In this study Pergularia daemia unripe fruits were used to synthesize zinc oxide nanoparticles (Pd-ZnONPs). UV-vis Spectroscopy detected the production of ZnONPs. XRD, FTIR, SEM, and TEM studies were used to characterize the synthesized Pd-ZnONPs. Aedes aegypti (Ae. aegypti) third instar larvae were analyzed to diverse concentrations of Pd-unripe fruit extract and Pd-ZnONPs for 24 hours to assess the larvicidal effect. Mortality was also detected in Ae. aegypti larvae under laboratory conditions, with corresponding LC50 and LC90 values of 11.11 and 21.20 µg/ml respectively. As a result of this study, the levels of total proteins, esterases, acetylcholine esterase, and phosphatase enzymes in the third instar larvae of Ae. aegypti were significantly lower than the control. These findings suggest that Pd-ZnONPs could be used to suppress mosquito larval populations.

With a growing consciousness of the importance of nature stewardship, researchers are focusing their efforts on utilizing renewable energy, particularly solar energy, to address environmental concerns. In this context, photocatalysis has long been viewed as one of the most promising cleaning methods. Hence, we have prepared a sunlight-active phytol-assisted ZnO-TiO2 nanocomposite (PZTN) for photocatalytic bacterial deactivation and dye degradation process. The PZTN-photocatalysis effectively deactivated the bacterial pathogens as well as malachite green dye within 240 min under direct-sunlight. Moreover, this will be the first complete study on safety level assessment of photocatalytically-remediated water through toxicity studies. The obtained results evidenced that photocatalytically-deactivated bacteria and MG-dye showed to have no toxic effects, signifying that the PZTN-photocatalyzed water seems to be extremely safe for the environment. As a result of this research, we suggest that the PZTN could be a promising sunlight-active photocatalyst for environmental water treatment. On the other hand, biofouling is a ubiquitous phenomenon in the marine environment. Bacteria are the first organisms to foul surfaces and produce biofilms on man-made submerged materials. Interestingly, PZTN-coated PVC plastic-films effectively disallowed biofilms on their surface. This part of this research suggests that PZTN coated PVC-plastics are the best alternative for biofouling management.

ZnO nanoparticles (NPs) synthesized using haemocyanin (Hc-ZnONPs) purified from haemolymph of Penaeus semisulcatus were characterized using various techniques. HR-TEM and SEM microscopy indicated Hc-ZnONPs had a typical size of 20-50 nm and were spherical. The objective of current investigation was to assess the effects of dietary supplementation of Hc-ZnONPs on the development and activity of digestive and metabolic enzymes, as well as the antioxidant levels in P. semisulcatus. Trial basal diets were supplemented with Hc-ZnONPs at rates of 0, 10, 20, 40, 60, and 80 mg kg-1 (dry feed weight) and were fed to P. semisulcatus for 30 d. For 60 mg kg-1 Hc-ZnONPs-supplemented feed, significantly (P < 0.05) enhanced endurance, development, and activity of the digestive enzyme were observed. The enzymatic antioxidants and metabolic enzymes activities in the muscle exhibited no significant changes when 10-60 mg kg-1 Hc-ZnONPs-supplemented feed was fed to P. semisulcatus. Conversely, feeding the P. semisulcatus with 80 mg kg-1 Hc-ZnONPs produced a harmful outcome, with significant increase in the enzymatic antioxidants and metabolic enzymes. Consequently, 80 mg kg-1 Hc-ZnONPs was identified as lethal to P. semisulcatus. Hence, it is proposed that the diet of P. semisulcatus can be supplemented with up to 60 mg kg-1 Hc-ZnONPs for improving the endurance, development and immunity.

This study unveils the in vitro and in vivo antibiofilm potential of 2,6-Di-tert-butyl-4-methylphenol (DTBMP) from Chroococcus turgidus against Vibrio spp. In the preliminary study, cell free culture supernatant (CFCS) of C. turgidus inhibited the violacein production in biomarker strain Chromobacterium violaceum and its mutant strain CV026 in a dose dependent manner. The effective biofilm inhibitory concentration (BIC) of pure compound DTBMP from C. turgidus was identified as 250 µg/ml concentration in tested Vibrio species. Furthermore, DTBMP proved to effectively inhibit the bioluminescence production in V. harveyi and other biofilm related virulence traits such as exopolysaccharides (EPS) production, hydrophobicity index, swimming and swarming motility at its BIC concentration in three major pathogenic vibrios: V. harveyi, V. parahaemolyticus and V. vulnificus. The antibiofilm potential of DTBMP was validated through light, confocal laser scanning and scanning electron microscopic analyses. In addition, the non-bactericidal effect of DTBMP was determined through growth curve and 2,3-bis (2-methyloxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide (XTT) assay. Real-time PCR studies revealed the down-regulation of master quorum sensing (QS) regulator genes of V. harveyi such as luxR, luxS, luxP, luxQ and luxO on treatment with DTBMP. In vivo results confirmed that DTBMP augmented the survival rate of Litopenaeus vannamei larvae up to 75, 88 and 66% upon infection with V. harveyi, V. parahaemolyticus and V. vulnificus, respectively. The results of this study ascertain the promising effects of DTBMP as an antibiofilm agent, which could be positively explored to treat biofilm-associated vibrios infections in aquaculture.

In this study, the pathogenicity of GFP tagged Vibrio parahaemolyticus Dahv2 and the protective effect of the probiotic strain, Bacillus licheniformis Dahb1 was studied on the Asian catfish, Pangasius hypophthalmus. The experiment was carried out for 24 days with three groups and one group served as the control (without treatment). In the first group, P. hypophthalmus was orally infected with 1 mL of GFP tagged V. parahaemolyticus Dahv2 at two different doses (10(5) and 10(7) cfu mL(-1)). In the second group, P. hypophthalmus was orally administrated with 1 ml of the probiotic B. licheniformis Dahb1 at two different doses (10(5) and 10(7) cfu mL(-1)). In the third group, P. hypophthalmus was orally infected first with 1 mL of GFP tagged V. parahaemolyticus Dahv2 followed by the administration of 1 mL of B. licheniformis Dahb1 (combined treatment) at two different doses (10(5) and 10(7) cfu mL(-1)). The growth, immune (myeloperoxidase, respiratory burst, natural complement haemolytic and lysozyme activity) and antioxidant (glutathione-S-transferase, reduced glutathione and total glutathione) responses of P. hypophthalmus were reduced after post infection of GFP tagged V. parahaemolyticus Dahv2 compared to control. However, after administration with the probiotic B. licheniformis Dahb1 at 10(5) cfu mL(-1), P. hypophthalmus showed significant increase in the growth, immune and antioxidant responses compared to 10(7) cfu mL(-1). On the otherhand, the growth, immune and antioxidant responses of P. hypophthalmus infected and administrated with combined GFP tagged Vibrio + Bacillus at 10(5) cfu mL(-1) were relatively higher than that of GFP tagged V. parahaemolyticus Dahv2 and control groups but lower than that of probiotic B. licheniformis Dahb1 groups. The results of the present study conclude that the probiotic B. licheniformis Dahb1 at 10(5) cfu mL(-1) has the potential to protect the P. hypophthalmus against V. parahaemolyticus Dahv2 infection by enhancing the growth, immune and antioxidant responses. The probiotic B. licheniformis Dahb1 would be effectively used in the treatment of aquatic diseases for improvement of aquaculture industry.

In the present study, we purified two prophenoloxidases (proPO) from haemolymph of green tiger shrimp, Penaeus semisulcatus by gel fermentation chromatography using blue Sepharose matrix. The two purified prophenoloxidase macromolecules are of about 76 and 75 kDa determined through SDS-PAGE and named as Penaeus semisulcatus prophenoloxidase I (PSproPO I) and Penaeus semisulcatus prophenoloxidase II (PSproPO II). It was further characterized by X-Ray Diffraction (XRD), Fourier Transform Infrared Spectroscopy (FTIR), Circular Dichroism (CD) and High Performance Liquid Chromatography (HPLC) analysis. The purified PSproPO I and PSproPO II showed the strongest agglutination titre against human erythrocytes compared to goat RBC. The PSproPO I and PSproPO II showed phagocytic activity against yeast Saccharomyces cerevisiae and encapsulation activity against Sepharose CL 6B beads compared to CM Sepharose and Sodium alginate beads. The functional analysis of purified PSproPO I and PSproPO II showed enhanced PO activity when added with the triggering molecules such as pathogen associated molecular patterns (PAMPs), metals and chemicals. In addition, eluted fraction containing PSproPO I and PSproPO II showed antibiofilm activity against Gram positive and Gram negative bacteria. The above results concluded that no significant differences were found between the purified PSproPO I and PSproPO II immune indices and functions. This study might provide a sensitive platform to understand more about the critical roles of PSproPO I and PSproPO II in crustacean immune system.

Four strains of N-hexanoyl-L-homoserine lactone (AHL)-degrading Pseudomonas spp., named PsDAHP1, PsDAHP2, PsDAHP3, and PsDAHP4 were isolated and identified from the intestine of Fenneropenaeus indicus. PsDAHP1 showed the highest AHL-degrading activity among the four isolates. PsDAHP1 inhibited biofilm-forming exopolysaccharide and altered cell surface hydrophobicity of virulent green fluorescent protein (GFP)-tagged Vibrio parahaemolyticus DAHV2 (GFP-VpDAHV2). Oral administration of PsDAHP1 significantly reduced zebrafish mortality caused by GFP-VpDAHV2 challenge, and inhibited colonisation of GFP-VpDAHV2 in the gills and intestine of zebrafish as evidence by confocal laser scanning microscope and selective plating. Furthermore, zebrafish receiving PsDAHP1-containing feed had increased phagocytic cells of its leucocytes, increased serum activities of superoxide dismutase and lysozyme. The results suggest that Pseudomonas aeruginosa PsDAHP1 could protect zebrafish from V. parahaemolyticus infection by inhibiting biofilm formation and enhancing defence mechanisms of the fish.
