RESUMO

One of the problems of burn treatment is a creation of conditions providing most valuable skin rehabilitation. An experimental model of burn wound healing in a 0.9% NaCl solution is proposed. Synthetic activity of rat epidermis cells in the process of burn wound healing under a scab and in liquid environment was studied by luminescent microscopy. The effect of a 0.9% NaCl solution involves an increase of the basal layer cell synthetic activity of regenerating epidermis, and keeping a high level of this activity of hair follicle epithelial cells for a long time. Tissue-preserving effect of the 0.9% NaCl solution on burns healing has been confirmed in these results.

RESUMO

When the wound surface formed as a result of amputation of the tail tip in newborn rats was placed in sterile 0.9% aqueous solution of sodium chloride until full epithelialization of the defect, the epithelium moved onto the blood clot covering the wound. The blood clot was then substituted for connective tissue, into which the traumatized vertebra protruded. The vertebra restored its anatomical integrity, and this led to partial regrowth of the tail. A skin regenerate was formed on the apical tail surface with characteristic features of the intact skin of this locality. In the control animals, no regeneration of the vertebra epiphysis took place and a scar was formed at the stump end.

RESUMO

Peculiarities of formation of connective tissue framework of granulation tissue and scar were studied using light optical and scanning electron microscopy in rats during all-layer skin wound healing in interscapular region. Formation of granulation tissue and scar was accelerated after daily 20 minutes treatment with 0.2% trypsin and 0.1% ronidase solutions as compared to that in control. Regenerates are characterized by different morphological properties. Trypsin promotes quick formation of collagen fibre fascicles with their following thickening and gradual exfoliation. Ronidase action leads to small diameter collagen fibres formation and their following organization into larger fascicles. The peculiarities established are associated with different mechanisms of enzyme effect on reparative processes.

Extent of the completeness of the tail skin restoration was studied in 30 outbred albino male rats after the induction of full-thickness defect 10 x 5 mm in size with the restoration process running under different conditions: without any effects (control group), with the constant perfusion, of the wounded surface with 0.9% sodium chloride solution up to the complete wound epithelialisation (experimental group 1), with the moisturing of the wounded surface by the gauze napkin, plentifully moisted with the 0.9% sodium chloride solution up to the complete epithelialisation (experimental group 2). Two months after the operation the defect control part was represented by the connective tissue scar in the control group and in the experimental group 2. Structures, resembling the terminal regions of the sebaceous glands, are developing from the newly formed epithelium in the experimental 1. The composition of the regenerate connective tissue closely resembles that of the intact derm. Thus, the constant perfusion of the wounded surface with the isotonic sodium chloride solution promotes the organotypic skin regeneration.

Epidermis cells synthetic activity was studied by a luminescent microscopy method. Different conditions of skin wound healing in intrascapular region were investigated on 84 rats-males. Daily effect of 0.2% trypsin or 0.1% ronidase solution on the healing surface for 20 min has been shown to increase basal layer cells synthetic activity of forming epidermis in comparison with control. However, maximum raise of epidermis cells functional activity after ronidase influence was observed already on the 4th day, while after trypsin action--only on the 7th. Epithelial cells synthetic activity decreases several days before total skin defect epithelialization. In all experiments their activity stabilizes not less than one month after the operation. We have come to a conclusion that the mechanism of healing acceleration due to proteolytic enzymes depends on basal layer cells synthetic activity of forming epithelium.

RESUMO

A cicatrix formed in the process of posttraumatic skin regeneration is characterized by a higher cell adhesion power in the upper, and, especially, in the middle epidermis stratum. The cell adhesion power indices in these strata, and cell mitotic activity of basal epidermis stratum near the cicatrix depend on conditions of connective tissue formation in the wound defect. The effect of physiological solution and trypsin on the wound process results in a moderate rising of intercellular contact strength and in decreasing in the number of dividing basal stratum cells, while addition of ronidase solution results in a sharp rising of the corneous scale adhesion strength, and in establishing the straight correlation between this index and the speed of epidermocyte production.
