RESUMO

Rapid identification of bacterial species in patient samples is essential for the treatment of infectious diseases and the economics of health care. In this study, we investigated an algorithm to improve the accuracy of bacterial species identification with fluorescence spectroscopy based on autofluorescence from bacteria, and excitation wavelengths suitable for identification. The diagnostic accuracy of each algorithm for ten bacterial species was verified in a machine learning classifier algorithm. The three machine learning algorithms with the highest diagnostic accuracy, extra tree (ET), logistic regression (LR), and multilayer perceptron (MLP), were used to determine the number and wavelength of excitation wavelengths suitable for the diagnosis of bacterial species. The key excitation wavelengths for the diagnosis of bacterial species were 280 nm, 300 nm, 380 nm, and 480 nm, with 280 nm being the most important. The median diagnostic accuracy was equivalent to that of 200 excitation wavelengths when two excitation wavelengths were used for ET and LR, and three excitation wavelengths for MLP. These results demonstrate that there is an optimum wavelength range of excitation wavelengths required for spectroscopic measurement of bacterial autofluorescence for bacterial species identification, and that measurement of only a few wavelengths in this range is sufficient to achieve sufficient accuracy for diagnosis of bacterial species.

Biological nitrogen fixation (BNF) is important to sustain nitrogen fertility of paddy soil and rice yield, while could be affected by nitrogen fertilization. Iron-reducing bacteria, Anaeromyxobacter and Geobacter, are newly found diazotrophic bacteria predominant in paddy soil. Experimental field of this study is a long-term (35 years) nitrogen fertilized (6.0 g N/m2/year) and unfertilized paddy field, where ca. 70% of rice yield was obtained yearly in nitrogen unfertilized plot (443 ± 37 g/m2) compared to fertilized plot (642 ± 64 g/m2). Effects of long-term nitrogen fertilization/unfertilization on soil properties related to BNF were investigated with special reference to diazotrophic iron-reducing bacteria. Soil chemical/biochemical properties, soil nitrogen-fixing activity, and community composition of diazotrophic bacteria were similar between nitrogen fertilized and unfertilized plot soils. In both plot soils, Anaeromyxobacter and Geobacter were the most predominant diazotrophs. Their nifD transcripts were detected at similar level, while those of other general diazotrophs were under detection limit. It was concluded that long-term use/unuse of nitrogen fertilizer in this field did not affect the predominance and nitrogen-fixing activity of diazotrophic iron-reducing bacteria, composition of other general diazotrophs, and the resulting soil nitrogen-fixing activity. BNF, primarily driven by diazotrophic iron-reducing bacteria, might significantly contribute to sustain soil nitrogen fertility and rice yield in both plot soils. Appropriate soil management to maintain BNF, including diazotrophic iron-reducing bacteria, will be important for sustainable soil nitrogen fertility and rice production.

RESUMO

A counter-propagating laser-beam platform using a spherical plasma mirror was developed for the kilojoule-class petawatt LFEX laser. The temporal and spatial overlaps of the incoming and redirected beams were measured with an optical interferometer and an x-ray pinhole camera. The plasma mirror performance was evaluated by measuring fast electrons, ions, and neutrons generated in the counter-propagating laser interaction with a Cu-doped deuterated film on both sides. The reflectivity and peak intensity were estimated as ∼50% and ∼5 × 1018 W/cm2, respectively. The platform could enable studies of counter-streaming charged particles in high-energy-density plasmas for fundamental and inertial confinement fusion research.

Formation of a desirable submillimeter-scaled assembled structure of particles in the colloid is a difficult subject in colloidal chemistry. Herein, a submillimeter-scaled ordered assembled structure consisting of highly anisotropic two-dimensional plate-like particles, niobate nanosheets, was obtained through an optical manipulation technique that was assisted by a scattering-force-induced stream. A 532 nm continuous wave laser beam with a power of 400 mW was used to illuminate a liquid crystalline niobate nanosheet colloid from the bottom side of a sample cell, inducing the stream of oriented nanosheets toward the upper side of the sample cell. As a result, a 200 µm ordered assembled structure consisting of oriented nanosheets was formed. The assembled structure was also characterized by two-dimensional anisotropy, reflecting that the highly anisotropic morphologies of each nanosheet and the shape of that structure were dependent on the polarization of incident illumination. This study has revealed a new noncontact and on-demand way to obtain submillimeter-scaled ordered anisotropic colloidal assembled structures of nanosized particles such as nanosheets, contributing to fundamental materials science and expanding the utilities of nanosheets.

The shape of an instance hole (keyhole) created via a high-power laser was measured using a low-coherence interferometer with the following parameters: repetition rate, 10 MHz; center wavelength, 1550 nm; absolute spatial resolution, 10 µm; and measurement range, 5 mm. The keyhole was created on a 3-mm-thick stainless-steel plate using a high-power laser with 8-kW peak power and 1070-nm center wavelength. The cross-sectional area of the keyhole was measured to be 0.42 mm × 0.78 mm (width × depth) using the interferometer, and its side dimension was 0.46 mm × 0.78 mm (width × depth).

Pellet injection and repetitive laser illumination are key technologies for realizing inertial fusion energy. Numerous studies have been conducted on target suppliers, injectors, and tracking systems for flying pellet engagement. Here we for the first time demonstrate the pellet injection, counter laser beams' engagement and neutron generation. Deuterated polystyrene (CD) bead pellets, after free-falling for a distance of 18 cm at 1 Hz, are successfully engaged by two counter laser beams from a diode-pumped, ultra-intense laser HAMA. The laser energy, pulse duration, wavelength, and the intensity are 0.63 J per beam, 104 fs, and 811 nm, 4.7 × 10(18) W/cm(2), respectively. The irradiated pellets produce D(d,n)(3)He-reacted neutrons with a maximum yield of 9.5 × 10(4)/4π sr/shot. Moreover, the laser is found out to bore a straight channel with 10 µm-diameter through the 1-mm-diameter beads. The results indicate potentially useful technologies and findings for the next step in realizing inertial fusion energy.

RESUMO

Fluorescence correlation spectroscopy is a powerful technique for studying the structures and dynamics of living cells. Dynamic light scattering (DLS) is also used to study dynamic characteristics and it has the potential to measure cell dynamics. However, it is difficult to apply DLS to highly scattering media. In this article, we review low-coherence dynamic light scattering (LC-DLS). It strongly suppresses the influence of multiple scattering and has a greater potential for measuring cell dynamics than conventional DLS. The properties of LC-DLS are described theoretically and experimentally. Measurement of the diffusion coefficients of macromolecules in turbid media and interparticle and molecular interactions by LC-DLS is demonstrated.

The hydrodynamics of Brownian particles close to a wall is investigated using low-coherence dynamic light scattering. The diffusion coefficient of the particles in a suspension is measured as a function of distance from the wall. A sudden reduction in the diffusion coefficient near the interface is clearly observed using this method. The theoretically predicted wall-drag effect is experimentally confirmed when the influence of the spatial resolution due to the finite coherence length of the light source is accounted for. The space-dependent dynamics of Brownian particles under the wall-drag effect is obtained for the first time using our spatially resolved dynamic light scattering technique.

We are concerned with dynamic properties of interacting Brownian particles in concentrated colloidal suspensions. An effective diffusion coefficient measured by the modulated in phase low-coherence dynamic light scattering technique is investigated as a function of the volume fraction. The experimental results are compared with the numerical ones calculated under both the Cohen-de Schepper approximation for hydrodynamic interaction and the Percus-Yevick approximations for structural effect. It is confirmed that the Brownian motion of particles in the range of volume fraction from 0.01 to 0.2 is mainly dominated by the hydrodynamic interaction rather than the structural effect, which can be described well by the Cohen-de Schepper approximation.

The path-length-resolved power spectrum of a time-varying scattered light field measured by a time-of-flight method or low-coherence interferometry is evaluated by a new numerical simulation algorithm. The path-length-resolved power spectrum is theoretically derived by combining diffusing-wave-spectroscopy theory and radiative-transfer theory. The proposed algorithm, using the Monte Carlo method, is used to determine the scattering configurations and numerically calculate the power spectrum. The path-length distribution, path-length-dependent scattering order distribution, and path-length-resolved power spectrum are demonstrated numerically over all scattering orders. The resultant power spectra agree with experimental results measured by the low-coherence-dynamic-light-scattering method.

Single-scattering spectroscopy by use of a low-coherence interferometer is introduced to measure the power spectra of light scattered from extremely dense colloidal suspensions. The power spectrum of a heterodyne component can be obtained by subtraction of the power spectrum of a homodyne component from the measured power spectrum. The heterodyne power spectrum for light scattered from the medium is shown to coincide with the single-scattering spectrum to a depth of up to a few times the mean-free path length. Therefore single-scattering spectroscopy is newly proposed as a means by which to analyze the characteristics of extremely dense colloidal suspensions.
