OBJECTIVE: The aim of the study is to examine associations between paternal age at childbirth and offspring cardiovascular risk factors in adolescence. STUDY DESIGN: This is a cross-sectional study. METHODS: Data from the 2007-2016 Korea National Health and Nutrition Examination Survey was used. A total of 4,096 adolescents were included in the final analysis, and their information on blood pressure, fasting glucose level, and lipid profile was collected. Multiple linear regression models were applied to evaluate the effect of paternal age on cardiovascular risk factors with adjustment for potential confounders. RESULTS: The median age of participants was 13 years, and 53.2% were male. Maternal and paternal ages were closely correlated with each other, and older parents had older offspring. Paternal age was positively associated with levels of total cholesterol (p = 0.033) and triglycerides (p = 0.042) after adjusting for confounders. CONCLUSION: This nationwide study shows that advanced paternal age is associated with a less favorable lipid profile in a dose-dependent manner.

BACKGROUND: Numerous studies have shown that osteoporosis is common in kidney transplant recipients. However, the change in bone mineral density after kidney transplantation (KT) is not fully understood. METHODS: Thirty-nine kidney transplant recipients with bone densitometry at pretransplant and 24 months after KT were reviewed. RESULTS: The recipients' median age (44.5 ± 10.7 years) and dialysis duration before KT (4.2 ± 3.4 years) were recorded. The T-scores of the lumbar spine and femur neck at 24 months after KT were positively associated with the respective pretransplant T-score (P < .001 in the lumbar spine and P < .001 in the femur neck). However, the T-score after KT did not show significant change (P = .680 in lumbar spine, P = .093 in femur neck). Changes in the T-scores of the lumbar spine and femur neck over 24 months (delta T-score) were negatively associated with the respective pretransplant T-scores (P = .001 in lumbar spine, P = .026 in femur neck). Changes in the T-scores of the lumbar spine and femur neck over 24 months (delta T-score) were also associated with the pretransplant T-scores after the adjustment of other variables. CONCLUSION: The change of bone mineral density was related with pretransplant bone mineral density. Careful follow-up of bone densitometry for KT recipients was needed.

RESUMO

Esophageal carcinoids are exceedingly rare and only a few limited cases have been reported. Because knowledge about esophageal carcinoid is based primarily on case reports, the treatment of choice has not been settled. We report here on a localized carcinoid tumor in the esophagogastric junction. As our initial biopsy was nearly equal for the excision, we only performed coagulation ablation therapy instead of a wide excision which is traditionally recommended. The patient is free of recurrence so far during 50-month follow-up. Our report suggests that esophageal carcinoids may be not particularly associated with a poor prognosis.

PURPOSE: It is important to identify a 'metabolically unhealthy obese' subset with higher cardiovascular risk among obese individuals. We investigated the associations between the GNB3 C825T polymorphism and obesity-related metabolic risk factors among Korean obese women. METHODS: This study was a sub-investigation of a double-blind randomized controlled trial that examined the additive effect of or list at on weight loss with sibutramine. A sample of 111 obese women were divided into T-carriers (CT/TT) or a homozygous CC group, according to the presence of the 825T allele at GNB3. These groups were compared to determine their associations with obesity-related metabolic risk factors, i.e., fasting plasma glucose, serum lipids, serum insulin/insulin resistance, and abdominal fat amounts. RESULTS: The allele frequencies of the GNB3 polymorphism were C allele = 59.5% and T allele = 40.5%. The T allele was found to be significantly associated with greater visceral fat and higher serum lipids, and these significances remained robust after adjusting for potential covariates. CONCLUSIONS: The GNB3 825T polymorphism is significantly associated with greater visceral fat and higher serum lipids in Korean obese women and it suggests that the GNB3 C825T is a determinant of obesity-related metabolic traits in this population.

With the aim to effectively enhance the payload for nanocarriers, supersaturated deltamethrin (SSD) solution was prepared using the solvent-mediated method to produce lipid nanocarriers by a combination method of homogenization and sonication. In this study, deltamethrin was used as an active ingredient, corn oil was used as a lipid medium, soybean lecithin and Tween-80 were used as surfactants. At 25°C, the solubility of deltamethrin in SSD solution prepared by solvent-mediated method was 3.4 times and 1.5 times higher than that in saturated deltamethrin (SD) solution and that in SSD solution prepared by thermal-mediated method, respectively. Therefore, compared to the use of SD solution for the production of nanocarriers, the use of SSD solution significantly enhanced the payload, while keeping the encapsulation efficiency high. Particularly, nanocarriers produced from SSD solution prepared by solvent-mediated method had the higher payload of 2.1 times and 4.4 times compared with preparations using SSD solution prepared by thermal-mediated method and SD solution, respectively.

No clinical studies on the lipolytic effect of guanine nucleotide-binding protein ß3 subunit gene (GNB3) 825T polymorphism have been performed. This study was a subinvestigation of a 12-week randomized controlled trial (NCT01184560) for the additive effect of orlistat on sibutramine treatment. The analysis involved 101 obese females aged 18-49 years, genotyped at the GNB3 825 locus. To exclude any influence from potential confounders, we used an analysis of covariance model. After the intervention, fat mass proportion in total weight loss was significantly lower in subjects with a T allele than in those without a T allele (p = 0.034). GNB3 825T allele was associated with blunted fat mass reduction in obese females.

With the aim to establish a novel nanocarrier system with higher payload and higher photo-protection for deltamethrin (active ingredient) compared to solid lipid nanoparticles (SLN), nanostructured lipid carriers (NLC) were prepared by combining hot homogenization and sonication with different ratios of corn oil (liquid lipid) and beeswax (solid lipid). Compared to SLN, the incorporation of corn oil gave a higher payload, slower release rate and higher photo-protection for deltamethrin. Particularly, compared to beeswax solid lipid nanoparticles (BSLN), NLC gave a higher payload (approximately 1.79 times) with a high encapsulation efficiency (83.6%), higher photo-protection in direct photolysis (1.8 times) after 12 h exposure of ultraviolet light (UV) and in indirect photolysis (1.37 times) after 2 h exposure of UV in 2% (v/v) acetone solution used as a photosensitizer. This study demonstrated that novel nanocarrier possesses a wide range of applicability in protecting photo-labile compounds for the crop, food and pharmaceutical industries.

Chitosan-coated nano-liposomes containing etofenprox were prepared by ultrasonic homogenization (UH) and a combined use of UH and electro-spraying. The physicochemical properties of the resulting samples were examined and compared. The two methods yielded similar values and tendencies, except for encapsulation efficiency that differed by an average of 15%. In the coating process, as the chitosan concentration increased (0.1-0.5%, w/v) and the degree of deacetylation increased (chitosans A, B and C), the surface charge of the nano carrier likewise increased and carrier size distribution was altered. The encapsulation efficiency as measured by gas chromatography decreased slightly with the increasing chitosan concentration (0.1-0.5%, w/v). The results indicate that diverse preparation conditions could affect the physicochemical properties of the resulting nano carrier systems.

Chitosan-coated nanoliposomes containing etofenprox or alpha-cypermethrin prepared by ultrasonic homogenization maintained a size distribution in the nanometre range. Nanoliposomes were constructed using different types and concentrations of chitosan to regulate the mean size and surface charge. As the chitosan concentration (0.1-0.5%, w/v) and the degree of deacetylation increased, surface charge also increased. The encapsulation efficiency and release profile were measured by gas chromatography. Encapsulation efficiency decreased slightly as chitosan concentration increased (0.1-0.5%, w/v). As the intrinsic surface charge or concentration of the coating material increased, the release period of the entrapped core material was extended (chitosans A and B; 0.1 and 0.3%, w/v). The results indicate that diverse preparation conditions could affect the physicochemical properties and release profile of the resulting nanocarrier systems.

In evaluating toxicity, one of the most important factors is the administration method, because it can affect the exposure and absorbance level of the test article, and, consequently, influence the interpretation of toxicity test results. Continuous intravenous (IV) administration is a widely used administration method for anti-cancer drugs in clinical settings. Previous studies have reported the toxic effects of the test article following repeated IV dosing of CKD-602, a novel camptothecin-derivative anti-tumor agent that was developed by Chong Kun Dang Pharmaceutical Corporation in Seoul, Korea. However, CKD-602-related toxicities induced by IV infusion administration have not yet been evaluated, although the drug is more widely used in clinical settings. In the present study, CKD-602 was administered using a continuous IV infusion pump and using repeated IV administration at doses of 0, 0.003, or 0.01 mg/kg/day for 4 weeks to compare and evaluate the drug-induced toxicities using the two different administration methods. Higher mortality, more severe clinical symptoms, increased complete blood count, serum biochemistry, and histopathology were demonstrated when CKD-602 was administered using the 4-week continuous IV infusion pump method compared with the repeated IV administration method. Based on these results, we conclude that the administration of CKD-602 using the 4-week continuous IV infusion pump method can elicit more severe toxicity than that using 4-week repeated IV dosing method. Thus, more attention should be paid to the administration of CKD-602 using continuous IV infusion in the clinical setting.

Nano carrier systems were prepared by forming self-assembled liposomes having a size distribution in the nano range through use of an ultrasonic homogenizer. Phosphatidylcholine and cholesterol were utilized as an amphiphilic compound and a shape stabilizer, respectively. The size of prepared samples was decreased (up to 150 nm) by elevating ratio of lecithin and extending homogenization time (2 to approximately 6 min). After secondary coating with alginic acid (0.1, 0.3 and 0.5%, W/V), size was remarkably changed in the range of +/-30 nm and zeta-potential was altered (only chitosan coating (molecular weight: 30 000 Da, 0.2%, W/V): 10.3 mV, Alginic acid coating (0.5%, W/V) after the chitosan coating: -21.8 mV). The low molecular weight chitosan (0.1%, W/V)-coated nano-liposomes had a lower absolute value of zeta-potential than the high molecular weight chitosan (0.1%, W/V)-coated nano-liposomes. The encapsulation efficiency was measured by gas chromatography. The efficiency was decreased slightly by elevating chitosan concentration (0.1 to approximately 0.5%, W/V).
