Objectives: To investigate the effects of the oestradiol (ES) pulsed bone marrow-derived mesenchymal stem cells (BM-MSC) to treat adjuvant-induced arthritis in Wistar rats. Materials and Methods: BM-MSCs were pulsed with ES (0, 10,100, and 1000 nM) for 24 hr. RA was induced by collagen and Freund's Complete Adjuvant into the base of the tail of Wistar rats. Results: The least effective concentration of ES that can promote potent anti-inflammatory properties in the MSC population is 100 nM. At this concentration, ES increases the inhibition of the polyclonal T lymphocyte proliferation, production of IDO, IL-10, Nitric oxide, and TGF-ß, and expression of CXCR4 and CCR2 mRNA in the MSC population. Accordingly, the RA rats were treated with 2×106 MSCs or ES-pulsed MSCs (100 nM) on day 10 when all animals had developed signs of RA. ES-pulsed BM-MSCs reduced the severity of RA more profoundly than treatment with BM-MScs alone. The ability of ES-pulsed BM-MSCs to reduce symptoms and RA markers like CRP, RF, and nitric oxide was comparable to that of prednisolone. Prednisolone was more successful in reducing inflammatory cytokines than treatment with ES-pulsed BM-MSCs. ES-pulsed BM-MSCs were more successful in increasing anti-inflammatory cytokines than treatment with Prednisolone. The ability of ES-pulsed BM-MSCs to decrease the level of nitric oxide was comparable to that of prednisolone. Conclusion: ES-pulsed BM-MSCs may be a helpful strategy in RA control.

This study investigates the fabrication of films based on a polymer containing nanoclay, sodium diacetate (SDA), and tert-butyl hydroquinone (TBHQ) for packaging rainbow trout fillets. The films were prepared by the addition of 2% SDA (SDA film), 2% TBHQ (TBHQ film), and a combination of both (1% SDA + 1% TBHQ) into polyethylene polymer (93.00%) and montmorillonite nanoclay (5.00%). A film lack of nanoclay, SDA, and TBHQ was prepared and considered a control film. A film was also prepared by the addition of 95 g polyethylene + 5 g nanoclay (Nanoclay). Morphological properties of the films were investigated by a scanning electron microscope (SEM). In vitro antioxidant properties and antibacterial activities of the films and their effects as the coating on fish samples were evaluated against Listeria monocytogenes, Salmonella typhimurium, and Escherichia coli. The effects of films on oxidative stability, antibacterial activity, pH, total volatile basic nitrogen (TVBN), and total viable count (TVC) of fish samples were assessed. The SEM results showed the homogenous dispersion of SDA and TBHQ into films. The SDA, TBHQ, and ST films showed antibacterial activity against L. monocytogenes, S. typhimurium, and E. coli compared with the control film as the coating and under in vitro conditions (p < 0.05). The TBHQ and ST films exhibited higher antioxidant activity and prevented the oxidation as the coating. The films prepared from the SDA, TBHQ, and ST prevented an increase in TVC and TVBN (p < 0.05). The ST films can prevent spoilage in fish samples and can be utilized in the food industry. PRACTICAL APPLICATION: We successfully prepared films with the help of nanoclay, sodium diacetate (SDA), and tert-butyl hydroquinone (TBHQ) on polyethylene for packaging fish fillets. Films containing SDA, TBHQ, and nanoclay showed antibacterial activity and prevented spoilage. The films can be used for packaging fish fillets.

The eukaryotic expression plasmid of M2e-Hsp70 is a candidate M2e-based DNA vaccine. In order to evaluate the immunization potential of this construct, Specific Pathogen Free chickens were immunized either intramuscularly or orally. Mutant Salmonella typhimurium was used as carrier for oral delivery of the M2e-Hsp70 construct. M2e-specific humoral and cellular responses were tested by ELISA and Lymphocyte Proliferation Assay, respectively. Our results indicate that both humoral and cellular immune responses are conferred against M2e-Hsp70 plasmid in either of the intramuscular or oral routes of administration; however, these responses are significantly higher in intramuscular injection in contrast to oral administration. When it comes to mass vaccination of commercial chicken flocks oral administration is preferred due to the ease of application as well as its capability of eliciting mucosal, humoral and cellular immune responses; so measurements should be taken to improve the immunization potency of our orally delivered DNA vaccine.
