RESUMO

Catalase (CAT) is an enzyme to scavenge H2O2 and to protect peroxidation of cell wall lipid and lipoproteins. The increase in its activity has been observed in several diseases as an compensating reaction. The decrease in its activity may be related to some pathophysiologies of diseases. However, there has been few reports on the measurement of serum CAT activity. We formerly reported the method to measure CAT accurately with an automated analyzer. In the present experiment, we measured CAT in various kinds of diseases and healthy controls by this method. By an analysis of serum samples which were obtained from randomly selected 1,051 patients at Chibune Hospital for 12 months (from January to December, 1993), we obtained following results. The CAT of neuromuscular, cardiovascular and metabolic diseases were significantly high (p < 0.01). The CAT of psychiatric and otorhinopharyngologic diseases were significantly low (p < 0.01). There was significant relationship between CAT and some biochemical markers. CAT is thought as a protector against tissue injuries or circulatory insufficiencies. These results indicate that CAT increases in some kinds of diseases as a protection reaction to the injury, and that the decrease of CAT may cause some kinds of diseases.

RESUMO

The ethanol extract of Psoraleae Fructus (Psoralea corylifolia L.) was found to have cytotoxic activity against L929-cells in cell culture. The active compound was isolated by column chromatography on silica gel and identified as bakuchiol by means of spectral evidence. The cytotoxic activity of bakuchiol in cell culture was observed in short time and found to be unreversible. The mechanism of the cytotoxic activity was considered to be due to an injury of cell membrane from electron microscopic observation and hemolytic activity.

Bakuchiol is a major component of Corylifoliae Fructus (Psoralea corylifolia L.) and has been clarified to have cytotoxic activity. The chemical structure-cytotoxic activity relationship of bakuchiol was investigated by means of cytotoxic activity of synthesized analogues of bakuchiol and phenol. It was proved that an alkyl group was necessary for cytotoxic activity. But the double bonds in the unsaturated hydro-carbon group exerted but little influence on the cytotoxic activity. The cytotoxic activity of bakuchiol was the strongest as compared with that of the analogues examined.

RESUMO

A simple technique using high performance liquid gel permeation chromatography for determination of IgG in cerebrospinal fluid (CSF) is described. Accurate determination of IgG was achieved by precolumn derivatization with fluorescein isothiocyanate-conjugated anti-human IgG Fab, a substance that readily yields a fluorescent immune complex. Immune complexes separated as a well-delineated peak in the column void volume, and were measured by the fluorescence of the column eluates (Ex = 490 nm, Em = 520 nm). The results were reproducible with a deviation of less than 2% and a good linear relationship over the range of 2.0-24.5 mg/dl. The IgG concentration assayed by the column method correlated well with values obtained by laser nephelometry.

RESUMO

The application of high performance liquid gel permeation chromatography (HPL-GPC) and immunofluorescence for the estimation of a protein was studied using human IgG as a model system following separation of the fluorescent immune complex from free fluorescent-labelled antibody on a column of HPL-GPC. Using FITC-labelled anti-human IgG Fab fragment and HPL-GPC, nanogram levels of IgG could be determined within 5 min of mixing the antigen and antiserum. HPL-GPC immunofluorescence analysis is sensitive specific, simple, rapid and highly versatile.
