Current feed formulation and evaluation practices rely on static values for the nutritional value of feed ingredients and assume additivity. Hereby, the complex interplay among nutrients in the diet and the highly dynamic digestive processes are ignored. Nutrient digestion kinetics and diet × animal interactions should be acknowledged to improve future predictions of the nutritional value of complex diets. Therefore, an in silico nutrient-based mechanistic digestion model for growing pigs was developed: "SNAPIG" (Simulating Nutrient digestion and Absorption kinetics in PIGs). Aiming to predict the rate and extent of nutrient absorption from diets varying in ingredient composition and physicochemical properties, the model represents digestion kinetics of ingested protein, starch, fat, and non-starch polysaccharides, through passage, hydrolysis, absorption, and endogenous secretions of nutrients along the stomach, proximal small intestine, distal small intestine, and caecum + colon. Input variables are nutrient intake and the physicochemical properties (i.e. solubility, and rate and extent of degradability). Data on the rate and extent of starch and protein hydrolysis of different ingredients per digestive segment were derived from in vitro assays. Passage of digesta from the stomach was modelled as a function of feed intake level, dietary nutrient solubility and diet viscosity. Model evaluation included testing against independent data from in vivo studies on nutrient appearance in (portal) blood of growing pigs. When simulating diets varying in physicochemical properties and nutrient source, SNAPIG can explain variation in glucose absorption kinetics (postprandial time of peak, TOP: 20-100 min observed vs 25-98 min predicted), and predict variation in the extent of ileal protein and fat digestion (root mean square prediction errors (RMSPE) = 12 and 16%, disturbance error = 12 and 86%, and concordance correlation coefficient = 0.34 and 0.27). For amino acid absorption, the observed variation in postprandial TOP (61 ± 11 min) was poorly predicted despite accurate mean predictions (58 ± 34 min). Recalibrating protein digestion and amino acid absorption kinetics require data on net-portal nutrient appearance, combined with observations on digestion kinetics, in pigs fed diets varying in ingredient composition. Currently, SNAPIG can be used to forecast the time and extent of nutrient digestion and absorption when simulating diets varying in ingredient and nutrient composition. It enhances our quantitative understanding of nutrient digestion kinetics and identifies knowledge gaps in this field of research. Already useful as research tool, SNAPIG can be coupled with a postabsorptive metabolism model to predict the effects of dietary and feeding-strategies on the pig's growth response.

Food production has a major impact on environmental emissions, climate change and land-use. To reduce this impact, the circularity of future food production systems is expected to become increasingly important. In a circular food system, crop land is primarily used for plant-based food production, while low-opportunity cost feed materials (LCF), i.e. crop residues, co-products of the food industry, grass from marginal land and food waste form the basis of future, animal feeds. Animal diets thus contain much less cereals and soybean meal and include a higher proportion of diverse co-products, residues and novel human-inedible ingredients. These diets are characterised by a lower starch content, and a higher content of fibre, protein, fat, and phytate compared to present diets. In this review, possible consequences of the development towards a more circular food system for the type, volume and nutritional characteristics of feed materials and complete feeds are addressed and related research questions in the area of animal nutrition, physiology and metabolism are discussed. Additional attention is given to possible effects on intestinal health and gut functionality and to (bio)technological processing of LCF to improve their suitability for feeding farm animals, with a focus on the effects in pigs and poultry. It is concluded that an increased use of LCF may limit the use of presently used criteria for the efficiency of animal production and nutrient utilisation. Development of characteristics that reflect the efficacy and efficiency of the net contribution of animal production in a circular food system is required. Animal scientists can have an important role in the development of more circular food production systems by focussing on the optimal use of LCF in animal diets for the production of animal-source food, while minimising the use of human-edible food in animal feed.

Providing pigs a diet that matches their nutrient requirements involves optimizing the diet based on the nutrient digestibility values of the considered feed ingredients. Feeding the same quantity of a diet to pigs with similar BW but with different requirements, however, can result in a different average daily gain (ADG) and backfat thickness (BF) between pigs. Digestibility may contribute to this variation in efficiency. We investigated variation in feed efficiency traits in grower-finisher pigs associated with variation in faecal digestibility values, independent of feed intake at the time of measuring faecal digestibility. Considered traits were ADG, average daily feed intake (ADFI), feed conversion ratio (FCR), BF and residual feed intake (RFI). Feed intake, BW, and BF data of one hundred and sixty three-way crossbreed grower-finisher pigs (eighty female and eighty male) were collected during two phases, from day 0 of the experiment (mean BW 23 kg) till day 56 (mean BW 70 kg) and from day 56 to slaughter (mean BW 121 kg). Pigs were either fed a diet based on corn/soybean meal or a more fibrous diet based on wheat/barley/by-products, with titanium dioxide as indigestible marker. Faecal samples of one hundred and five pigs were collected on the day before slaughter and used to determine apparent faecal digestibility of DM, ash, organic matter (OM), CP, crude fat (CFat), crude fibre (CF), and to calculate the digestibility of nonstarch polysaccharides (NSPs) and energy (E). The effects of diet, sex and covariate feed intake at sampling (FIs) on faecal digestibility values were estimated and were significant for all except for CFat. Faecal digestibility values of each individual pig determined at the day before slaughter, corrected for diet, sex and FIs, were used to estimate their association with ADG, ADFI, FCR, BF, and RFI. In the first phase, a one percent unit increase in faecal digestibility of DM, ash, OM, E, CP, CFat, CF, NSP, and Ash individually was related to 0.01-0.03 unit reduction in FCR and 6-23 g/day reduction in RFI. A unit increase in CP digestibility was related to 0.1 mm increase in BF and 10 g/day increase in ADG. In the second phase, a one percent unit increase in faecal digestibility of DM, CP and Ash was related to a decrease of 16-20 g/day in RFI. In conclusion, the relationship between variation in feed efficiency traits and faecal digestibility values is different across the developmental stages of a pig.

Requirements for energy and particular amino acids (AAs) are known to be influenced by the extent of immune system stimulation. Most studies on this topic use models for immune system stimulation mimicking clinical conditions. Extrapolation to conditions of chronic, low-grade immune system stimulation is difficult. We aimed to quantify differences in maintenance energy requirements and efficiency of energy and protein used for growth (incremental energy and protein efficiency) of pigs kept under low (LSC) or high sanitary conditions (HSC) that were fed either a basal diet or a diet with supplemented AA. Twenty-four groups of six 10-week-old female pigs were kept under either LSC or HSC conditions for 2 weeks and fed a diet supplemented or not with 20% extra methionine, threonine and tryptophan. In week 1, feed was available ad libitum. In week 2, feed supply was restricted to 70% of the realized feed intake (kJ/(kg BW)0.6 per day) in week 1. After week 2, fasting heat production (FHP) was measured. Energy balances and incremental energy and protein efficiencies were measured and analyzed using a GLM. Low sanitary condition increased FHP of pigs by 55 kJ/(kg BW)0.6 per day, regardless of diet. Low sanitary condition did not alter the response of faecal energy output to incremental gross energy (GE) intake, but it reduced the incremental response of metabolizable energy intake (12% units), heat production (6% units) and energy retained as protein (6% units) to GE intake, leaving energy retained as fat unaltered. Incremental protein efficiency was reduced in LSC pigs by 20% units. Incremental efficiencies for energy and protein were not affected by dietary AA supplementation. Chronic, low-grade immune stimulation by LSC treatment increases FHP in pigs. Under such conditions, the incremental efficiency of nitrogen utilization for body protein deposition is reduced, but the incremental efficiency of absorbed energy for energy or fat deposition is unaffected.

Rheological properties of digesta play a role in digesta passage kinetics through the gastrointestinal tract, in turn affecting nutrient absorption kinetics. Therefore, we studied the effects of diet viscosity on digesta passage and physicochemical properties in pigs. Twenty male growing pigs (35 kg body weight at the start) were assigned to one of five diets with increasing dietary concentrations of ß-glucans (BG; from 0 % to 10 %), in exchange for maize starch. After a 17-day adaptation period, pigs were euthanised and the mean retention time (MRT) of digesta solids (TiO2) and liquids (Cr-EDTA) in the stomach, and proximal and distal half of the small intestine was quantified. In the stomach, the MRT of liquids, but not of solids, increased when dietary BG level increased (6 min per % dietary BG, P = 0.008 and R2 = 0.35). Concomitantly, stomach DM content (5 g/kg per % dietary BG, P < 0.001 and R2 = 0.53) and apparent digesta viscosity (56 Pa × s at 1/s shear rate per % dietary BG, P = 0.003 and R2 = 0.41) decreased. In the proximal half of the small intestine, no effects of dietary BG level were observed. In the distal half of the small intestine, water-binding capacity (WBC) of digesta increased (0.11 g/g digesta DM per % dietary BG, P = 0.028 and R2 = 0.24) and starch digestibility decreased (0.3% per % dietary BG, P = 0.034 and R2 = 0.23) when dietary BG level increased. In the colon, apparent digesta viscosity at 45/s shear rate increased (0.1 Pa × s per % dietary BG, P = 0.03 and R2 = 0.24) in the proximal half of the colon, and digesta WBC increased (0.06 g/g digesta DM per % dietary BG, P = 0.024 and R2 = 0.26) in the distal half of the colon when dietary BG level increased. To conclude, increasing dietary BG level caused the MRT of liquids, but not that of solids, to increase in the stomach, resulting in reduced separation of the solid and liquid digesta fractions. This caused dilution of the stomach content and reduction in digesta viscosity when dietary BG levels increased. Effects of dietary BG level on physicochemical properties in the proximal small intestine were absent and may have been due to a low DM content. The WBC of digesta in the distal small intestine and colon increased when dietary BG level increased, as did apparent digesta viscosity in the proximal colon. This likely reflects the concentration of BG in digesta when moving through the gastrointestinal tract.

In current feed evaluation systems, the nutritional value of protein sources in diets for pigs is based on the ileal digestibility of protein and amino acids, which does not account for the kinetics of protein digestion along the gastrointestinal tract. The objective of the present study was to determine the in vitro protein digestion kinetics of different protein sources (soya bean meal (SBM), wheat gluten (WG), rapeseed meal (RSM), whey powder (WP), dried porcine plasma protein, yellow meal worm larvae and black soldier fly larvae (BSF)). Protein sources were incubated with pepsin at pH 3.5 for 0 to 90 min and subsequently with pancreatin at pH 6.8 for 0 to 210 min at 39°C. The in vitro protein digestion kinetics were described as the kinetics of nitrogen (N) solubilisation and the release of low molecular weight peptides (LMW) (<500 Da). The N solubilisation rate ranged from 0.025 min-1 for BSF to 0.685 min-1 for WP during the incubation with pepsin, and from 0.027 min-1 for RSM to 0.343 min-1 for WP during the incubation with pancreatin. The release rate of LMW peptides ranged from 0.027 min-1 for WG to 0.093 min-1 for WP during the incubation with pepsin, and from 0.029 min-1 for SBM to 0.385 min-1 for WP. Black soldier fly larvae showed a similar release rate of LMW peptides as WP during the incubation with pancreatin. At the end of the sequential incubation with pepsin (90 min) and pancreatin (210 min), WG and WP showed the highest percentage of N present in LMW peptides relative to total N (78% and 79%, respectively), whereas SBM showed the lowest (35%). In conclusion, protein sources for pig diets show substantial differences in in vitro protein digestion kinetics as measured by the kinetics of N solubilisation and the release of LMW peptides. The rate of release of LMW peptides was not correlated to the rate of N solubilisation for each of the protein sources evaluated.

Lowering protein level in diets for piglets urge to have knowledge on the piglet's requirements for essential amino acids (AA) and their interactions. The present studies aimed to determine the interaction between the dietary level of valine (Val) and tryptophan (Trp) and the effect of AA imbalance at two levels of dietary Val on the growth performance of post-weaning piglets. In Experiment 1 (duration 4 weeks), the effects of supplementation of free l-Val (1.0 g/kg) and/or l-Trp (0.5 g/kg) in a low-CP diet (CP 17.7%), marginal in Trp and Val, was studied in a 2×2 factorial design and using an additional reference treatment (CP 19.5%). In Experiment 2 (duration 5 weeks), the influence of a stepwise increase in excess supply of isoleucine (Ile), histidine (His) and leucine (Leu), up to 10, 10% and 30% relative to their requirement values respectively, was evaluated at 60% or 70% standardized ileal digestible (SID) Val relative to SID lysine, using a 3×2 factorial design. In Experiment 1, over the whole experimental period, feed intake (FI) was affected by dietary Trp level (P<0.05) and feed conversion ratio (FCR) by both the level of Trp and Val in the diet (both P<0.05). Increasing Trp level increased FI and decreased FCR while increasing dietary Val level reduced FI and increased FCR. For BW gain (BWG), there was an interaction between dietary level of Trp and Val (P<0.05). Valine supplementation decreased BWG using a diet marginal in Trp, whereas it increased BWG when using a Trp sufficient diet. Piglets fed the low-CP diet with adequate levels of Val and Trp showed at least same performance compared to piglets fed the high CP reference diet. In Experiment 2, increasing dietary Val improved FI and BWG (P<0.001) and tended to improve FCR. Dietary AA excess for Ile, His and Leu reduced FI and BWG (P<0.05) and only affected FCR (P<0.01) in the 1st week of the study. Dietary level of Val and AA excess did not show interactive effects, except for FCR over the final 2 weeks of the study (P<0.05). In conclusion, an interaction exists between dietary supply of Val and Trp on the zootechnical performance of post-weaning piglets and dietary AA excess for Ile, Leu and His, reduces growth performance of piglets in low-protein diets, independent of the dietary level of Val.

Safety evaluation of a muramidase produced by a Trichoderma reesei strain (safe lineage), expressing a muramidase gene isolated from Acremonium alcalophilum is presented. Intended use in feed of this enzyme is as digestive aid in broiler chickens. Muramidase 007, was non-mutagenic and non-clastogenic in vitro, and no adverse effects were observed in 90-day subchronic toxicity studies in rats at doses up to 1132 mg TOS/kg body weight/day. The enzyme did not exhibit, in vitro, skin, nor eye irritation potential. Acute aquatic toxicity evaluated on daphnia and algae showed absence of effect of the enzyme at the standard doses tested. Muramidase 007 was fully tolerated by broiler chickens in a 6-weeks tolerance study showing no adverse effects in any of the dietary treatments (0, 1×, 5× and 10× maximum recommended dose). In conclusion, Muramidase 007 is found to be toxicologically inert, and there are no worker's safety concerns if standard precautions are instituted and a non-dusty formulation is employed. Muramidase 007 is well tolerated by the target species (broiler chickens) and cause no harm to the environment. The beneficial safety evaluation of Muramidase 007 is in line with this type of enzyme that is found ubiquitously in nature.

There is growing evidence that requirements for particular AA increase when pigs are kept under low sanitary conditions. The extent to which reduction in growth performance is related to these increased requirements is unclear. To evaluate this relationship, an experiment (2 × 2 × 2 factorial arrangement) was performed with 612 male pigs (9 per pen) kept under low sanitary conditions (LSC) or high sanitary conditions (HSC) and offered ad libitum access to either a normal CP concentration diet (NP; 17, 15, and 15% CP for the starter, grower, and finisher phase, respectively) or a low CP concentration diet (LP; 20% CP reduced relative to NP for each phase), each of which containing a basal AA profile (AA-B) or a supplemented AA profile (AA-S). The supplemented diet type contained 20% more Met, Thr, and Trp relative to Lys on an apparent ileal digestible basis compared with the basal diet type. Pigs were followed for a complete fattening period and slaughtered at a targeted pen weight of 110 kg. Haptoglobin concentrations in serum (0.92 g/L for LSC and 0.78 g/L for HSC) and IgG antibody titers against keyhole limpet hemocyanin (3.53 for LSC and 3.08 for HSC) collected in the starter, grower, and finisher phases and pleuritis scores at slaughter (0.51 for LSC and 0.20 for HSC) were greater for LSC pigs compared with HSC pigs ( ≤ 0.01), illustrating that sanitary conditions affected health conditions. The ADG and G:F were greater for HSC pigs compared with LSC pigs ( ≤ 0.01). The number of white blood cells (WBC) was higher in (AA-S)-fed pigs compared with (AA-B)-fed pigs when kept at LSC but not at HSC [SS (sanitary conditions) × AA interaction, = 0.04]. Pigs fed NP had a lower number of WBC compared with pigs fed LP ( = 0.02). The number of platelets in pigs fed AA-S diets was higher compared with pigs fed AA-B diets ( ≤ 0.01). A 20% reduction in dietary supplementation of Met, Thr, and Trp relative to Lys decreased G:F more in LSC pigs than in HSC pigs (interaction, = 0.03), illustrating that dietary requirements for these AA differ depending on sanitary conditions. This study, performed under practical conditions, shows that AA requirements are dependent on sanitary conditions. Furthermore, supplementation of diets with particular AA may improve performance, especially under poor hygienic conditions. Dietary protein concentration as well as Met, Thr, and Trp supplementation can modify immune status, which may influence resistance to subclinical and clinical diseases.

The intestinal epithelium glycocalyx sugar motif is an important determinant of the bacterial-host interaction and may be affected in pigs by gut microbiota and by blood group genotype. The aim was to study the effect of intestinal association with different microbiota and A0 blood group genotypes on the expressed glycomic pattern in the small intestine. Twelve caesarean-derived pigs previously associated with a simple association (SA) or complex association (CA) microbiota were selected at 26 to 37 d of age. In each subject, different jejunal loops were perfused for 8 h with enterotoxigenic K88 (ETEC), ETEC fimbriae (F4), (LAM), or a saline control. The piglets were genotyped for A0 blood group and the glycomic profile was evaluated by microscopic screening of lectin binding: peanut agglutinin (PNA), which is galactose specific; agglutinin I (UEA), which is fucose specific; lectin II (MALii), which is sialic acid specific; concavalin A, which is mannose specific; soybean agglutinin (SBA), which is -acetyl-galactosamine specific; and wheat germ agglutinin (WGA), which is -acetyl-glucosamine specific. A0 pigs had fewer UEA-positive cells, MALii-positive cells ( < 0.001), and SBA-positive cells ( < 0.10) than 00 pigs. Simple association pigs had more SBA positive cells ( < 0.01) than CA pigs. Enterotoxigenic K88-perfused intestinal loops had fewer UEA-positive cells ( < 0.01) and WGA positive cells ( < 0.001) cells and more PNA positive cells (only in SA pigs, < 0.01). No effects of introduction of F4 and LAM in the intestinal lumen were observed. The porcine A0 blood group genotype and the luminal presence of ETEC strongly affected the jejunal mucosa glycomic pattern profile whereas an early oral simple or complex microbial association had limited effects. Pig genetic background has relevance on the cross talk between intestinal epithelium glycocalyx sugar motif and ETEC and, ultimately, on the gut microbial colonization in later life.

The evolution of hyper-prolific pig breeds has led to a higher within-litter variation in birth weight and in BW gain during the nursery phase. Based on an algorithm developed in previous research, two populations from a pool of 368 clinically healthy piglets at 6 weeks of age were selected: a low (LP) and a high (HP) performing population and their development was monitored until the end of the nursery phase (10 weeks of age). To understand the cause of the variation in growth between these populations we characterized the LP and HP piglets in terms of body morphology, behaviour, voluntary feed intake, BW gain, and apparent total tract and ileal nutrient digestibility. Piglets were housed individually and were fed a highly digestible diet. At selection, 6 weeks of age, the BW of LP and HP piglets were 6.8±0.1 and 12.2±0.1 kg, respectively. Compared with the LP piglets the HP piglets grew faster (203 g/day), ate more (275 g/day) from 6 to 10 weeks of age and were heavier at 10 weeks (30.0 v. 18.8 kg, all P<0.01). Yet, the differences in average daily gain and average daily feed intake disappeared when compared per kg BW0.75. Assuming similar maintenance requirements per kg BW0.75 the efficiency of feed utilization above maintenance was 0.1 g/g lower for the LP piglets (P=0.09).The gain : feed ratio was similar for both groups. LP piglets tended to take more time to touch a novel object (P=0.10), and spent more time eating (P<0.05). At 10 weeks, LP piglets had a higher body length and head circumference relative to BW (P<0.01). Relative to BW, LP had a 21% higher small intestine weight; 36% longer length, and relative to average FI, the small intestinal weight was 4 g/kg higher (both P=<0.01). Apparent total tract and ileal dry matter, N and gross energy digestibility were similar between groups (P>0.10). We concluded that the low performance of the LP piglets was due to their inability to engage compensatory gain or compensatory feed intake as efficiency of nutrient utilization and feed intake per kg BW0.75 was unaffected. LP piglets tend to be more fearful towards novel objects. The morphological comparisons, increased body length and head circumference relative to BW imply that LP piglets have an increased priority for skeletal growth.

Postnatal (muscle) growth potential in pigs depends on the total number and hypertrophy of myofibers in skeletal muscle tissue. In a previous study an algorithm was developed to predict piglet BW at the end of the nursery period (10 wk of age) on the basis of BW at birth, at weaning, and at 6 wk of age. The objective of this study was to determine whether the differences in growth performance between poor (PP) and high (HP) performing piglets could be the result of different skeletal muscle properties. Therefore, from a total of 368 piglets (offspring from Hypor sows bred to TOPIGS sires) 2 groups with a divergent growth performance were selected at 6 wk of age: HP (n = 20, predicted BW at 10 wk of age 26.8-30.9 kg) and PP (n = 20, predicted BW at 10 wk of age 16.0-22.9 kg). Piglets were euthanized at 10 wk of age, and samples of the semitendinosus muscle (STN) were collected for histochemistry and gene expression analysis using quantitative PCR (qPCR). At 10 wk of age, realized BW did not differ from predicted BW in either group (P > 0.880). The HP piglets exhibited greater ADG and ADFI from 6 to 10 wk and greater BW at birth and 6 and 10 wk of age (P ≤ 0.002) compared with the PP piglets, whereas G:F ratio was similar (P = 0.417). Superior growth performance of HP piglets was associated with a 1.27-fold higher IGF1 plasma concentration at 10 wk compared with the PP piglets (P = 0.044). The greater weight and muscle cross-sectional area of STN in HP piglets was due to a 1.20-fold increase in total muscle fiber number (TFN; P = 0.009) and 1.34-fold increase in fiber cross-sectional area (FCSA; P = 0.004) compared with the PP piglets. The number of myonuclei per red and intermediate fiber was greater in HP piglets (P ≤ 0.097), but the nucleus-to-cytoplasm ratio was unaffected by the performance group (P = 0.861). The mRNA expression of proliferating cell nuclear antigen (PCNA), paired box 7 (PAX7), myogenic factor 5 (MYF5), and myogenic differentiation factor (MYOD) did not differ between groups (P ≥ 0.327). However, IGF2-specific mRNA expression was numerically higher in the HP piglets (P = 0.101). The greater myofiber number, the higher degree of myofiber hypertrophy, and the increased muscular mRNA expression of IGF2 indicate that HP piglets exhibit a greater capacity for lean accretion and may grow faster until market weight. In summary, pigs that were selected for predicted high BW at 10 wk of age using a complex selection model had a superior muscularity in terms of greater TFN and FCSA, which may be of advantage for lean mass accretion in later life and for meat quality.

Two experiments were conducted to develop a simplified dose-response technique to estimate the Lys requirement of individual, meal-fed growing pigs. In Exp. 1, we studied adaptation processes that occur during such a dose-response study in meal-fed pigs, and in Exp. 2, we studied the accuracy of this simplified technique to estimate changes in Lys requirement estimates of pigs following changes in energy intake. In Exp. 1, the effect of the Lys supply strategy on the Lys requirement was assessed in 14 barrows fed an increasing [low to high (LH)] or decreasing [high to low (HL)] total Lys supply, with total Lys levels varying from 0.36 to 1.06 g/MJ DE in 7 equidistant steps of 4 d each. Urinary urea and ammonia excretion and whole body N turnover were measured. In Exp. 2, the accuracy of the dose-response technique to determine a shift in Lys requirement was assessed in 20 barrows fed at either 2.2 [low energy (LE)] or 2.7 [high energy (HE)] times the energy requirements for maintenance, with total Lys supply decreasing from 1.10 to 0.37 g Lys/MJ DE in 9 equidistant steps of 3 d each. In Exp. 1, a lower increment in protein synthesis, breakdown, and whole body N turnover with increasing dietary Lys supply was observed in LH pigs than HL pigs (P < 0.01) and the estimated Lys requirement was 0.06 g/MJ DE greater (P = 0.01) in LH pigs than HL pigs. These results indicated that pigs at a decreasing Lys supply strategy require less time for metabolic adaptation to a change in Lys supply than those at an increasing Lys supply. In Exp. 2, the estimated Lys requirement was 2.6 g/d greater (P < 0.001) in HE pigs than LE pigs. The variation in AA requirement estimates between individual pigs was low (4.9% in LH pigs and 3.0% in HL pigs in Exp. 1 and 8.1% in LE pigs and 6.0% in HE pigs in Exp. 2). The present studies indicated that a dose-response technique with a decreasing Lys supply in time and a step length of 3 d with urinary N excretion as response criteria provides a simple, accurate technique to quantitatively estimate a change in AA requirements of individual meal-fed pigs.

In pig (Sus Scrofa) production, within-batch variation in bw gain of piglets during the nursery period (up to 10 wk of age) can be high and is of high economic importance. Homogeneity of BW within batches of animals is important as it influences the efficiency of use of the grower and finisher facilities, and provides an extra value for the fattening farms. In the current study, factors for a light BW at the end of the nursery period of pigs were determined by analyzing datasets from 3 different swine research centers in the Netherlands and France. The entire dataset contained information on 77,868 individual piglets born between 2005 and 2010. Body weight was determined at different time points over the pre- and post-weaning phase, and sex, season of birth, litter information (litter size at day of birth and after cross-fostering, number of piglets born alive per litter, number of total born littermates, sow parity number), cross-fostered animals (yes or no), and pen group size over the post-weaning period were recorded. A risk factor analysis approach was used to analyze the datasets to determine factors that predict piglet bw at the end of the nursery period. Body weight at the end of the nursery period corrected for age was mainly determined by season (P < 0.001), birth weight (BiW, P < 0.001), weaning weight (WW, P < 0.001), and BW at 6 wk of age (P < 0.001). These variables were consistent among datasets and explained approximately 70% of the overall variation in BW at the end of the nursery period. Litter information did not significantly (P > 0.05) contribute to explaining the BW at the end of the nursery period. To discard the possibility of intrauterine growth retarded piglets (IUGR) being the reason for the influence of BiW as an explanatory factor in the regression model, a further analysis was performed on the effect of this category of piglets on the results of the regression analysis. Overall, it was concluded that the bw of piglets at the end of the nursery phase is mainly determined by season, sex, birth, WW, and BW at 6 wk of age. Piglets with a BiW greater than the mean biw minus 2.5 times the sd have the potential to compensate during the subsequent phases of growth.

Enterotoxigenic Escherichia coli (ETEC) expressing K88 (F4) adhesins are associated with post-weaning diarrhoea in piglets. Different grain fractions from pea (Pisum sativum) and faba bean (Vicia faba) were tested in vitro for their capacity to counteract aetiological factors, which contribute to the development of diarrhoea. In detail, adhesion of E. coli O149:K91:K88ac (ETEC K88ac) to grain legume products, intended to impair the colonization of the host, was studied as well as interference with receptor binding of the pathogen's heat-labile enterotoxin LT, intended to reduce toxin-inflicted gut cell damage. When comparing different pea and faba bean products tested for their binding capacity of ETEC K88ac, especially pea hulls, but also whole pea meal, starch-enriched and protein-enriched pea meal, and digestion-resistant pea hull and meal fractions showed a higher binding of ETEC K88ac than faba bean products. In contrast to the ETEC K88ac adhesion results, bean hulls proved more effective than pea hulls in preventing GM1 receptor binding of LT. Previous small intestinal segment perfusion experiments we performed with ETEC K88ac-challenged piglets indicated that both pea and bean hulls have the potential for successful application in diarrhoea prophylaxis and treatment, which is in agreement with and refined by our detection of their different modes of functioning.

The possible beneficial effects of surplus dietary Trp (+5 g of Trp/kg of diet) on factors related to stress, immunology, behavior, and N retention were investigated in postweaning piglets (approximately 15 kg of BW) challenged for 10 d with intravenous bacterial lipopolysaccharide (from Escherichia coli). Two diets fed restrictively (732 kJ of NE/kg of BW(0.75)/d) were compared, 1) a basal diet (apparent ileal digestible Trp = 1.9 g/kg; the recommended amount of Trp to warrant near-optimal growth in nonendotoxemic piglets), and 2) a Trp-enriched basal diet (+5 g of free l-Trp/kg), with 8 individually housed piglets per diet. Pooled salivary cortisol, but not plasma cortisol sampled at euthanasia, showed a tendency (P = 0.07) toward reduced concentrations in the Trp group (1.1 vs. 1.4 ng/mL; pooled SE = 0.1 ng/mL). Plasma C-reactive protein was reduced (P = 0.04) in the Trp group (0.9 vs. 5.0 mg/L; pooled SE = 1.3 mg/L), but haptoglobin, IL-6, tumor necrosis factor α, and lipopolysaccharide-induced fever were similar between the 2 dietary treatments. Physical activity related to approaching a human showed a tendency (P = 0.08) toward increased latency time in the Trp group (101 vs. 60 s; pooled SE = 16 s), but the times spent standing, sitting, and lying were similar between dietary treatments. The ADFI, ADG (346 vs. 302 g/d; pooled SE = 14 g/d; P = 0.11), body N retention (11.6 vs. 11.0 g/d; pooled SE = 0.2 g/d; P = 0.18), and G:F (0.55 vs. 0.49; pooled SE = 0.03; P = 0.17) were not different between the groups fed Trp and the basal diet. In conclusion, surplus dietary Trp had limited effects on stress, immunology, behavior, and N retention in a pig model of systemic endotoxemia.

In a small intestinal segment perfusion (SISP) study in pigs, effects were studied of intestinal perfusion of ground faba beans (Vicia faba), faba bean hulls, or saline on intestinal net fluid absorption in intestinal segments either challenged or not with an enterotoxigenic Escherichia coli (ETEC). After an 8-h perfusion, piglets were euthanized and small intestinal mucosa samples were collected for analysis of expression level of a selected set of genes (APOC3, TIMP1, AQP8, MMP1, MUC13, and PAP) using real-time quantitative PCR (qPCR). Perfusion with ground faba beans and faba bean hulls and challenge with ETEC affected (P < 0.05) expression of several of genes in the intestinal mucosa. Expression of APOC3, TIMP1, AQP8, MMP1, and PAP was correlated with net fluid absorption in the small intestine of pigs.

The present study was designed to develop a model in piglets that allows the investigation of the effects of postnatal association with a simple or a complex microbiota on gut health and development. Thirty piglets from 2 sows were obtained by caesarean delivery (day 0) and were equally divided over 2 treatment groups housed in separate clean, nonsterile rooms. All piglets received orally a simple microbiota consisting of Lactobacillus amylovorus, Clostridium glycolicum, and Parabacteroides spp. on days 1, 2, and 3 after birth. On day 3 and 4 the piglets received either a complex microbiota by providing them with a fecal inoculant of an adult sow [complex association (CA)] or a placebo inoculant [simple association (SA)]. Fecal microbiota composition, as determined by denaturing gradient gel electrophoresis and by pig intestinal tract chip (PITChip) analysis of 16S rRNA genes (days 3, 5, 7, 14, and 28), was less diverse in the SA group compared to the CA group. A difference in fecal microbiota composition between treatments persisted until the end of the study. It was concluded that the composition of microbiota in feces of cesarean delivery-derived piglets is influenced by bacterial association in the first days after birth. Differences in fecal microbiota composition between piglets exposed to a simple or complex inoculum at early age persisted for at least 3 wk.

The aim of the study was to evaluate the requirement for Trp in relation to diet composition in piglets in the period after weaning (BW range of 9 to 24 kg). Two Trp-deficient [relative to the Dutch (CVB, 1996) and NRC (NRC, 1998) requirement values for piglets of 10 to 20 kg of BW] basal diets were formulated: one based on corn and soybean meal and a second one based on wheat, barley, soybean meal, peas, and whey powder [10.0 g/kg of apparent ileal digestible (AID) Lys; 1.4 g/kg of AID Trp; 1.5 g/kg of standardized ileal digestible (SID) Trp]. Both basal diets were supplemented with 0.3, 0.6, and 0.9 g of l-Trp per kg of diet to obtain diets with 1.7, 2.0, and 2.3 g of AID Trp per kg (1.8, 2.1, and 2.4 g of SID Trp per kg), respectively. Each of the 8 treatments was evaluated in 8 replicates (pens with 8 male or female piglets). Average daily feed intake, ADG, and G:F were measured as response criteria. Over the 28-d experimental period, ADG and G:F were greater for the treatments on the wheat/barley diet compared with those on the corn/soybean meal and were increased by the level of Trp in the diet (P < 0.05). Average daily feed intake was only increased by the level of Trp supplementation (P < 0.05). Increasing the Trp level increased ADFI for the corn/soybean meal diet up to 2.3 g of AID Trp per kg (2.4 g of SID Trp per kg) and up to 2.0 g of AID Trp per kg (2.1 g of SID Trp per kg) in the wheat/barley diet (P < 0.05). For both diet types supplementation of free l-Trp increased the G:F up to 1.7 g of AID Trp per kg (1.8 g of SID Trp per kg). Nonlinear regression analysis of the response curves for ADFI using an exponential model for estimating a requirement value for Trp (defined as the Trp level resulting in 95% of the maximum response) revealed a requirement estimate of 2.3 g of AID Trp per kg for the corn/soybean meal-based diet and 2.1 g of AID Trp per kg for the wheat/barley-based diet, equivalent to 2.4 and 2.2 g of SID Trp per kg of diet, respectively. For ADG, a requirement estimate of 2.1 g of AID Trp per kg for both types of diets was derived, equivalent to 2.2 g of SID Trp per kg of diet. The Trp requirement for young piglets seems to be greater than indicated by some commonly used recommendations and does not seem largely dependent on diet ingredient composition.

An experiment was conducted to determine the effects of yeast culture (YC) and modified yeast culture [YC + cell wall product (CWP) containing mannan oligosaccharides] in pig diets on the performance, gut integrity, and blood cell composition of weanling pigs and to determine whether these dietary supplements could replace antimicrobial growth promoters (AGP) in pig diets. A total of 480 weanling pigs (27 d old and 7.8 +/- 0.1 kg of BW) were assigned to 1 of 4 experimental treatments: 1) diets without AGP or YC (control diet); 2) control + AGP; 3) control + 0.125% YC; and 4) control + 0.125% YC + 0.2% CWP. Piglets were fed experimental diets for 5 wk after weaning. Blood samples were collected from 8 piglets at weaning and from 8 piglets per treatment on d 14 and 35 after weaning for blood cell composition. These piglets were slaughtered for measurement of villous length and crypt depth in the jejunal mucosa and microbial profiling on the intestinal digesta. Average daily gain (P = 0.06) and G:F (P = 0.02) were improved for piglets that were fed the supplemented diets compared with piglets that were fed the control diet. Average daily feed intake was unaffected by dietary treatment. Performance was similar in piglets fed diets supplemented with AGP, YC, and YC + CWP. Blood cell composition, villous length, crypt depth, and microbial composition in the gut were unaffected by dietary treatment, but they were affected by time after weaning. Red blood cells, hemoglobin, hematocrit value, mean cell volume, mean cell hemoglobin, percentage of lymphocytes in the leukocyte population, villous length, and crypt depth were greater (P < 0.05) at 5 wk after weaning than at 2 wk after weaning. Eosinophils (P = 0.06) in the leukocyte population tended to be greater at 5 wk after weaning. Concentration of neutrophils in the leukocyte population and percentages of CD4 and CD8 cells were lower (P < 0.02) at 5 wk after weaning. The CD4:CD8 ratio (P = 0.07) tended to be lower at 5 wk after weaning. Results suggest that yeast culture could be an alternative to AGP in the diets of weanling pigs and that addition of CWP to diets containing YC would not improve the performance or health of weanling pigs above that of YC alone. Thus, more insight into the mode of action of YC is needed.
