after B-SCS show improvements across all evaluated psychological measures with the largest impact observed on catastrophizing and depression (the affective component of pain processing). These pain-related beliefs and behaviors, and not pain intensity, have been shown to put patients at greatest risk of a poor prognosis and quality of life.

A serial harvest was conducted every 28 d from 254 to 534 d on feed (DOF) to quantify changes in growth and composition of calf-fed Holstein steers (n = 115, initial body weight (BW) = 449.2 ± 19.9 kg). One-half were supplemented with the ß-2 adrenergic agonist zilpaterol hydrochloride (ZH; 8.33 mg/kg 100% dry matter (DM) basis) during the final 20 d followed by a 3-d withdrawal prior to harvest; the remainder was fed a non-ZH control (CON) ration. Five steers were randomly selected and harvested after 226 DOF which served as a reference point for modeling purposes. Fabricated carcass soft tissue was ground, mixed, and subsampled for proximate analysis. Moreover, following the traditional method of rib dissection which includes the 9th, 10th, and 11th rib contained within the IMPS 103 primal, the relationship of carcass chemical composition to 9-10-11 rib composition was evaluated. Carcasses in this investigation had more (P < 0.01) separable lean, fat, ash, and moisture concomitant with less bone and ether extract than rib dissections. However, protein levels were similar (P = 0.27) between carcasses and rib dissections. Using regression procedures, models were constructed to describe the relationship of rib dissection (RD) composition including separable lean (RDSL), separable fat (RDSF), separable bone (RDSB), ether extract (RDEE), protein (RDP), moisture (RDM), and ash (RDA) with carcass composition. Carcass lean (CL), carcass fat (CF), and carcass bone (CB) were correlated (P < 0.01) with RDSL, RDSF, and RDSB with simple r values of 0.41, 0.71, and 0.50, respectively. Chemical composition of the rib and carcass, carcass ether extract (CEE), carcass protein (CP), carcass moisture (CM), and carcass ash (CA) were correlated (P ≤ 0.01) with simple r values of 0.75, 0.31, 0.66, and 0.37, respectively. Equations to predict carcass fatness from rib dissection variables and ZH supplementation status were only able to account for 50 and 56%, of the variability of CF and CEE, respectively. Overall, the relationships quantified and equations developed in this investigation do not support use of 9/10/11 rib dissection for estimation of carcass composition of calf-fed Holstein steers.

Pseudomonas fluorescens are known bacterial pathogens in fish. The P. fluorescens group contains at least nine different bacterial species, although species from fish have rarely been differentiated. Two isolated fish kills affecting wild bluegills, Lepomis macrochirus Rafinesque, and pumpkinseed sunfish, Lepomis gibbosus (Linnaeus), occurred in the spring of 2015 during cool water temperatures (12.5°C-15.5°C). Disease signs included severe bacteraemia with rare gross external signs. Pure bacterial cultures isolated from kidneys of all affected fish were identified as P. fluorescens using the API 20NE system, while no bacteria were isolated from asymptomatic fish. To further identify the species of bacterium within the P. fluorescens complex, genetic analysis of the 16S rRNA, rpoD and gyrB genes was conducted. DNA sequences of bacterial isolates from both mortality events were identical and had close identity (≥99.7%) to Pseudomonas mandelii. Although likely widespread in the aquatic environment, this is the first report of a bacterium closely resembling P. mandelii infecting and causing disease in fish. The bacterium grew at temperatures between 5°C and 30°C, but not at 37°C. It is possible that infections in fish were a result of immunosuppression associated with spring conditions combined with the psychrotrophic nature of the bacterium.

Understanding the maximum slaughter size for calf-fed Holstein steers based on hip-height has become a contemporary issue in the beef processing industry. Increased carcass size, in terms of both weight and length, has outpaced the ability of some abattoirs to handle the larger animals. Moreover, some abattoirs have begun rejecting animals that exceed 147.3 cm (58 inches) at the hip, creating a challenge for Holstein cattle feeders. The objective of this study was to quantify the skeletal growth rate of calf-fed Holstein steers fed in confinement. Hip-height of calf-fed Holstein steers ( ≤ 135) was measured every 28 d from 226 to 422 d on feed. Hip-height was a dependent variable modeled via linear regression procedures utilizing days of age and BW as independent variables. Additionally, logistic regression was used to estimate the probability of a steer exceeding a hip-height of 147.3 cm (58 inches) from independent variables of days of age and BW. The linear relationship of BW to hip-height had an adjusted value of 0.7112 (Hip-height, cm = [0.0593 × BW, kg] + 109.00) and on average the calf-fed Holstein steers grew 1.0 cm for each 16.9 kg of BW gain during the finishing phase. The 10%, 50%, and 90% probability of a steer exceeding 147.3 cm (58 inches) of hip-height was achieved at 563, 653, and 743 kg of BW, respectively. The linear relationship of days of age to hip-height had an adjusted value of 0.6687 (Hip-height, cm = [0.0937 × days of age] + 104.4) and the calf-fed Holstein steers grew 1.0 cm for each 10.7 d of age during the finishing phase. The 10%, 50%, and 90% probability of a steer exceeding 147.3 cm (58 inches) of hip-height was estimated to occur at 408, 459, and 510 d of age, respectively. Knowledge of Holstein steer growth rate in relation to BW and age may allow for more accurate sorting to prevent oversized cattle arriving at the abattoir and subsequent discounts or being rejected for slaughter.

Holstein steers ( = 110) were fed zilpaterol hydrochloride (ZH) for 0 or 20 d before slaughter during a 280-d serial harvest study. Cattle were harvested every 28 d beginning at 254 d on feed (DOF) and concluding at 534 DOF. After slaughter, carcasses were chilled for 48 h and then fabricated into boneless closely trimmed or denuded subprimals, lean trim, trimmable fat, and bone. Inclusion of ZH increased cold side weight (CSW) by 10.3 kg ( < 0.01; 212.7 vs. 202.4 kg [SEM 1.96]) and saleable yield by 10.4 kg ( < 0.01; 131.9 vs. 121.5 kg [SEM 1.16]) in calf-fed Holstein steer carcasses. Additionally, saleable yield as a percentage of CSW increased ( ≤ 0.01) by 2.19% (62.64 vs. 60.45% [SEM 0.22]) for cattle supplemented with ZH. Subprimal weights were heavier ( ≤ 0.05) from cattle that received ZH except for the bottom sirloin ball tip, back ribs, and outside skirt regardless of slaughter endpoint. Yield of top round, bottom round, and knuckle was increased ( ≤ 0.01) following ZH supplementation by 0.37, 0.24, and 0.18%, respectively. Yield of the top sirloin butt, strip loin, and tenderloin was increased ( ≤ 0.01) concurrent with ZH supplementation by 0.18, 0.11, and 0.09%, respectively. Regarding the rib primal, the rib eye roll tended ( = 0.08) to had increased yield (2.80 vs. 2.72% [SEM 0.03]) with ZH supplementation; both back ribs and blade meat exhibited increased ( ≤ 0.04) yields of 0.04%. Relative to the chuck primal, increased ( ≤ 0.03) yields of shoulder clod, pectoral meat, and mock tender were observed (0.13, 0.07, and 0.04%, respectively). Yield changes for subprimal brisket, plate, and flank were limited to increased ( < 0.01) proportion of flank steak and elephant ear (cutaneous trunci), 0.07 and 0.04%, respectively. Feeding duration notably altered ( ≤ 0.01) weights and percentages of all subprimals except the brisket. Saleable yield increased ( ≤ 0.01) by 0.192 kg/d with additional DOF. Moreover, trimmable fat and bone increased ( ≤ 0.01) by 0.146 and 0.050 kg/d, respectively. These data illustrate improved saleable meat yields for calf-fed Holstein steers supplemented with ZH and provide the beef industry knowledge of fabrication yield changes throughout a wide range of harvest endpoints.

An indirect calorimetry trial examined energy metabolism, apparent nutrient digestibility, C retention (CR), and N retention (NR) of cattle supplemented with zilpaterol hydrochloride (ZH). Beef steers ( = 20; 463 ± 14 kg) blocked ( = 5) by weight and source were individually fed and adapted to maintenance energy intake for 21 d before allotment to ZH (90 mg/steer∙d) or no ß-adrenergic agonist treatment (control [CONT]) for 20 d (455 ± 14 kg at the start of treatment). Respiration chambers = 4 were used to quantify heat production (HP) during maintenance (d 12 to 16 of the ZH period) and fasting heat production (FHP; d 19 to 20 of ZH period; total 4 d of fast). Steers were harvested after a 6-d ZH withdrawal and carcasses were graded 24 h after harvest. Control cattle lost more BW ( < 0.01; 9 kg for CONT and 2 kg for ZH-treated) during maintenance whereas the BW loss of ZH-treated steers was greater ( < 0.01; 9 kg for ZH-treated and vs. 4 kg, for CONT) during FHP; no differences ( ≥ 0.76) were detected for G:F, ADG, and end BW. No differences in DMI, apparent nutrient digestibility, O consumption, or CH production ( ≥ 0.12) were detected; however, ZH-treated cattle had greater CO production during maintenance ( = 0.04; 23.6 L/kgBW for ZH-treated and 22.4 L/kg BW for CONT). Digestible energy and ME did not differ ( ≥ 0.19); however, urinary energy was greater ( = 0.05; 0.091 Mcal for CONT and 0.074 Mcal for ZH-treated) in CONT cattle. Steers treated with ZH tended to have greater HP ( = 0.09; 12.44 Mcal for ZH-treated and 11.69 Mcal for CONT), but the effect was reduced on a BW basis ( = 0.12; 0.126 Mcal/kg BW0.75 for ZH-treated and 0.120 Mcal/kg BW0.75 for CONT vs. 0.120 Mcal/kg BW). No treatment difference in FHP was observed ( ≥ 0.32) although CO production (L/steer) increased with ZH treatment ( = 0.04; 1,423 L/steer for ZH-treated and 1,338 L/steer for CONT). Control cattle excreted more ( = 0.05) N in urine (39.8 g/d for CONT and 32.4 g/d for ZH-treated); therefore, NR ( = 0.07; 22.14 g/d for ZH-treated and 14.12 g/d for CONT steers) tended to be greater for ZH-fed steers. Steers treated with ZH lost more C via CO ( = 0.04; 1,036.9 g/d for ZH-treated and 974.3 g/d for CONT) although total CR did not differ ( ≥ 0.23). Empty BW, HCW, and harvest yields (g/kg empty BW) were not different ( ≥ 0.13), whereas ZH increased dressed yield ( = 0.02; 62.12 % for ZH-treated and 60.65% for CONT) and LM area ( = 0.02; 77.81 cm for ZH-treated and vs. 70.90 cm for CONT). Separable carcass lean and actual skeletal muscle protein (SMP) were increased with ZH ( ≤ 0.04; 201.6 and 41.2 kg, respectively for ZH-treated and 196.0 and 38.4 kg, respectively for CONT). Results from this trial indicate that ZH treatment increased ( = 0.03) SMP and tended ( ≥ 0.07) to increase NR and modify HP during maintenance by increasing CO production.

A 2 × 11 factorial treatment structure was applied in a completely randomized experimental design to investigate differences in noncarcass tissue among serially harvested Holstein steers. Steers ( = 110) were randomly assigned to 1 of 2 dietary treatments: a ration supplemented with zilpaterol hydrochloride (ZH) fed at a rate of 8.3 mg/kg DM for 20 d followed by a 3-d withdrawal or a control ration with no ZH included in the diet. Within treatment, steers were assigned to harvest groups of 254, 282, 310, 338, 366, 394, 422, 450, 478, 506, or 534 d on feed (DOF) prior to initiation of the trial. Cattle fed ZH realized an empty BW (EBW) increase ( ≤ 0.03) of 2.8% (644.2 vs. 626.4 kg [SEM 5.4]) and a HCW increase of 5.0% (429.1 vs. 408.4 kg [SEM 4.0]) with a concomitant 12% reduction (45.1 vs. 51.2 kg [SEM 3.1]) in gastrointestinal contents and 2.1 percentage unit increase in dressed carcass yield (62.1 vs. 60.0% [SEM 0.01]). Additionally, ZH supplementation decreased (P ≤ 0.03) the absolute weight of the liver and kidneys by 0.3 and 0.1 kg, respectively. When noncarcass components were expressed on an empty body basis (g/kg EBW), reductions ( ≤ 0.01) in the limbs (18.8 vs. 19.5 g/kg EBW [SEM 0.1]), hide (81.1 vs. 78.1 g/kg EBW [SEM 0.7]), liver (14.2 vs. 13.2 g/kg EBW [SEM 0.2]), kidneys (2.6 vs. 2.3 g/kg EBW [SEM 0.04]), small and large intestines (74.9 vs. 69.6 g/kg EBW [SEM 1.2]), and gastrointestinal tract (119.8 vs. 113.4 g/kg EBW [SEM 1.3]) were observed with ZH supplementation. Additionally, there was a tendency ( = 0.07) for the proportion of total offal to be reduced (253.2 vs. 247.4 g/kg EBW [SEM 2.5]) with ZH supplementation. Empty BW and HCW linearly increased ( < 0.01) by 1.16 and 0.758 kg/d ( < 0.01), respectively, with additional DOF. The weight of the liver and intestines linearly increased ( < 0.01) by 0.007 and 0.133 kg/d ( < 0.01), respectively, with additional DOF. These data indicate the magnitude of change in noncarcass tissues that can be expected when calf-fed Holstein steers are supplemented with ZH.

This experiment was designed to study the effect of days on feed (d 225-533) and zilpaterol hydrochloride (ZH) supplementation on Holstein steer ( = 110) performance and feeding behavior as part of a serial slaughter trial. Steers were randomly assigned to 1 of 11 harvest groups with 10 steers ( = 5 control and = 5 ZH; ZH at 8.33 mg/kg diet) harvested each 28 d. Steers were weighed every 28 d (d 225, 253, 281, 309, 337, 365, 393, 421, 449, 477, 505, and 533); individual daily meal consumption data for each steer were recorded using GrowSafe technology. In the pretreatment period, dry matter intake expressed a negative quadratic relationship with days on feed (DOF) {DMI = -5.7120 + (0.08370 x DOF)- (0.00011 x DOF); Adj. = 0.2574; RMSE = 0.25 75; 0.01}. A linear increase in BW ( < 0.01) occurred during the pretreatment 308 d period from 466 to 844 kg, {BWend = 137.61 + (1.4740 x DOF); Adj. = 0.8819; RMSE = 37.06; < 0.01}, whereas ADG and G:F decreased linearly. Dry matter intake per meal exhibited a quadratic relationship over days on feed and peaked ( < 0.01) during d 365 to 392 at 1.065 kg coinciding with the highest numerical daily DMI (11.19 kg). Daily consumption visit duration differed ( < 0.01) during the 308 d period, with a low of 52.29 min (d 337-364) and a high of 55.59 min (d 365-392). Consumption rate peaked at 714 g/min (d 337-364) and exhibited a quadratic relationship to DOF. The difference ( < 0.04) in DMI between control and ZH treated cattle across all 11 harvest groups averaged 0.575 kg. Moreover, ZH treatment resulted in decreased ( 0.01) DMI per meal event of 0.093 kg. Gain to feed tended to improve ( = 0.06) with ZH treatment by 0.017 kg gain per kg feed relative to the control cattle. Daily bunk, consumption, and meal visit durations were influenced by ZH during the 20 d treatment period ( = 0.01); the average difference between control and ZH supplemented cattle over the 308 d trial was 9.09, 8.71, and 11.39 min per d, respectively. The data collected in this trial indicate live growth performance and feeding behavior were impacted by both DOF and ZH supplementation.

Vitamin D (D3) supplementation may be used to increase tenderness in beef from cattle fed zilpaterol hydrochloride (ZH). The study was arranged as a 2 × 2 factorial with fixed effects of ZH (no ZH or ZH fed at 8.3 mg/kg DM for 20 d with a 3-d withdrawal) and D3 (no D3 or 500,000 IU D3·steer·d for 10 d prior to harvest). Cattle ( = 466) were harvested in 2 blocks on the basis of BW with subsequent collection of carcass data. Full loins and inside rounds ( = 144 of each subprimal) were collected for fabrication of 5 steaks from the longissimus lumborum (LL), gluteus medius (GM), and semimembranosus (SM), which were aged for 7, 14, 21, 28, or 35 d. Warner-Bratzler shear force (WBSF) was used to evaluate mechanical tenderness of LL, GM, and SM steaks at all aging periods. Slice shear force (SSF) analysis was conducted on only 14- and 21-d LL steaks. No interactions ( > 0.05) between ZH and D3 occurred throughout the entire study. Supplementing ZH resulted in increased HCW ( < 0.01), larger LM area ( < 0.01), and improved calculated yield grades ( < 0.01) with decreases in fat thickness ( = 0.02) and marbling scores ( = 0.05). Supplementation with D3 increased calculated yield grade ( < 0.01) and decreased ( = 0.01) rib eye area. Feeding ZH increased ( ≤ 0.05) WBSF of LL steaks at each postmortem age interval, whereas D3 had no effect ( > 0.05) on WBSF or SSF of LL steaks. Like for WBSF, ZH supplementation increased SSF values at 14 and 21 d postmortem ( < 0.01) compared with those for non-ZH steaks. There was an interaction between ZH and postmortem age ( < 0.01) for WBSF of LL steaks. At 7 d LL steaks from ZH steers sheared over 0.6 kg greater than non-ZH steaks; however, by 21 d this difference was reduced to an average of 0.2 kg. Differences in distribution between LL steaks below 3.0 kg from non-ZH and ZH-fed cattle were also notable ( ≤ 0.05) through 21 d of aging. At 35 d postmortem a high proportion of LL steaks (68.5%) from ZH-fed steers required less than 3.0 kg to shear. Supplementation with ZH and D3 had no impact ( > 0.05) on WBSF values of GM steaks. Feeding ZH did not alter WBSF of SM steaks, but at 28 d D3 increased ( = 0.04) WBSF values. Shear force in ZH steaks was not effectively reduced by feeding D3 for 10 d to steers prior to harvest. Aging, however, was an effective method of reducing initially greater shear force values in LL steaks and, to a lesser degree, GM steaks from ZH-fed cattle.

Angus crossbred steers (n = 40; 563 ± 44 kg) were used to examine the effects of handling method and fat thickness on the blood chemistry and physiology of market steers. Steers were blocked by backfat (BF) thickness and were randomly assigned to treatment groups: low-stress handling (LSH) and aggressive handling (AH). Cattle were then ran¬domly assigned to one of 5 blocks containing 4 steers from the LSH and AH treatments. Steers in the LSH treatment were walked and AH cattle were run through a course of 1,540 m. Blood samples were obtained via jugular venipuncture before handling (BASE), at 770 m (LAP1), at 1,540 m (LAP2), and at1 h (1H) and 2 h (2H) after finishing the course. Blood samples were analyzed for plasma lactate (LAC), creatinine kinase (CK), base excess (BE), blood pH (pH), serum cortisol (CORT) concentrations, and venous carbon dioxide (PvCO2) and oxygen (PvO2) pressures. Heart rate (HR), respiratory rate (RR), and rectal temperature (TEMP) were measured at the same intervals. Cattle in the AH treatment had greater ( < 0.05) LAC than those in LSH at BASE (4.1 vs. 3.0 mmol/L), LAP1 (16.5 vs. 2.3 mmol/L), LAP2 (22.3 vs. 2.4 mmol/L), 1H (7.2 vs. 2.7 mmol/L), and 2H (4.0 vs. 2.5 mmol/L), respectively. Creatinine kinase and RR were not different (P > 0.14). Blood pH in AH cattle was decreased compared with that in LSH cattle ( < 0.05) at LAP1 (7.25 vs. 7.45) and LAP2 (7.19 vs. 7.48) but was not different ( > 0.13) at BASE, 1H, or 2H. Heart rate and TEMP were increased in AH cattle compared to LSH ( > 0.01). Serum cortisol was increased ( < 0.05) in AH compared to that in LSH cattle at LAP1 (87.5 vs. 58.9 nmol/L), LAP2 (144.4 vs. 93.1 nmol/L), and 1H (113.5 vs. 53.1 nmol/L). Although RR was not differ¬ent between LSH and AH, PvCO2 was decreased in AH compared to that in LSH ( < 0.05) at LAP2 (30.6 vs. 39.3 mmHg) and PvO2 was increased at LAP1 (42.7 vs. 33.5 mmHg) and at LAP2 (51.5 vs. 36.6 mmHg). Lactate was increased in AH cattle in the thicker BF group at 1H ( < 0.05), and blood pH was decreased at LAP1, LAP2, and 1H ( < 0.05) compared to the thinner BF cohorts. Four AH steers became exhausted (EXH) and did not complete the course. Increased CK, decreased PvCO2, and muscle tremors occurred in EXH steers compared to non-exhausted AH cohorts. Results of this study show that AH causes physiologic and blood chemistry changes in steers, which can be potentially detrimental to cattle, emphasizing the need for low-stress handling practices.

Serial harvests were conducted using Holstein steers ( = 110) fed zilpaterol hydrochloride (ZH) for 0 or 20 d prior to harvest. Steers were harvested in 28-d increments beginning at 254 d on feed (DOF) and ending at 534 DOF. After harvest and a 36-h chill period, carcasses were evaluated using grading methods standard for the United States (USDA), Canada (Canadian Beef Grading Association [CBGA]), and Japan (Japanese Meat Grading Agency [JMGA]). No ZH treatment differences ( = 0.81) were detected for 12th-rib fat thickness; however, additional DOF resulted in a daily linear increase ( < 0.01) of 12th-rib fat thickness by 0.004 cm/d. Longissimus muscle area was increased ( < 0.01) by 8.7 cm with ZH supplementation and linearly increased ( < 0.01) 0.08 cm2/d with additional DOF. Calculated USDA yield grade (YG) decreased ( < 0.01) 0.33 units due to ZH treatment and linearly increased ( < 0.01) 0.009 units/d. Steers supplemented with ZH exhibited increased ( < 0.01) CGBA LM width; however, no difference ( = 0.37) was detected in CGBA LM length. No ZH treatment differences ( = 0.64) were observed for CBGA fat class; however, CGBA fat class linearly increased ( < 0.01) by 0.01 units/d. No ZH differences ( ≥ 0.17) were detected for the CBGA estimated lean percentage or YG equations. Evaluation for JMGA occurs at the sixth and seventh rib interface; LM area was 4.6 cm2 greater ( = 0.02) for cattle supplemented with ZH and linearly increased ( < 0.01) by 0.07 cm2/d with additional DOF. Subcutaneous fat thickness was not different among ZH treatments ( = 0.10) but linearly ( < 0.04) increased ( < 0.01) by 0.005 cm/d with additional DOF using the JMGA grading method. No difference ( ≥ 0.21) was calculated between ZH treatments or DOF for JMGA estimated yield. No ZH treatment differences ( = 0.85) were detected in USDA marbling score; however, marbling linearly increased ( < 0.01) 0.07 units/d. These data illustrate the impact of ZH and increasing DOF on economically important carcass grading outcomes used in the USDA, CBGA, and JMGA grading programs.

Two studies evaluated sorting and feeding zilpaterol hydrochloride (ZH) on feedlot performance and carcass characteristics in randomized block-designed finishing trials. In Exp. 1 (initial BW 342 ± 10 kg, = 1,000), 5 treatments included an unsorted non-ZH fed negative control (-CON), an unsorted ZH fed positive control (+CON), and 3 treatments in which the heaviest 20% within the pen were sorted and marketed 28 d early and the remaining 80% were fed ZH. The 20% were identified at the beginning (EARLY), 100 d from slaughter (MIDDLE), or 50 d from slaughter (LATE). Because of sorting, the remaining steers in sorted treatments were fed 14 d longer than -CON and +CON. Average days on feed for control treatments were 165 and 173 d for the EARLY, MIDDLE, and LATE treatments. In Exp. 2 (initial BW 376 ± 29 kg, = 1,400), 4 treatments included -CON; +CON; an early weight sort fed ZH (1-SORT) with the heaviest 20% identified at d 1 and sorted 50 d from harvest and marketed 14 d before -CON and +CON, with the remaining 80% of the pen fed 7 d longer than -CON and +CON; and a 4-way sort 50 d from harvest fed ZH (4-SORT) with steers sorted into HEAVY, MID-HEAVY, MID-LIGHT, and LIGHT groups marketed -14, 0, +7, and +28 d from -CON and +CON, respectively. Average days on feed for control treatments were 154 and 157 d for the 1-SORT and 159 d for 4-SORT. Steers were fed Zilmax at 8.3 mg/kg DM for 20 d followed by a 3 d withdrawal. In Exp. 1, steers fed +CON had 13 kg greater (P < 0.01) HCW than steers fed -CON. Steers sorted EARLY, MIDDLE, and LATE had 28, 25, and 24 kg heavier ( P< 0.01) HCW than -CON steers, respectively. Carcass weight SD was greater (P = 0.01) for +CON than -CON but was not different (P = 0.17) between -CON and ZH sorted treatments. Percentage of overweight carcasses (454 kg) was greater (P ≤ 0.05) in sorted treatments than in -CON. In Exp. 2, HCW for +CON was 15 kg heavier (P < 0.01) than that for -CON, and HCW for 4-SORT was greater (P < 0.02) than that for +CON. Carcass weight SD was not different (P > 0.10) between +CON and -CON, whereas carcass weight SD of 4-SORT was reduced (P < 0.01) compared with that of -CON and +CON. Steers fed ZH had a greater percentage of carcasses over 454 kg than steers fed -CON (P < 0.01). Although not statistically different (P = 0.27), the percentage of carcasses over 454 kg was reduced by 28% for 4-SORT compared with +CON. Feeding ZH increases carcass weight, but sorting reduces variation, allowing further increases in carcass weight while minimizing overweight carcasses.

Systemic lupus erythematosus (SLE) and lupus nephritis (LN) have strong concomitance with cardiovascular disease that cannot be explained fully by typical risk factors. We examined the possibility that serum or urine expression of adipokines may act as biomarkers for LN, as these proteins have been associated previously with cardiovascular disease as well as SLE. Antibody arrays were performed on serum and urine from lupus patients and matched controls using a cross-sectional study design. From the initial array-based screening data of 15 adipokines, adiponectin, leptin and resistin were selected for validation by enzyme-linked immunosorbent assay (ELISA). Correlations were determined between adipokine expression levels and measures of disease activity or lupus nephritis. The expression of adiponectin and resistin was increased in both sera and urine from LN patients, while leptin was increased in LN patient sera, compared to matched controls. Serum resistin, but not urine resistin, was correlated with measures of renal dysfunction in LN. Serum resistin expression may be useful as a marker of renal dysfunction in patients with LN, although longitudinal studies are warranted. Further studies are necessary to determine if resistin has functional consequences in LN.

A database of daily feed deliveries for steers and heifers fed at 3 commercial feedyards in Kansas between January 1, 2010, and January 31, 2012 (n = 1,515 pens), was used to investigate the prevalence and extent of changes in DMI after initiation of feeding zilpaterol hydrochloride (ZIL) at 8.3 mg/kg (DM) for 20 d. Season affected the percentage of pens experiencing a decrease in DMI post-ZIL (P < 0.01), but there were significant (P < 0.01) season × sex, season × feedyard, season × pre-ZIL DMI, season × days post-ZIL, and season × period post-ZIL interactions. Average DMI decreased within 1 d after initiation of ZIL feeding in all seasons; however, this initial decrease was greater (P < 0.01) in the summer (-0.30 kg) and winter (-0.27 kg) than in the spring (-0.05 kg) or fall (-0.06 kg). The decrease in DMI averaged across all days post-ZIL was greater in summer than during other seasons for both steers and heifers, and the change in intake was greater in steers than heifers in all seasons but fall. Size of intake change within each season varied by feedyard and by season. The percentage of pens that had a large DMI decrease (≥ 0.9 kg/d) was greatest during the summer (33%), and the percentage of pens with no decrease was the least (15%); during the fall, 34% of pens had no DMI decrease and only 8% of pens had a large decrease in DMI. Intake before ZIL initiation affected size and prevalence of DMI decrease; with increasing pre-ZIL DMI, the percentage of pens with a decrease increased from 62% for pens with pre-ZIL DMI of less than 7.7 kg/d to 82% for pens consuming greater than 10.5 kg/d pre-ZIL (P < 0.01). Of those pens with greater than 10.5 kg/d pre-ZIL DMI, 27% had DMI decrease of greater than 1.4 kg/d compared to only 3% for pens consuming <8.7 kg/d pre-ZIL. The average dosage of ZIL consumed per animal with an average DMI of 7.3, 8.2, 9.1, 10.0, and 10.9 kg/d was calculated to be 61, 68, 76, 83, and 91 mg/animal daily, which may be related to the differences in DMI decrease. Pre-ZIL DMI contributed to DMI decrease during ZIL administration, but the increased occurrence and size of DMI decrease during the summer may indicate an additional physiological mechanism.

British × Continental steers (n = 168; 7 pens/treatment; initial BW = 362 kg) were used to evaluate the effect of dose/payout pattern of trenbolone acetate (TBA) and estradiol-17ß (E2) and feeding of zilpaterol hydrochloride (ZH) on serum urea-N (SUN), NEFA, IGF-I, and E2 concentrations and LM mRNA expression of the estrogen (ER), androgen (ANR), IGF-I (IGF-IR), ß1-adrenergic (ß1-AR), and ß2-adrenergic (ß2-AR) receptors and IGF-I. A randomized complete block design was used with a 3 × 2 factorial arrangement of treatments. Main effects were implant (no implant [NI], Revalor-S [REV-S; 120 mg TBA + 24 mg E2], and Revalor-XS [REV-X; 200 mg TBA + 40 mg E2]) and ZH (0 or 8.3 mg/kg of DM for 20 d with a 3-d withdrawal). Steers were fed for 153 or 174 d. Blood was collected (2 steers/pen) at d -1, 2, 6, 13, 27, 55, 83, 111, and 131 relative to implanting; LM biopsies (1 steer/pen) were collected at d -1, 27, 55, and 111. Blood and LM samples were collected at d -1, 11, and 19 relative to ZH feeding. A greater dose of TBA + E2 in combination with ZH increased ADG and HCW in an additive manner, suggesting a different mechanism of action for ZH and steroidal implants. Implanting decreased (P < 0.05) SUN from d 2 through 131. Feeding ZH decreased (P < 0.05) SUN. Serum NEFA concentrations were not affected by implants (P = 0.44). There was a day × ZH interaction (P = 0.06) for NEFA; ZH steers had increased (P < 0.01) NEFA concentrations at d 11 of ZH feeding. Serum E2 was greater (P < 0.05) for implanted steers by d 27. Serum trenbolone-17ß was greater (P < 0.05) for implanted steers by d 2 followed by a typical biphasic release rate, with a secondary peak at d 111 for REV-X (P < 0.05) implanted steers. Implanting did not affect mRNA expression of the ANR or ER, but the IGF-IR and the ß1-AR and ß2-AR were less (P < 0.05) for REV-S than NI at d 55 and ß2-AR mRNA was less (P < 0.05) for REV-S than for REV-X. Expression of the IGF-IR and the ß1-AR at d 111 was greater (P< 0.05) for REV-X than for REV-S and NI at d 111, and the ß2-AR was less (P< 0.05) for REV-S than for REV-X. Feeding ZH did not affect mRNA expression of the ß1-AR and ß2-AR. Both implanting and feeding ZH decreased SUN, but a greater dose of TBA + E2 did not result in further decreases. In addition, feeding ZH increased serum NEFA concentrations. Metabolic changes resulting from implanting and feeding ZH may aid in explaining steer performance and carcass responses to these growth promotants.

Mechanical portioning tests were performed on beef rib, strip loin, tenderloin, and top sirloin subprimals obtained from calf-fed Holstein steers to characterize the influence of zilpaterol hydrochloride (ZH), ractopamine hydrochloride (RH), or no ß-adrenergic agonist (ßAA; CON) on subprimal and steak yield. In addition, ßAA effects on tenderness, composition, and raw and cooked color of steaks from the aforementioned strip loin subprimals were characterized. At 14 to 15 d (ribs, tenderloins, and top sirloin) or 16 d (strip loin) postmortem, subprimals were portioned into steaks using a mechanical portioning machine. The appropriate variables were measured before and after portioning to determine ßAA influence on trimmed and untrimmed subprimal weight, subprimal length (rib only), steak weight and yield, and steak thickness (rib only). Steaks obtained from the strip loin subprimals were subjected to analysis of raw instrument color (L*, a*, b*), proximate composition, and pH. In addition, strip steaks were aged (16 or 23 d) before analysis of cooked internal color, Warner-Bratzler shear force (WBSF), and slice shear force (SSF). Briefly, ZH supplementation increased (P < 0.01) the weight of all subprimals when compared to CON. Furthermore, subprimals from CON animals consistently had fewer and lighter steaks (P ≤ 0.04) than subprimals from ZH-fed steers. Additionally, raw steaks from ZH cattle were a less vivid red (lower a* and saturation index values; P < 0.01) when compared to CON and RH steaks, which did not differ (P > 0.05). There was no interaction between ßAA treatment and postmortem aging length for WBSF or SSF (P > 0.10). However, CON steaks (3.25 kg) had lower WBSF values (P < 0.05) than ZH or RH steaks (3.68 and 3.67 kg, respectively). Regardless, aging for 23 d vs. 16 d resulted in decreased WBSF and SSF (P < 0.01) for all ßAA treatments. Although differences were numerically small, evaluations indicated the internal cooked surfaces of ZH and RH steaks were less red (P < 0.05) than CON steaks. Overall, these data reemphasize increased subprimal weights due to ßAA supplementation, particularly ZH. However, the data are not indicative of increased steak yield due to ßAA supplementation. Furthermore, the data demonstrate ßAA supplementation increases the shear force of calf-fed Holstein strip steaks regardless of postmortem aging period. However, no differences in shear force between the ßAA treatments (ZH or RH) were noted.

Holstein steers (n = 2,275) were assigned to 1 of 3 treatments: 1) a control diet containing no ß-agonists, 2) a diet that contained zilpaterol hydrochloride (ZH; 8.3 mg/kg [100% DM basis]) for 20 d with a 3-d withdrawal period before harvest, and 3) a diet that contained ractopamine hydrochloride (RH; 30.1 mg/kg [100% DM basis]) for 28 d before harvest. No differences (P ≥ 0.18) were detected between treatments for initial BW, BW at d 28, or DMI. Final BW, BW gain for the last 28 d, total BW gain, ADG for the last 28 d, and overall ADG were greater (P < 0.05) for steers fed ZH or RH than for steers fed the control diet. Additionally, G:F for the last 28 d and G:F for the entire trial was increased (P < 0.02) for steers fed ZH (0.147, 0.147) or RH (0.153, 0.151) compared to steers fed the control diet (0.134, 0.143), respectively. Steers fed ZH or RH had HCW that were 15.5 and 8.2 kg heavier (P ≤ 0.01) and LM areas that were 7.1 and 2.3 cm(2) larger (P < 0.01) than control cattle. Steers fed ZH also had dressed carcass yields that were 1.3% to 1.5% greater and USDA calculated yield grades that were decreased 0.16 to 0.23 units compared to RH and control steers. No differences (P ≥ 0.39) were found between treatments for marbling score, fat thickness, and percentage KPH. Steers fed ZH had an increased (P ≤ 0.04) percentage of yield grade 1 and 2 carcasses (15.1, 55.0) and a reduced (P ≤ 0.02) percentage of yield grade 3 carcasses (27.1) compared with those fed RH (10.5, 49.1, 36.1) or the control diet (9.0, 47.4, 36.4), respectively. Additionally, ZH-fed steers had a decreased (P ≤ 0.04) percentage of yield grade 4 and 5 carcasses (2.8) compared with steers fed the control diet (6.9). Steers fed ZH had an increased (P ≤ 0.01) percentage of USDA Select grading carcass (31.0%) and a decreased (P ≤ 0.01) percentage of USDA Choice grading carcasses (65.0%) compared with steers fed RH (25.8%, 70.2%) and no ß-agonist (24.8%, 72.0%), respectively. Feeding either ß-agonist to calf-fed Holstein steers increased live performance through increased BW, BW gain, and ADG. Furthermore, supplementing calf-fed Holstein steers with ZH provides greater improvements in HCW, LM area, and yield grade components, with a slight decrease in quality grade when compared to calf-fed Holstein steers supplemented with RH.

Beef steers (n = 1,914) were assigned to 1 of 3 ß-adrenergic agonist (ßAA) supplementation treatments-zilpaterol hydrochloride (ZH; 8.3 mg/kg of DM for 20 d with 3-d withdrawal), ractopamine hydrochloride (RH; 308 mg·head(-1)·d(-1) for 28 d), or no ßAA (CON)-to determine the effects on consumer eating quality. Strip loins (n = 1,101; CON = 400, RH = 355, and ZH = 346) were obtained and fabricated into 2.5-cm-thick steaks for proximate, Warner-Bratzler shear force (WBSF), slice shear force (SSF), and consumer analyses; steaks were aged until 14 or 21 d postmortem. Fat and moisture contents were not affected by ßAA supplementation (P > 0.05), but strip steaks from steers fed ZH had more protein (P < 0.01) than those from steers fed CON or RH, which were similar. An interaction between ßAA and aging was observed (P < 0.01) for WBSF but not SSF. Within steaks aged 14 d, ZH steaks required the most force to shear, RH steaks were intermediate, and CON steaks had the lowest WBSF values; however, RH steaks had a stronger response to aging than CON or ZH steaks, resulting in the lowest WBSF values at 21 d. Slice shear force values were greater (P < 0.01) in steaks from steers fed ZH than in steaks from steers fed CON or RH, which did not differ. Following shear force analyses, steaks within 2 SD of each treatment mean for WBSF were selected randomly for consumer assessment of eating quality. Consumer testing (n = 400; 200/postmortem aging period) was arranged in a 3 × 3 factorial representing 3 quality grades (Select, Low Choice, and Premium Choice) and 3 treatments (ZH, RH, and CON). In steaks aged 14 d, ßAA supplementation affected (P < 0.01) tenderness, flavor, and overall liking and tenderness acceptability, resulting in lower consumer scores for ZH than CON and RH; however, juiciness, flavor, and overall acceptability were similar (P > 0.05). In steaks aged 21 d, feeding ßAA influenced (P < 0.01) only tenderness and juiciness scores. Despite these differences, ßAA did not affect (P > 0.05) acceptability. Quality grade impacted (P < 0.01) all traits and acceptability in steaks aged 14 and 21 d. In 14-d steaks, Premium Choice typically was scored higher than Low Choice or Select; however, consumers rated 21-d Low Choice and Premium Choice similarly-both receiving greater scores than Select. Consumers detected several differences in eating quality at 14 d because of ßAA supplementation. Increasing aging from 14 to 21 d mitigated differences in shear force and tenderness scores because of feeding ZH, so that tenderness and overall acceptability were similar between ZH, RH, and CON.
