RESUMO

The effects of acute and chronic ethanol administration on neurotensin-like immunoreactivity (NTLI) were investigated in discrete regions of the rat brain. Rats were administered a single dose (5 g/kg) of ethanol or rendered ethanol-dependent by repeated forced drinking of a 20% ethanol solution (6-15 g/kg) for 4 days and tested while still intoxicated or at 12-19 h (W-1) or 7 days (W-2) after ethanol withdrawal. Increases in NTLI were seen in the caudate-putamen in the toxic, W-1 and W-2 groups and decreases were observed in the midbrain in the toxic and W-1 groups. It was assumed that ethanol produced the changes of NTLI in the caudate-putamen and midbrain through its actions upon the dopaminergic system.

RESUMO

Anti-idiotypic monoclonal antibodies that interact with the binding site of sigma receptors were generated. First, BALB/c mice were immunized with a haloperidol-bovine serum albumin conjugate, and monoclonal anti-haloperidol antibodies that recognize the piperidinyl moiety of haloperidol molecule were obtained. Second, for generation of anti-idiotypic antibodies, BALB/c mice were immunized with the anti-haloperidol monoclonal antibodies coupled to keyhole limpet hemocyanin. Anti-idiotypic antisera and three hybridomas secreting anti-idiotypic monoclonal antibodies were obtained. All of them were shown to inhibit [3H]haloperidol binding to the anti-haloperidol antibodies. The anti-idiotypes were potent in displacing the binding of [3H]haloperidol to rat brain sigma receptors. Furthermore, they significantly immunoprecipitated the sigma receptors from a detergent-solubilized preparation. These findings demonstrate the generation of anti-idiotypic monoclonal antibodies specifically interacting with membrane-bound and solubilized sigma receptors.

The electroencephalograms (EEGs) of 22 patients suffering from hereditary DRPLA were studied. Epileptoform patterns were observed in 14 patients (63.6%) with epileptic seizures. The epileptoform patterns most frequently observed were those atypical spike-wave complexes. Slow wave bursts were seen in 18 patients (81.8%). Photosensitivity was revealed in six (27.3%) patients, all of whom presented progressive myoclonus epilepsy (PME) syndrome. Abnormal background activity was evident in 17 (77.3%) patients. These abnormalities in EEG were more severe in patients in the PME type than those of the A (ataxia) and AE (ataxia and epilepsy) types.
