RESUMO

The influence of the new cardioprotector flocalin was investigated on the culture of rat's neonatal cardiomyocytes during anoxia-reoxygenation modelling. The mechanisms of apoptosis and necrosis were investigated under influence of ATP-sensitive potassium (K(ATP)) channels activation and in conditions of blocking of the L-type calcium (VGCCs) channels. Flocalin was added in the culture medium in the dose 5 and 20 microM at 2 minutes before anoxia (30 minutes) and following reoxygenation (60 minutes). These doses are near to: the first dose means the opening of K(ATP) channels and the second one means the IC50 block of VGCCs. It is discovered that in dose 5 microM of flocalin drew the change of correlation of living, necrotizing and apoptizing cells drew side-shifting living. The number of live cells was almost the same like in control (experiments without anoxia-reoxygenation modelling). The opening of K(ATP) channels decreases necrosis in two times and fully prevented development of apoptosis which was induced anoxia-reoxygenation modelling. Flocalin depressed the apoptosis of neonatal cardiomyocytes so that he was on to 36% less than in control group (without anoxia-reoxygenation). But in the high dose (20 microM) that provokes not only K(ATP) channels opening but also IC50 block of VGCCs cardioprotection was not detected after modelling of anoxia-reoxygenation. The last can be investigation both enough strong activating of the potassium channels and by investigation of both factors are opening of potassium and inhibition of VGCCs channels and, accordingly, substantial diminishing of level of introcellular Ca2+ and violation of metabolic processes yet to anoxia-reoxygenation. Thus, small doses of flocalin, that induce moderate opening of K(ATP) channels significantly decrease the number of necrotic and apoptotic cells in culture of rat neonatal cardiomyocytes induced by anoxia-reoxygenation.

Clinical physiological monitoring of 32 men aged 25-60 years with chronic stoneless cholecystitis in the phase of remission found that in 14 patients 75-85 minutes after drinking the bioactive water Naftussya (3 ml/kg, t(0) 18-20 degrees C) the plasma level of total triiodothyronine (T3) was not significantly changed, while in 9 patients it was reduced to 0.26 +/- 0.10 nM/1 (by 11%). In the other 9 patients the level increased to 0.29 +/- 0,04 nM/1 (by 17%). The changes in T3 level correlated negatively with the changes in absolute (r = -0.55) and relative (r = -0.47) power spectral density (PSD) of ultra very low-frequency components of heart rate variability (HRV). These changes correlated positively (r = 0.46) with the changes in relative PSD of low-frequency components of HRV. Subject to a weak correlation with HRV parameter pNN50 (r = -0.21) canonical correlation coefficient (R) between changes in HRV parameters and T3 reaches 0.68 (p = 0.002). On the other hand, the changes in T3 correlated with its initial level (r = -0.46), as well as initial levels of moda (r = -0.40), its amplitude (r = 0.30) and absolute PSD of low-frequency components HRV (r = -0.33), i.e. thyrotropic effects conditionized these parameters by 30% (R = 0.55; p = 0.038). The method ofdiscriminant analysis has identified 14 primary parameters that allowed predicting the thyrotropic effect of Naftussya water with 100% accuracy.

RESUMO

The distribution of thiamine-binding and thiamine triphosphatase activity typical of thiamine-binding proteins was studied in intracellular structures of rats liver and kidneys. It was found that the fraction of microsomes has the highest rate of specific thiamine-binding activity amide fractions of subcellular structures that was isolated using differential centrifugation in the both organs. Hydrolysis of thiamine triphosphate (pH 7.4) was also extremely active in these structures. The results of our research allow to make a conclusion that subcellular structures precipitated as fraction of microsomes (endoplasmic reticulum and vesicled parts of plasma membranes) are the sites of the most probable localisation of thiamine-binding proteins of liver and kidneys.

The effects of both arachidonic acid (AA) and inhibition of its metabolism on the antibody-induced changes in the mechanical contraction and action potentials of the isolated papillary muscles were studied. It has been shown that antibody-dependent induction of AA oxidation led to the initiation of Ca2+ entry through voltage-dependent Ca2+ channels of a myocyte. Inhibition of the lipoxygenase way of AA peroxidation prevented Ca2+ overload of cardiomyocytes more than inhibition of the cyclooxygenase way, but less than blocking Ca2+ channels.

RESUMO

The study of thiamine-binding proteins (ThBP) isolated from liver and kidneys of rats was held in order to find out the peculiarities and physiological role of the ThBP isolated earlier from the rat brain. It was demonstrated that ThBP from liver and kidneys of rats as well as ThBP from rat brain described earlier, were bifunctional: on an equal footing with ability to bind thiamine specifically, they show an ability to hydrolyse the phosphoric esters of thiamine selectively. The ThBP of these tissues (liver, kidneys and brain) didn't differ by the molecular weight, but differed by the enzymatic activities. The molecular weight of ThBP was estimated to be 100 kDa by gel-filtration; 63 kDa and 35 kDa by sodium dodecylsulfate gel electrophoresis. Specific thiamine-binding activity increases as follows: ThBP from rat brain < ThBP from rat liver < ThBP from rat kidneys.

RESUMO

Action of antibodies specific to the myocardial sarcolemma of rat on contraction of the isolated cardiac muscle was investigated. It is found that initially antibodies cause stimulation of the cell function (an increase in force or frequency of contractions). Then a slow and progressive inhibition of phasic contractions and development of tonic tension and contracture are observed. Those different effects indicate that antibodies induce an increase in intracellular Ca2+ concentration. The direct measurements have shown that the Na(+)-K(+)-pump activity is depressed by the action of antibodies. Possibly one of the mechanisms of inhibition is lipid peroxidation of the membrane under the influence of antibodies.
