RESUMO

The paper is concerned with the experimental data on a ERP signal size in two blood plasma metalloproteins--ceruloplasmin and transferrin and their correlations after intraperitoneal hydrocortisone and corticotropin injections (5 mg and 2.5 U/100 g body weight, respectively) to Wistar adult male rats and after bilateral adrenalectomy. A conclusion has been made that antioxidant activity of the ceruloplasmin-transferrin complex in rat blood plasma is increased with a rise of the corticotropin level but it is decreased with hydrocortisone loading.

RESUMO

Intensity of 3H-dopamine uptake in hypothalamic synaptosomes was similar both in intact rats and in the animals which received a single administration of bromocriptine at a dose of 5 mg/kg within 4 hrs. Within 4 days after the drug administration incorporation of the label was increased 1.22-fold and 1.28-fold in intact and adrenalectomized rats, respectively. Alterations in dopamine uptake in hypothalamic synaptosomes caused by bromocriptine appear to be one of dopaminergic-dependent effects of the drug in the animal brain structures.

RESUMO

The effect of nakom, containing the dopamine precursor levodopa, on the activity of the hypothalamo-hypophysial-adrenal system (HHAS), the content of dopamine in the hypothalamus and the cerebral hemisphere cortex and cAMP in tissues, was studied in experiments on rats. It was revealed, that the preparation in the 2 mg per 100 g dose has a marked inhibiting action on the activity of the HHAS, that is expressed in 2 h by the drop of the level of ACTH, corticosterone, aldosterone in blood that is maintained for 10 hours and disappears 24 hours after the administration. One of the possible mechanisms of the inhibiting action of the preparation on HHAS activity is the increase of dopamine content in the hypothalamus and cerebral hemisphere cortex. The inhibiting action of the preparation on HHAS activity is dose-dependent. The inhibiting influence of nakom on the HHAS activity is realized through the adenylate cyclase system.

RESUMO

Specific insulin binding with insulin receptors of fatty acid plasma membranes is established to be intensified 10 days after adrenalectomy in rats due to an increase in the receptor number. Hydrocortisone administered for 10 days in a dose of 1 mg per 100 g of body mass to adrenalectomized rats for substitution therapy and to intact ones for 14 days in a dose of 5 mg per 100 g of body mass to induce hypercorticism inhibits the expression of insulin receptors of fatty plasma membranes because of their number and affinity for the hormone. The data obtained confirm information on an inhibitory effect of glucocorticoids on the expression of insulin receptors.

A study was made of the effect of adrenalectomy (10 days after operation), administration of hydrocortisone to adrenalectomized rats (1 mg per 100 g bw a day for 10 days, starting from the 1st day after operation), and hydrocortisone-induced hypercorticoidism (5 mg per 100 g bw a day for 14 days) on the parameters of insulin receptors of liver plasma membranes. Depiction of the obtained data in the form of the Sketchard-Meites-Rose dependencies has shown that adrenalectomy increases the concentration and affinity of the receptors for the hormone. Administration of hydrocortisone to adrenalectomized rats reduces the concentration of the receptors, while hypercorticoidism decreases their affinity without affecting the concentration. The data obtained suggest an indirect action of glucocorticoids on the insulin receptors in liver plasma membranes.
